A Study on Electro-optical Characteristics in Three Kinds of Liquid Crystal Display Operating Mode

  • In this study, we investigated response characteristics of liquid crystal display (LCD) with different operating mode of nematic liquid crystals (NLCs) such as 45 $^{circ} twisted nematic (TN), 67.3 $^{circ} TN and electrical controlled birefringence (ECB) on the rubbed polyimide (PI) surface with side chains. The pretilt angles generated on polyimide surfaces of the three kinds of LCD operating modes were about 12 $^{circ} that was higher than those of conventional TN-LCOs. Also, the Electro-optical (EO) performance of these LCOs showed stable condition. Low transmittance of the 45 $^{circ} TN and 67.3 $^{circ} TN cell on the rubbed PI surface were measured by using low cell gap d. The fast response time in ECB cell among the three kinds of LCD operating modes was achieved. Also, thermal ability of fast 90 $^{circ} TN-LCD was investigated. The threshold voltage and the response time of thermal stressed TN-LCOs showed the same performances on no thermal stressed TN-LCOs. There was little change of value in these TN cells. However, the transmittances of TN-LCOs on the rubbed PI surface decreased while increasing thermal stress time. Therefore, the thermal stability of TN-LCD was decreased by the high thermal stress for the long duration.

The Preparation for Sintered Body of $CeO_2$ Based Complex Oxide in Low Temperature Solid Oxide Fuel Cells Using Colloidal Surface Chemistry (콜로이드 계면화학을 이용한 저온형 고체전해질용 $CeO_2$계 복합 산화물의 소결체 제조)

  • In this study, the dispersion stability of CeO2 based complex oxide was studied, and density, porosity, and microstructure of green body were investigated using colloid surface chemistry to manufacture the Gd2O3 doped CeO2 solid electrolyte in an aqueous system. To prepare the stable slurry for slip casting, the dispersion stability was examined as a function of pH using ESA(electrokinetic sonic anplitude) analysis. The dynamic mobility of particles was enhanced with anionic and cationic dispersant were added the amount of 0.5wt% respectively, but pH value in slurries didn't move to below 6.0 because of the influence of dopants. This phenomenon also appeared in the CeO2-Y2O3 and CeO2-Sm2O3 systems, so it could be inferred that rare earth dopants such as Gd2O3, Sm2O3 and Y2O3 not only have the similar motion with changing pH in an aqueous system but also can be dissolved in the range of pH 6.0∼6.5. In CeO2-Gd2O3 system, when the anionic dispersant was added the amount of 0.5wt% and pH value in slurries was fixed at 9.5, the green body density was 4.07g/㎤, and the relative density of sintered body was 95.2%. It could be inferred from XRD analysis that Gd3+ substituted into Ce4+ site because there was no free Gd2O3 peak.

Analysis of the Segmental Reinforced Retaining Wall Behavior by Field Monitoring (현장계측을 통한 블럭식 보강토 옹벽의 거동분석)

  • Geogrid reinforced soil structures with segmental block facing have been increased since 1990's, because of the convenience of installation and the flexible appearance. In this paper, the behavior of the segmental reinforced retaining wall was analysed with the results of field monitoring. The height and length of reinforced wall are 12m and 25m, respectively. The field measurement equipments are horizontal and vertical earth pressure cells, settlement plate, strain gauge, inclinometer, and displacement pin. Based on the field monitoring, the horizontal earth pressure was approximately 0.3times higher than that of the theoretical method and the maximum tensile strength of reinforcement was 26.2kN/m. The displacement of facing wall was 23mm at the point of 7.1m height of the wall and toward the wall facing. The results of the study indicate that the segmental reinforced retaining wall is in a stable condition because of good compaction & reinforcement effects, and long period of construction time. Finally, the computer program of SRWall is very useful tool to design the segmental reinforced retaining wall.

Analysis on the Substrate Specificity and Stability of Hansenula polymorpha Alcohol Oxidase (Hansenula polymorpha 알코올 산화효소의 기질특이성 및 안정성 분석)

  • An alcohol oxidase from Hansenula polymorpha was strongly induced when cells were grown with 0.5% methanol supplementation as the carbon source. The induced Hansenula polymorpha alcohol oxidase was purified to electrophoretic homogeneity by using DEAE-Sephacel and Mono Q column chromatographys. The enzyme oxidized mainly primary aliphatic alcohols and exhibited high substrate specificity towards ethanol and methanol. The activity of the enzyme optimally proceeded at pH 8.5 and $50^{\circ}C$. The midpoint of the temperature-stability curve for the enzyme was approximately $52^{\circ}C$ and the enzyme was not completely inactivated even at $65^{\circ}C$ temperature. The enzyme showed resistance toward detergents and highly stable over 7 weeks of storage condition. This Hansenula polymorpha alcohol oxidase may be useful for the enzymatic determination of alcohol and for the industrial production of alcohols and aldehydes.

  • A novel 8 V DC power source with an external series-parallel connection of 50 Dye-Sensitized Solar Cells(DSSCs) has been proposed. One DSC has the optimized length to width ratio of $5.2{\times}2.6$ cm and an active area 8 $cm^2$($4.62{\times}1.73$ cm) which attained a conversion efficiency of 4.2%. From the electrochemical impedance spectroscopic analysis, it was found that the resistance elements related to the Pt electrode and electrolyte interface behave like that of diode and the series resistance corresponds to the sum of the other resistance elements. In addition, the TEMoo mode pulsed Nd:YAG laser beam is used to improve the incident photon to current efficiency(IPCE) of DSSC. From this result, this novel 8V-0.38A DC power source shows stable performance with an energy conversion efficiency of about 4.5% under 1 sun illumination(AM 1.5, Pin of 100 $mW/cm^2$).

