내독소내성 마우스에서 Nitric Oxide 생성에 미치는 림프구 부전 (Impaired Functions of Lymphocytes on Nitric Oxide Production in Endotoxin- Tolerant Mice)

  • 본 연구에서는 내독소내성 상태에 있는 마우스 세포의 싸이토카인 생성능을 측정하기 위하여 대식세포-림프구 공동배양계를 이용한 nitric oxide (NO) 생성을 조사하였다. 마우스 복강 대식세포에 lipopolysaccharide (LPS)와 interferon-g (IFN-g)를 처리시 NO 생성이 증가되었으며, tumor necrosis factor-a (TNF-a) 또한 LPS처럼 NO 합성을 자극하는 것을 관찰할 수 있었다. 한편, 대식세포를 비장세포와 공동배양시, LPS 단독처리만으로도 NO 합성이 증가되었다. 반면, 2.5 mg/kg LPS로 전처리하고 치사량의 LPS를 2차 투여한 마우스의 경우, 마우스의 치사 및 혈중 TNF-a와 IFN-g가 증가되지 않았다. 또한 LPS-내성 마우스로부터 분리한 대식세포를 정상 비장세포와 공동배양시 LPS에 의한 NO생성이 일어나지 않았으며, 외래 TNF-a에 의한 NO 생성도 일어나지 않았다. 이와 아울러 정상 대식세포와 LPS 내성 마우스로부터 분리한 비장세포를 공동배양하였을 때, LPS 자극으로 인한 NO 생성이 일어나지 않았으며, 이러한 억제현상은 외래 IFN-g 또는 IFN-g 생성을 촉진시키는 concanavalin A (ConA)에 의해서 다시 역전되었다. 이러한 결과는 대식세포 뿐만 아니라 림프구도 LPS 내성에 관여하는 것을 보여준다고 사료된다. INF-g는 TNF-a 발현을 증가시키기 때문에, 림프구의 INF-g 합성 감소는 LPS에 내성을 보이는 대식세포의 TNF-a 합성 저하와 상호작용으로 내독소내성 상태를 유도하며 과도한 염증반응을 억제하는 것으로 사료된다. 따라서 LPS 내독소내성은 중환자의 심각한 패혈증에 대한 예방법으로 활용될 수 있을 것으로 기대된다.

용각산의 면역조절 및 백혈병세포의 아폽토시스에 미치는 효과 (Effect of Yonggak-san on the Immuno-regulatory and Apoptosis of Leukemia cells)

  • The purpose of this research was to investigate the effect of Yonggak-san (YGS) on the immune reaction and apoptosis of leukemia cells. Administration of YGS(500 mg/kg) enhanced proliferation of splenocytes, thymocytes and mesenteric lymph node cells, and also YGS accelerated subpopulation of splenic Band T, thymic T and mesenteric lymph node-T lymphocytes, especially significantly increased CD4+-TH cells in BALB/c mice. YGS accelerated phagocytic activity and production of nitric oxide in peritoneal macrophages. YGS induced apoptosis of transplanted-L1210 cells in vivo, increased apoptotic cell death of cultured-L1210 and/or Molt4 human leukemia cells, decreased of mitochondrial transmembrane potential of both cells in vitro. These results suggest that YGS have an immune-regulatory effect and anti-cancer property.

은교산의 면역계에 미치는 효과 (Effects of Eunkyo-San on the Immune System)

  • The purpose of this research was to investigate the effect of Eunkyo-San(EKS) on the immune system. Administration of EKS(500 mg/kg) enhanced viability of splenocytes and thymocytes in BALB/c mice, and also EKS increased of splenic B, T lymphocytes and thymic T lymphocytes, significantly increased CD4 positive TH cells. EKS markedly enhanced the production of -interferon in mice serum. EKS accelerated the production of nitric oxide in peritoneal macrophages. These results suggest that EKS have an immune-enhancing activity.

