Reproductive Cycle of the Venus Fish, Aphycypris chinensis (왜몰개, Aphyocypris chinensis의 생식주기)

  • The gonadosomatic index (GSI), hepatosomatic Index (HSI), egg size distribution, gonad development and reproductive characteristics of venus fish, Aphyocypris chinensis, were examined during March 1997- March 1998 in agricultural waterways of the Sedo-myun, Puyo county, Chungnam Province, Korea. Annual reproductive cycle was classified into the following five successive phases by monthly changes in GSI and the characteristics of ovary: quiescent phase (January- February), secondary growing and mature phase (March-May), ripe and spawning phase (June-July), degenerating and resting phase (August-September) and primary growing phase (October-December). The hepatosomatic index (HSI) showed clear seasonal pattern with two separate peaks. However, it exhibited a negative correlation to changes of GSI values. The regression analysis suggested that fecundity showed a strong positive linear relationship ($r^2\;=\;0.91$, n = 34) with body weight. The sex ratio of female to male was 1.4 : 1 in the natural population during the study. The minimum reproductive size of female and male venus fish was 38 mm and 33 mm in fork length, respectively.

  • We investigated the reproductive cycles of two freshwater fishes, Moroco oxycephalus and M. lagowskii, in Korea. Seasonal changes in gonadosomatic index (GSI) and gonads were investigated histologically from April 1998 to April 1999. The reproductive cycles of two species were not shown any differences. The reproductive cycle can be divided into 5 phases : phase I (spent phase), phase II (immature phase), phase III (early developing phase), phase IV (late developing phase), and phase V (ripe phase). In phase I, the gonads of two species began to lose distinctly their weights from mid April, and reached the lowest GSI in late July (phase II). In September, the GSI values of testis and ovary increased very slowly (phase III) and gonadal developments rested during the winter season (phase IV). In March, the GSI values of M. oxycephalus and M. lagowskii began to increase, and reached the maximum in April (phase V). From the cyclic changes in the GSI and histological analyses, the spawning period was between mid April and mid May.

Naturally Collection and Development until Yolk Absorption of Domestic Walleye Pollock Theragra chalcogramma Fertilized Eggs and Larvae (국내 명태 Theragra chalcogramma 자연채란과 난황흡수까지의 난 발생)

  • We collected and reared Theragra chalcogramma walleye pollock brood-stock for use in natural spawning tests and undertook to obtain domestic pollock via fertilized egg capture, development of fertilized eggs, and absorption of yolk sac after hatching. Whole pollock were caught with trammel and set nets and immediately placed in a deep-sea water tank. Adults were the most common pollock age group (43.0%; n = 86) among the 254 pollock captured in March 2014 with 57.9% (n = 147) being captured off Southern Gosung, Korea. The main spawning period of pollock is February (spawning phase of 91% of pollock). From the deep-sea tank, we collected 1640 mL of naturally fertilized eggs (~820,000 eggs) from 12 spawning events occurring between February 4 and 22 2015. The floating/ live eggs were maintained in deep-sea water tanks at $5.5{\pm}0.2^{\circ}C$. Egg size was $1.5{\pm}0.03mm$. Six hours after fertilization the eggs were at the 2 cell stage, and the eggs hatched approximately 340 hours after collection. At hatching, larval length and yolk sac area were $5.2{\pm}0.25mm$ and $9.5{\pm}1.00mm^2$ (100%), respectively. Four days after hatching, the yolk sac area was $2.2{\pm}0.53mm^2$ ($23.1{\pm}5.55%$). This is the first report of collection of naturally fertilized eggs from pollock and their subsequent hatching while held in an indoor deep-sea water tank. The results suggest that such collection could assist in the recovery of pollock resources and the possibility of domestic rearing of cultivated larvae.

