  • The purpose of this research was to investigate the effect of Maekmoondong-tang(MMDT) on the activity of immune cell. The addition of MMDT(10 ㎍/㎖) enhanced the proliferation of cultured-splenocytes and thymocytes. And administration of MMDT(250, 500 ㎎/㎏) accelerated subpopulation of splenic T lymphocytes in BALB/c mice. In contrast, the treatment of the high concentration (500 ㎎/㎏)of MMDT were decreased thymic T lymphocytes. Administration of MMDT(250, 500 ㎎/㎏) eminently enhanced the production of IFN-γ. And MMDT did not affect the cell viability of Jurkat leukemia cells. These results suggest that MMDT have a immunoregulatory effect via enhanced cell mediated immunity

  • In order to investigate the of effects of nalbuphine on immune system in mice, we examined the various immunological parameters. After single oral administration of nalbuphine (130, 260, 390 mg/kg, i.p.) to female ICR mite, the weights of bodies and organs (thymus, spleen, liver, kidney), and hematological parameters were examined on day 2, 4, 6, and 8. The increased rate of body weight, relative weight of organ, and hematological parameters in nalbuphine -treated groups, were not significantly changed when compared with control group. However, number of WBC was decreased by the treatment of nalbuphine. To assess the effects of nalbuphine on humoral immune responses, splenic IgM plaque forming cell (PFC) and serum IgM were assayed. When nalbuphine wat administered after immunization with SRBC, but not before immunization, splenic IgM PFC and ,serum IgM level against SRBC were significantly lowered in a dole -dependent manner. These results indicate that the suppressive effects of nalbuphine on primary humoral immune response may be dependent on the timing of its administration relative to the initial antigenic sensitization.

  • The purpose of this research was to investigate the effect of Sojagangqi-tang(SJGQT) on the immune cell activity. The addition of SJGQT enhanced the proliferation of cultured-mice splenocytes and thymocytes. Administration of SJGQT(250 mg/kg) accelerated the subpopulation of splenic T lymphocytes especially CD/sup 4+/-TH cells in BALB/c mice. But high concentration(500 mg/kg) of SJGQT decreased the splenic T, B lymphocytes and thymic Tc (CD/sup 8+/) lymphocytes. Oral administration of SJGQT(250 mg/kg) significantly enhanced the production of IFN-γ and IL-4 in mice serum. And also, the addition of SJGQT(100 ㎍/ml) inhibited the proliferation of cultured-Jurkat leukemia cells in vitro. These results suggest that SJGQT have a cellular immuno-modulatory effect and anti-cancer property action

  • Effects of swainsonine (SW;8${\alpha}$, ${\beta}$-indolizidine-${\alpha}$, $2{\alpha}$8-triol from Locoweed) on the humoral immune responses of lipopolysaccharide (LPS) wer studied in ICR mice. Mice were divided into 4 groups (10 mice/group), and LPS was given to each mouse 1 hr after i.p. injection with 3.7 mg/kg of swainsonine, by i.p. injection twice a week for 14 days at a dose of 2 mg/kg. Humoral immune responses were evaluated by hemagglutination (HA) titer and splenic plaque forming cells (PFC). The results of this study were summarized as follows: Mice administrated each of LPS and SW showed significant enhancement of the weight ratios of spleen to body, HA titer, 2-mercaptoethanol-resistant HA(MER-HA) titer and PFC compared with those in controls. However, the LPS plus SW treatment decreased HA titer, MER-HA titer and PFC corresponding to humoral immunity, as compared with those in the mice treated with LPS alone. These findings indicated that LPS significantly enhanced humoral immune responses, but their enhancement effects were lowered somewhat by SW.

