  • A study was conducted to compare and determine the incidence of ultrastructural alterations in the testes of Philippine carabaos and crossbred buffaloes. Thirteen Philippine carabao bulls and twenty five crossbred male buffaloes were used in this study. Testicular biopsy was used to get tissue samples which were prepared for histologic evaluation using the electron microscopy method. There was no significant difference in Sertoli cell alterations between Philippine carabaos and crossbred buffaloes. However, more crossbred buffaloes (40%) had both Sertoli cell and spermatogenic cell alterations which were significantly higher compared to the 7.7% occurrence in Philippine carabaos. Sertoli cells of crossbred buffaloes exhibited intracavitary structures and exaggerated infoldings of the nuclear envelope (36%), nuclear bleb (16%), and intracytoplasmic vacuolations (16%). Philippine carabaos exhibited few ultrastructural alterations which were mainly intracytoplasmic vacuolations in Sertoli cells (15%).

Evolutionary History of Two Paralogous Glyceraldehyde 3-Phosphate Dehydrogenase Genes in Teleosts

  • Glyceraldehyde 3-phosphate dehydrogenase(GAPDH) is a key enzyme for carbohydrate metabolism in most living organisms. Recent reports and our own searches of teleost species in publicly available genomic databases have identified at least two distinct GAPDH genes in a given species. The two GAPDH genes are located on the same chromosome in teleosts, whereas they are located on the different chromosomes in mammals. Thus, we reconstructed a phylogenetic tree to better understand the evolutionary history of the GAPDH genes in the vertebrate lineage. Our phylogenetic analysis revealed unambiguously that the two GAPDH genes of teleosts are phylogenetically closely affiliated to one of the cytosolic GAPDH and spermatogenic GAPDH-S of mammals. This indicates that the two paralogous GAPDH genes shared a common ancestor and subsequently underwent a gene duplication event during early vertebrate evolution. However, GAPDH-S of teleosts showed significant differences in the polypeptide residues and tissue distribution of its mRNA transcripts from that of mammals, implying they have undergone a different history of functionalization.

  • Testicular development of the male African lungfish, Protopterus annectens (Owen) was investigated histologically. The testis was found to be an elongated structure that possessed two distinct portions: an anterior spermatogenic part that was made up of a system of testicular tubules and a posterior vesicular part that invaded the kidney tissue. Spermatogenesis can be divided into five stages; primary spermatogonia, secondary spermatogonia, spermatocyte, spermatids and spermatozoa. Based on the gonadosomatic index (GSI) and histological changes observed, the reproductive cycle can be divided onto four distinct stages: resting and quiescent (December to February), growing (March to June) ripe and spent (July to August) and postspawning (September to November). The GSI was the maximum on July when reproductive cells were mature, ripe and ready for spawning; and the minimum in August after fish spawned.

북방산개구리의 정자형성주기에 관한 연구

  • 북방산개구리의 성체 수컷들 년중 채집하여 gonadosomatic index(GSI)와 정자형성과 정 및 정소내 testosterone양의 변화를 조사하였다 정자형성은 8월에 시작되어 9월에 가장 활발하게 진행되었으며 이때 GSI도 가장 높았고 세정관의 단면적도 가장 넓었다 번식기(2월) 이후 일정기간 동안에는(3월-7월) 정자형성이 정지되었으며, GSI나 세정관의 단면적도 최저치를 나타내었다. 정소내 testosterone양은 11월부터 급격히 증가하여 2월에 가장 높았으며 3월부터 급격히 감소되어 10월 까지 매우 낮은 농도를 유지하였다. Interstitial cell의 수도 동면기간이 찰동기 보다 훨씬 많았으며 핵의 크기도 동면기간이 훨씬 컷다. 따라서 testosterone의 생성 증가와 interstitial cell의 활성화가 일치함을 알 수 있었다. 본 결과들은 북방산개구리의 정 소에서는 정자형성과정이 불연속적으로 일어난다는 것과 2월 이후에는 testosterone의 양이 급격히 줄어드는 것으로 보아 이 개구리의 번식기가 2월임을 확인해 주고 있다.

Preventive Effects of Vitamin E and Catechin on Spermatogenic Disturbance Induced by Di-(2-Ethylhexyl) Phthalate in Rat (쥐에서 Di-(2-Ethylhexyl) Phthalate로 유발된 정자 형성 장애에 대한 Vitamin E와 Catechin의 예방 효과)

  • The purpose of the present study was to determine the preventive effects of the two antioxidant vitamin E and catechin on DEHP-induced disturbance of spermatogenesis in male rats. Rats at 4 weeks of age were randomly allocated into five groups with 20 animals per group. The first group was not any administrated as control. The second group was administrated DEHP (2 g/kg) daily for 14 days. The third group was administrated vitamin E (500 IU/kg) following DEHP treatment by the same method (daily for 14 days). The fourth group was administrated catechin (200 mg/kg) following DEHP treatment by the same method. The fifth group was co-administrated vitamin E (500 IU/kg) and catechin (200 mg/kg) following DEHP treatment by the same method. In order to determine the preventive effects, we examined pathological changes of testis with apoptotic index, and characteristics of sperm with computer assisted sperm analysis (CASA). Vitamin E and catechin supplementation were significantly prevented the testicular atrophy, apoptosis of germ cells in the seminiferous tubules and abnormal rate of sperm. Moreover, sperm concentration, viability and motility was significantly recovered in groups of alone and along with vitamin E and catechin. The results suggest that preventive effects of alone and along administration of vitamin E and catechin on DEHP-induced testicular atrophy damages have been demonstrated.

