Determination of Seed Protein and Oil Concentration in Kiddny Bean by Near Infrared Spectroscopic Analysis (근적외 분광분석법을 이용한 강낭콩 종실단백질 및 지방의 비파괴 분석)

  • Near infrared spectroscopy (NIRS) is a rapid and accurate analytical method for determining the composition of agricultural products and feeds. An important merit of the NIRS analytical system is consistent predictions across instruments. However, proper calibration is the most important factor for a NIRS analytical system. Forty samples were obtained from Kyonggi-do Agricultural Research and Extension Services, and used to develop calibrations for crude protein content and crude oil content. Calibrations equations were developed using multiple linear regression (MLR). Accuracy and precision of NIRS predictions were adequate for quality measurement for the two constituents in kidney bean seed. In calibration sample sets (N=30), multiple correlation coefficient between NIR and lab measurements is 0.90 for seed, 0.97 for powder in seed protein concentration and 0.40 for seed and 0.92 for powder in seed oil concentration, respectively. It is concluded that NIRS method is suitable for the determination of seed composition in whole kidney bean.

Prediction of Nutrient Composition and In-Vitro Dry Matter Digestibility of Corn Kernel Using Near Infrared Reflectance Spectroscopy

  • Nutritive value analysis of feed is very important for the growth of livestock, and ensures the efficiency of feeds as well as economic status. However, general laboratory analyses require considerable time and high cost. Near-infrared reflectance spectroscopy (NIRS) is a spectroscopic technique used to analyze the nutritive values of seeds. It is very effective and less costly than the conventional method. The sample used in this study was a corn kernel and the partial least square regression method was used for evaluating nutrient composition, digestibility, and energy value based on the calibration equation. The evaluation methods employed were the coefficient of determination ($R^2$) and the root mean squared error of prediction (RMSEP). The results showed the moisture content ($R^2_{val}=0.97$, RMSEP=0.109), crude protein content ($R^2_{val}=0.94$, RMSEP=0.212), neutral detergent fiber content ($R^2_{val}=0.96$, RMSEP=0.763), acid detergent fiber content ($R^2_{val}=0.96$, RMSEP=0.142), gross energy ($R^2_{val}=0.82$, RMSEP=23.249), in vitro dry matter digestibility ($R^2_{val}=0.68$, RMSEP=1.69), and metabolizable energy (approximately $R^2_{val}$ >0.80). This study confirmed that the nutritive components of corn kernels can be predicted using near-infrared reflectance spectroscopy.

  • Considerable attention has been focused on the applications of flexible liquid crystal (LC)-based displays because of their many potential advantages, such as portability, durability, light weight, thin packaging, flexibility, and low power consumption. To develop flexible LCDs that are capable of delivering high-quality moving images, like conventional glass-substrate LCDs, the LC device structure must have a stable alignment layer of LC molecules, concurrently support uniform cell gaps, and tightly bind two flexible substrates under external tension. However, stable LC molecular alignment has not been achieved because of the layerless LC alignment, and consequently high-quality images cannot be guaranteed. To solve these critical problems, we have proposed a PDMS pixel-wall based bonding method via the IB irradiation was developed for fasten the two substrates together strongly and maintain uniform cell gaps. The effect of the IB irradiation on PDMS with PI surface was also evaluated by side structure configuration and a result of x-ray photoelectron spectroscopic analysis of PDMS interlayer as a function of binder with substrates. large number of PDMS pixel-walls are tightly fastened to the surface of each flexible substrate and could maintain a constant cell gap between the LC molecules without using any other epoxy or polymer. To enhance the electro-optical performance of the LC device, we applied an alignment method that creates pretilt angle on the PI surface via ion beam irradiation. Using this approach, our flexible LCDs have a contrast ratio of 132:1 and a response time of about 15 ms, resulting in highly reliable electro-optical performance in the bent state, comparable to that of glass-substrate LCDs.

  • Tunable diode laser absorption spectroscopy was performed for analysis of the H$_2$$^{18}$ O/H$_2$$^{16}$ O isotope ratio of a water sample which was enriched by the membrane distillation method. In order to improve the signal-to-noise ratio, the wavelength modulation spectroscopic method was used with a lock-in amplifier. The fringe noise could be suppressed by using the FFT (Fast Fourier Transform) lowpass filter and the optimization of the modulation depth of the laser frequency. The maximum deviation of $\delta$-value was measured to be$\pm$4$\textperthousand$.