산수유의 면역조절작용 (Immunoregulatory Action of Comus officinalis Sieb.et Zucc)

  • The purpose of this research was to investigate the immunoregulatory effect, apoptosis of L 1210 leukemia cells of Comus officinalis Sieb.et Zucc. The proliferation of cultured splenocytes and thymocytes were enhanced by the addition of SSY. Splenic, thymic and mesenteric lymph node-T lymphocytes, especially TH cells was significantly increased in SSY-administered (p.o. for 7 days) mice. SSY treatment induced the apoptosis of L1210 mouse leukemia cells. In addition, SSY accelerated the phagocytic activity and nitric oxide production in peritoneal macrophages. These results suggest that SSY have an immuno-regulatory property and anti-cancer effect.

Immunomodulatory Effect of Acidic Polysaccharide Fraction from Korean Red Ginseng (Panax ginseng)

  • Effects of red ginseng acidic polysaccharides (RGAP) on immune system were studied. The proliferation of spleen cells was induced by RGAP treatment per se. Cotreatment of lipopolysaccharide $(100\;{\mu}g/ml)$ or concanavalin A $(1\;{\mu}g/ml)$ with RGAP further stimulated the spleen cell proliferation. BALB/c mice treated with RGAP showed a slight splenic hyperplasia and increased antibody forming cell response to sheep red blood cells. Flow cytometry analysis revealed an influx of macrophages in the mice treated with RGAP.

  • The oral administration of Aurantii nobilis pericarpium (ANP) extract and Aurantii immaturi pericarpium (AIP) extract suppressed the cell viability of both thymocytes and splenocytes in BALB/c mice. The ANP extract (500 mg/kg) enhanced the population of $B220^+$ cells, and the AIP also enhanced the population of B220+ and Thy-1+ cells in splenocytes. The AIP extract enhanced the population of $CD4-CD8^+$ cells in splenic T-lymphocytes. However, the ANP did not affect, whereas the AIP enhanced the phagocytic activity and the nitric oxide production in peritoneal macrophages.

방풍통성산(方風通聖散) 전탕액(煎湯液)의 면역(免疫) 조절작용(調節作用) (Immunoregulative Action of Bangpoongtongsungsan)

  • The purpose of this research was to investigate effects of Bangpoongtongsungsan water extract(BTSE) on the immune reaction, anti-allergy action and anti-inflammatory action in BALB/c mice. The administration of BTSE (500mg/kg) enhanced the cell viability of thymocytes and the population of helper T cells in splenic T-lymphocytes. BTSE suppressed the production of nitric oxide, but enhanced the phagocytic activity in peritoneal macrophages. BTSE enhanced hemagglutination titer in mice. BTSE inhibited passive cutaneous anaphylaxis induced by egg albumin in rat, the lethal anaphylaxis induced by platelet activating factor and compound 48/80 in mice, and then inhibited the degranulation of peritoneal mast cells induced by compound 48/80. BTSE did not inhibit Arthus reaction, but inhibited the delayed type hypersensitivity induced by SRBC and contact dermatitis induced by DNFB. BTSE inhibited the acute hind paw edema induced by histamine after 30 minutes, the permeability of evans blue into peritoneal cavity induced by acetic acid and the writhing syndrome induced by acetic acid. These results suggest that BTSE has an immunopotentiative action, anti-allergy action and anti-inflammatory action via the inhibition of histamine release.

