Hepatoprotective Effect of Stamen Extracts of Mesua ferrea L. against Oxidative Stress induced by $CCl_4$ in Liver Slice Culture Model

  • Stamens of Mesua ferrea L. are a well-known herbal drug used in Indian System of Traditional Medicine to treat various diseases. The claimed activity of this plant part is necessitated to investigate antioxidant and hepatoprotective activity. Authenticated plant sample was extracted with hexane, ethanol (EtOH) and water (aq.) using ASE 100 accelerated solvent extractor. Antioxidant activity was evaluated by means of different in vitro assays. Hepatoprotective effect was investigated on carbon tetrachloride induced oxidative stress in liver slice culture model. Cytotoxic marker lactate dehydrogenase (LDH) released in culture medium and the activity of lipid peroxidation along with antioxidant enzymes (AOEs) namely superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) were estimated. Hexane and EtOH extracts were significantly inhibited DPPH, NO, SOD and $ABTS^+$ radical in dose dependent manner. The trade of phenol content was: aq. extract < hexane extract < EtOH extract. A significant correlation was shown by total phenol content and free radical scavenging activity of extracts. The culture system treated with hexane extract, EtOH extract or ascorbic acid exhibited significant depletion in LDH, lipid peroxidation, antioxidative enzymes SOD, CAT and GR. Hexane extract and EtOH extracts of stamen of M. ferrea protected liver slice culture cells by alleviating oxidative stress induced damage to liver cells.

An Unrecorded Species from Korean Flora: Vallisneria spinulosa (Hydrocharitaceae) (한국산 미기록 식물: 낙동나사말(자라풀과))

  • We report Vallisneria spinulosa S. Z. Yan (Hydrocharitaceae) as an unrecorded species from the flora of Korea. This aquatic vascular plant occurs in reservoirs, streams, and ditches at Changnyeong, Changwon, and Gimhae which belong to Nakdong river basin. Vallisneria spinulosa is distinguishable from V. natans (Lour.) Hara already known to Korean flora by the presence of turion and hairs at base of stamen, adnation between pistil and staminodia, the number of stamen, and shape of fruit and seed. V. spinulosa is similar to V. desnseserrulata Makino in the viewpoint of distribution (Japan and southern China), but V. spinulosa has three to five epidermal wings on the surface of seed.

The Effects of Nelumbo nucifera on Utraviolet-B Irradiated human Keratinocytes (연잎, 연꽃, 연꽃 수술 추출물이 UVB 자외선 조사에 의한 각질형성세포의 보호 및 피부 노화 방지에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the anti-aging effects on cultured human keratinocytes with Nelumbo nucifera extracts. Methods : Each parts of leaves, flowers and stamen were extracted with water or 70% ethanol. These extracts were tested for cell viability on HaCaT cells (human keratinocyte line) by MTT assay. We investigated the effects of Ultraviolet-B (UVB) irradiation on cytotoxicity and lipid peroxidation in cultured skin keratinocytes. Results : The ethanol extract of Nelumbo nucifera flowers showed maximun cell viability as 111.39% in 30 ug/ml concentration. The water extracts of stamen, flowers, leaves showed cell viability as 107.12, 101.65, 101.46%, respectively. HaCaT keratinocytes were survived 63.06% at $20mJ/cm^2$ UVB irradiation. The cell membrane lipid peroxidation was measured by accumulation malondialdehyde (MDA). The levels of MDA were decreased by the ethanol extract of Nelumbo nucifera flowers and the water extracts of stamen. Conclusions : These finding suggest that the ethanol extract of Nelumbo nucifera flowers prevent anti-aging effects on cultured human keratinocytes during UVB irradiation.

Antioxidative Effect and Melanogenesis of Nelumbo nucifera Stamen Extract on Cultured Human Skin Melanoma Cells Injured by Hydrogen Peroxide (연꽃수술추출물이 과산화수소로 손상된 배양 인체피부흑색종세포에 대한 항산화효과 및 멜라닌화에 미치는 영향)

  • To examine the antioxidative effect and melanogenesis of Nelumbo nucifera stamen (NNS) extract on hydrogen peroxide $H_2O_2$ induced cytotoxicity in cultured human skin melanoma cells (SK-MEL-3), cell adhesion activity (CAA), tyrosinase inhibitory activity and total amount of melanin synthesis were measured by colorimetric assay. In this study, $H_2O_2$ significantly decreased CAA, and $CAA_{50}$ value of $H_2O_2$ was determined at 30 uM. In the antioxidative effect, NNS extract increased cell adhesion activity which was decreased by $H_2O_2$ induced cytotoxicity, and also, tyrosinase activity and total amount of melanin were decreased by NNS extract. These results suggested that $H_2O_2$ was highly toxic on cultured human skin melanoma cells and NNS extract showed the antioxidative and inhibitory effect of melanogenesis by the increased CAA, and the decresed tyrosinase activity and total amount of melanin synthesis.

Structures and Development of Floral Organs in Bupleurum falcatum L. (시호의 화기구조 및 화기내 각 기관의 발육과정)

  • We investigated the structures of floral organs and the developmental process of each floral organs in Bupleurum falcatum. The overall size of a floral was about 2mm. The lengths of ray, pedicel, pistil and stamen were 22.5mm, 3.6mm, 1.0mm and 1.3mm respectively. The ovary surface was 0.9mm in length and 1.4mm in width. And the developmental periods of each floral organs were as follows; 1 through 6 days in stamen emergence, 6 through 9 days in petal detachment and pistil emergence, 9 through 16 days in pistil ma­turation, and above 16 days in pistil degeneration after onset of flowering. This plant was admitted to be a allogamous plant, especially with the protandry form of dichogamy.

