Yield and Quality of Black Soybean (Glycine max L.) in Paddy Field under Different Sowing Dates (검정콩 논재배에서 파종시기가 수량 및 종실 특성에 미치는 영향)

  • Recently, Black soybean (Glycine max L. Merr.) is being cultivated in paddy fields instead of rice. However, research related to the effective sowing date is insufficient in paddy fields. This study aimed to identify the sowing date for stable cultivation of black soybean by investigating its yield and seed characteristics in a paddy field. In the study, cultivation experiment with five different sowing dates (May 25, June 10, June 25, July 10, July 25) were conducted in 2019 and 2020. Days from sowing to flowering can be shortened by delaying the sowing date. In the present study, the yield of black soybean in paddy fields was the highest with June 10 as the sowing date and was calculated as 224 kg·10a-1 and 200 kg·10a-1 in 2019 and 2020, respectively. However, the highest values of seed coat cracking was 51.1±5.1% and that of total anthocyanin contents was the 3.99±0.72 mg/g, both of which were observed in 2020 for the experiment with May 25 as the sowing date. Regression analysis showed a positive correlation (R2=0.9312) between soil water contents and seed coat cracking rate during the flowering period. Hence, the soil water contents during the flowering period would have a negative effect on the seed coat development.

Absorption and Accumulation of Sr-90 by Rice and Soybean and Its Soil-to-Plant Transfer Coefficients (벼와 콩에 의한 Sr-90 흡수.축적 및 토양-작물체간 전이계수)

  • Radio-tracer experiments on the Sr-90 absorption by rice and soybean from a sandy-loam soil of pH 6.35 treated with 5.2 and 31.2Bq Sr-90 per g-soil were carried out through pot cultivations. Sr-90 absorption rates of both crops increased till the mature stage when the rates were about 1.0%. Concentrations in their whole tops, however, decreased or changed little as they grew. Sr-90 concentrations in plant parts of both crops increased with the increase of those in soil. Soil-to-plant transfer coefficients of Sr-90 for rice and soybean at mature stages ranged, on the dry weight basis, from 0.07(unpolished seed) to 3.67(lean and from 0.86(seed) to 9.26(leaf), respectively. Only the unpolished rice seed showed a significant difference in the coefficient with 0.17 in 5.2Bq treatment and 0.07 in 31.2Bq treatment. Sr-90 retention rates of the upper 15cm soil after crop harvests were about 80% Sr-90 absorptions had no effect on the plant growth and yield of the crops.

Effect of Soil Mulching after Green Manual Crops on Control of Common Scab and Yield Characteristics of Fall Potato (녹비작물재배후 플라스틱필름 멀칭에 따른 가을감자의 더뎅이병 방제 및 수량특성 변화)

  • This experiment was conducted to investigate the changes of rate of infected common scab and yield characteristics of fall potato(Solanum tuberosum L.) where green manual crops had been previously cultivated, crushed and tilled and P\ulcornerE transparent vinyl film had been mulched and tunneled to solar heating of soil from May 21. 1998 to July 28. 1998. The total yields of green manual crops which had been previousely cultivated were as followed order : pioneer 855F(64.3MT/ha), soybean(25.0MT/ha), red clover and orchardgrass. The average below-ground temperature at the depths of 5, 10 and 20cm were 54, 45 and 44$^{\circ}C$ during the mulching period, respectively The rate of infected area per potato tuber of common scab decreased by solar heating the soils with mulching after the soybean and red clover cultivation as with previous croppings. Plant height, SPAD(soil plant analysis development) reading, fresh weight of stems, and fall potato's tuber yields tended to increase by the cultivation of green manual crops and mulching of P\ulcornerE film. Fall potato's tuber yields were remarkably affected in the plot of soybean, red clover and pioneer 855F+cultivation of fall potato. T-N, K and Ca contents of fall potato(stem and tuber) also tended to increase by the cultivation of green manual crops and mulching of P\ulcornerE film. T-N, K and Ca contents of soil tended to increase after the cultivation of green manual crops and mulching of P\ulcornerE film and then reduced to contents of soil before green manual planting after potato harvesting.

