Highly Pure Culture of Photosynthetic Bacteria for Pilot-scale Production (광합성세균의 파이롯트 단위 생산을 위한 고순도 배양)

  • Lab-scale and pilot-scale productions of photosynthetic bacteria, that were able to efficiently treat wastewater from aquacultural farm, were experimented for their highly-pure culture. The results of experiments in a gas pack reactor, an anaerobic flask and a flask using $N_2$ gas as N-source showed that only photosynthetic bacteria formed red colonies on agar plate and their purity was over 90% in a colony, observed under a microscope. It was found that the basal medium could most promote the growth of photosynthetic bacteria, confirmed by experiments of serial cultures on various media. Under the culture conditions, the specific growth rate was found to be $0.18h^{-1}$ from the culture in 5L bioreactor and the same value could be obtained in pilot-scale production.

Improving Biomass Productivity of Freshwater microalga, Parachlorella sp. by Controlling Gas Supply Rate and Light Intensity in a Bubble Column Photobioreactor (가스공급속도 및 광도조절을 이용한 담수미세조류 Parachlorella sp.의 바이오매스 생산성 향상)

  • The objective of the present study was to improve the biomass productivity of newly isolated freshwater green microalga Parachlorella sp. This was accomplished by culture conditions optimization, including CO2 concentration, superficial gas velocity, and light intensity, in 0.5 L bubble column photobioreactors. The supplied CO2 concentration and gas velocity varied from 0.032% (air) to 10% and 0.02 m/s - 0.11 m/s, respectively, to evaluate their effects on growth kinetics. Next, to maximize the production rate of Parachlorella sp., a lumostatic operation based on a specific light uptake rate (qe) was applied. From these results, the optimal CO2 concentration in the supplied gas and the gas velocity were determined to be 5% and 0.064 m/s, respectively. For the lumostatic operation at 10.2 µmol/g/s, biomass productivity and photon yield showed significant increases of 83% and 66%, respectively, relative to cultures under constant light intensity. These results indicate that the biomass productivity of Parachlorella sp. can be improved by optimizing gas properties and light control as cell concentrations vary over time.

Anaerobic Digestion of Distillery Wastewater in a Two-phase UASB System (이상 UASB 공정을 이용한 주정폐수의 혐기성소화)

  • A two-phase UASB system was operated for high-rate treatment of concentrated distillery wastewater. The phase separation was obtained by adjusting pH in each reactor. When influent SS concentration was 4.1/g/l, the first phase UASB reactor was effectively operated up to the loading rate of 16.5kg $COD/m^3.day$, producing 3.9g HAc/l.day. In the methanogenic UASB reactor, loading rate up to 44kg $COD/m^3.day$ could be applied while removing 80% of influent COD with a specific gas production of 16.5 l/l. day. After the formation granular sludge in both reactors, it was possible to maintain the appropriate pH in the first phase only by recirculating the effluent from methanogenic phase without the addition of alkaline chemicals.

Photoproduction of Hydrogen from Acetate by Rhodopseudomonas: Effect of Culture Conditions and Sequential Dark/Light Fermentation

  • Rhodopseudomonas palustris P4 can produce $H_2$ either from CO by water-gas shift reaction or from various sugars by anaerobic fermentation. Fermentative $H_2$ production by P4 is fast, but its yield is relatively low due to the formation of various organic acids. In order to increase $H_2$ production yield from glucose, P4 was investigated for the photo-fermentation of acetate which is a major by-product of fermentative $H_2$ production. Experiments were performed in batch modes using both light-grown and dark-grown cells. When the dark-grown P4 was challenged with light and acetate, $H_2$ was produced with the consumption of acetate after a lag period of 25 h. $H_2$ production was inhibited when a nitrogen source, especially ammonium, is present. When the dark-fermentation broth containing acetate was adopted for photo-fermentation with light-grown cells, $H_2$ production and concomitant acetate consumption occurred without a lag period. The $H_2$ yield was estimated as 2.4 - 2.8 mol $H_2/mol$ acetate and the specific $H_2$ production rate was as 9.8 ml $H_2/g$ cell${\cdot}$h, The fact that a single strain can perform both dark- and light-fermentation gives a great advantage in process development Compared to a one-step dark-fermentation, the combined dark- and light-fermentation can increase the $H_2$ production yield on glucose by two-fold.

