Effects of Different Sources of Dietary n-3 Fatty Acids on Immune Response and Eicosanoids Production (급원이 다른 n-3 지방산이 흰쥐에서 면역반응과 Prostaglandin $E_2$ 및 Leukotriene $B_4$ 생성에 미치는 영향)

  • This study was planned to compare the effects of source of dietary n-3 fatty acids, i.e., tuna oil and perilla oil, on mitogenesis and production of prostaglandin E$_2$ and leukotriene B$_4$ in rats. Weaning male Sprague-Dawley rats were fed 3 different experimental diets for 4 weeks(Control: beef tallow 50% + sesame oil 50%, FO : beef tallow 50% + sesame oil 25% + tuna oil(27% docosahexaenoic acid) 25%, PO : beef tallow 50% + sesame oil 25% + perilla oil 25%). Food intakes were higher in FO group than in other groups, but body weight gains, food efficiency rates and weights of spleen were not different among groups. Proliferation of splenocyte to PWM(pokeweedmitogen) was higher in FO and PO group than control group. But there was no difference between FO and PO group. Response to ConA was not different among three groups. Serum PGE$_2$ levels were higher in control group than other groups. Serum LKB$_4$ levels were not different among groups. Therefore, it seemed that n-3 fatty acids increased the immune response by means of decreasing the PGE$_2$ production.

Effects of Plantago asiatica L. on antioxidative activities and lipid levels in hyperlipidemic Sprague-Dawley(SD) rats (질경이(Plantago asiatica L.) 추출물이 고지혈 유발 흰쥐의 항산화활성 및 지질농도에 미치는 영향)

  • This study was carried out to investigate the effects of Plantago asiatica L. extract on anti oxidative potential, free radical generation and the lipid levels in rats. Sprague-Dawley(SD) rats were divided into two groups based on their diet, as follows: the AIN-76 diet (control group), and the modified AIN-76 diet(cholesterol 0.5%) with 0.5% P. asiatica extract, for 7 weeks. The body weight and teed efficiency ratios of the two groups did not significantly differ. The antioxidative potentials more significantly increased in the group that was fed P. asiatica extract than in the control group(p<0.05). There was no difference in the rate of free radical generation, though. The weights of the organs, such as heart, kidney, liver, and spleen, of the rats in the two groups did not differ, though. The ratio of the HDL cholesterol to the total cholesterol in the P. asiatica group was significantly higher than that in the control group, and the other serum lipid parameters (total cholesterol, HDL cholesterol, triglyceride, and phospholipids) did not differ between the two groups. These results imply that supplementation with P. asiatica extract may improve the antioxidant potential and decrease the lipid levels in the blood.

Therapeutic Antiallergy Effect of Fermented Soy Curd by Pediococcus inopinatus Y2 (항알레르기에 대한 Pediococcus inopinatus Y2 유산균 발효 두유 커드의 치료 효과)

  • The use of fermented soy curd as a functional substance has been actively studied due to the anti-inflammatory and antiallergic activity of soybean protein hydrolyzate by enzymes of lactic acid bacteria. The present study investigated the potential of soy curd as a treatment for atopic dermatitis (AD). Pediococcus inopinatus Y2 (P. inopinatus Y2) lactic acid bacteria were inoculated into soy milk and fermented ($30^{\circ}C$, 24 hr). Changes in body weights, ear thicknesses, IgE concentrations, and weights of immune organs in ICR female mice were quantified. Moderate weight gain occurred in most of the groups. The ear thickness was lowest in the untreated group (no group), and it was allergic and thickened in the phthalic anhydride (PA) treatment group. Based on visual observations, as compared with the skin condition of the PA-induced AD group, the skin condition of the animals in the fermented soy curd (FSC) group was improved. Therefore, FSC by lactic acid bacteria seemed to improve AD. Based on the comparison of the weights of the spleen, thymus, and lymph nodes, as well as the results of the IgE analysis, soy milk, in addition to FSC, had a therapeutic effect. However, the antiallergy effects of FSC in terms of AD were far superior to those of soy milk. These results indicated that FSC can be used as a treatment for AD.

