Effect of Water Extract of Sparasis crispa on the Expression of TNF-α, iNOS and IL-1β Genes in RAW 264.7 Cells (꽃송이버섯 추출물이 RAW 264.7 세포에서 TNF-α, iNOS, IL-1β 유전자 발현에 미치는 영향)

  • Purpose : The purpose of this study was to examine the anti-inflammatory effects of Sparassis crispa (SC). SC is a well-known traditional herbal remedy and its mushroom is used for treatment of inflammation. Many diseases that are increasing recently have characteristics of inflammatory diseases. Researchers are finding bioactive substances from natural products that can promote treatment and prevention of inflammation. We investigated the effect of water extracted from SC on the expression of effector genes involved in the function of RAW 264.7 cells. Methods : Effects of RAW 264.7 cells on cell viability, antioxidation, and mRNA expression were examined using water extracts from SC. A 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay was performed to determine the effect of water extracts from SC on cell viability in RAW 264.7 cells. Inflammation of RAW 264.7 cells induced by lipopolysaccharide (LPS) treatment and expression levels of inflammatory cytokine TNF-α, iNOS and IL-1β gene were analyzed using quantitative reverse transcription PCR (qRT-PCR) analysis. Results : The MTS assay was performed on RAW 264.7 cells after treatment with various concentrations of water extracts of SC. Treatment of RAW 264.7 cells with water extracts from SC and LPS at a concentration of 0.125, 0.5 mg/㎖ for twenty four hours promoted mRNA expression of TNF-α, iNOS and IL-1β. Conclusion : MTS assay was applied to RAW 264.7 cells after various concentrations of water extracts of SC. Through experimental demonstration of anti-oxidant and anti-inflammatory effects of water extracts from SC, we suggest that SC is a valuable material for the prevention and treatment of various inflammatory diseases.

Site Characteristics and Vegetation Structure of the Habitat of Cauliflower Mushroom (Sparassis crispa) (꽃송이버섯 자생지의 입지특성 및 식생구조)

  • From 2006 to 2008, natural habitat of Sparassis crispa were surveyed for investigating ecological conditions at sixteen different sites in Korea. The investigated sites showed very wide altitudinal distribution ranged from 240 meters to 1,100 meters above the sea level. In general, S. crispa showed distinct feature of heart-rot fungi as it occurs on soils around the stems of larch (Larix kaempferi) and Korean white pine (Pinus koraiensis). But it also could be found on stems or on the stumps of dead trees, which indicated that the fungus might have several invasion routes and capabilities to grow on various ecological conditions. All of the sixteen sites were pure stands consisted with conifers such as larch or Korean white pine. The dominant tree layer showed $15.3{\sim}38.0$ meters for tree heights, thicker than twenty centimeters for the diameter at breast height (DBH), and all of them were older than thirty years. Since the stands were pure stand, species diversity of trees in the sites was extremely low. While the dominant tree layer showed only pure coniferous stand, the co-dominant tree layer, shrub layer and herbaceous layer showed more diverse features with higher Shannon-Wiener (H') indices. Soil texture of thirteen sites among sixteen investigated sites were loamy soils, and the contents of organic matter in soil were more or less higher than general forest soils in Korea with $3.79{\sim}14.32%$. The cation exchange capacity (CEC) was also relatively higher than general forest soils with $16.1{\sim}27.2$ cmol+/kg. The data indicated that the cauliflower mushroom occurring sites were relatively fertile than general forest soils. The soils were acidic with pH ranged from 4.2 to 5.2, which were typical features for conifer stands in Korea.

Lipolytic Effect of Sparassis crispa Extracts in Differentiated 3T3-L1 Cells and High Fat Diet-induced Obese Mice (분화된 3T3-L1 세포와 비만유도 쥐에 꽃송이버섯 추출물의 지질분해 효과)

  • The purpose of this study was to investigate whether water extracts of Sparassis crispa (SC) have anti-obesity effects. Treatment of mature adipocytes with SC caused a decrease in lipid accumulation (assessed by Oil Red O staining) and an increase in glycerol release. Mice were induced to obesity by a high fat diet (45% fat in total kcal) and experimental groups were treated with two different dosages of SC extracts, a low SC (LSC, 100 mg/kg/day) or high SC (HSC, 300 mg/kg/day). SC extracts were administered by gavages for 10 weeks in the experimental groups, while the control group was fed with distilled water. The body weight gain of mice fed SC was significantly reduced (11.88% lower in LSC, 14.54% lower in HSC) compared to the control group. Additionally, there were significantly reduced serum levels of triglycerides (13.57% lower in LSC, 19.46% lower in HSC), total cholesterol (32.22% lower in LSC, 24.67% lower in HSC) and glucose (28.85% lower in LSC, 25.82% lower in HSC) in mice fed SC compared to the control group. Hepatic triglycerides in mice fed SC were lower (9.68% lower in LSC, 14.24% lower in HSC) than the control group and total cholesterol levels were also lower in mice fed SC (38.72% lower in LSC, 35.20% in HSC). These results demonstrate that the water extract of SC may enhance lipolysis and up-regulate the expression of lipolytic enzymes in vitro and reduce body weight in vivo. These significant effects were found for both low and high doses of SC treatment, and suggest SC can be used as potential therapeutic substances for the prevention and treatment of obesity.

