A study on the practicability of genotyping and cytotoxicity of Actinobacillus actinomycetemcomitans isolated from periodontal patients (Actinobacillys actinomycetemcomitans의 유전자형의 유용성과 세포독성에 관한 연구)

  • The purpose of this study was to evaluate the genotypic characterization of Actinobacillus actinomycetemcomitans (A. actinomycetemcomitans) using arbitrarily primed polymerase chain reaction (AP-PCR), to investigate the cytotoxicity of both clinical isolates and standard strains of A. actinomycetemcomitans for the human Jurkat T cells, and to measure the osteoclastogenic cytokines released by Jurkat cells infected with these bacterial strains. The random sequence primer 15 and 16 could distinguish different AP-PCR profiles between clinical isolates of A. actinomycetemcomitans. A. actinomycetemcomitans significantly suppressed Jurkat cell viability in time dependent fashion and the results of DNA fragmentation assay indicated that this bacterial species induced apoptosis in Jurkat cells undergoing apoptosis released the osteoclastogenic cytokine, IL-1$\beta$, IL-6, TNF-$\alpha$. These data support the hypothesis that induction of apoptosis is at least one essential step in A. actinomycetemcomitans induced local immunosuppressive pathway, and that A. actinomycetemcomitans can modulate the immunomodulatory cell population in the periodontal tissue by inducing T cell death through apoptosis, and that apoptosis of local resident T cells may play an active role in bone resorption by releasing osteoclastogenic cytokines, e.g. IL-1$\beta$, IL-6, TNF-$\alpha$.

Interleukin-8 production and interleukin-8 mRNA expression induced by lipopolysaccharides from Prevotella intermedia and Prevotella nigrescens in monocyte-derived macrophages (Prevotella intermedia 및 Prevotella nigrescens의 지질다당질이 대식 세포에서의 Interleukin-8 생성에 미치는 영향)

  • Purpose: Interleukin-8 (IL-8) is an important mediator of immune and inflammatory reactions and is produced by a variety of different cell types. This study was undertaken to investigate the effects of lipopolysaccharides (LPSs) from Prevotella intermedia and Prevotella nigrescens, the major causes of inflammatory periodontal disease, on the production of IL-8 and the expression of IL-8 mRNA in differentiated THP-1 cells, a human monocytic cell line. Methods: LPSs from P. intermedia ATCC 25611 and P. nigrescens ATCC 33563 were prepared by the standard hot phenol-water method. THP-1 cells were incubated in the medium supplemented with phorbol myristate acetate to induce differentiation into macrophage-like cells. Results: We found that LPS preparations from P. intermedia and P. nigrescens can induce IL-8 mRNA expression and stimulate the release of IL-8 in differentiated THP-1 cells without additional stimuli. Conclusions: There are no previous reports of the ability of P. intermedia and P. nigrescens LPS to stimulate the release of IL-8, and the present study clearly shows, for the first time, that LPSs from P. intermedia and P. nigrescens fully induced IL-8 mRNA expression and IL-8 production in differentiated human monocytic cell line THP-1. The ability of P. intermedia and P. nigrescens LPS to promote the production of IL-8 may be important in the pathogenesis of inflammatory periodontal disease.

Investigation into the mechanism of action of Moringa oleifera for its anti-asthmatic activity

  • In the present investigation, we studied the effect of alcoholic extract of Moringa oleifera (M. oleifera) seed kernels on various experimental models of bronchial asthma. Significant (P < 0. 05) increase in preconvulsion time was observed due to pretreatment with M. oleifera when the guinea pigs were exposed to either acetylcholine (Ach) or histamine aerosol. This bronchodilating effect of M. oleifera was comparable to ketotifen fumarate. Spasmolytic effect of M. oleifera was also observed by dose dependent inhibition of ideal contractions induced by Ach, 5HT, histamine and $BaCl_2$. Alcoholic extract of M. oleifera produced significant dose dependent protection by egg albumin and compound 48/80 induced mast cell degranulation. Pretreatment with alcoholic extract of M. oleifera also decreased carrageenan induced rat paw edema, which was comparable to that of standard diclofenac sodium. Minimum inhibitory concentration for alcoholic extract of M. oleifera was low as compared to cold-water extract and hot water extract when antimicrobial activity was tested against various respiratory pathogens like Escherichia coli (E. coli), Staphylococus aureus (S. aureus) and pseudomonas aeruginosa (P. aeruginosa). Our data suggest that antiasthmatic activity of M. oleifera seed kernels may be due to its bronchodilator, anti-inflammatory, mast cell stabilization and antimicrobial activity.