Formation of Gametophytes and Development of Zygotic Embryo in Dicentra spectabilis (금낭화(Dicentra spectabilis)의 배우체 형성 및 접합자배의 발달)

  • This study was carried out to obtain the basic informations on the characteristics of gametophytes formation and embryo developement in Dicentra spectabilis. Microspore mother cells developed from archesporial cells, start meiosis when flower bud length reaches around 1 mm, formed tetrahedral type tetrad. The 4 microspores were separated. They were developed to male gametophytes, respectively. Megaspore mother cells were observed when flower bud length was $4{\sim}5\;mm$. The developemental type of megaspore was polygonum and embryo sac was amphitropous. Three large and distinctive antipodals did not degenerated and remained after embryo sac was developed. When the male and female gametophytes was fully developed, the length of stamen and style was very similar or stamen was shorter about 0.5 mm than that of style. This result indicates that self-fertilization can be occurred in this species. After fertilization, developing zygotic embryos showed various stages of development from globular to cotyledonary embryos, and zygotic embryo in seed scattering time seemed to have an early cotyledonary stage.

Screening of Plant Extracts with Cholinesterase Inhibition Activity (콜린 에스테라제 저해효과 보유 식물 추출물 탐색)

  • This study was conducted to select candidates from plant resources for the purpose of improving or treating Alzheimer's disease, a type of dementia. One hundred and eighty-four plant extracts at a final concentration of $100{\mu}g/ml$ were screened to determine their capacity to inhibit acetylcholinesterase (AChE) by in vitro assay. From this AChE assay, seven plant extracts - including methanol ext. and water ext. of Phellaodendron amurense Rupr. (bark), methanol ext. of Nelumbo nucifera Gaertn (stamen/ovary), methanol ext. of Persicaria tinctoria H. GROSS (flower), methanol ext. of Coptis chinensis (rhizome), ethanol ext. of Cinnamomum cassia Blume(bark) and ethanol ext. of Carthamus tinctorius L. (fruit) - showed effective inhibition activity ranging from 18.7% to 63.1%. The selected extracts were testified their inhibition activities on AChE and BuChE (butyrylcholinesterase) at concentrations of 25, 50, 100, $200{\mu}g/ml$. In the AChE assay, five extracts including methanol ext. of Nelumbo nucifera Gaertn. (stamen/ovary), methanol ext. of Persicaria tinctoria H. GROSS (flower), methanol ext. of Coptis chinensis (rhizome), methanol ext. and water ext. of Phellaodendron amurense Rupr. (bark) showed inhibition activity of 15.0%~73.5%, 19.5%~63.5%, 81.6%~58.5%, 69.9%~80.5%, and 54.8%~78.3%, respectively, at concentrations of 25, 50, 100, $200{\mu}g/ml$. In the BuChE assay, the extracts of Nelumbo nucifera Gaertn. (stamen/ovary), Persicaria tinctoria H. GROSS (flower), and Coptis chinensis (rhizome) showed inhibitory capacities of 58.9~81.6%, 45.8%~72.4%, and 33.1%~55.4% at concentrations of 25, 50, 100, $200{\mu}g/ml$, respectively. In conclusion, it is suggested that Nelumbo nucifera Gaertn. (stamen/ovary), Persicaria tinctoria H. GROSS, Coptis chinensis (rhizome) and Phellaodendron amurense Rupr. (bark) could be selected as candidate materials for improving or treating Alzheimer's disease on the basis of further study.

  • The analysis in external morphology of Astilbe including three endemic species in Korea was undertaken. From this study, five species and three varieties of Astilbe in Korea were redescribed. Their scientific names and Korean names were also consulted. Korean Astilbe could be grouped in two Series-Series 1. Simplicifoliae; Astilbe simplicifoliae and Series 2. compositae; A. taquetii, A. microphylla, A. chiensis var. chinensis, A. chinensis var. paniculata, A. chinensis var. davidii, A. divaricata, A. koreana. Interspecific relationships of eight taxa were represented as a toxonomic model by the considering their external morphology-pubescence on floral axis and surface of leafblade, inflorescence type, ratio of petal and stamen length-and distributional range in Korea and neighboring countries.

  • Floral morphology of Magnoliaceae and Illiciaceae in Korea is investigated. Magnoliaceae has long and elongate floral axis where numerous stamens and pistils are pistils are spirally arranged, whereas Illiciaceae has short floral axis and pistils are arranged in a single whorl on the axis. The stamens are basically similar in both families; winged and broad filament, prominant connective, long sporangium, and no clear limitation between fertile and sterile parts, which are primitive characters in stamen. The ovaries are the conduplicate type, in both, with decurrent stigma, stigmatic surface confined to stylar position, and no distinct style. Magnoliaceae has one locule and two ovules, whereas Illiciaceae has one locule and one ovule. The ovule is anatropous or hemianatropus, and has bitegmic integument and crassinucellate nucellus.

  • A new species of Corydalis sect. Corydalis (Fumariaceae), C. misandra B.U.Oh, is described from the provinces of Gyonggi-do and Gangwon-do in Korea. The new species is distinct from its close relative C. namdoensis in having elliptic or linear leaflet shape, entire or slightly dentate bract apex, broad and rhomboidal lower outer petal, 14 papillate stigma, oblong stamen and linear capsule with one rowed seeds, etc.