  • A navigation aid buoy is a kind of safety facility for maritime navigation with a purpose of leading the vessels for navigating, docking and sail off. An advanced rechargeable battery is required for stable power supply for navigation aid buoy as the high magnitude LED lamps, real time location/control for navigation aids and e-Navigation support systems with maritime climate observation equipments have recently been deployed. This study is focused on the lithium battery, especially lithium polymer battery which is believed to be safer than the other types of batteries. The lithium polymer battery reviewed in this study is designed with $LiFePO_4$-based cathode, which has superior safety features to the oxide-based cathodes. Besides, a 3.6kWh battery pack has been built with the above-mentioned unit cells for the purpose of comparative research with lead acid battery system.

  • The antibacterial protein gene, nuecin was expressed in Sf9 cells using baculovirus expression vector system (BEVS). The antibactetial activity of mature nuecin against Pectobacterium carotovorum subsp. carotovorum, Ralstonia solanacearum and Pseudomonas tolaasii was significantly high, demonstrating that nuecin had a wider antibacterial spectrum on gram negative and positive bacteria. The result appears to be superior to other antibacterial peptide, attacin. The nuecin was purified by SP-sepharose and Mono Q HR ion-exchange chromatography, and then by Superdex 200 HR 10/30 column. The purified nuecin is quite stable at 80$\^{C}$ and 100$\^{C}$ for several hours of incubation and in a wide pH range (pH 2-12).

Utilization of the Bombyx mori Hypothetical Protein 32 Promoter for Efficient Transgene Expression

  • For stable germline transformation, the promoter of Bombyx mori cytoplasmic actin gene (BmA3) has been used for ubiquitous expression of transgenes. So far, no strong promoter is available for ubiquitous expression in B. mori, excluding BmA3 promoter. To identify more powerful promoter than previously reported BmA3 promoter, we isolated 9 clones that show stronger signal compared to BmA3 by a dot blot hybridization. Among these 9 clones, we focused on one clone which has high amino acid homology (85%) with hypothetical protein 32 gene of Lonomia obliqua. This clone, named bHp32 (B. mori hypothetical protein 32) was ubiquitously expressed in all tissues and developmental stage of fifth instar B. mori larvae. As result of promoter assay using dual luciferase assay system, we found the highest transcription activity region (-1,200/+220) in the 5'-flanking region of bHp32 gene, which has 42-fold more intensive promoter activity than BmA3 promoter. Moreover, the bHp32 promoter was normally regulated in Bm5, Sf9, and S2 cells. Therefore, we suggest that bHp32 promoter may be used more powerful and effectively for transgene expression in various insects containing B. mori as a universal promoter.

Changes of the goneral components and acid value in market milks according to the preserving periods and temperature (시유의 보존기간 및 온도에 따른 일반 유성분 및 산가의 변화)

  • This study was carried out to investigate the changes of physical and chemical properties according to the preserving periods and temperature in heat-treated market milks ($130^{\circ}C$, 2 sec). The market ilks were divided into 3 experimental groups, that is $4^{\circ}C$, $15^{\circ}C$ and $30^{\circ}C$ according to the preserving temperature during 7 days. There were analysed fat, protein, lactose, total solids, solids-not-fat, specific gravity and acid value of market milks in each experimental group. These observations indicated that heat-treated market milks ($130^{\circ}C$, 2 sec) may be acceptable physically and chemically after 7 days at $4^{\circ}C$. The results obtained were summarized as follows ; 1. Total microorganisms in market milks were found 12$\times$$10^1$ cells at 1st day Coliform and pathogenic bacteria were not detected. 2. Milk fats tended to decrease 3.69% to 3.48% according to the time lapsed in all experimental groups without concerning to the storage temperature. 3. Milk proteins were slowly decreased 3.18% to 3.13% according to the time lapsed without concerning to the storage temperature. 4. Milk lactose was slightly increased 4.41% to 4.55% in all experimental groups. 5. Specific gravity in market milks was 1.0316 to 1.0299, and showed no changes according to the preserving periods and temperature. 6. Total solids and solids-not-fat in market milks were slightly decreased 12.30 to 12.05 and 8.55 to 8.40. 7. Acid values were dramatically increased 0.1289 to 0.3116 at $30^{\circ}C$ storage group, while tended to be stable at $4^{\circ}C$ storage group for 7 days.

The SL1 Stem-Loop Structure at the 5′-End of Potato virus X RNA Is Required for Efficient Binding to Host Proteins and forViral Infectivity

  • The 5′-region of Potato virus X (PVX) RNA, which contains an AC-rich, single-stranded region and stem-loop structure 1 (SL1), affects RNA replication and assembly. Using Systemic Evolution of Ligands by EXponential enrichment (SELEX) and the electrophoretic mobility shift assay, we demonstrate that SL1 interacts specifically with tobacco protoplast protein extracts (S100). The 36 nucleotides that correspond to the top region of SL1, which comprises stem C, loop C, stem D, and the tetra loop (TL), were randomized and bound to the S100. Remarkably, the wild-type (wt) sequence was selected in the second round, and the number of wt sequences increased as selection proceeded. All of the selected clones from the fifth round contained the wt sequence. Secondary structure predictions (mFOLD) of the recovered sequences revealed relatively stable stem-loop structures that resembled SL1, although the nucleotide sequences therein were different. Moreover, many of the clones selected in the fourth round conserved the TL and C-C mismatch, which suggests the importance of these elements in host protein binding. The SELEX clone that closely resembled the wt SL1 structure with the TL and C-C mismatch was able to replicate and cause systemic symptoms in plants, while most of the other winners replicated poorly only on inoculated leaves. The RNA replication level on protoplasts was also similarly affected. Taken together, these results indicate that the SL1 of PVX interacts with host protein(s) that play important roles related to virus replication.
