Effects of Protein Functionality on Myofibril Protein-Saccharide Graft Reaction

  • The myofibril protein (MP) isolate-saccharide graft reactions was prepared using the Maillard reaction with saccharides. The effects of various saccharides on protein functionality and quality of the Maillard reaction were investigated and compared with those of MP. The grafting degree of the MP isolate-saccharide graft reaction was significantly higher in the reducing sugar-treated groups (lactose, glucose, fructose, and palatinose). The browning intensity of the MP isolate-saccharide graft reaction with fructose, sucrose, and erythitol was higher than that observed in the control reaction (p<0.05). MP that reacted with reducing sugars (glucose, fructose, palatinose, and lactose) had fainter bands than MP that reacted with non-reducing sugars (sucrose, erythitol, trehalose, sorbitol, and xylitol). MPs conjugated with glucose exhibited higher protein solubility. The palatinose and lactose treatments were maximum in water binding capacity, though no significant difference in oil binding capacity among the saccharide treatments was observed. The emulsion stability of the MP isolate-saccharide graft reaction with palatinose and erythitol was higher than that of the control reaction. Therefore, reducing sugars have good protein functionality in the MP isolate-saccharides graft reaction.

Development of a High Strength Manufacturing Technology for the Shock Absorber Base Assembly Using Friction Welding (마찰용접을 이용한 고강도 쇼크업소버 베이스 어셈블리의 제조 기술 개발)

  • The shock absorber base assembly is one of the parts in the shock absorber equipment that controls the vehicle movement. It absorbs the shock and vibration to guarantee riding stability and comfort. It demands strength, reliability and strict airtightness of the welded section because the shock absorber base assembly is a container which resists pressure and needs durability by being filled with gas and oil. However, the current engineering needs a lot of production time, has a high cost and shows a low production rate. These problem due to the eight production processes, four of which are spot welding, reinforcement welding like metal active welding (MAG), prior process of the base assembly cap and tube for precision and pressing. We will analyze the manufacturing processes of the base assembly and suggest an improved manufacturing method that uses frictional welding. The results will show that the new method of the frictional welding is better than the previous welding technique. Through the use of this concept of frictional welding, the welding conjunction will be strengthened, measurements will be more precise, and the cost and the number of processes will be reduced.

Microstructural Changes of Mayonnaise during Storage (마요네즈 저장 중 미세구조의 변화)

  • The microstrutural changes of mayonnaise during storage were examined by light microscopy (LM) and scanning electron microscopy (SEM). Fresh mayonnaise was composed of heterogenous population of dispersed spherical oil droplets and droplet size was normally distributed with one mode. During storage at $60^{\circ}C\;and\;-10^{\circ}C$, a shift in droplet size distribution toward larger droplets was observed, as a result of coalescence of lipid droplets. Turbidimetric study also confirmed that coalescence was occurring during this accelerated aging treatments. Measurements obtained from SEM microgrphs provided better determination of smaller droplets and resulted in lower mean diameter of droplets than those obtained from LM. From these results, SEM was found to be an advantageous method of examining emulsion products as compared to LM, providing a better resolution of small droplets and a more representative view of droplet distribution, as dilution of the sample was avoided.

Effects of Jerusalem Artichoke Powder and Sodium Carbonate as Phosphate Replacers on the Quality Characteristics of Emulsified Chicken Meatballs

  • Today incorporation of natural ingredients as inorganic phosphate replacers has come into prominence as a novel research topic due to health concerns about phosphates. In this study, we aimed to investigate the quality of emulsified chicken meatballs produced with Jerusalem artichoke powder (JAP), either alone or in combination with sodium carbonate (SC) as sodium tripolyphosphate (STPP) replacers. The results showed that naturally dried JAP showed favorable technological properties in terms of water-oil binding and gelling. Emulsion batters formulated with JAP-SC mixture showed lower jelly and fat separation, higher water-holding capacity and higher emulsion stability than control samples with STPP. In final product, incorporation of JAP-SC mixture increased moisture and reduced lipid and energy values, and kept the pH value similar to control. Added JAP lead to increments in $b^*$ values whereas decreases $L^*$ values. Cook yield was similar to control in phosphate-free samples formulated with JAP-SC mix. Either low or medium ratios of JAP in combination with SC managed to protect most of the sensory parameters, while sensory scores tend to decrease in samples containing high levels of JAP. Addition of JAP to formulations presented samples that have equivalent behavior to phosphates in terms of lipid oxidation. In conclusion, our study confirms that utilization of JAP in combination with SC had promising effects as phosphate replacers by presenting natural solutions and providing equivalent quality to standard phosphate containing products.