Experimental study on the effects of EVA(Ethylene Vinyl Acetate) for solar cell's long-term life (EVA(Ethylene Vinyl Acetate) 수지가 태양전지의 장기적인 수명에 미치는 영향에 관한 실험적 연구)

  • In this study, analysed the characteristics of power drop and surface damage in solar cell through high temperature and humidity test in the 3 case of EVA(ethylene vinyl acetate) and 2 case ribbon thickness. The solar cells were tested during the 500hr in $85^{\circ}C$ temperature and 85% relative humidity conditions, that excerpted standard of PV Module(KS C IEC-61215). Through the EL(Electroluminescence) shots, specimen's surface have partialy damaged. Before and after high humidity and high temperature test, ribbon thickness $200{\mu}m$ EVA1 case power drop rate was 8.463%, EVA2 case was 6.667%, EVA3 case was 6.373%. In the ribbon thickness $250{\mu}m$ EVA1 case power drop rate was 6.521%, EVA2 case was 8.517%, EVA3 case was 6.019%. EVA3 case was the lowest power and FF(fill factor) drop rate at the 2 case of ribbon thickness, because EVA3 is laerger than EVA1 and EVA2 in thickness, elongation and tensile strength.

  • The integration of FC (Fuel Cell) and GSHP (Ground Source Heat Pump) hybrid system could produce a synergistic advantage in thermal and electric way. This study intends to analyse the economical aspect of a FC integrated GSHP hybrid system compared to the conventional system which is consisted with a boiler and a chiller. Based on the hourly simulation, the study indicated that GSHP system and FC+GSHP hybrid system could reduce the energy consumption on a building. The method of the economic assessment has been based on IEA ECBCS Annex 54 Subtask C SPB(Simple Payback) method. The SPB was calculated using the economic balanced year of the alternative system over the conventional (reference) system. The SPB of the alternative systems (GSHP and FC+GSHP) with 50% initial incentive was 4.06 and 26.73 year respectively while the SPB without initial incentive of systems was 10.71 and 57.76 year.

Origin of Milled Wood Lignin : An Immunocytochemical Approaches in Combination with Transmission Electron Microscopy (면역전현법(免疫電顯法)에 의한 마쇄(磨碎) 리그닌의 기원(起源)에 관한 연구(硏究))

  • Present work was undertaken to investigate the origin of milled wood lignin(MWL) in the wood cell wall using immunocytochemical techniques, which can provide the information on the localization of specific antigens(MWL in the present study) to be examined. Spruce MWL dissolved in DMSO and emulsified with Freund adjuvant was injected directly into the mouse spleen. The animals were boostered at two-week intervals after the initial immunization. Blood samples were purified in standard procedures. The characteristics of antibodies against MWL were tested by indirect ELISA. Visualization of MWL was carried out using conventional indirect immunogold-labelling methods on the ultrathin sections of spruce wood. Immuno-TEM observations showed that the immunogold probes were selectively attached to secondary cell walls of spruce wood. The most intense labelling was frequently observed in the S2 layer. In contrast, gold labelling in the lignin-rich regions, such as middle lamella and cell corner was not found. The immuno-TEM provides an indication that spruce MWL originates from the S2 layer.