큰눈물버섯(Psathyrella velutina) 균사배양물로부터 분리한 단백다당체 PVMP의 면역활성 (Immunoactivities of PVMP, a Protein-polysaccharide Fraction Isolated from Mycelial Culture of Psathyrella velutina)

  • In the previous report, we described the marked antitumor and immunomodulatory activities of PVp, a protein-polysaccharide fraction of a Korean wild mushroom Psathyrella velutina. In this study, a protein-polysaccharide fraction, PVMP, was prepared from the shake-cultured mycelia of the same mushroom and its immunoactivities as well as chemical compositions were investigated. At 200 $\mu\textrm{g}$/ml, PVMP weakly stimulated the BALB/c mouse splenic lymphocytes to form lymphoblasts and upregulated the expression of CD25 molecules, but failed to stimulate peritoneal macrophages. In chemical analysis these two protein-polysaccharide fractions were found to be quite different in that the carbohydrate contents of PVMP and PVP, respectively, was 85.3% and 41.2%. These results reveals that PVMP, unlike PVP, is a moderate immunostimulator on the immune system.

브로콜리로부터 분리한 추출물의 In vitro 면역증진 활성평가 및 화학적 특성 (Chemical Properties and Assessment of Immunomodulatory Activities of Extracts isolated from Broccoli)

  • For the purpose of developing new immunomodulatory agents from broccoli, ethanol extract (BCEE), hot water extract (BCHW), and crude polysaccharide (BCCP) were isolated from broccoli, and their immunomodulatory activities and chemical properties were examined. In the in vitro cytotoxicity analysis, BCHW and BCCP did not affect the growth of tumor cells and normal cells. Murine peritoneal macrophages stimulated with BCCP showed higher production of IL-6, IL-12, and $TNF-{\alpha}$ cytokines than those stimulated with BCHW. Also, BCHW and BCCP did not show proliferation of splenic lymphocytes. In the in vitro assay for intestinal immunomodulatory activities, only BCCP enhanced GM-CSF secretion and the bone marrow cell-proliferating activity via cells in Peyer's patches at $1,000{\mu}g/mL$. Also, BCHW mainly contained 33.7% neutral sugars, such as arabinose, glucose, and galactose, and 30.7% uronic acid, and BCCP consisted of 42.6% neutral sugars, including arabinose, galactose, and glucose, and 50.5% uronic acid. The above results lead us to conclude that crude polysaccharide (BCCP) isolated from broccoli causes considerably high cytokine production in peritoneal macrophages and bone marrow cell proliferation, and the polysaccharide extraction process is indispensable for separation of new immunomodulatory agents from broccoli.

Inhibitory Effect of Electroacupuncture on Murine Collagen Arthritis and its Possible Mechanisms

  • The influence of electroacupuncture (EA), a traditional Chinese medical treatment, on type Ⅱ collagen-induced arthritis (CIA) was examined in DBA/1J mice in vivo. Mice were immunized intradermally twice at the 3-week interval with bovine type Ⅱ collagen(C Ⅱ). EA stimulation, begun on the 21 simultaneously with the second immunization, was applied at the acupoint equivalent to GV4 three times a week for 3 weeks. The results showed that EA delayed the onset, attenuated the severity of arthritis, and reduced the anti-collagen antibody level. Furthermore, we investigated the impact of EA on the productions of endogenous $interleukin-1{\Beta}$ (IL-1 beta) and prostaglandin E2 (PGE2), and the levels of IL-1 beta mRNA in splenocytes and synovial tissues from C Ⅱ immunized mice on the 45 and cyclooxygenase-2 (COX-2) mRNA in lipopolysaccharide (LPS)-stimulated macrophages of normal mice by using reverse transcriptase-polymerase chain reaction (RT-PCR). EA stimulation significant inhibited the concentrations of splenic endogenous IL-1 beta and serum PGE2. The expression of IL-1 beta mRNA in spleen cells was obviously down-regulated and that in synovial tissues was modestly affected by EA. COX-2 mRNA was highly expressed in cultured peritoneal macrophages when stimulated with LPS. Previous treatment with EA also reduced LPS-stimulated induction of COX-2 mRNA. These data suggest that EA has an inhibitory effect on murine CIA, and the partial mechanism of its therapeutic result may be attributed to inhibiting the productions of IL-1 beta and PGE2 by suppression the IL-1 beta and COX-2 gene activations.