SMEDDS (Self-MicroEmulsifying Drug Delivery System) As An Intraurethral Prostaglandin E1 Delivery System

  • Prostaglandin $E_1\;(PGE_1)$ was formulated as two self-microemulsifying drug delivery systems (SMEDDS) composed of Cremophor $EL^{(R)}$ or Cremophor $ELP^{(R)}$ as a surfactant, ethanol as a cosurfactant and Labrafac $CC^{(R)}$ as an oil to develop liquid preparation for the treatment of erectile dysfunction. In pseudo-ternary phase diagram, viscous gel area and microemulsion area were defined. In the measurement of viscosity, the viscosity of two formulations increased gradually upon the addition of water and it decreased from the water contents over 40%. With excessive water, the present systems formed a microemulsion spontaneously. From these results, rte could expect that the present liquid $PGE_1$ SMEDDS formulations might stay within the urethra in the viscous state when contacting the moisture of the urethra and can be easily eliminated by urination. In long-term stability study, we could select one formulation more stable at the shelf storage condition of $4^{\circ}C$.

  • The casing has been used as a protective conduit during all phases of drilling operations and productions for the oil and gas industries. The casing is manufactured in various diameters, wall thicknesses, lengths, strengths, and connections. When the casing is designed, it has to be considered to withstand a variety of forces, such as collapse, burst, and tensile failure, as well as chemically aggressive brines. Once the casing is damaged, serious problems in geothermal well have been detected continuously. Therefore, this paper describes the casing design for stability of geothermal well to determine influence of casing parameters on the strength and load. In addition, the casing design program was developed. The estimated collapse, burst, tension and depth pressure can provide benefit in the casing design and completion method. This program provides a safety factor and predicts the casing stress more easily.

Change of Functional Properties and Extraction of Sesame Meal Protein with Phytase and Protease (Phytase와 Protease 혼합처리에 의한 참깨박 단백질 용출 및 기능성 변화)

  • To extract insoluble proteins and improve functional properties of sesame meal proteins was treated with phytase and protease from Aspergillus sp. It was found that the optimum pH, optimum temperature, optimum treatment time and optimum unit of enzyme for extraction of protein were pH $10{\sim}12$ (alkaline), $60^{\circ}C$, 11 hr. and 900 units of phytase and 60 units of protease, respectively. The foaming capacity, foaming stability, oil absorption and water absorption of sesame meal protein after treatment with phytase and protease were increased as compared to the control.

Development of Propofol-Ioaded Microemulsion Systems for Parenteral Delivery

  • The aim of the present study was to develop the aqueous parenteral formulation containing propofol using o/w microemulsion systems. Propofol itself was chosen as the oil phase and its content was fixed to 1$\%$, w/w. Pseudoternary phase diagrams were constructed to obtain the concentration range of surfactant and cosurfacatnt and the optimum ratio between them for microemulsion formation. Consequently, the suitability of the chosen microemulsion system as a parenteral formulation was evaluated from the stability and hemolysis tests on that. Among the surfactants and cosurfactants screened, the mixture of Solutol HS 15-ethyl alcohol (5/1) showed the largest o/w mocroemulsion region in the phase diagram. When 1 $\%$ (w/w) of propofol was solubilized with 8$\%$ (w/w) of Solutol $HS^{circledR}$??? 15-ethyl alcohol (5/1), the average droplet size (150 nm) and the content of propofol in the systems were not significantly changed at 40$^{circ}C$ for 8 weeks. The hemolysis test showed that this formulation was nontoxic to red blood cells. In conclusion, propofol was successfully solubilized with the o/w microemulsion systems.

  • A total of 10,317 hours of the KMTNet telescope time were allocated for the predefined science programs, and 7,765 hours have been used for science exposures in Chile, Australia and South Africa last year. The success rate of science observation has increased from 70.7% to 75.3%, and the system operation rate has also increased from 97.6% to 99.6%. There were many improvements in mechanical parts of the dome structure and telescope system, and newly installed filter driers of the CCD camera increased the stability of the system by preventing contamination of oil in the gas line in advance. In order to prepare for the time domain astronomy and multi messenger astronomy era, a pilot program was designed and is now being tested. It targets for fast follow-up observations of optical transient events, however it runs during twilight times only so it does not interrupt any granted science program. A total of 32 SCI papers were published using the system in 2018 and it is a good indication of the high science performance of KMTNet. The selection process of the next observation programs starting from October 2020 and its timeline will be discussed in this meeting.

Effect of Mixing Route and Temperature on Formation of Nanoemulsions (나노에멀젼 형성에서의 혼합 경로와 온도의 영향)

  • In this work, we compared the emulsion states having the same composition of liquid paraffin/ Span 80-Tween 80/ pure water and the different mixing paths and temperatures. Routes reaching the final composition in three component phase diagram were composed of three different ways. The average particle size of the emulsion prepared from the different mixing routes showed a significant difference and decreased as the mixing temperature was increased. However, the mixing route affected more in the size of the emulsions than mixing temperature.