Radioprotective Effect of Mesna on Mouse Testis (Mesna의 쥐 고환에 대한 방사선 보호 효과)

  • Mesna has been used with ifosfamide to prevent urotoxicity in the treatment of testicular cancers. This drug also protected the toxicities of adriamycin without compromising cytostatic activity. With an idea of radioprotective role of sulfhydryl group of radioprotectors and of mesna decreasing the toxic effect of adriamycin which produces free radicals, mesna and radiation were administered to mice to study the protective effect of this drug and to identify the difference in regenerative capacity of the germ cells in the testis between radiation-treated and both mesna-and radiation-treated groups. The shape and numbers of spermatogenic cells in the seminiferous tubules were examined every week after irradiation. In both groups, initial reduction and later recovery in germ cell numbers and shape was observed. The lowest germ cell number was found around three weeks after irradiation. Mean germ cell number of the mesna-treated group was significantly higher than radiation-treated group at all observed periods (p<0.05). More competent regeneration was present in mesna-treated group. These results suggest that mesna protect the testis from radiation injury. Further study will be necessary to identify whether mesna protects other tissues from radiation and it does not hamper tumor control.

Effects of Allii tuberosi Semen Extract Solution on Reproductive Capacities in Mice (농도별(濃度別) 구자(?子) 투약(投藥)이 수컷 생쥐의 생식능력(生殖能力)에 미치는 영향(影響))

  • Purpose : These studies were undertaken to evaluate the effects of Allii tuberosi Semen (ATS) on the spermatogenic abilities such as concentration, motility and morphological normality of sperm from the testis and the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Materials and Methods : We used the 8-week-old mice and administered the 0.2 ml extract solution of ATS in the different concentration (0.1 mg/ml, 1 mg/ml, 10 mg/ml and 100 mg/ml) once a day for 60 days. The control group was administered the distilled water in the same way. After the administration of each extract solution, we examined the number of total, motile and normal sperm, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Also we observed the histological changes of isolated testis. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods. Results : The concentration of total sperm, the motility and normality of spermatozoa were significantly increased in ATS groups, especially in 1 and 10 mg/ml groups, compared to control group. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the ATS groups compared to the control group, respectively. Also, the activity of hyaluronidase was significantly increased in the ATS groups compared to the control group. In the antioxidant activity analysis, the activity of testicular peroxidase was significantly increased in the ATS groups compared to the control group, especially in 1 mg/ ml group. The activity of testicular catalase was increased in ATS groups. Conclusion : This study shows that ATS has the beneficial effect on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase and testicular peroxidase. We can suggest that ATS extract solution be useful for the treatment of male sexual dysfunctions and infertility.

Misuse of testosterone replacement therapy in men in infertile couples and its influence on infertility treatment

  • Objective: We investigated the clinical characteristics of men with testosterone replacement therapy (TRT)-induced hypogonadism and its effect on assisted reproductive technology (ART) in infertile couples. Methods: This study examined the records of 20 consecutive male patients diagnosed with azoospermia or severe oligozoospermia (< 5 × 106/mL) who visited a single infertility center from January 2008 to July 2018. All patients were treated at a primary clinic for erectile dysfunction or androgen deficiency symptoms combined with low serum testosterone. All men received a phosphodiesterase 5 inhibitor and TRT with testosterone undecanoate (Nebido®) or testosterone enanthate (Jenasteron®). Patients older than 50 years or with a chronic medical disease such as diabetes were excluded. Results: The mean age of patients was 37 years and the mean duration of infertility was 16.3 ± 11.6 months. At the initial presentation, eight patients had azoospermia, nine had cryptozoospermia, and three had severe oligozoospermia. Serum follicle-stimulating hormone levels were below 1.0 mIU/mL in most patients. Three ongoing ART programs with female factor infertility were cancelled due to male spermatogenic dysfunction; two of these men had normal semen parameters in the previous cycle. After withholding TRT, serum hormone levels and sperm concentrations returned to normal range after a median duration of 8 months. Conclusion: TRT with high-dose testosterone can cause spermatogenic dysfunction due to suppression of the hypothalamic-pituitary-testicular axis, with adverse effects on infertility treatment programs. TRT is therefore contraindicated for infertile couples attempting to conceive, and the patient's desire for fertility must be considered before initiation of TRT in a hypogonadal man.