  • Liquid hydrogen has the best storage capacity per unit mass and is economical among storage methods for using hydrogen as fuel. As the demand for hydrogen increases, the need to develop a storage and supply system of liquid hydrogen is emphasizing. In order to liquefy hydrogen, it is necessary to pre-cool it to a maximum inversion temperature of -253℃. The Gifford-McMahon (GM) refrigerator is the most reliable and commercialized refrigerator among small-capacity cryogenic refrigerators, which can extract high-efficiency hydrogen through liquefied hydrogen production and boil of gas re-liquefaction. Therefore, in this study, the optimal conditions for liquefying gas hydrogen were sought using the GM cryocooler. The process was simulated by PRO/II under various cooling capacities of the GM refrigerator. In addition, the flow rate of hydrogen was calculated by comparing with specific refrigerator capacity depending on the pressure and flow rate of a refrigerant medium, helium. Simulations were performed to investigate the optimal values of the liquefaction flow rate and compression pressure, which aim for the peak refrigeration effect. Based on this, a liquefaction system can be selected in consideration of the cycle configuration and the performance of the refrigerator.

Operation of UASB Reactor for Treatment of Dairy Wastewaters (유가공폐수 처리를 위한 UASB 반응조 운전)

  • The performance and the operational problems of UASB(Upflow Anaerobic Sludge Blanket) reactor for treatment of dairy wastewaters were investigated. Synthetic milk wastewater was successfully treated up to the loading rate of 3.9kg $COD/m^3.day$, with a specific gas production rate of 1. 23 I/I. day and a COD removal efficiency of over 90%. However, the sludge rising was observed at the loading rate of 2.1kg $COD/m^3.day$, due probably to the formation of scum layer at the surface of settling compartment. The BMP(biochemical methane potential) of raw milk wastewater and ice cream wastewater, measured by using SBT(serum bottle test), were 0.135 and 0.66ml $CH_4/mg\;COD_{added}$, respectively. The sludge activity increased more than 8 times from 0.159g $COD-CH_4/g$ VSS. d during 90 days of operation.

  • The anaerobic bacterium P. productus was known to produce acetate from CO, C02 and H2. In this research the acetate f formation from waste gas was studied. For this research, kinetic parameter study on CO conversion were carried out. From t this study maximum CO conversion rate of 39.3 mmol/L . hr . 0.0 and Km of 0.578 atm were obtained. Also the effect of c CO refreshment, N source, initial pH and c비ture temperature on acetate formation were studied. Acetate formation in 5L lab s scale fermenter was tested and specific acetate production rate of 0.48 g/L-hr-O.O. was obtained and the acetate c concentration was 21 g/L.

NOx Reduction Characteristics of Ship Power Generator Engine SCR Catalysts according to Cell Density Difference (선박 발전기관용 SCR 촉매의 셀 밀도차에 따른 NOx 저감 특성)

