Development of Physical Cell Lysis Using a Spiked CNT Membrane for Polyhydroxybutyrate Recovery (폴리하드록시부틸레이트 회수를 위한 물리적 세포 파쇄용 돌기형 탄소나노튜브 분리막 제작)

  • Conventional extraction methods for polyhydroxybutyrate (PHB), a sustainable alternative to petroleum-based plastics, cause a decrease in molecular weight and a change in properties. In this work, we developed a method to extract PHB accumulated in microorganisms by physical disruption through filtration using a spiked carbon nanotube (CNT) membrane with functionalized CNT. In addition, filtration of the PHB-containing microbial solution was performed to confirm PHB extraction, which was found to be 4% more efficient than chloroform, the most used extraction method. These results indicate that the spiked CNT membrane has potential in the bioplastics recovery process.

Evaluation of Accuracy and Precision of Analysis of Metals with Polyvinyl Chloride Membrane Filters (PVC 여과지를 이용한 금속 분석방법에 대한 정확도와 정밀도 평가)

  • Objectives: This study was conducted to evaluate the accuracy and precision of airborne metal analysis using polyvinyl chloride(PVC) membrane filter by pretreatment methods. Methods: A total of 75 spiked PVC samples for Cr, Fe and Mn ranged from 6 ug/sample to 40 ug/sample were used to evaluate recovery rates for three pretreatment methods: acid extraction, hot plate ashing and microwave digestion. For Mn, an additional 75 spiked mixed cellulose ester(MCE) membrane filters were analysed to compare the recovery rates of PVC samples. All samples were analysed with an inductively coupled plasma optical emission spectrometer(ICP-OES) and manganese samples were additionally analyzed by atomic absorption spectrometer(AAS). Results: The overall mean recovery rates of PVC samples for Cr, Fe and Mn were 90% or higher regardless of pretreatment methods, but there were statistically significant differences in recovery rates for Cr(p<0.05) and Mn(p<0.01) samples by pretreatment methods. The biases and the coefficient variations of PVC samples for three metals pretreated with three kinds of pretreatment methods ranged from 1.7% to 4.7% and from 1.6% to 6.5%, respectively. The manganese PVC samples pretreated by microwave digestion and analyzed with ICP-OES had the lowest bias at 1.9% and also showed lower bias than the bias for MCE samples, 2.7%. Conclusions: In order to accurately analyze the metals sampled with PVC membrane filters, microwave digestion and ICP-OES can be recommended.

Accuracy and Precision of Microwave Oven Digestion/Atomic Absorption Spectrophotometry for Analyzing Airborne Chromium Collected on MCE Filter in Plating Operation (도금공정 크롬시료 분석을 위한 Microwave Oven Digestion/Atomic Absorption Spectrophotometry 방법의 정확도 및 정밀도 평가)

  • The purpose of this study was to evaluate the analytical accuracy and precision of microwave oven digestion/atomi absorption spectrophotometry (AAS) for analyzing airborne chromium collected on mixed cellulose ester membrane (M filter from the work environment, and to compare the accuracy and the precision with those of the National Institute for Occupational Safety and Health (NIOSH) Method #7024 hot plate digestion/AAS method. For this study, field air sample pairs were collected from a electroplating process, and spiked samples in a laboratory were prepared and using these samples. Two digestion methods were comp; and evaluated in terms of recovery rate and bias as indices of accuracy and coefficient of variation as a index of precision. The results and conclusions are as follows. In spiked samples, the accuracies (% mean recoveries) of hot plate/AAS and microwave oven/AAS method were 97.19%, 97.1%, respectively, and the precisions (pooled respectively, and the precisions (pooled coefficient of variance, $CV_{pooled}$) 6.93% and 3.88%, respectively. The biases of hot plate ani microwave oven methods were 4.56 - 14.7% and 2.22 - 7.42% respectively. There was no statistically significant difference between hot plate and microwave oven methods recovery rates of spiked samples (p>0,05). Also, no statistically significant difference was shown among the concentrations of air samples determined by two method (p>0.05). In conclusion, microwave oven/AAS method h excellent accuracy and precision, and advantages such as time-saving and simple procedure in comparison with the classical NIOSH method. Therefore, this method can be use widely to analyze airborne chromium collected on MCE filter from the work environments.