Electronic Structure of Ce-doped ZrO2 Film: Study of DFT Calculation and Photoelectron Spectroscopy

  • In this study, we evaluated the change of electronic structure during redox process in cerium-doped $ZrO_2$ grown by sol gel method. By sol-gel method, we could obtain cerium-doped $ZrO_2$ in high oxygen partial pressure and low temperature. After post annealing process in nitrogen ambient, the film is deoxidized. We used spectroscopic and theoretical methods to analysis change of electronic structure. X-ray absorption spectroscopy (XAS) for O K1-edge and Density Functional Theory (DFT) calculation using VASP code were performed to verify the electronic structure of the film. Also, high resolution x-ray photoelectron spectroscopy (HRXPS) for Ce 3d was carried out to confirm chemical bond of cerium doped $ZrO_2$. Through the investigation of the electronic structure, we verified as followings. (1) During reduction process, binding energy of oxygen is increase. Simultaneously, oxidation state of cerium was change to 4+ to 3+. (2) Cerium 4+ and cerium 3+ states were generated at different energy level. (3) Absorption states in O K edge were mainly originated by Ce 4+ $f_0$ and Ce 3+, while occupied states in valance band were mainly originated from Ce 4+ $f_2$.

ANALYZING ISUAL SPECTROPHOTOMETER DATA USING A TWO-COLOR DIAGRAM METHOD

  • Transient luminous events (TLEs; sprites, elves, jets and etc.) are lightning-related optical flashes occurring above thunderstorms. Since the first discovery of sprites in 1989, scientists have learned a great deal about the morphological, spectroscopic and electromagnetic characteristics of TLEs through ground and spacecraft campaigns. However, most of the TLE studies were based on events recorded over US High Plains. To elucidate the possible biasing effects, space-borne observations are needed and have their merits. Imager of sprites and Upper Atmospheric Lightning (ISUAL) on the FORMOSAT-2 satellite is the first instrument to carry out a true global measurement of TLEs from a low- earth orbit. In this short paper, we apply a common astronomical data analysis technique, two-color diagram, on the ISUAL spectrophotometer (SP) data. By choosing appropriated bandpasses and converting the measured flux of TLEs into the unit of magnitude, two-color diagrams of TLEs can be constructed. We demonstrate that two-color diagrams, which were constructed from the narrow-band spectrophotometer data, can be used to classify different types of TLEs and trace their temporal evolution. The amount of reddening due to Earth's atmosphere can also be estimated from two-color diagrams assembled from the broad-band spectrophotometer data.

Sensing Technology for Rapid Detection of Phosphorus in Water: A Review

  • Purpose: Phosphorus is an essential element for water quality control. Excessive amounts of phosphorus causes algal bloom in water, which leads to eutrophication and a decline in water quality. It is necessary to maintain the optimum amount of phosphorus present. During the last decades, various studies have been conducted to determine phosphorus content in water. In this study, we present a comprehensive overview of colorimetric, electrochemical, fluorescence, microfluidic, and remote sensing technologies for the measurement of phosphorus in water, along with their working principles and limitations. Results: The colorimetric techniques determine the concentration of phosphorus through the use of color-generating reagents. This is specific to a single chemical species and inexpensive to use. The electrochemical techniques operate by using a reaction of the analyte of interest to generate an electrical signal that is proportional to the sample analyte concentration. They show a good linear output, good repeatability, and a high detection capacity. The fluorescence technique is a kind of spectroscopic analysis method. The particles in the sample are excited by irradiation at a specific wavelength, emitting radiation of a different wavelength. It is possible to use this for quantitative and qualitative analysis of the target analyte. The microfluidic techniques incorporate several features to control chemical reactions in a micro device of low sample volume and reagent consumption. They are cheap and rapid methods for the detection of phosphorus in water. The remote sensing technique analyzes the sample for the target analyte using an optical technique, but without direct contact. It can cover a wider area than the other techniques mentioned in this review. Conclusion: It is concluded that the sensing technologies reviewed in this study are promising for rapid detection of phosphorus in water. The measurement range and sensitivity of the sensors have been greatly improved recently.