Immune-enhancing effects of a traditional herbal prescription, Kyung-Ok-Ko (전통적인 한방 처방 경옥고의 면역 증강 효과)

  • Objectives : A traditional herbal prescription, Kyung-Ok-Ko (KOK), has long been used in oriental medicine as an invigorant for age-related diseases, such as amnesia and stroke. However, the beneficial value of KOK for immune responses is largely unknown. Based on the above mentioned effects of KOK, other previous reports, and its use in traditional medicine, we hypothesized that KOK displays beneficial effects against methotrexate (MTX)-induced immune suppression. Methods : We investigated the effects of KOK (0.6 g/kg/day, oral (p.o.)) on deteriorated immunity caused by MTX (2 mg/kg/day, p.o.) in an immune suppression mouse model. MTX was fed to mice once a day for 7 days. After the immune responses of the mice deteriorated by MTX treatment, KOK in water was fed to the mice once a day for 14 days. We then measured the expression levels of various cytokines, such as T helper cell (Th1, Th2) cytokines, and the number of immune cells, such as spleen T cells, B cells, and macrophages. Results : The data showed that MTX decreased Th1 profiles (interferon $(IFN)-{\gamma}$, interleukin (IL)-2, IL-12) and the number of immune cells, and increased Th2 profiles (IL-4, IL-5, IL-13), which were normalized significantly by post-administration of KOK. However, there was no significant difference in body-weight gain between MTX- and KOK-treated mice. Conclusion : These results indicate that KOK has immune-enhancing functions and reduces immunotoxicity of MTX, suggesting that supplementation with KOK will improve immune responses clinically and be useful for the prevention of immune-related diseases.

  • This study assessed the impact of the toltrazuril derivative N-(4-(4-Fluorophenoxy)-3-methylphenyl) acetamide on natural cytotoxic cell (NCC) activity of olive flounder, Paralichthys olivaceus spleen. Five groups of fifteen olive flounder, comprising non-treatment and vehicle control groups, were randomly assigned. N-(4-(4-Fluorophenoxy)-3-methylphenyl) acetamide was injected intramuscularly at doses of 120, 150 and 200 mg/kg body weight; a total of ten injections were given over the course of 30 days. The NK activity of flounder splenic cells was evaluated against YAC-1, mouse lymphoma cells or HINAE cells with a choice of co-cultivation times of 4 or 18 hrs. In case of YAC-1 co-culture we observed a significant increase in cytotoxicity at a dose of 200 mg/kg, up to 3.06 times more than that of the control group. Only the trial with the 4 hrs co-culture produced a significant difference in the HINAE cell experiment; the experimental group at the 200 mg/kg dose exhibited the maximum cytotoxicity, demonstrating 2.3 times more cytotoxicity than the control group. Furthermore, the expression level of IL-12b was markedly induced in the group with 200 mg/kg, which was 6.62 times greater than that of the control group. In terms of the altered NK cell activity, the repeated high doses of N-(4-(4-Fluorophenoxy)-3-methylphenyl) acetamide can cause changes in the normal performance of immune function.

Comparison of Resistance to Fowl Typhoid among Crossbreed (산란계 합성종의 가금티푸스 저항성 비교 연구)