Effect of Veterinary Antibiotics on the Growth of Lettuce

  • Veterinary antibiotics (VAs) has been used to treat animal disease and to increase body weight. However, released VAs in the soil via spreading of compost can transport to plant and affect its growth. Main purpose of this research was i) to monitor VAs concentration in plant and ii) to evaluate inhibition effect of VAs residuals on the plant growth. Red lettuce (Lactuca sativa) was cultivated for 35 days in the pot soil spiked with 3 different concertation (0.05, 0.5, $5.0mg\;kg^{-1}$) of chlortetracycline (CTC) and sulfamethazine (SMZ). After 35 days of cultivation, concentration of CTC and SMZ in the plant was measured. Residual of CTC and SMZ was only quantified at the range of $0.007-0.008mg\;kg^{-1}$ and $0.006-0.017mg\;kg^{-1}$ in the leaf and root respectively when high concentration ($5.0mg\;kg^{-1}$) of antibiotic was spiked in the soil. Leaf length and root mass was statistically reduced when $0.05mg\;kg^{-1}$ of CTC was spiked in the soil while no statistical difference was observed for SMZ treatment. This result might indicated that high $K_{ow}$ and $K_d$ value are the main parameters for inhibiting plant growth. Antibiotics that has a high $K_{ow}$ causing hydrophobicity and easy to bioaccumulate in the lipid cell membrane. Also, antibiotics that has a high $K_d$ properties can be sorbed in the root causing growth inhibition of the plant. Overall, management of VAs should be conducted to minimize adverse effect of VAs in the ecosystem.

Removal Process of Metallic Impurity for Silicon Surface Detergent by Ion Exchange (실리콘 표면처리에 있어서 이온교환 막에 의한 금속불순물의 제거공정)

  • HF purification performance of an ion exchange membrane(IEM) was evaluated with 0.5% HF spiked with 10ppb of Fe, Ni and Cu nitrates. The result show that after less than five turnovers through an IEM, the metallic impurity concentration drops below 1ppb. The decrease rate can be fitted to a model assuming the experimental tanks to be continuously stirred tank reaction and that the metallic impurity concentration after the IEM is a function of the single-pass purification efficiency of the membrane, the concentration before purification and the metals desorbed form the IEM. The Concentration after purification was investigated up to a cumulative Fe loading of 300ppb in the 23 liter recirculated loop. It increases linearly vs. cumulative loading and can be explained by the Langmuir theory resulting in a purification efficiency at the equilibrium of close to 99.5% in this loading regime.

Rejection property of geosmin and 2-Methylisoborneol (MIB) with high concentration level at multi stage nanofiltration (NF) membrane system (다단 나노여과 공정에서 고농도 geosmin 및 2-Methylisoborneol (MIB)의 제거특성)

  • Algal problem in drinking water treatment is being gradually increased by causing deterioration of water supplies therefore, especially taste and odor compounds such as geosmin and 2-MIB occur mainly aesthetic problem by its unpleasant effects resulting in the subsequent onset of complaints from drinking water consumer. Recently, geosmin and 2-MIB are detected frequently at abnormally high concentration level. However, conventional water treatment without advanced water treatment processes such as adsorption and oxidation process, cannot remove these two compounds efficiently. Moreover, it is known that the advanced treatment processes i.e. adsorption and oxidation have also several limits to the removal of geosmin and 2-MIB. Therefore, the purpose of this study was not only to evaluate full scale nanofiltration membrane system with $300m^3/day$ of permeate capacity and 90% of recovery on the removal of geosmin and 2-MIB in spiked natural raw water sources at high feed concentration with a range of approximately 500 to 2,500 ng/L, but also to observe rejection property of the compounds within multi stage NF membrane system. Rejection rate of geosmin and 2-MIB by NF membrane process was 96% that is 4% of passage regardless of the feed water concentration which indicates NF membrane system with an operational values suggested in this research can be employed in drinking water treatment plant to control geosmin and 2-MIB of high concentration. But, according to results of regression analysis in this study it is recommended that feed water concentration of geosmin and 2-MIB would not exceed 220 and 300 ng/L respectively which is not to be perceived in drinking tap water. Also it suggests that the removal rate might be depended on an operating conditions such as feed water characteristics and membrane flux. When each stage of NF membrane system was evaluated relatively higher removal rate was observed at the conditions that is lower flux, higher DOC and TDS, i.e., $2^{nd}$ stage NF membrane systems, possibly due to an interaction mechanisms between compounds and cake layer on the membrane surfaces.

