Assessment of the Effect of Substrates from Different Wood Species on the Cultivation of Edible Mushroom

  • Mushroom is very important to rural dwellers due to the various economic, health and nutritional benefits that can be derived from its consumption, sales and utilization. Hence, this experiment was carried out with a view to evaluating the influence of locally available substrate i.e., sawdust of different wood species on the cultivation of Pleurotus ostreatus (oyster mushroom). Pleurotus ostreatus was grown on sawdust (indigenous and exotic species) using two pretreatment methods, which included hot water treatment to remove (a water-soluble extract) and non-extracted sawdusts (substrate). The result of this study showed that there was a significant difference in the effect of different wood species selected. Parkia biglobosa produced a better yield (71.06 g), followed by Anogeissus leiocarpus (53.17 g) and Gmelina arborea (51.39 g) in a non-treated unextracted states. In the treated samples, Parkia biglobosa has the highest yield (50.61 g) followed by Anogeissus leiocarpus (49.28 g) and Gmelina arborea (45.00 g). Thus, Parkia biglobosa and Anogeissus leiocarpus produced a higher yield of mushroom either in a treated or non-treated state and they could be recommended for growing oyster mushroom (Pleurotus ostreatus).

전복느타리버섯(Pleurotus cystidiosus O.K.Miller)의 인공재배에 관한 연구 (Studies on the Artificial Cultivation of Pleurotus cystidiosus O.K.Miller, the Abalone Mushroom)

  • 이 시험은 전복느타리버섯의 재배적 특성을 구명하므로써 우리나라 환경에 적합한 인공재배 기술을 확립하기 위한 기초적인 자료를 얻고자 일련의 시험을 수행하였다. 그 결과, 공시균주 중 ASI2079가 수량 및 품질면에서 가장 우수하였다. 공시균주들의 재배시험 결과, 톱밥 병재배시 미송톱밥에 첨가제로 미강을 사용했을 때 미강 20%에서 수량이 좋았으며 초발이 소요일수도 빠른 편이었다. 또한 병재배용 용기는 850 ml 병재배가 수량이나 회수율이 좋았으며 균긁기를 하지않은 것이 초발이 소요일수도 빨랐고 수량도 좋았다. 또한 P.P. 봉지를 이용한 전복느타리버섯의 재배 시험 결과, 배지재료는 절단볏짚을 첨가하지 않은 폐면 단용구가 수량이 높았으며, 전복느타리버섯의 재배시기는 여름재배가 가장 적정하다고 생각된다. 자동화와 원활한 유통을 목적으로 할 때는 병재배를 하는것이 좋고 일반 느타리 재배사에서 재배시에는 봉지재배가 유리하다고 생각된다. 따라서 본 전복느타리버섯 재배 시험결과는 고온기 버섯 재배가 가능하여 버섯 연중재배에 기여할 것으로 본다.

  • 잎새버섯을 병재배 새로운 소득 품목으로 개발하고자 적정 발이방법을 구명하기 위한 연구결과는 다음과 같다. 발이는 모든 처리구에서 6일째 개시가 되었으나, T7(균긁기후 역상)은 9일로 다른 처리보다 3일 늦었다. 발이율은 T3(페트리디쉬)과 T7이 98.9%로 가장 높고, 전체 재배기간은 T7이 51일로 다른 처리구의 46~47일보다 3일 늦은 것으로 나타났다. 수량성은 T3과 T7이 각각 110g과 112g으로 우수하였으나, T7의 방법이 자실체의 발생도 균일하고, 품질도 우수하여(장단비 0.89) 잎새버섯 병재배에 적합한 발이방법으로 판단되었다.

꽃송이버섯 액체종균배지 및 배양조건에 따른 균사 배양 특성 (Mycelial growth characteristics of Sparassis latifolia according to liquid media and incubation conditions)

  • 꽃송이버섯 안정생산을 위해 액체종균배지조성 및 배양조건에 관한 연구결과, 물엿을 8Brix%로 조절하고 질소원으로 Yeast extract를 0.2%첨가한 배지에서 균사생장량이 우수하여 적합 배지로 선발하였다. 통기량에 따른 $CO_2$ 발생량은 배양 9일까지 증가하다 감소하였고, 통기량이 높을수록 균체직경은 작아지고 배지감소율은 증가하여 통기량 0.9vvm이상에서 배지감소율이 급증하여 꽃송이버섯 액체종균에 적합한 통기량은 0.3~0.6 vvm 이 적합하였다. 또한, 배양기간일 길수록 균체량이 많았으나, $CO_2$ 발생량이 높고 배지감소율이 낮은 9~11일을 적합 배양기간으로 설정하였다.

목이버섯 봉지재배 배지 개발 (Development of bag culture medium of Auricularia auricula)

  • 목이버섯 봉지재배의 배지 선발은 톱밥85%를 참나무 80%+포플러20%의 중량비로 혼합하였고 이 때 필요한 부재료는 면실박10%와 밀기울5%를 혼합한 배지였다. 개발한 배지자실체 수량검정을 한 결과 버섯생육기간은 대조에 비해 33일 단축되고 수량도 25%증수되었다.

  • 1. 사철느타리버섯(Pleurotus florida)는 퇴적시(堆積時)에 미강(米糠)을 볏짚건물중(建物重)의 5%로 첨가(添加)하여 퇴비(堆肥)더미 정상부(頂上部)의 온도(溫度)가 $60^{\circ}C$일 때 3회퇴적(回堆積)한 발효(醱酵)볏짚부지(部地)에서 가장 높은 자실체(子實體) 수확(收穫)을 얻을 수 있었다. 2. 사철느타리버섯의 종균재식(種菌栽植)재직량(栽直量)을 볏짚건물중(建物重)의 10%로, 부지(部地)와 혼합재식(混合栽植)하였을 때 자실체(子實體) 수량(收量)이 가장 높았다. 3. 한국(韓國)에서 사철느타리버섯의 재배기간(栽培期間)은 9월(月)부터 익년(翌年) 7월중순(月中旬)까지였다.