  • Glucan has been shown to have a significant role in the activation of the immune system, including increased activity of macrophages and so on. Sparassis latifolia (formerly S. crispa) is an edible mushroom abundant in dietary fiber and widely known to contain high levels of ${\beta}$-glucan. In the present study, fermentation broths containing soluble ${\beta}$-glucan were prepared by fermentation with mushrooms with four Lactobacillus species (L. plantarum subsp. Plantarum, L. acidophilus, L. helveticus, and L. delbrueckii subsp. Bulgaricus). After culturing four Lactobacillus spp. in MRS broth, each Lactobacillus was inoculated into MRS broth containing S. latifolia powder 5% (w/v) at $37^{\circ}C$ in an anaerobic incubator for five days. It showed that the ${\beta}$-glucan contents were different in each fermentation sample. The suitable conditions for the preparation of mushroom fermentation broths were investigated and discussed.

Effect of a Hot Water Extract of Sparasis Crispa on the Expression of Tight Junction-Associated Genes in HaCaT Cells (꽃송이버섯 열수추출물이 HaCaT의 세포 연접 관련 유전자의 발현에 대한 영향)

  • Purpose : Keratinocytes are the main cellular components involved in wound healing during re-epithelization and inflammation. Dysfunction of tight junction (TJ) adhesions is a major feature in the pathogenesis of various diseases. The purpose of this study was to identify the various effects of a Sparassis crispa water extract (SC) on HaCaT cells and to investigate whether these effects might be applicable to human skin. Methods : We investigated the effectiveness of SC on cell HaCaT viability using MTS. The antioxidant effect of SC was analyzed by comparing the effectiveness of ABTS to that of the well-known antioxidant resveratrol. Reverse-transcription quantitative polymerase chain reaction (qRT-PCR) is the most widely applied method Quantitative RT-PCR analysis has shown that SC in HaCaT cells affects mRNA expression of tight-junction genes associated with skin moisturization. In addition, Wound healing is one of the most complex processes in the human body. It involves the spatial and temporal synchronization of a variety of cell types with distinct roles in the phases of hemostasis, inflammation, growth, re-epithelialization, and remodeling. wound healing analysis demonstrated altered cell migration in SC-treated HaCaT cells. Results : MTS analysis in HaCaT cells was found to be more cytotoxic in SC at a concentration of 0.5 mg/㎖. Compared to 100 µM resveratrol, 4 mg/㎖ SC exhibited similar or superior antioxidant effects. SC treatment in HaCaT cells reduced levels of claudin 1, claudin 3, claudin 4, claudin 6, claudin 7, claudin 8, ZO-1, ZO-2, JAM-A, occludin, and Tricellulin mRNA expression by about 1.13 times. Wound healing analysis demonstrated altered cell migration in SC-treated HaCaT cells and HaCaT cell migration was also reduced to 73.2 % by SC treatment. Conclusion : SC, which acts as an antioxidant, reduces oxidative stress and prevents aging of the skin. Further research is needed to address the effects of SC on human skin given the observed alteration of mRNA expression of tight-junction genes and the decreased the cell migration of HaCaT cells.

Determination and Characterization of Inhibitory Activity of Mycelial Extracts from Several Korean Mushrooms against Helicobacter pylori (한국산 버섯추출물의 Helicobacter pylori에 대한 항균력 검색과 특성)

  • The effect of hot water and methanol extracts of 9 species from Korean mushrooms (Fomitopsis pinicola, Fomitella traxinea, Codyceps militaris, Phellinus linteus, Coriolus versicolor, Sparassis crispa, Ganoderma lucidum, Fomes fomentarius, Agaricus blazei) on the growth of Helicobacter pylori were examined using a Mueller-Hinton agar diffusion method. Hot water ($121^{\circ}C$) extracts from fruit bodies of F. traxinea, C. militaris, P. linteus, C. versicolor and F. pinicola showed $10{\sim}15\;mm$ inhibition zone against H. pylori. Methanol extracts of F. pinicola showed 44 mm inhibition zone, but another extracts showed no inhibition. Early fractions of DEAE-Sephadex A-25 column chromatography of methanol extracts from F. pinicola showed high inhibition activity against H. pylori.

Safety Evaluation of Kyungokgo-gamibang Administration Based on Hematological, Biochemical, Protein, and Lipid Profiles in Dogs

  • Kyungokgo-gamibang, Kyungokgo with Iksuyongjingo and Sparassis crispa, is a traditional Korean medicine used for restorative effects. This study aimed to evaluate the safety of Kyungokgo-gamibang in healthy beagle dogs. In the single-dose oral toxicity study, three beagle dogs were orally administered 2,000, 1,000, and 500 mg/kg of Kyungokgo-gamibang and were observed for 14 days. In the repeated-dose oral toxicity study, nine healthy dogs were orally administered 0.2g/kg of Kyungokgo-gamibang (n = 3, low-dose group), 1 g/kg of Kyungokgo-gamibang (n = 3, high-dose group), or normal saline (n = 3, control group) twice a day for 8 weeks. The hematological, serum biochemical, urine, protein, and lipid profiles were evaluated to investigate the adverse effects of the Kyungokgo-gamibang. During the study period, the dogs demonstrated no clinical signs and the hematological, serum biochemical, urine, protein, and lipid analyses revealed unremarkable findings. The study results suggest that Kyungokgo-gamibang can be safely administered to dogs without any adverse effects.