Development of Techniques for Evaluating the Virus Removal Rate using Adenovirus (아데노바이러스를 이용한 바이러스 제거율 평가를 위한 기법 개발)

  • Waterborne infectious disease is induced by several pathogenic microbes such as bacteria, viruses and protozoans, and the cases caused by viral infection is currently increasing. Water treatment process could reduce the number of virus in the water, but there were many difficulties to completely remove the virus particles from water. Therefore, the membrane separation technology which was reported to effectively remove pollutants from raw water has attracted increasing attention and demand. Since its efficiency has been introduced, demands for evaluation method toward the membrane filtration process are increasing. However, progression of the method development is slow due to the difficulties in cultivation of several waterborne viruses from animal models or cell culture system. To overcome the difficulties, we used adenovirus, one of the commonly isolated pathogenic waterborne viruses which can grow in cell culture system in vitro. The adenovirus used in this study was identified as human adenovirus C strain. The adenovirus was spiked in the raw water and passed through the microfiltration membrane produced by Econity, a Korean membrane company, and then the viral removal rate was evaluated by real-time PCR. In the results, the amount of virus in the filtered water was decreased approximately by 5 log scale. Because coagulant treatment has been known to reduce filtering function of the membrane by inducing fouling, we also investigated whether there was any interference of coagulant. In the results, we confirmed that coagulant treatment did not show significant interference on microfiltration membrane. In this study, we found that waterborne virus can be effectively removed by membrane filtration system. In particular, here we also suggest that real-time PCR method can rapidly, sensitively and quantitatively evaluate the removal rate of virus. These results may provide a standard method to qualifying membrane filtration processes.

A Dipstick-Type Enzyme-Linked Immunosorbent Assay for the Detection of the Insecticide Fenitrothion in Food Samples

  • In a previous study, we obtained polyclonal antibodies against the organophosphorus insecticide fenitrothion and developed an enzyme-linked immunosorbent assay (ELISA) for this pesticide. Using these antibodies and an enzyme tracer, a direct competitive ELISA method specific for fenitrothion using a dipstick format was developed. Dipstick ELISA using antibodies to fenitrothion immobilized on an Immunodyne membrane allowed the quick visual detection of fenitrothion at concentrations above $10\;{\mu}g/L$. The $IC_{50}$ value of dipstick ELISA using reflectance detection was $27\;{\mu}g/L$ with a detection limit of $2\;{\mu}g/L$. The recovery of fenitrothion from spiked lettuce and rice samples using the dipstick ELISA ranged from 87-107%.

Characterization and Food Application of a Potentiometric Biosensor Measuring $\beta$-Lactam Antibiotics

  • $\beta$-Lactam antibiotics such as penicillin G, amoxicillin, and ampicillin were determined by a potentiometric biosensor system which exploited penicillinase immobilized on Immobilon cellulose nitrate membrane and a flat-bottomed pH electrode-as the biological component and transducer. The optimum reaction buffer for maximum sensitivity was found as 2 mM of sodium phosphate buffer (pH 7.2). The detection limit of the biosensor could be extended to 1 $\mu{M}$ of the analytes by increasing the enzyme loading for immobilization to 100 units/$m\ell$. The model samples spiked with each of the standard penicillins were measured for their biosensor responses and HPLC peak area, resulting in the relative responses of 82.1-103.5% and 79.5-106.1% for the biosensor method along with HPLC analysis, respectively. This result showed a good precision of the current biosensor method for screening the penicillin compounds.

Detection of Fish Pathogenic Viruses in Seawater Using Negatively Charged Membranes (Negatively Charged Membrane을 이용한 해수 중 어류질병바이러스의 검출)

  • After an outbreak of viral disease in an aquafarm, release of virus (es) from infected fish into environmental seawater has been suspected. In the present study, we utilized a negatively charged membrane (HA type) as an efficient method for concentration and detection of fish pathogenic viruses, specifically, megalocytivirus and viral hemorrhagic septicemia virus (VHSV) present in field-collected seawater samples or inoculated into seawater artificially. Positively charged viruses adsorbed onto the negatively charged membrane and were eluted with 1 mM NaOH (pH 10.5) following rinsing with 0.5 mM $H_2SO_4$ (pH 3.0). Megalocytivirus and VHSV particles isolated using anegatively charged HA membrane from seawater inoculated with each virus at a concentration of 10 viral particles/mL were of sufficient quantity to show positive results in atwo-step PCR (or RT two-step PCR); however, despite it being negatively charged, a cellulose acetate (CA) membraneshowed negative results. In quantitative PCR, the detection limits of the HA membrane for megalocytivirus and VHSV in seawater were 1.20E+00 viral particles/mL and 1.22E+01 viralparticles/mL, respectively. The calculated mean recovery yields from 1 L seawater spiked with known concentrations of megalocytivirus and VHSV particles were 28.11% and 23.00%, respectively. The concentrate of a 1-L sample of culturing seawater from the aquatank of flounder suffering from VHSV showed clear positive results in PCR when isolated with an HA, but not a CA, membrane. Thus, viral isolation using an HA membrane is a practical and reliable method for detection of fish pathogenic viruses in seawater.