  • Objectives: The object of this study was to observe antitumor, anticachexia and immunomodulatory effects of Shipyeukmiyeugi-eum(SYM) on human breast cancer cell, MCF-7, xenograft Balb/c nu-nu nude mice. Methods: Three different dosages of SYM-125, 250 and 500 mg/kg were orally administered once a day for 28 days from 11 days after tumor cell inoculation, and the changes on the body weights, tumor volume and weights, weights of spleen and popliteal lymph node and epididymal fat, serum IL-6 and IFN-${\gamma}$ levels, NK cell and peritoneal macrophage activities, splenic TNF-${\alpha}$, IL-$1{\beta}$ and IL-10 contents were observed. In addition, histopathological observations of apoptotic cell, spleen, popliteal lymph node and cervical brown adipose were also detected. The results were compared with a potent cytotoxic estrogen receptor antagonist, Tamoxifen 20 mg/kg treated mice. Results: Tumor volumes and weights were decreased without cytotoxic effects on the both MCF-7 and MCF-10A cells as results of all three different dosages of SYM treatment. And weights of body, spleen, popliteal lymph node, epididymal fat, serum IFN-${\gamma}$, NK cell, peritoneal macrophage activities, splenic TNF-${\alpha}$, IL-$1{\beta}$ and IL-10 contents were increased with decrease of serum IL-6. At histopathological observations, apoptotic tumor cells, spleen, popliteal lymph node and cervical brown adipose tissue were increased. That means tumor-related immunosuppress and cachexia were markedly inhibited by SYM treatment as compared with tumor-bearing mice. On the other hand, Tamoxifen showed marked cytotoxic effects against MCF-7 and MCF-10A, decreases of tumor volume and weights, and increases of apoptotic tumor cells and related decreases of tumor cell volumes, but tamoxifen markedly deteriorated the tumor-related immune-suppress and cachexia. Conclusions: The results obtained in this study suggest that SYM showed favorable anticancer effects and anticachexic effects on the MCF-7 cell xenograft through immunomodulatory effects. SYM did not induce any cytotoxic effects against both normal and cancer cells.

  • To investigate the preventive effects of melatonin (MLT) on the immunotoxicity of cadmium acetate[Cd(AC)2] in ICR mice, Mlt(10,50mg/kg as cadmium) were orally administered to mice once a day (5:00, PM) for 28 consecutive days. Cadmium(Cd) test solution was also administered at 25mg/kg of cadmium through the same route 2hr after administration of MLT daily, Mice were immunized and challenged with sheep red blood cells (SRBC). Immune functions evaluated were delayed type hypersensitivity (DTH) response, mitogenic response, and flow cytometry analysis. The results of these studies were summarized as follows ; DTH response was abnormally increased in mice treated with Cd alone. DTH response was normally depressed in mice treated with Cd plus MLT along with the increase of MLT doses. The mitogenic response of splenic T cell to Con A and that of B cells to LPS was remarkably increased by MLT treatment as compared with treatment of Cd alone In case of CD 8+ cells, the slight increase was observed in MLT treatment. Splenic T cells and B cells were significantly increased by MLT treatment as compared with treatment with Cd alone. These results suggest that MLT has significant preventive effects on the immunotoxic status induced by Cd exposure.

  • Pollen has been used for Prevention or treatment of certain diseases such as diabetes, arthritis, or cancer in traditional medicine. In addition, pollen is under investigation as a host cell for a gene expression. This study was undertaken to evaluate the immunologic safety of pollen intake. BALB/c mice were administered with 500, 50,5, or 0.5 mg/kg bw of lily pollen for five times a week for four weeks through gastric intubation. Comparing the control mice administered with distilled water, no significant changes were observed in body weight gain, weight of liver, spleen, lung, and his-topathological findings of liver and kidney of the mice groups administered with the pollen. Plasma level of IgG1, IgG2a, and IgE was not different among the groups. When splenic B lymphocytes were stimulated in vitro with lipopolysaccharides for 7 days, level of IgGl and IgGwa produced in the culture supernatants was not significantly different among the groups. Furthermore, no significant alteration was observed in IL-4 and $IFN{\gamma}$ producing ability with splenic T lymphocytes stimulated in vitro with phytohemagglutinins for 48 hours between the pollen-administered and the control mice. Overall, this study suggests that the lily pollen intake is Inducing no significant modulation of humoral and cell-mediated immunity in mice.

