DNA Structural Perturbation Induced by the CPI-Derived DNA Interstrand Cross-linker : Molecular Mechanisms for the Sequence Specific Recognition

  • The highly potent cytotoxic DNA-DNA cross-linker consists of two cyclopropa[c]pyrrolo[3,4-3]indol-4(5H)-ones insoles [(+)-CPI-I] joined by a bisamido pyrrole (abbreviated to "Pyrrole"). The Pyrrole is a synthetic analog of Bizelesin, which is currently in phase II clinical trials due to its excellent in vivo antitumor activity. The Pyrrole has 10 times more potent cytotoxicity than Bizelesin and mostly form DNA-DNA interstrand cross-links through the N3 of adenines spaced 7 bp apart. The Pyrrole requires a centrally positioned GC base pair for high cross-linking reactivity (i.e., $5^1$-T$AT_2$A*-$3^1$), while Bizelesin prefers purely AT-rich sequences (i.e., $5^1$-T$AT_4$A*-$3^1$, where /(equation omitted) represents the cross-strand adenine alkylation and A* represents an adenine alkylation) (Park et al., 1996). In this study, the high-field $^1$H-NMR and rMD studies are conducted on the 1 1-mer DNA duplex adduct of the Pyrrole where the 5′(equation omitted)TAGTTA*-3′sequence is cross-linked by the drug. A severe structural perturbation is observed in the intervening sequences of cross-linking site, while a normal B-DNA structure is maintained in the region next to the drug-modified adenines. Based upon these observations, we propose that the interplay between the bisamido pyrrole unit of the drug and central C/C base pair (hydrogen-bonding interactions) is involved in the process of cross-linking reaction, and sequence specificity is the outcome of those interactions. This study suggests a mechanism for the sequence specific cross-linking reaction of the Pyrrole, and provides a further insight to develop new DNA sequence selective and distortive cross-linking agents.

Fracture behaviors of tunnel lining caused by multi-factors: A case study

  • The cracking and spalling caused by fracture of concrete lining have adverse impacts on serviceability and durability of the tunnel, and the subsequent maintenance work for damaged structure needs to be specific to the damaging causes. In this paper, a particular case study of an operational tunnel structure is presented for the serious cracking and spalling behaviours of concrete lining, focusing on the multi-factors inducing lining failure. An integrated field investigation is implemented to characterize the spatial distribution of damages and detailed site situations. According to results of nondestructive inspection, insufficient lining thickness and cavity behind lining are the coupled-inducement of lining failure bahaviors. To further understanding of the lining structure performance influenced by these multiple construction deficiencies, a reliable numerical simulation based on extended finite element method (XFEM) is performed by using the finite element software. The numerical model with 112 m longitudinal calculation, 100 m vertical calculation and 43 m vertical depth, and the concrete lining with 1450 solid elements are set enrichment shape function for the aim of simulating cracking behavior. The numerical simulation responses are essentially in accordance with the actual lining damaging forms, especially including a complete evolutionary process of lining spalling. This work demonstrates that the serious lining damaging behaviors are directly caused by a combination of insufficient thickness lining and cavity around the surrounding rocks. Ultimately, specific maintenance work is design based on the construction deficiencies, and that is confirmed as an efficient, time-saving and safe maintenance method in the operational railway tunnel.

An Intelligent Wireless Sensor and Actuator Network System for Greenhouse Microenvironment Control and Assessment

  • Purpose: As application-specific wireless sensor networks are gaining popularity, this paper discusses the development and field performance of the GHAN, a greenhouse area network system to monitor, control, and access greenhouse microenvironments. GHAN, which is an upgraded system, has many new functions. It is an intelligent wireless sensor and actuator network (WSAN) system for next-generation greenhouses, which enhances the state of the art of greenhouse automation systems and helps growers by providing them valuable information not available otherwise. Apart from providing online spatial and temporal monitoring of the greenhouse microclimate, GHAN has a modified vapor pressure deficit (VPD) fuzzy controller with an adaptive-selective mechanism that provides better control of the greenhouse crop VPD with energy optimization. Using the latest soil-matrix potential sensors, the GHAN system also ascertains when, where, and how much to irrigate and spatially manages the irrigation schedule within the greenhouse grids. Further, given the need to understand the microclimate control dynamics of a greenhouse during the crop season or a specific time, a statistical assessment tool to estimate the degree of optimality and spatial variability is proposed and implemented. Methods: Apart from the development work, the system was field-tested in a commercial greenhouse situated in the region of Punjab, India, under different outside weather conditions for a long period of time. Conclusions: Day results of the greenhouse microclimate control dynamics were recorded and analyzed, and they proved the successful operation of the system in keeping the greenhouse climate optimal and uniform most of the time, with high control performance.