Simple Analysis for Interaction between Nanoparticles and Fluorescence Vesicle as a Biomimetic Cell for Toxicological Studies

  • With continuing progress of nanotechnologies and various applications of nanoparticles, one needs to develop a quick and fairly standard assessment tool to evaluate cytotoxicity of nanoparticles. However, much cytotoxicity studies on the interpretation of the interaction between nanoparticles and cells are non-mechanistic and time-consuming. Here, we propose a simple screening method for the analysis of the interaction between several AgNPs (5.3 to 64 nm) and fluorescence-dye containing vesicles ($12{\mu}m$) acting as a biomimetic cell-membrane. Fluorescence-dye containing vesicle was prepared using a fluorescence probe (1,6-diphenyl-1,3,5-hexatryene), which was intercalated into the lipid bilayer due to their hydrophobicity. Zeta potential of all materials except for bare-AgNPs (+32.8 mV) was negative (-26 to -54 mV). The morphological change (i.e., rupture and fusion of vesicle, and release of dye) after mixing of the vesicle and AgNPs was observed by fluorescence microscopy, and fluorescence image were different with coating materials and surface charge of x-AgNPs. In the results, we found that the surface charge of nanoparticles is the key factor for vesicle rupture and fusion. This proposed method might be useful for analyzing the cytotoxicity of nanoparticles with cell-membranes instead of in vitro or in vivo cytotoxicity tests.

An Approximate Analysis of Cell Loss Probability of ATM Multiplexer with Homogeneous MPEG Video Sources (동일한 MPEG 비디오원 입력에 대한 ATM 다중화기 셀손실률 근사분석)

  • For VBR video traffic, Motion-Picture Experts Group(MPEG) coding algorithm was adopted as the standard coding algorithm by International Telecommunication Union(ITU). In this paper, we propose a traffic model of an MPEG coded video traffic in frame level and cell level, and develop an approximate model for evaluating performance of a ATM multiplexer with homogeneous MPEG video sources by considering burst-level variation of aggregated traffics. For homogeneous MPEG video traffics which are frame-synchronized, the performance of the ATM multiplexer is influenced by source correlation at the multiplexing time. When sources are highly correlated, we decompose the aggregated cell streams by the frame-type and model multiplexing process during a frame time as n*D/D/1/K queueing model and suggest an approximate method for obtaining CLP of the ATM multiplexer. In the case that sources are highly correlated, the solution has the meaning of the upper bounds of performance of the ATM multiplexer. For the verification of our model, we compare the solution of our model with simulation resets. As the number of sources increases. The CLP obtained from our model approaches to simulation results, and gives upper bounds of simulation results.

  • Dye-sensitized solar cell (DSC) has been considered as a possible alternative to current silicon based p-n junction photovoltaic devices due to its advantages of high efficiency, simple fabrication process and low production cost. Numerous researches for high efficient DSC in the various fields are under way even now. Among them, the compact layer, which prevents the back electron transfer between transparent conductive oxides and the redox electrolyte, is fabricated by various methods such as a ZnO dip-coating, $TiCl_4$ dip-coating, and Ti sputtering. In this study, we tried to fabricate the $TiO_2$ compact layer by the spin-coating method using aqueous $TiCl_4$ solution. The effect of the spin-coating method was checked as compared with conventional dip-coating method. As a result, DSC with a spin-coated compact layer had 33.4% and 6% better efficiency than standard DSC and DSC with a dip-coated compact layer.

Analysis of Phosphatidylinositol 3,4,5-Trisphosphates of PTEN Expression on Mammalian Cells

  • The goal of this study is to find an experimental condition which enables us to perform enzymatic studies on the cellular behavior of PTEN (phosphatase and tensine homolog) through identification of molecular species of phosphatidylinositol 3,4,5-trisphosphates and their quantitative analysis in a mammalian cell line using mass spectrometry. We initially exployed a two-step extraction process using HCl for extraction of phosphatidylinositol 3,4,5-trisphosphates from two mammalian cell lines and further analyzed the extracted phosphatidylinositol 3,4,5-trisphosphates using tandem mass spectrometry for the identification of them. We finally quantified the concentration of phosphatidylinositol 3,4,5-trisphosphates using internal standard calibration. From these observation, we found that HEK 293-T cells is a good model to examine the enzymatic behavior of PTEN in a cell, and the minimum amount of phosphatidylinositol 3,4,5-trisphosphates is more than 50 pmol for quantification in a mass spectrometer. These results suggest that the well-optimized experimental conditions are required for the investigation of the cellular PTEN in terms of the catalytic mechanism and further for the detailed identification of cellular substrates.