Inhibition Effect on Root Rot Disease of Panax ginseng by Crop Cultivation in Soil Occurring Replant Failure (윤작물 재배에 의한 인삼 뿌리썩음병 발생 억제 효과)

  • To study the effect of crop rotation on the control of ginseng root rot, growth characteristics and root rot ratio of 2-year-old ginseng was investigated after the crops of 18 species were cultured for one year in soil contaminated by the pathogen of root rot. Fusarium solani and Cylindrocarpon destructans were detected by 53.2% and 37.7%, respectively, from infected root of 4-year-old ginseng cultivated in soil occurring the injury by continuous cropping. Content of $NO_3$, Na, and $P_2O_5$ were distinctly changed, while content of pH, Ca, and Mg were slightly changed when whole plant of crops cultured for one year were buried in the ground. All of EC, $NO_3$, $P_2O_5$, and K were distinctly increased in soil cultured sudangrass, peanut, soybean, sunnhemp, and pepper. All of EC, $NO_3$, $P_2O_5$, and K among inorganic component showed negative effect on the growth of ginseng when they were excessively applied on soil. The growth of ginseng was promoted in soil cultivated perilla, sweet potato, sudangrass, and welsh onion, while suppressed in Hwanggi (Astragalus mongholicus), Deodeok (Codonopsis lanceolata) Doraji (Platycodon grandiflorum), Gamcho (Glycyrrhiza uralensis), Soybean. All of chicory, lettuce, radish, sunnhemp, and welsh onion had effective on the inhibition of ginseng root rot, while legume such as soybean, Hwanggi, Gamcho, peanut promoted the incidence of root rot. Though there were no significant correlation, $NO_3$ showed positive correlation, and Na showed negative correlation with the incidence of root rot.

  • A series of experiments were conducted on procedures of $N_2$ fixation measurements with soybean (Glycine max. L.) root nodules by acetylene reducing assay and estimation of amounts of $N_2$ fixed by soybean root nodules grown in the pot. The results are summarized as follows; 1. In case of nitrogen fixing activities were determined by means of acetylene reducing method, more appropriates results were obtained with direct determination of root nodules which was adhering from root under 2 hours after incubation, while more fluctuated data was obtained with root nodules determined under separated conditions. 2. The weight of soybean nodules was linearly increased from 17 days to 55 days after planting, whereas, the amounts of $N_2$ fixation from soybean nodules were Peaked at 38 days after planting, and declined thereafter. 3. When calculated upon the basis of the amounts of $N_2$ fixed as measured by acetylene reducing activities in single soybean plant, it was found that the average amount of symbiotic $N_2$ fixation falls in the range of 80 kg N per ha.

Effect of the Density of Heterodera glycines on Soybean Yield (콩씨스트선충(Heterodera glycines)의 밀도가 콩 수량에 미치는 영향)

  • This study was conducted to investigate the yield loss of soybean by initial density of Heterodera glycines in soil. A soybean cultivar named 'Cheongja5ho' was cultivated as test material in Dangjin city, Chungcheongnam-do, Korea. Each seed was planted with distance of 25 cm in 35 plots by initial density of egg of the cyst nematode on June 21st, and allowed to harvest on November 12st in 2019. As a results, the yield of soybean decreased sharply when as the initial density of eggs of cyst nematode increased. The initial density of eggs in soil affected largely on the decreased yields of the soybean by more than 20% in above 2,000 eggs, and 30% in above 6,000 eggs of the cyst. Beyond the 6,000 eggs, the yield loss was more slowly decreased compared with the above cases. Eggs of H. glycines are widely distributed in Korea, resulting in the serious soybean yield loss by H. glycines.

Selection of Tolerant Plant Species using Pot Culture for Remediation of Explosive Compounds Contaminated Soil (포트 재배에 의한 화약물질 오염토양 정화용 내오염성 식물 선정)