  • Pollen has been used for prevention or treatment of certain diseases such as diabetes arthritis or cancer in traditional medicine. Among various pollens, pine tree pollen is known to relieve hypertension, suppress fatty liver progression, and facilitate the digestion, but its immunological activities are less known. To evaluate immunological reactivities and immunotoxicities of pine tree pollen, BALB/c mice were administered to the poller through oral route. Pine tree pollen suspended in distilled water or extracted with methanol has been administered at the concentration of 0, 10, or 100 mg/kg five days per week for four weeks. Polyclonal activation of splenic T cells with phytohemagglutinins did not induce a significant difference in IL-4 and $IFN_{\gamma}$ production between the pollen-administered mice groups and the control mice. Furthermore, polyclonal activation of splenic B cells with lipopolysaccharides did not result a significant difference in IgG1 and IgG2a production among the groups. These findings imply that the intake of pine tree pollen does not bring any humoral and cellular immune-dysrequlation. Whereas, viability of Listeria monocytogenes was suppressed in the mice administered with 100 mg/kg bw methanol extract, indicating the potential ability of pine tree pollen to enhance cell-mediated immunity mediated by type-1 helper T cells. In addition, aberrant upregulation of plasma IgG1 level was observed in the pollen-administered mice, which suggests a possibility of allergic response induction through the pine tree pollen uptake. Overall, pine tree pollen-mediated modulation of humoral or cellular immunity is worthy of further systematic investigation.

In the expression of CD3e/CD4, CD3e/CD8, and B220 in peripheral blood mononuclea cells (PBMCs) of mice, GRT had no significant change to the normal group in CD4. However, GRT showed an increase to the normal group in CD8 and GHT in the only $1\mu\textrm{g}/ml$ category showed an increase to the normal group in B220. 4. In the cytotoxic effect of GRT on A549, SK-OY-3, SK-MEL-2 and MCF-7 cells, there was no significant cytotoxic effect compared to the control group. 5. In the inhibitory effect on the activity of DNA topoisomerase I, GHT in the $10\mu\textrm{g}/ml$ category showed the inhibitory effect on the activity of DNA topoisomerase I in proportion to the concentration. 6. In the T/C% in ICRmice bearing S-180, GHTtreated group showed 123.7% of T/C% compared to the control group. 7. In the inhibitory effect of cell adhesive of A549 Cells and SK-OV-3 Cells to complex extracellular matrix, GRT in the only $100\mu\textrm{g}/ml$ category showed the significant inhibitory effect compared to the control group. 8. In the inhibitory effect on lung colonies, GHT showed the significant inhibitory effect on lung colonies compared to the control group. 9. In the change of lung tissue, GHT showed a significant decrease of lung cancer growth, interalveolar fibrosis and hyaline material compared to the control group. In the development of lymphocyte around lung cancer cells and lung parenchymal, GHT showed the significant inducement efficacy compared to the control group. 10. In CAM assay, the antiangiogenic activity of GHT showed 30%. 11. In the effect on MMP-2 and MMP-9 gene expression in the RT1080 cell line, GHT had no significant inhibitory effect on MMP-2 and MMP-9 gene expression compared to the control group. According to the above results, it could be suggested that GHT has an antitumor activity and antimetastatic activity.

  • Effect of age and dietary fatty acid composition on immune responses were investigated in Sprague-Dawley male rets. The animals weighing 88.6$\pm$2.2g were fed 10% dietary fat (W/W, 20% of calorie) with P/S ratio of 0.5, 1 and 2, and in each group, there were three different levels of n-6/n-3 fatty acid ratio; 2, 4 and 8(3$\times$3). The experimental periods were 1 month, 6 months and 12 months. The results of this study were; 1) Weights of thymus and spleen were significantly reduced with increasing age, moreover thymus weights were reduced with increasing the degree of unsaturation in dietarty fatty acid at 12 month. 2) The proportion of splenic lymphocyte in the total T-cell was increased with increased with age. The proportion of helper T-cell was not changed, while the proportion of suppressor T-cell was decreased. Thus at 12 month, the ratio of helpe $r^pressor T-cell was appeared to increase significantly, and showed the tendency to increase by consuming the low amount of dieraty n-3 fatty acid. 3) Proliferation stimulated by Con A or PWM reduced significantly at 12 month in which are high in dietary P/S ratio, representing the similar pattern with decrease in thymus weight. 4) Natural killer cell activities were shown significantly higher at 1 month than those at 6 month or 12 month.th.