Ultrastructures of Oocyte Development and Electrophoretic Patterns of the Yolk Protein Following HCG Treatment in Korean Native Catfish (Silurus asotus)

  • During the rapid phase of gonadal development of the freshwater teleost, the catfish (Silurus asotus), the influence of hCG upon the inducement of final oocyte maturation and spawning was investigated electrophoretically and ultrastructurally. The electrophoretic patterns obtained were different in the presence and absence of some of the major or minor zones, because of the hormone level in catfish. The vitellogenin of hormone-treated fish was stained more intensively than that of sham-treated fish. These proteins showed some minor or main bands of egg extracts which migrated at positions corresponding to molecular weights of approximately 90,000. However, the thickness of electrophoretic band in molecular weight for hCG-treated fish was slightly lower than that for saline control. It seemed the plasma protein with molecular weight of approximately 45,000 in hCG-treated fish disappeared. In contrast to the control fish, the ovaries in the catfish treated with hCG shows a marked ultrastructural change under the electron microscope. No dilated profiles were seen in the granulosa cells of the mature oocyte before ovulation. After germinal vesicle breakdown (GVBD), the zona radiata interna (ZRI) becomes more compact, and there is a loss of all the processes from the pore canals. There is a wide space between the vitelline membrane and zona radiata. Also, during final maturation, the microvillar processes from the oocyte are seen no longer to penetrate deeply into the extracellular spaces of the overlying granulosa cells, and the reticulate patterns of the zona radiata interna becomes occluded, giving the zona radiata a more solid appearance. It has been possible to initiate 100% oocyte maturation in yolk granules and follicles in vivo by treatment with hCG and a high water temperature ($27^{\circ}C$). In hCG-treated fish, the percentages of successful artificial fertilization and hatching were maximal at 15 h after a single injection. It seems clear that a long acting preparation containing hCG can be successfully used in prespawning fish to advance the final events of gonadal maturation and initiate spawning. Further studies are necessary to evaluate the potential of hCG to either stimulate or inhibit the reproductive development of fish at other stages of the seasonal reproductive cycle.

  • To clarify the annual reproductive cycle of the banded catfish, Pseudobagrus fulvidraco (Richardson), the seasonal changes in histological aspect of gonad and liver were examined. The adult fish was raptured from the upper stream of Young-San river, Chunnam in each month from May 1992 to June 1993. Based on the annual changes in GSI (gonadosomatic index), HSI (hepatosomatic index), CF (condition factor) and histological aspects of the gonads, the annual reproductive cycle were classified into 5 periods as follows: 1) Growing phase (from April to early May): The value of GSI increased and the size of oocytes in perinucleolus stage in oocytes increased gradually. Spermatogonia were developed actively from the epithelial tissues of seminiferous tubules. 2) Maturing phase (from Hay to early June): GSI levels increased rapidly in both sex. Oocytes at various developmental stages were observed. Appearance of active spermatogenesis were observed. 3) Mature and spawning phase (from June to August): High values of GSI remained static and oocytes accumulated significant quantitis of yolk globules. 4) Degenerating phase (from September to November): GSI levels decreased and ovaries were filled mostly with oocytes at the perinucleolus stage. Hepatic cells accumulated significant amounts of lipid droplets. 5) Resting period (from December to March) : Low values of GSI were kept and the size of oocytes at the perinucleolus stage did not increase. Spermatogenesis was not observed.

GROWTH OF SOUTH AND WEST COAST PACIFIC OYSTER SPATS(CRASSOSTREA GIGAS) AFTER CROSS-TRANSPLANTATION (남해산 및 서해산 참굴(Crassostrea gigas)종패의 상호 이식 후의 성장)

  • As an attempt to determine if the morphological differences between the southern and western oysters are due simply to the local ecological factors or are based on their fundamental genetic nature, oyster seeds produced in 1968 at Tong-Young, Ye-Chun, and Ko-Hung on the south coast and at Kan-Wol-Do on the west coast were cross-transplanted during May of 1969 to compare their growth. The spats were placed in plastic baskets which permitted free water flow through and the baskets hung from a wooden rack located at a tidal zone of less than I hour exposure at a depth chosen to keep the baskets submerged in water at all times. Twice a month the growth of the spats were measured along with the air and water temperature and salinity. The early summer spats, which were $17-240\%$ larger, in size, than the late summer spats at the time of cross-transplantation, grew more slowly than the late summer spats when exposed to identical environmental conditions, shortening the initial gap to a $5-20\%$ level as the first year of the growth phase came to an end in December. The growth of the Kan-Wol-Do spats lagged considerably behind the southern spats at all localities tested, whereas there were no significant differences among the latter groups. This suggests that the morphological differences between southern and western Pacific oysters in Korea are a manifestation of genetic variety and that Pacific oysters cultured along the south coast are of an identical variety as they are commonly believed to be. The seasonal changes in temperature and salinity even during rainy season in both the southern and western coastal areas are well within the range suitable for successful spawning, and spat fall. However, since the results were based on twice-a-month measurements with no data covering the critical period before and after spawning, they can only serve to indicate at best the general pattern of changes in the environmental conditions of each growing area.

