Waterlogging Effects on Nitrogen Accumulation and $N_2$ Fixation of Supernodulating Soybean Mutants

  • Soybean is sensitive to waterlogging stress, leading to reduce their growth and yield significantly. The objective of this study was to characterize the relative sensitivities of biomass accumulation and specific nodule activity under waterlogging stress between supernoduating mutants, 'SS2-2' and 'Sakukei 4' and their wild-type soybeans, 'Sinpaldalkong 2' and 'Enrei', respectively. Flooding treatment was performed to soybean plants grown in a pot by waterlogging for 15 days from the beginning bloom(R1) stage under natural light. The nodule number and weight were considerably decreased by waterlogging stress. The bleeding sap rate of waterlogging soybean plants was decreased by 78-80% in supernodulating mutants and 65-74% in their wild types compared to control plants. The relative ureide-N content was also decreased by waterlogging and the reduction was high in supernodulating mutants. This may cause the massive reduction of shoot and root dry weight and leaf area in waterlogged soybean plants. There was a varietal difference in response to the waterlogging stress. During the waterlogging, supernodulating mutants maintained higher spad value than their wild types. Particularly, the difference between soybean varieties was clear in low rank leaves from the top. Also, supernodulating mutants showed a weak waterlogging tolerance than their wild types. Under waterlogging conditions, massive nodules were considerably destroyed and specific nodule activity after waterlogging may not be recovered when compared to their wild-type soybeans. Supernodulating mutants showed lower seed yield than their wild types in waterlogging conditions.

Comparison of antioxidant activities in Korean black soybean landraces

  • Black soybean with the anthocyanin in the seed coat is known to have higher antioxidant activity than the yellow soybean. This study was carried out to evaluate antioxidant activity of 231 Korean black soybean landraces which conserved at RDA gene bank. Antioxidant activities were measured using DPPH, TPC, TFC, ABTS, and FRAP assay. DPPH showed wide variations, ranging from 16.4 to 200.4($IC_{50}$). TPC, TFC, ABTS, and FRAP were ranged from 0.8 to 13.2 mg gallic acid equivalent/g (mg GAE/g), 0.15 to 0.82 mg quercetin equivalent/g (mg QE/g), 2.0 to 8.3 mg ascorbic acid/g (mg ASC/g) and 0.2 to 3.1 mg ASC/g, respectively. Among 231 Korean black soybean landraces, IT177715 showed the highest antioxidant activity in DPPH assay. In TPC, TFC, ABTS, and FRAP assays, IT274975, IT274551, IT167725, and IT178047 showed the highest antioxidant activity, respectively. In Relative Antioxidant Capacity Index (RACI), IT178047 showed the highest antioxidant activity, while IT177197 the lowest. This study will be able to provide useful data to select black soybean landraces with high antioxidant activity.

Elucidation of Cause of Cotyledon Black-Decay of Soybean Sprout by Bean Bug, Riptortus clavatus

  • Cotyledon Black Decay (CBD) on soybean sprout mimics the black spot due to microbial infection. CBD, not visible or predictable at seedlot state, for some reason, shows up exclusively on cotyledon of soybean sprout during sprouting process. Such an incidence rate fluctuated from 0.8 to 19.5% over three years from 2004. We suspected some pod-infecting anthracnose fungi and/or pod-blight pathogen, or pod-sucking bean bug, one of the major pests of soybean, might have involved, of which we ruled out fungal pathogen because it was preventable through heat treatment, a proven method for seedlot disinfestation. The healthy seeds artificially fed by bean bug for one to seven days were sprouted, and 6 to 41% of the soybean sprout revealed the CBD mimic to those occurred in soybean sprout from previous commercial seedlot screening experiments. This finding is the first report to confirm that bean bug damage to pod at $R_8$ stage is directly responsible for the CBD, which did not concur with any other deleterious effects on sprouting such as reduction in hypocotyls elongation and rooting except unsightly sprout quality. However, earlier feeding either at green pod or greenish yellow pod stage ($R_6$ -early $R_7$ stage) resulted in rather severe damages, which strikingly reduced hypocotyls growth to about one forth to about two third, as well as the reduction in rates of seed germination.