Green Synthesis of Silver Nanoparticles Using Cell Extracts of Anabaena doliolum and Screening of Its Antibacterial and Antitumor Activity

  • In the present work, we describe a simple, cheap, and unexplored method for "green" synthesis of silver nanoparticles using cell extracts of the cyanobacterium Anabaena doliolum. An attempt was also made to test the antimicrobial and antitumor activities of the synthesized nanoparticles. Analytical techniques, namely UV-vis spectroscopy, X-ray diffraction, Fourier transform infrared (FTIR) spectroscopy, transmission electron microscopy (TEM), and TEM-selected area electron diffraction, were used to elucidate the formation and characterization of silver-cyanobacterial nanoparticles (Ag-CNPs). Results showed that the original color of the cell extract changed from reddish blue to dark brown after addition of silver nitrate solution (1 mM) within 1 h, suggesting the synthesis of Ag-CNPs. That the formation Ag-CNPs indeed occurred was also evident from the spectroscopic analysis of the reaction mixture, wherein a prominent peak at 420 nm was noted. TEM images revealed well-dispersed, spherical Ag-CNPs with a particle size in the range of 10-50 nm. The X-ray diffraction spectrum suggested a crystalline nature of the Ag-CNPs. FTIR analysis indicated the utilization of a hydroxyl (-OH) group in the formation of Ag-CNPs. Ag-CNPs exhibited strong antibacterial activity against three multidrug-resistant bacteria. Additionally, Ag-CNPs strongly affected the survival of Dalton's lymphoma and human carcinoma colo205 cells at a very low concentration. The Ag-CNPs-induced loss of survival of both cell types may be due to the induction of reactive oxygen species generation and DNA fragmentation, resulting in apoptosis. Properties exhibited by the Ag-CNP suggest that it may be used as a potential antibacterial and antitumor agent.

A Study on the Viability of Human Dermal Fibroblast Cell by Media for Ni-Cr alloy elution (치과용 Ni-Cr합금 용출배지에 의한 인간 피부 섬유아세포 성장도 관찰 연구)

  • Purpose: Standards of alloy for porcelain fused to metal crown be classified by metallic factor and biological factor. Metallic factors consist of stability of alloy composition and mechanical strength and surface characteristics for chemical bond. Biological factors be considered properties of metallic elements and problems originated by toxicity and hypersensitive reaction. Alloys considered such controversial points are the most suitable alloy for dental instrument. Method: Alloys added Be and Nb using Ni-Cr alloy which has been widely used for dental instrument be selected and classified experimental group. Non-addition Be and Nb to Ni-Cr alloy classify control group and addition Be alloy is Be-experimental group, addition Nb alloy is Nb-experimental group. Specimens for cytotoxicity analysis gave effect to washing and sterilization. and then made an experiment on elution with cell medium after disinfection. It conducted specimens within cell medium with 24hours, 48hours, 72hours, respectively. It cultured human dermal fibroblast(HDF) using cell medium for cytotoxicity test and then investigated elution rate through spectroscopic analysis by MTT-assay. Result: As results of cytotoxicity test by MTT-assay, cultured cell rate of VII measured more low numerical value within elution medium for 24hours focused on control group. Also, cultured cell rate of K3 alloys observed low value for 48hours, 72hours than value of control group. Conclusion: According to final result that synthesize above results, Ni-Cr alloy added Be and Ni has little difference in Cytotoxicity by MTT-assay.

Application of Infrared Spectroscopical Techniques for Investigation of Archaeological Woods (적외선(赤外線)(IR) 분광법(分光法)에 의한 고목재(古木材) 성상(性狀)의 심지(深知))

  • Infrared (IR) spectroscopic techniques for the analysis of wood samples and the absorbance spectra of solid woods were presented. KBr pellets were prepared by throughly mixing approximately 300 mg of dried KBr and 1 mg of finely milled wood powder extracted with ethanol-cyclohexane previously. This mixture was made into a transparent disc by means of a pellet-making die (10 ton/$cm^2$ for 10 min). This IR techniques were applied for the analysis of archaeological wood samples. The most notable difference in the IR spectra between the recent and the archaeological waterlogged woods is that the absorption band centered at $1,730cm^{-1}$ was significantly diminished in the waterlogged ones. Total loss of absorption in $1,730cm^{-1}$ might be mainly due to the result of hemicellulose degradation. Another feature indicated by IR spectral comparision are that the degraded waterlogged wood samples showed 1) the increased intensity of the 1,600, 1,500 and $1,270cm^{-1}$ due to the residual lignin and the increased intensity at 1,470 and $1,425cm^{-1}$ due to the degradation of hemicellulose and 2) to the emergence of single band around $1,050cm^{-1}$ instead of three bands at 1,110, 1,060 and $1,040cm^{-1}$ in recent wood due to the degradation of cellulose crystalline. It was revealed from the IR examinations that the first change of wood in the waterlogged situation was the lysis of hemicellulose and the second the lysis of cellulose. It was also suggested that IR spectroscopy could serve a fast method for the investigation on the chemical characteristics of archaeological wood samples.