  • The fiber reinforced composite material is widely used in the multi-industrial field where the weight reduction of the infrastructure is demanded because of their high specific modulus and specific strength. Pressure vessels using this composite material in comparison with conventional metal vessels can be applied in the field where lightweight and the high pressure are demanded from the defense and aerospace industry to rocket motor case due to the merits which are energy cutdown the weight reduction and decrease of explosive damage preceding to the sudden explosion which is generated by the pressure leakage condition). In this paper, for nonlinear finite element analysis of E-glass/epoxy filament winding composite pressure vessel receiving an internal pressure, the standard interpretation model is developed by using the ANSYS, general commercial software, which is verified as the accuracy and useful characteristic of the solution based on Auto LISP and ANSYS APDL. Both the preprocessor for doing exclusive analysis of filament winding composite pressure vessel and postprocessor that simplifies result of analysis have been developed to help the design engineers.

The Effects of Digital Transformation Intention of Small Traders in Domestic Traditional Industries (국내 전통산업 소상공인의 디지털 전환의도에 미치는 영향)

  • With the advent of the fourth industrial revolution, the digital transformation craze has intensified in diverse industries, such as finance, automotive, health care and agriculture. However, relative to industry demand, there is only few domestic research on digital transformation of small traders and no one has implemented empirical study on this field at all. Thus, the study conducted an empirical study to measure the digital transformation intention of domestic small traders. In the early days of domestic business start-ups, we searched for factors affecting the digital conversion intention of small business owners and designed an integrated framework to explain the causal relationship between factors. This study not only expanded and refined the scope of related research but also suggested the direction of another research that was not presented in previous research. However, there are limitations in many ways as much as the initial stage of a specific field of small business owners. There are limitations in explaining the overall perception of Korean small business owners. Therefore, in future studies, it is necessary to study more deeply by presenting more specific and various variables. In addition, although the number of respondents was sufficient in the process of empirical analysis, it is considered that the respondents are not enough to universalize the research result as a traditional small business person who did not convert all digital. It is necessary to carry out a comparative analysis by dividing the two groups in more detail and more specifically in the future study. The result of this study can suggest the direction for accelerating small traders' digital transformation intention in terms of practical view, and provide useful references which can support the theoretical foundation for researchers who will perform future related research in terms of academic view.

  • Ultrasound images are noninvasive, can be observed in real time, have no radiation exposure, do not cause pain, and are not restricted in use depending on the patient's prosthetic implant or medical condition. Since the use of ultrasound in the dental field was first applied for tooth preparation in 1957, the use of diagnostic ultrasound for the first time in 1963 has been reported. Currently, it is used in the diagnosis of soft tissue lesions such as malignant tumor or salivary gland disease, fine needle aspiration test, temporomandibular joint disease, lymph node metastasis, measurement of muscle thickness and inflammatory diseases, differentiation of periapical cyst and granulation tissue, measurement of periodontal tissue thickness. The ultrasound image can be visualized in real time. The clinician can explain the structure to the patient while consulting the patient and consult the patient. When injecting the drug into a specific site or aspirating a specific site or substance, So that it can be confirmed and practiced. Recently, ultrasonic equipment specialized in the dental field has been developed and marketed, and it is expected that the use of ultrasonic waves will become active in the dentistry. In the future, development of popular equipment with size and frequency suitable for dental diagnosis and various researches on maxillofacial ultrasonic anatomy. If clinical studies are continuously carried out to demonstrate efficacy, ultrasound is expected to aid in accurate diagnosis and treatment throughout the dentistry.

