The experiment was carried out to determine the effects of soil condition on the agronomic characters and aromatic substances in cultivated Codonopsis Lanceolata. An increase of planting densities of Codonopsis Lanceolata. from 1 to 5 plant per pot resulted in the smaller root width, but root yield increased from 109.8g to 286.9g per 1/200pot. The width growth as soil water contents were higher in surplus soil water than that of cultivated deficit soil water. The contents of water, reducing sugar, crude protein and malic acid were higher in cultivated wild than in cultivated upland soil. The higher contents of volatile essential oils as 3-rnethylpentanoic acid and squalene was obtained at the normal soil moisture.
The rare edible and medicinal fungus Antrodia cinnamomea has a substantial potential for development. In this study, Illumina HiSeq 2000 was used to sequence its transcriptome. The results were assembled de novo, and 66,589 unigenes with an N50 of 4413 bp were obtained. Compared with public databases, 6,061, 3,257, and 2,807 unigenes were annotated to the Non-Redundant, Gene Ontology, and Kyoto Encyclopedia of Genes and Genomes databases, respectively. The genes related to terpene biosynthesis in the mycelia of A. cinnamomea were analyzed, and acetyl CoA synthase (ACS2 and ACS4), hydroxymethylglutaryl CoA reductase (HMGR), farnesyl transferase (FTase), and squalene synthase (SQS) were found to be upregulated in XZJ (twig of C. camphora) and NZJ (twig of C. kanehirae). Moreover, ACS5 and 2,3-oxidized squalene cyclase (OCS) were highly expressed in NZJ, while heme IX farnesyl transferase (IX-FIT) and ACS3 were significantly expressed in XZJ. The differential expression of ACS1, ACS2, HMGR, IX-FIT, SQS, and OCS was confirmed by real-time quantitative reverse transcription PCR. This study provides a new concept for the additional exploration of the molecular regulatory mechanism of terpenoid biosynthesis and data for the biotechnology of terpenoid production.
This study aims to demonstrate the effect of squalene (SQ), one of the natural chelator, on the ultrastructural changes in the mouse liver caused by $HgCl_2$. A total of 40 healthy ICR that weighted 30 gm $({\pm}2gm)$ was used for experiment. The experimental group was divided into two groups; group A and B. The group A administrated $HgCl_2$ (4.0mg/kg) to the intraperitoneal. The group B administrated $HgCl_2$ (4.0 mg/kg) to the intraperitoneal treated with SQ (180 mg/kg, 2 times/day). Each group was observed at 24, 48, 72, 96 hours after injected $HgCl_2$. The results were as follows: 1. Group A Nucleus showed condensation of nuclear membrane at the 24 hours. At the 48 hours, observed distinct condensation. But nuclear membrane be seen relative rounded-shape at the 96 hours. At overall the time, inner cavity of mitochondria swollen and development of cristae weakened. Also electron density of matrix was a little low. At the 72 hours, destruction of the inner and outer membrane of mitochondria observed occasionally. Swelling of inner cavity of rER and destruction of lamellae be found from 24 hours to 72 hours, but at the 96 hours, only some swelling 2. Group B Nuclear membrnae and chromatin be seen normal shape at overall the time. Mitochondria showed destruction of the inner membrane until the 48 hours, but mostly normal shapes. Electron density showed high on the all groups. RER be found swelling of inner cavity at the 24 and 48 hours, but found typical lamellae and observed a number of transfer vesicles around rER at the 72 and 96 hours. These results suggest that squalene attenuates the toxic effect of the $HgCl_2$ in the mouse liver.
In order to investigate the effects of elicitors on the growth and ginsenoside biosynthesis of ginseng hairy roots, we treated Panax ginseng hairy root with various concentrations of Haliotidis concha according to different time course. Haliotidis concha supplement increased the biomass and ginsenoside accumulation at 10 mg/L concentration. The growth rate of hairy root under a lighter concentration was greater than hairy root treated with a denser concentration. The highest content and productivity of ginsenosides appeared at 2 weeks after the treatment of 10 mg/L Haliotidis concha. The gene expression of squalene synthase, squalene epoxidase, dammarenediol synthase, cycloartenol synthase, $\beta$-amyrin synthase in hairy roots of ginseng were examined by RT-PCR. The Haliotidis concha treatment resulted in the obvious accumulation of the mRNA of triterpene biosynthesis in Panax ginseng hairy root as compared with the control. In this study, Haliotidis concha acts as a kind of elicitor for the production of ginsenosides.