  • Outlines for plasma $estradiol-17\beta$, components, electrophoretic patterns, and ultrastructural changes were obtained in female rainbow trout (Oncorhynchus mykiss) during the seasonal reproductive cycles. Plasma $estradiol-17\beta$ under the natural conditions, exhibited distinct seasonal variation, peaking very late in vitellogenic season during September, decreasing gradually the halt of spawning in December, and ultimately falling during the early stages of seasonal ovarian recrudescence in February and March. This change in $estradiol-17\beta$ appeared to stimulate vitellogenin production as evidenced by increases in plasma calcium, phosphorus, glucose, albumin and total protein levels. The electrophoretic patterns of late maturing or spawning oocytes were stained more intensively than those of late perinucleolus oocytes (molecular weights of approximately 70,000 and 200,000). Two protein bands were found in the SDS-PAGE separation, coincident with the $estradiol-17\beta$ hormone peak. Gonadosomatic indices (GSI) significantly increased from October to January, and showed the highest peak in January, coinciding with the numerically abrupt increase of ripe ova in female. A positive correlation (r=0.701, p<0.01) was established between plasma $estradiol-17\beta$ levels and the gonadosomatic index during the prespawning. The highest level of hepatosomatic index (HSI) observed in December. During the breeding season (December), the gonadotropes were large and filled with GTH-containing inclusions such as granules and globules. The vitellogenic phase began as late perinurleolus oocytes became transformed into early maturing oocytes through the accumulation of yolk, and oocytes reached the late maturing stages as the ooplasm was completely packed with yolk. Marked ultrastructural changed in the granulosa cells during nuclear migration involve the dilation of the rough endoplasmic reticulum and the appearance of the rod-shaped mitochondria with tubular cristae. Microvilli (finger-like projections), from the zona radiata and from the oocyte grew, and made contact with each other in the pore canals of the zona radials during vitellogenesis, but were withdrawn as the zona radiata became more compact and devoid of pore canals during oocyte maturation. The zona radiata grew to a tripartite structure such as an outer thin homogeneous layer, and two inner thick helicoidal layers (zona radials interna and zona radiata externa). Under the normal conditions, the ovarian follicle influenced the histological development and periodical secretion of the hormones , sufficient for a oogenesis and gonadal steroid production.

Growth and Production of Mactra veneriformis (Bivalvia) on the Songdo Tidal Flat, West Coast of Korea (서해 송도 갯벌에서의 동죽(Mactra veneriformis: Bivalvia)의 성장과 생산)

  • The present study was performed to determine th growth and production of Mactra veneriformis on the Songdo tidal flat off Inchon. west coast of Korea. Monthly samples were collected from Marc 1989 to September 1990. The size distributions of shell length showed unimodal patterns from March to August 1989. The binodal pattern after September 1989 indicated a recruitment of young clams during this period. The density fluctuated irregularly, but showed a decreasing pattern over time on the whole. The density of the year class 1989 increased markedly after February 1990. The growth in shell length showed three phases during a year, a fast growth phase in spring, a slow growth phase in summer and autumn, and a lag phase in winter. The annual increment in shell length reduced with age. The flesh dry weight showed a peak in late spring just before the spawning period, and another peak in autumn. The length of annuli and the back-calculated flesh dry weight were used to describe the annual growth. The growth in shell length fitted to the von Bertalanffy model and the flesh dry weight to the Gompertz model. The absolute growth rate of the shell length reduced gradually with age. The year class 1985 had the highest L, and the yeat class 1986 the lowest L. The annual growth in weight showed a typical sigmoid curve. The growth rate was maximum at the age of 2 to 3. Year classes of 1984 and 1985 had higher values of W and higher absolute growth rates than those of 1986 and 1987. Maximum absolute growth rates of 1986 and 1987 year classes were recorded at younger ages than 1984 and 1985 year classes. The annual relative growth rates of length and weight decreased exponentially with age. Biomass in flesh dry weight increased till spring and thereafter decreased progressively. Maximum biomass was 134.6g m/SUP -2/ in August 1990. The biomass of 1989 yeat class occupied highest proportion in total biomass. Annual production in flesh dry weight from March 1989 to March 1990 was 67.9 g m/SUP -2/ year/SUP -1/. Cumulative production of 1989 year class was highest among all year classes, and that of year class 1984 was lowest. The production is high in spring and autumn, and very low in summer and winter.