Feature Extraction by Neural Network for On-line Recognition of Korean Characters (온라인 한글인식을 위한 특징추출 신경망에 관한 연구)

  • This paper describes a feature extraction process by using a multi-layer neural network and is applied to the Korean stroke pattern for on line hand written character recognition, In the first layer the features are detected during the writing process and in the second layer the stroke specific features are extracted. A modified Masking field algorithm for direction co9nstancy has been used in this neural network and the resulting action potential of stroke specific features represents statistical distribution of the features in the on-line input stroke pattern and these results can be used in the recognition of on-line hand written Korean characters successfully.

  • Recently, with the advent of the 4th industrial revolution, digital twins are emerging as an important technology that connects and integrates physical systems and cyber systems. In this article, we analyzed the major requirements of digital twins required for the construction of digital twins of power equipments in the energy field, focusing on the industry 4.0-based Asset Administration Shell(AAS). However, since not so many studies have been conducted yet on a common platform or demonstration model for implementing digital twins both domestically and internationally, digital twin requirements are analyzed with the consideration of digital twinning of power equipment in the energy field. Also, we suggested necessary procedures and specific functions of AAS to establish a smart energy digital twin in the future by analyzing the core requirements necessary for the construction and designing the AAS design for specific power equipment.

  • Prostate cancer ranks as the world's second most frequently diagnosed cancer among men, and is responsible for the fifth highest number of cancer-related deaths in this population. The development of effective diagnostic and therapeutic approaches for prostate cancer remains a major challenge in the field of oncology. Over the past few years, the prostate-specific membrane antigen (PSMA) has raised as a hopeful tracer for the diagnosis and treatment of prostate cancer.Various radioisotopes, such as 131I, 99mTc, 68Ga, and 177Lu, have been used to label PSMA analogues, with varying degrees of success. Among these, 68Ga-PSMA-11 and 177Lu-PSMA-617 have emerged as the most promising radioligands for clinical use. Recently, researchers have been exploring the use of other radioisotopes, such as 211At, 89Zr, 64/67Cu, and 203/212Pb, for the labeling of PSMA-targeted radioligands. These radioisotopes have unique properties that may offer advantages over existing radioligands, such as longer half-lives, higher specific activities, and different emission profiles. Efforts are currently underway to develop these radiopharmaceuticals and make them more widely available for clinical use. These exciting developments highlight the potential of PSMA-targeted radioligands for the diagnosis and treatment of prostate cancer, and provided significant implications for the management of this disease in the future. The current study aims to provide a comprehensive summary of the latest research and clinical applications of radiolabeled PSMA inhibitors for diagnoses and therapy of prostate cancer, emphasizing the exciting developments in the field and their potential impact on clinical practice.

Using US Patent Analysis to Monitor the Technological Trend in the Field of Gastrointestinal Microbiome - Implications on Korean Medicine Research and Development - (미국 특허분석으로 보는 장내 미생물 기술 발전 현황 - 한의학 연구 및 한의약 기술 발전에 주는 시사점 -)

  • Objectives: The purpose of this study was to provide direction for future research in the field of Korean medicine by analyzing microbiome based technologies emerging as a new diagnostic and treatment paradigm. Methods: To achieve the purpose of the study intellectual property data was used. After establishing citation network from registered microbiome-related US patents, citation network was analyzed by knowledge persistence-based main path approach to understanding technological trajectories. Furthermore, community detection algorithms were used to quantitatively identifying specific technological domain in a particular time period. Results: Results shows that early technologies in livestock industry contribute most to the recent patents. Knowledge in the patents flow through the path of food and beverage technological domain, and finally are inherited to the recent development of diagnosis, treatment and prevention technic. Conclusions: This study indicate that developing diagnostic tools which can link the composition of microbiome to specific diseases should be given high priority. Researches should lead to novel therapeutic strategies. Specifically, improving reliability of pattern identification and finding effective therapeutic compositions based on principles of Korean medicine is necessary.