  • The present study was conducted to compare the disease resistance to fowl typhoid among White Leghorn (WL) known as possessing greater genetic resistance to Salmonella, Rhode Island Red (RIR) susceptible to the disease and their mating crossbreed, WL ${\times}$RIR. Also, it was carried out to compare fowl typhoid-resistance among Hyline white${\times}$Hyline brown(HwHb), Hw${\times}$lsa brown(Hwlb), Isa white${\times}$lb(Iwlb), Iw${\times}$ Hb(IwHb) and Ib ${\times}$ Iw(IbIw) in order to select crossbreed chickens which have disease resistance to fowl typhoid and simultaneously produce the brown eggs which our domestic consummers prefer There was no death of WL chickens challenged with Sal. gallinarum at each 2, 5 and 8 wk of age, while average 86.2 to 64.1% of RIR chickens tested died at the same challenge age during the test period of 2 weeks. Their maiting crossbreed, WL ${\times}$ RIR were intermediate between the two breeds. weight gains of tested chickens were significantly higher in WL and WL ${\times}$RIR without difference between them than RIR. In recovery rate of tested strain of Sal. gallinarum from braun, heart, fiver, spleen and cecum of chickens challenged, WL ${\times}$RIR fell between the other two breeds, with somewhat higher rate then WL but much lower then RIR. In the disease resistance determination test using HwHb, Hwlb, Iwlb, IwHb and IbIw$.$ crossbreed chickens experimentally infected with Sal. gallinarum, it was recognized that all of crossbreeds tested developed marked resistance to fowl typhoid, based on the results judged by the measurement parameters of mortality, weight gain, recovery rate of challenged strain of bacteria, and positive rate of agglutination antibody, and HwHb had a tittle higher disease resistance then ethers.

Effects of Puerariae Flos on Antioxidative Activities and Lipid Levels in Hyperlipidemic Sprague-Dawley Rats (갈화(Puerariae Flos) 추출물이 고지혈 유발 흰쥐의 항산화활성 및 지질농도에 미치는 영향)

The ratio HDL cholesterol to total cholesterol in the Puerariae Flos group was significantly higher than in the control group, while the other serum lipid parameters (total cholesterol, HDL cholesterol, triglyceride, and phospholipids) were not different between the two groups. These results imply that supplementation of Puerariae Flos extracts may beneficially contribute to improve antioxidant potential and to decrease the lipid levels in the blood.

The Food Safety of Superfine Powder (Phellinus linteus) Processed by Nanomill in C57BL/6 Mice (C57BL/6 마우스에서 나노밀 가공된 초미세분말(상황버섯)의 식이 안전성 연구)

  • A officinal mushroom, Phellinus linteus (PL) has been known to exhibit potent biological activities including antioxidative and anticancer effect. PL is consumed as a type of powder or extract for the purpose of health promotion and disease treatment. Recently superfine PL products was commercialized according to the development of pulverizing technology such as nanomill, so the evaluation of food safety is suggested. This study was conducted to evaluate the food safety of superfine PL (SPL) through hematological, biochemical and histological examination in mice as compared with fine PL (FPL). In the particle size distribution in volume after nanomill processing, the mean diameter of SPL and FPL particles was 11.78 ${\mu}m$ and 216.1 ${\mu}m$, and d (0.5), the particle diameter measured at 50% of distribution was 5.5 ${\mu}m$ and 147.9 ${\mu}m$, respectively. As the result of body weight, food intake and the weight of organs, SPL group didn't show any statistical difference compared with FPL group and normal group (N). Hematological and biochemical values were also involved in the normal range, although ALT (N vs. FPL, P<0.001) and BUN (N vs. FPL, P<0.01; N vs. SPL, P<0.01) showed significance compared with N group but there are no significance between FPL and SPL group. In the result of histological examination with liver, kidney, spleen, and small and large intestine, abnormal findings such as inflammatory reaction and histological changes were not observed. Our results suggest that the oral intake of SPL diet is not harmful to the animal in the hematological, biochemical and histological aspects although particle size was reduced to the level of superfine. However, further study will be necessary to confirm the histological safety in relation to the gastrointestinal contact of superfine particles in the case of large amount and long-term intake.