Cultivation of Sparassis crispa on Several kinds of Medium Density and Particle Size of Sawdust-based Medium Made of Larix kaempferi (낙엽송 톱밥배지 밀도 및 입자크기에 따른 꽃송이버섯의 재배특성)

  • Cauliflower mushroom ($Sparassis$ $crispa$) is being cultivated as a functional mushroom since the mushroom contains larger amount of ${\beta}$-glucan than other edible or medicinal mushrooms. In this study, as a fundamental study for the cultivation process of cauliflower mushroom, we assayed the mycelial growth and the productivity of the mushroom cultivated on the sawdust-based medium made of larch ($Larix$ $kaempferi$) by 4 kinds of medium densities and 6 kinds of particle sizes. Low densities of sawdust-based media (0.68~0.72 g/$cm^3$) showed high variations in mycelial growth at early stage on sawdust-based medium. The coefficient of variance for the mycelial growth decreased from higher than 40% at the 3rd week to lower than 10% at the 7th week. High density of sawdust-based medium (0.80 g/$cm^3$) showed relatively lower variation in the mycelial growth at early stage with less than 30% of coefficient of variance, the high density of sawdust-based medium was thought to be quite stable compared to the lower densities of sawdust-based medium. From the viewpoint of mushroom productivity especially for the goods (excluding bottom of fruiting body), 0.76 g/$cm^3$ was better than any other densities; the return rates of fruiting body from each medium were 12.2, 13.6, 13.1, and 12.0% for 0.80, 0.76, 0.72, and 0.68 g/$cm^3$, respectively. By the way, it took about 40 days for harvesting from primordium formation with the sawdust-based medium excluded the particles less than 1 mm, while it took about 70 days with the medium including the particles less than 1 mm. The yield from the sawdust-based medium with the particles less than 1 mm was also quite less than any other sawdust-based medium especially for the goods. Therefore, we recommend that the sawdust-based medium with larch for the cultivation of cauliflower mushroom be prepared as 0.76 g/$cm^3$ in medium density with excluding the particles less than 1 mm.

Total sugar contents of edible and medicinal mushrooms comparative analysis by the extraction method (추출방법별 식용 및 약용버섯류의 총 당함량 비교분석)

  • This study was to comparative analysis that different methods of extraction about total sugar content which the major functional components of edible and medicinal mushrooms. Through the phenol-sulfuric acid method was measured that Hot water extraction, acid extraction, and alkaline extraction. Drawing the standard curve to obtain the concentration of the glucose solution and the sample by applying the measured absorbance on the standard curve. Total sugar contents was measured after extraction with different solvents.(Hot water, 70% EtOH, 80% MeOH). The level of total sugar in Cordyceps militaris is highest in the hot water extraction with 658.4(${\mu}g/g$) and Coriolus versicolor in 649.7(${\mu}g/g$) and 679.0(${\mu}g/g$) was found as the highest in the acid extraction and the alkaline extraction. C. militaris and ASI 7025 showed the highest level in the hot water extraction. Finally, C. versicolor, Sparassis crispa showed that higher than the other mushrooms at the 1hr, 2hr microwave extraction. It was varied total sugar content according to the different extraction solvent and the extraction method.

Effect of mycelial culture of cauliflower mushroom (Sparassis crispa) using LED lighting operation (LED 조명처리가 꽃송이버섯의 균사배양에 미치는 효과)

  • As a result of advenced research, Penicillium growth inhibition effect in media of cauliflower mushroom by different LED lighting color inhibited all treated groups, but blue wavelength treatment group was unfitted for culture of cauliflower mushroom due to lots of spore of penicillium. So, to investigated characteristics of mycelial growth of cauliflower mushroom according to different LED wavelength and LED wavelength color. As a results, all red wavelength treatment groups found highest mycelial growth tendency. Thus, mycelial growth investigated different quantity of red lighting wavelength conditions. The quantity of lighting wavelength was low intention, 1.41 ${\mu}mol/m^2S$ treatment group found highest mycelial growth. Effects of mycelial growth by subculture found difference of statistical in one time to carry out a subculture treatment group. Mycelial growth by different quantity of LED lighting in different media composition of wood chip media indicated highest trend in the Korean pine treatment groups. To cultured treatment group for 84th days found difference of statistical, when a quantity of LED lighting red wavelength 2.11 ${\mu}mol/m^2S$ treated in wood chip of the Korean pine media. In conclusion, good culture condition of cauliflower mushroom estimated quantity of red lighting wavelength 2.11 ${\mu}mol/m^2S$ in wood chip media of the Korean pine for 84th days.