Background : Exogenous lipoid pneumonia is caused by inhalation or aspiration of animal, vegetable or mineral oil. Most cases are ascribed to aspiration of oil in laxatives or nose drops Petroleum, another pure hydrocarbon used as a base in various medications, is occasionally involved. Especially animal oil produces severe tissue inflammatory reaction, but most patients present with only abnormal chest X-ray and no specific clinical symptoms or signs. Method: Seven patients, 3 males and 4 females, with exogenous lipoid pneumonia, who was hospitalized or referred to pulmonary division at Samsung Medical Center from December 1994 10 July 1996, were included. They hadn a history of laking shark liver oil(so-called "squalene") for varying period of time. We reviewed clinical, radioloic and pathologic findings. Result: Patients look 7 to 30 capsules of "squalene" a day for at least one month to 5 years. Six cases had chronic disease such as diabetes, hypertension, or cerebrovascular accident. Respiratory symptoms of mild fever, cough and sputum were present in 3 cases and in 3 cases there was no clinical symptoms and signs but abnormal findings by chest X - ray. The major radiologic findings by simple chest X - ray and computed tomography consisted of consolidation, infiltration involving mainly right middle and both lower lobes, and ground-glass opacity. Five of six bronchoscopic examinations demonstrated both lipid droplets floating on the surface of bronchoalveolar lavage fluid and Lipid-laden macrophages in bronchoalveolar lavage fluid or lung tissue. Follow-up chest X -ray showed improvement in 4 cases but no marked interval change in 3 cases after removal of exposure to "squalene". Conclusion: Shark liver oil can induce lipoid pneumonia in adults. In case of high clinical suspicion, confirmation of "squalene" use by careful history taking is required and bronchoscopy is helpful in diagnosis.

Salvia bucharica belonging to the family Lamiaceae (Labiatae), afforded a novel terpenoid (1) named as salvatrione. The structure of 1 was elucidated through extensive 2-D NMR experiments and the biogenetic pathway of 1 has also been proposed. On biogenetic grounds, 1 may be considered to be a pseudo-triterpenoid as it is derived from the coupling of mono/ diterpenic units and not from squalene.

Atypical furnculosis caused by atypical Aeromonas salmonicida, is an emerging problem of farming of rockfish (Sebastes schlegeli) in Korea. In this study, protection against atypical furunculosis was compared in rockfish vaccinated with A. salmonicida formalin killed cell (FKC) and adjuvant containing FKC. The formalin inactivated A. salmonicida vaccine provided a low protection of 20% and 10% relative percent survival (RPS) at 44 and 58 days post vaccination. However, addition of adjuvant (squalene and aluminum hydroxide) into inactivated A. salmonicida vaccine clearly enhanced the level of protection showing 70% and 50% RPS at 44 and 58 days post vaccination.
Terbinafine is a synthetic allylamine that is available in an oral formulation and is used at a dosage of 250mg/day. It is used as an active antifungal agent and inhibits the fungal enzyme squalene epoxidase, which leads to the accumulation of the sterol squalene, which is toxic to the organism. The purpose of the present study was to evaluate the bioequivalence of two terbinafine tablets, Lamisil (Novartis Korea Ltd.) and Terbinex (C-TRI Ltd.), according to the guidelines of Korea Food and Drug Administration (KFDA). Eighteen normal male volunteers, 26.00$\pm$2.57 year in age and 70.51$\pm$9.36 kg in body weight, were divided into two groups and a randomized 2${\times}$2 cross-over study was employed. After one tablet containing 125 mg of terbinafine was orally administered, blood was taken at predetermined time intervals and the concentrations of terbinafine in plasma were determined using HPLC with UV detector. Pharmacokinetic parameters such as AUC, $C_{max}$ and $T_{max}$ were calculated and ANOVA test was utilized for the statistical analysis of the parameters. The results showed that the differences in AUC, $C_{max}$ and $T_{max}$ between two tablets were -4.191%, 5.223% and -25.720%, respectively when calculated against the Lamisil, tablet. The powers (1-$\beta$) for AUC, $C_{max}$ and $T_{max}$ were 81%, 87% and below 60%, respectively. Minimum detectable differences(.il) at alpha=O.1 and 1-/3=0.8 were less than 20% (e.g., 19.72% and 17.77% for AUC and $C_{max}$, respectively). But minimum detectable differences($\Delta$) at alpha=0.1 and 1-$\beta$=0.8 for $T_{max}$ were more than 20% (e.g., 26.25%). The 90% confidence intervals were within $\pm$20% (e.g., -17.440∼9.06 and -6.713∼17.160 for AUC and $C_{max}$ respectively). But 90% confidence intervals for $T_{max}$ were not within $\pm$20% (e.g., -43.346∼8.083). Another ANOVA test was conducted for logarithmically transformed AUC and $C_{max}$. These results showed that there are no significant differences in AUC and $C_{max}$ between the two formulations: The differences between the formulations in these log transformed parameters were all for less than 20% (e.g., -4.19% and 5.22% for AUC and $C_{max}$, respectively). The 90% confidence intervals for the log transformed data were not the acceptance range of log 0.8 to log 1.25 in AUC but the acceptance range of log 0.8 to log 1.25 in $C_{max}$ (e.g., log 1.13∼log 1.50 and log 0.94-log 1.22 for AUC and $C_{max}$ respectively). The major parameters, AUC and $C_{max}$ met the criteria of KFDA for bioequivalence although $T_{max}$ did not meet the criteria of KFDA (1998 year) for bioequivalence, indicating that Onfran tablet is bioequivalent to Zofran tablet. But in another ANOVA test AUC did not meet the criteria of KFDA (2002) for bioequivalence but $C_{max}$ met the criteria of KFDA (2002 year) for bioequivalence.or bioequivalence.equivalence.equivalence.equivalence.