Effects of Liquid Culture of Agaricus blazei Murill on Lipid Metabolism and Enzyme Activities in Rats Fed High Fat Diet (신령버섯 균사체 액체배양액이 고지방 식이를 급여한 흰쥐의 지질대사 및 효소활성에 미치는 영향)

  • This study was performed to investigate the effects of liquid culture of Agaricus blazei on the lipid metabolism and enzyme activities in growing male rats. Sprague-Dawley rats were given four different types of diets for a succeeding period of 5 weeks, respectively: a normal diet group (7% corn oil), a high fat diet group (7% com oil + 15% lard), a 20 or 30% Agaricus diet groups (high fat diet + 20 or 30% Agaricus in water) according to the levels of Agaricus supplementation. The body weight gains, food intake, food efficiency ratios, and hepatic, kidney, spleen and pancreas weights of the rats fed 20 or 30% Agaricus diets were similar to those of the rats fed high fat diet. The epididymal fat pad weight of the rats fed high fat diet and 20 or 30% Agaricus diets were significantly higher than that of the rats fed normal diet. The concentrations of serum triglyceride, total cholesterol, LDL-cholesterol and HDL-cholesterol, and the activity of glutamic pyruvic transaminase in the rats fed 30% Agaricus diet were significantly lower than those in the rats 114 high fat diet. But the concentrations of hepatic total cholesterol and triglyceride of rats fed the 20 or 30% Agaricus diets were similar to those of rats fed the high fat diet. The HDL-oholesterol/total-cholesterol ratio of the rat fed 30% Agaricus diet was significantly higher than that of the rats fed high fat diet, The activity of glutamic oxaloacetic transaminase in the rats fed 20 or 30% Agaricus diets were similar to those in the rats fed high fat diet. There were no differences in the concentrations of total protein, hemoglobin and glucose, and the activities of alkaline phosphatase, and the atherogenic index in the serum among the experimental groups. These results showed that the 30% Agaricus diet feeding decreased the total cholesterol, the triglyceride and the LDL-cholesterol, and increased the HDL-oholesterol/total cholesterol ratio in serum of rats.

Effect of Dietary Crude Protein and Amino Acid Contents on Growth Performance and Carcass Characteristics in Korean Native Ducks (사료의 조단백질 및 아미노산 함량이 토종오리의 성장과 도체 특성에 미치는 영향)

  • This study was undertaken to assess dietary crude protein (CP) and amino acid (AA) concentrations for growth performance and carcass characteristics in Korean native ducks. In a $2{\times}3$ factorial arrangement, 1-d-old Korean native male ducks were allotted to 6 dietary treatments in a completely randomized design. Experimental diets contained 23 or 21% CP with 1.31/1.09, 1.21/1.00, 1.11/0.91 and 1.11/0.91, 1.02/0.83, 0.92/0.75 AA (Lysine/Total sulfur amino acid, Lysine/TSAA), respectively, from 0 to 3 wk of age. From 4 to 8 wk of age, experimental diets had 19 or 17% of diet; each contained 1.00/0.79, 0.94/0.75, 0.88/0.71 and 0.88/0.71, 0.82/0.67, 0.76/0.63 AA (Lysine/TSAA), respectively. Each dietary treatment has 6 replicates and feed and water were provided ad libitum. Body weight (BW), feed intake and uniformity were measured at 3 wk and 8 wk and carcass characteristics were evaluated at 8 wk of age. As CP increased from 21 to 23%, the BW and BW gain significantly increased (P<0.05) during 0 to 3 wk of age. From 4 to 8 wk of age, BW, feed intake, BW gain, feed conversion rations (FCR) and uniformity were not different (P>0.05) between treatments. Carcass yield and relative weights of liver, spleen, right breast and leg per 100 of BW were not different (P>0.05) between treatments. The meat color, shear force value, cooking loss and pH were not affected by dietary treatments (P<0.05). Korean native ducks require relatively low levels of dietary CP and AA for late growth and carcass yield due to low daily weight gain. This suggests the possible differences in CP and AA needs between Korean native ducks and commercial breeds from foreign breeding companies, especially late growth stage.