  • 영지버섯의 토양매몰재배에 대한 연작피해균인 노랑얼룩무늬병원 균의 피해를 경감하기 위한 영지버섯 톱밥재배의 시험결과는 다음과 같다. 영지버섯톱밥재배를 위한 최적 배지 및 혼합비율은 참나무톱밥75%+면실박10%+비트펄프15%를 혼합한 배지에서 균사생육이 양호하였다. 영지버섯의 자실체 발생을 위한 완경조건은 생육온도 $28{\sim}30^{\circ}C$, 광조건 1,000~1,500Lux, 이산화탄소는 1%이하 조건에서 자실체 발생 및 상품성이 우수 하였다.

느타리버섯 균상재배 중 배지내 미생물상의 변화 및 분포양상 (Changes and Distributional Pattern of Microflora in Cotton Waste Media of Oyster Mushroom Cultivation)

  • The diversity of microflora according to growth stage of Pleurotus ostreatus and the correlation between microbe and medium fermentation were investigated. In farmhouse I, the aerobic bacteria and fungi as longer of growing period were increased. And, thermophilic bacteria and fluorescent Pseudomonas sp. showed high density at the early stage of spawn inoculation. The thermophilic actinomycetes were distributed evenly during all the growing period, but mesophilic actinomycetes were not observed. In farmhouse II, thermophilic actinomycetes were not observed in fermented medium and density of fungi were suddenly increased at 60 days after spawn inoculation. And also, mushrooms can hardly be harvested due to Penicillium spp. After medium fermentation, density of aerobic bacteria, thermophilic bacteria, and fluorescent Pseudomonas sp. was higher at farmhouse I than those of farmhouse II. In farmhouse I, Bacillus sp. and Pseudomonas sp. dominated at early stage of mushroom growth but as time goes by, density of Bacillus sp. was higher than the others. And also, the kind of microbe showed a few at early stage of mushroom growth but increased as time goes by. In farmhouse II, Bacillus sp. was dominated at early stage of mushroom growth. And the growth of Bacillus sp. and Pseudomonas sp. showed intersect aspect each other in the farmhouse I but Bacillus sp. dominated during all growth periods in the farmhouse II.

Effects of Aeration of Sawdust Cultivation Bags on Hyphal Growth of Lentinula edodes

  • The effects of aeration through lid filters on the hyphal growth of Lentinula edodes (oak mushroom) in sawdust cultivation bags were investigated. The aeration treatment levels were traditional 27 mm hole cotton plugs, cotton balls and combinations of seven hole sizes ${\times}$ two hole positions (up and under) in the lids covering plastic bags containing 1.4 kg sawdust medium at 63% moisture that had been autoclaved for one hour and inoculated with sawdust spawn of L. edodes strain 921. Aeration treatment effects were measured based on the $CO_2$ concentration at the 15th wk, as well as the hyphal growth rate and degree of weight loss of bags every 14 days for 15 wk. In bags with traditional cotton plugs, the $CO_2$ concentration was $3.8{\pm}1.3%$, daily mean hyphal growth was $2.3{\pm}0.6mm$ and daily mean weight loss was $0.84{\pm}0.26g$. In the bags with 15 mm diameter holes, the $CO_2$ concentration was $6.0{\pm}1.6%$, daily hyphal growth was $2.8{\pm}0.2mm$ and daily weight loss was $0.86{\pm}0.4g$. The bags with 15 mm holes had a higher $CO_2$ concentration and lower water loss than bags with other hole sizes, but the hyphal growth was not significantly different from that of other bags. The weight loss of bags increased proportionally relative to the lid hole sizes. Taken together, these results indicate that traditional cotton plugs are economically efficient, but 15 mm hole lids are the most efficient at maintaining hyphal growth and controlling water loss while allowing $CO_2$ emissions.

Characteristics of mycelial growth and fruit body of Sparassis latifolia strains and selection of suitable incubation conditions in liquid spawn

  • Sparassis latifolia is called "Cauliflower Mushroom" and is known as an edible mushroom that has high content of ${\beta}$-glucan. Recently, artificial cultivation of S. latifolia has been done by bottle, plastic bag and wood cultivation in Korea. However it is not widely used because there are low incubation ratio and yield. For the high efficiency of production, we aim to find the superior strains and media for better mycelial and fruit body growth. First, we analyzed the genetic relationship among 31 strains and divided five groups with three kinds of URP primers. And then ten strains were selected from five groups based on the experiment of mycelial growth. The suitability of media for mycelial growth was different according to media type. The suitable solid and liquid media for mycelial growth of S. latifolia isolates were PDA and M2, respectively. In addition, with regard to C/N ratio, the mycelial growth increased even until C/N 160. Second, we investigated the production of fruitbody of the strains by plastic bag cultivation. The substrate was mixed with larch sawdust, corn flour, and wheat flour (8:1:1, v/v). Moisture content of substrate was controlled by about 60% with 10% molasses solution. Out of 31 strains, 19 strains formed primordia. The eight strains produced more than 140g/1kg in fresh weight. Third, molasses culture media was selected for the mycelial growth. And molasses suitable sugar content and input aeration were around 8Brix% and 0.3~0.6vvm, respectively. The longer the incubation period is, the more dried weight of mycelia increased, but medium volume decreased. Therefore, the best incubation period was 9 to 11 days depending on strains. In the future, research project entitled development of culture system and new variety for stable production of S. latifolia will be considered as a new item.