Captive breeding of endangered betta fish, Betta rubra, under laboratory conditions

  • Betta rubra, classified as endangered fish species by the International Union for Conservation of Nature (IUCN), has been successfully bred and raised in captivity for two generations under laboratory conditions. This study aimed to provide comprehensive information on the captive breeding of B. rubra, focusing on various parameters crucial for ex-situ conservation and domestication. The research involved breeding trials, embryo and larvae observation, first feeding experiments, larva and fry rearing trials, and the evaluation of growth and reproduction in two generations. The study revealed that the female B. rubra, with an average total length of 5.17 ± 0.15 cm and weight of 1.61 ± 0.06 g, produced an average of 73.67 ± 7.09 eggs, 34.33 ± 5.13 total larvae, and exhibited a hatching rate of 46.67 ± 5.77%. The embryogenesis process commenced on the day of spawning (dps) and continued until the eggs hatched at 6 dps. Larvae development and yolk absorption occurred from 0 to 6 days post-hatching (dph). The study also examined the impact of different initial feeding options, with chopped Tubifex resulting in the most significant in- crease (p < 0.05) in length. The growth pattern of B. rubra larvae showed slow initial growth during the first seven days, followed by a rapid exponential growth phase from day 8 to day 39. Two generations of B. rubra (G1 and G2) were successfully bred in captivity, with G2 showing a better tendency for growth in length and weight compared to G1. Notably, there were no significant differences (p > 0.05) in reproductive success between the wild-origin broodstock (G0), G1, or G2. This research contributes valuable insights into the captive breeding of B. rubra and its early life stages, offering critical information for the conservation and sustainable management of this endangered species. Further research is needed to explore the long-term effects of domestication on behavior, physiology, and phenotypic diversity.

  • Changes of the activities of the hepatic cells of female mud skipper, Boleophthalmus pectinirostris were investigated under transmission electron microscopy. Monthly changes of gonadosomatic index(GSI) and hepatosomatic index(HSI), variations of protein and nucleic acid contents(total RNA and DNA) of the liver tissues with the gonadal development phase were also studied. GSI began to increase from May(the growing stage), reaching the maximum value in late June(the mature stage), and then it began to decrease from late July(the degenerative stage), reaching the lowest value in late September. Monthly variations of HSI were negatively related to GSI. HSI decreased in the summer season when the ovary was getting mature and reached the maximum in mid October when the ovary was degenerating. In June(the mature stage), the female hepatic cells of the liver tissues became large and nuclei were hypertrophic. The amounts of glycogen particles and lipid droplets in the cells gradually decreased, while a number of granular endoplasmic reticulum increased. It was assumed that well-developed granular endoplasmic reticulum binding ribosomes are supposed to play the leading role in protein synthesis and deposition for vitellogenin in the cytoplasm. In July(the spawning period), glycogen particles and lipid droplets gradually increased, and then these substances were still observed in large quantity in August(the degenerative stage). The protein contents of the liver tissues with the gonadal phases of the ovaries were shown the maximum value($4.720{\pm}0.103\;mg/g$) in June, and afterwards gradually decreased being the minimum($3.640{\pm}0.130\;mg/g$) in July, and then gradually increased in August. The mean total RNA contents per gram of the liver tissues appeared the maximum($0.523{\pm}0.040\;mg/g$) in June, and afterwards gradually decreased to the minimum($0.158{\pm}0.006\;mg/g$) in July and slightly increased in August again. From these results, it could be assumed that protein contents were closely related to RNA contents. The mean total DNA contents per weight (gr) of the liver tissues appeared to be similar although there were some monthly fluctuations. The ratio of the mean total RNA/DNA were 0.745 in June, 0.262 in July, 0.341 in August respectively.