  • Nine plant species were selected through vegetation survey at three military shooting ranges at northern Gyeonggi Province. Plants were germinated in normal soil and three seedlings were transplanted to a bottom sealed pot containing sandy loam soils contaminated with either RDX (291 mg/kg) or TNT (207 mg/kg). Planted, blank (without plant), and control (without explosive compound) pots were grown in triplicate at a green house for 134 days. During cultivation, transplanted plants exhibited chlorosis and necrosis in flower and leaf by explosive toxicity and stress. Only three plants, Wild soybean, Amur silver grass, Reed canary grass, survived in TNT treated pot, while seven plant species except for field penny cress and jimson weed, thrived in RDX treated pot. Appreciable amount of TNT (61.6~241.2 mg/g-D.W.) was detected only in plant roots. Up to 763.3 mg/g-D.W. along with 4-amino-2,6-dinitrotoluene, an intermediate of TNT, accumulated in the root of wild soybean. In addition, azoxy compounds, abiotic intermediates of TNT, were detected in TNT treated soils. RDX absorbed average 1,839.95 mg/kg in shoot and 204.83 mg/kg in root. Most of TNT in plant was accumulated in underground part whereas RDX was localized in aerial part. Material balance calculation showed that more than 95% of the initial TNT was removed in the planted pots whereas only 60% was removed in the blank pot. The amount of RDX removed from soil was in the order of Amur Silver Grass (51%) > Chickweed (43%) > Evening primrose (38%). Based on the results of pot cultures, Amur silver grass and Reed canary grass are selected as tolerant remedial plants for explosive toxicity.

  • These studies was conducted to investigate the influence of ALLELOPATHIC effects of upland weeds on crops and soil microorganisms. The aqueous extract of weed residues were used to identify physiological controll of plant and microorganisms. The results obtained are as follows : 1. The aqueous extract of weeds controlled germination and growth of plant, especially the 5%-extract of Potulaca oleracea suppressed the growth of soybean about 8~12%. 2. The 5%-extract of Potulaca oleracea inhibited effect of phytosynthesis of soybean about 60%. 3. The move the concetration of phenolic compounds in soil, the less a number of actinomycetes and bacteria. 4. The number of soil microorganism(fusarium oxysporum spp.) were decresed as to the concentration of vanillic acid was increased.

Biosurfactant 생산 효모 Rhodotorula sp. G-1의 분리 및 Biosurfactant 생산

  • Some microorganisms including yeasts produce surface tension-decreasing biosurfactants. The strain G-1, the best producer of biosurfatants was isolated from the soil and identified as Rhodotorula sp., which was not discribed any report. The Rhodotorula sp. G-1 produced biosurfactant from vegetable oils, but failed to produce it from n-alkane or carbohydrate. Yeast extract was found to be more effective for the biosurfactant production as nitrogen source than any other inorganic nitrogen source. The composion of the optimal medium contained the following conponents: soybean oil 4%, glucose 2%, yeast extract 0.5%, KH$^{2}$PO$^{4}$ 0.1%, K$^{2}$HP0$^{4}$ 0.l%, MgSO$^{4}$ 5%, CaCl$^{2}$ 0.01%, NaCl 0.01%, pH 6.0. The surface tension activity was increased to 14% when, at first, the culture broth was fermented with only soybean oil as carbon sourse, and after 90 hours, feeded glucose, than that Of glucose and soybean oil added to it simultaneously. The maxium yield of the biosurfactant was about 15 g/l by after 90 hours, the feeding method of glucose.

Characterization of a Thermostable Protease from Thermophilic Bacillus amyloliquefaciens NS 15-4 (고온성 Bacillus amyloliquefaciens NS 15-4가 생산하는 내열성 Protease의 특성)

  • A thermophilic bacteria showing proteolytic activity against defatted soybean was isolated from soil. It was identified as Bacillus amyloliquefaciens based on its morphological and physiological characteristics. The Bacillus amyloliquefaciens NS 15-4 was cultivated at 50$\circ$C by rotary shaking in a medium containing defatted soybean. An extracellular protease from this strain was purified to homogeneity by ammonium sulfate precipitation, ion exchange, and hydrophobic interaction chromatographies. The molecular weight of the enzyme was estimated to be approximately 30,000 by SDS-PAGE and the N-terminal amino acid sequence of the enzyme was turned out to be AQSVPYGISQIKAPA. The optimum temperature and pH for the enzyme reaction were 60$\circ$C and 11, respectively, and its thermostability was increased by the addition of calcium ion. The enzyme was inactivated by phenylmethylsulfonylfluoride, suggesting it be a serine protease. Comparing with other commercial proteases, the enzyme showed relatively high proteolytic activity against defatted soybean, a water-insoluble protein substrate.