  • The selective catalytic reduction (SCR) is known as a very efficient method to reduce nitrogen oxides (NOx) and the catalyst performs reduction from nitrogen oxides (NOx) to nitrogen (N2) and water vapor (H2O). The catalyst, which is one of the factors determining the performance of the nitrogen oxide (NOx) ruduction method, is known to increase catalyst efficiency as cell density increases. In this study, the reduction characteristics of nitrogen oxides (NOx) under various engine loads investigated. A 100CPSI(60Cell) catalysts was studied through a laboratory-sized simulating device that can simulate the exhaust gas conditions from the power generation engine installed in the training ship SEGERO. The effect of 100CPSI(60Cell) cell density was compared with that of 25.8CPSI(30Cell) cell density that already had NOx reduction data from the SCR manufacturing. The experimental catalysts were honeycomb type and its compositions and materials of V2O5-WO3-TiO2 were retained, with only change on cell density. As a result, the NOx concentration reduction rate from 100CPSI(60Cell) catalyst was 88.5%, and IMO specific NOx emission was 0.99g/kwh satisfying the IMO Tier III NOx emission requirement. The NOx concentration reduction rate from 25.8CPSI(30Cell) was 78%, and IMO specific NOx emission was 2.00g/kwh. Comparing the NOx concentration reduction rate and emission of 100CPSI(60Cell) and 25.8CPSI(30Cell) catalysts, notably, the NOx concentration reduction rate of 100CPSI(60Cell) catalyst was 10.5% higher and its IMO specific NOx emission was about twice less than that of the 25.8CPSI(30Cell) catalysts. Therefore, an efficient NOx reduction effect can be expected by increasing the cell density of catalysts. In other words, effects to production cost reduction, efficient arrangement of engine room and cargo space can be estimated from the reduced catalyst volume.

Study on Rumen Cellulolytic Bacterial Attachment and Fermentation Dependent on Initial pH by cPCR (cPCR 기법을 이용한 초기배양 pH에 의한 반추위 섬유소 분해 박테리아의 부착 및 발효에 관한 연구)

  • The cPCR technique was used to monitor rumen fermentation and attachment of Fibrobacter succinogenes to cellulose at different pH in the in vitro culture medium. The target fragments of 16S rDNA(445 bp) were amplified from genomic DNA of F. succinogenes with specific primers and internal controls(205 bp) were constructed. Cell counts were estimated from the amounts of genomic DNA, which was calculated from cPCR results. F. succinogenes in pH 6.8 and 6.2 showed apparently higher attachment than in pH 5.8 during all incubation time. There were some difference between pH 6.8 and 6.2 in the degree of attachment, but the different was not significant (P>0.05). Cellulose degradation increased in process of incubation time and the increasing rate was higher when initial pH was higher. The pH in culture medium decreased regardless of initial pH in course of incubation time. After 24 h of incubation, medium pH was dropped by 0.24, 0.58 and 0.16 units from original medium pH 6.8, 6.2 and 5.8, respectively. More gas was produced at higher initial pH in the same manner as in cellulose degradation. In summery, Initial pH of rumen culture in vitro significantly influenced cellulose digestion, gas production, pH change and bacterial attachment. Especially, low pH(5.8) resulted in much lower bacterial attachment and fiber digestion compared to higher medium pH.

Influence of COD/Nitrate-N Ratio on Denitrification and Methanogenesis in Anaerobic Sludge (COD/Nitrate-N비가 혐기성 슬러지에서 탈질과 메탄화에 미치는 영향)

  • Simultaneous denitrification and methanogenesis at various influent COD/Nitrate-N(below C/N) ratio was investigated with two different anaerobic sludges of which one was inoculated with glucose only and the other was inoculated with glucose and nitrate. Three different C/N ratios of 30, 20, and 10 were applied with each anaerobic sludge in serum bottles agitated in a rotary shaker to monitor the time related changes of amount of gas production and gas composition. Glucose was used as a single carbon source, which was adjusted to 1,000 mg/L as COD and potassium nitrate was used as a nitrogen source to adjust the each C/N ratio. COD removal by methanogenesis was dominant at higher C/N ratios, while COD removal by denitrification was dominant at lower C/N ratios. With the sludges cultured at C/N ratios below 10, SMA and SDNR rising different carbon substrates were investigated. The most efficient carbon substrate was acetate and the highest SMA and SDNR were 0.76 g COD/g VSS day with pure anaerobic sludge and 1.38 g ${NO_3}^--N/g$ VSS day with sludge cultured at C/N ratio of 5, respectively.