나물콩 품종별 콩나물의 물리화학적 및 관능적 특성 비교 (Comparison of Physicochemical and Sensory Characteristics of Soybean Sprouts from Different Cultivars)

  • 국내 외산 나물콩 품종별 콩나물의 물리화학적 및 관능적 특성을 비교하였다. 백립중은 국내산이 캐나다산보다 월등히 켰으며, 장려품종이 재래품종에 비해 높게 나타났다. 또한 각 품종별 생장 수율은 백립중의 순서와 일치하였다. 수분흡수율은 초기 6시간까지 수분흡수가 급격히 일어나 종자 중량의 약 2.2배에 달하였으며 이후에는 거의 변화가 없었다. 아미노산 조성은 모든 품종에서 16종의 아미노산이 관찰되었으며, Asp, Glu, Lys 및 Arg의 함량이 총 아미노산의 50% 이상을 차지하였다. 지방산 조성은 품종에 따른 차이가 나타나지 않았으며 모든 품종에서 linoleic acid의 함량이 가장 높게 나타났다 무기질은 모든 품종에서 K와 P의 함량이 높고 Fe와 Zn의 함량이 낮게 나타났다 재배직후 배축의 압착강도는 은하가 가장 크고 준저리가 가장 작은 것으로, 자엽의 압착강도는 단엽이 가장 크고 캐나다산이 가장 작은 것으로 나타났다. 삶은 콩나물과 물에 대해 정량묘사분석한 결과 자엽의 노란정도, 자엽의 크기 등 6가지 특성에서 유의적인 차이를 나타내었다.

Object Detection Based on Deep Learning Model for Two Stage Tracking with Pest Behavior Patterns in Soybean (Glycine max (L.) Merr.)

  • Soybean (Glycine max (L.) Merr.) is a representative food resource. To preserve the integrity of soybean, it is necessary to protect soybean yield and seed quality from threats of various pests and diseases. Riptortus pedestris is a well-known insect pest that causes the greatest loss of soybean yield in South Korea. This pest not only directly reduces yields but also causes disorders and diseases in plant growth. Unfortunately, no resistant soybean resources have been reported. Therefore, it is necessary to identify the distribution and movement of Riptortus pedestris at an early stage to reduce the damage caused by insect pests. Conventionally, the human eye has performed the diagnosis of agronomic traits related to pest outbreaks. However, due to human vision's subjectivity and impermanence, it is time-consuming, requires the assistance of specialists, and is labor-intensive. Therefore, the responses and behavior patterns of Riptortus pedestris to the scent of mixture R were visualized with a 3D model through the perspective of artificial intelligence. The movement patterns of Riptortus pedestris was analyzed by using time-series image data. In addition, classification was performed through visual analysis based on a deep learning model. In the object tracking, implemented using the YOLO series model, the path of the movement of pests shows a negative reaction to a mixture Rina video scene. As a result of 3D modeling using the x, y, and z-axis of the tracked objects, 80% of the subjects showed behavioral patterns consistent with the treatment of mixture R. In addition, these studies are being conducted in the soybean field and it will be possible to preserve the yield of soybeans through the application of a pest control platform to the early stage of soybeans.

P34, 7S α' Subunit 및 Lectin 단백질이 없는 녹색자엽을 가진 검정콩 계통 개발 (Improvement of a Black Soybean Line With Green Cotyledon and Triple Null Alleles for P34, 7S α' Subunit, and Lectin Proteins)

  • 검정종피와 녹색자엽을 가진 콩 품종이나 유전자원은 눈의 피로 등 시신경 보호와 눈 질환 예방에 도움을 줄 수 있는 루테인 성분과 인체 내 활성산소 제거, 콜레스테롤 저하, 항산화 및 항암 작용, 면역증강, 노화 방지 등의 작용을 하는 안토시아닌 성분을 풍부하게 함유하고 있다. 그러나, 성숙 종자에는 알러지 유발 및 품질을 저하시키는 P34, 7S α' subunit 및 렉틴 단백질도 포함되어 있다. 따라서, 본 연구는 P34, 7S α' subunit 및 렉틴의 3가지 단백질이 부재하면서 검정종피와 녹색자엽을 가진 콩 계통을 개발하기 위하여 진행되었다. 갈색종피와 녹색자엽을 가지면서 7S α' subunit 및 렉틴 단백질이 부재한 모본과 검정종피와 녹색자엽을 가지면서 P34 및 렉틴 단백질이 부재한 부본과의 교배로 검정종피와 녹색자엽을 가진 157개의 F2 종자가 선발되었다. P34 및 7S α' subunit 단백질의 존재와 부재는 각각 3:1로 관찰되었고, 두 단백질 간에는 9:3:3:1의 분리비와 일치하여 서로 독립적임을 나타내었다. 두 단백질에 대하여 모두 열성(cgy1cgy1p34p34)을 보인 14개의 F2 종자로부터 형질이 양호하면서 P34, 7S α' subunit 및 렉틴 단백질이 모두 없고 검정종피와 녹색자엽을 가진 한 개의 개체가 선발되었다. 선발 개체의 F3 종자를 이용하여 3가지 단백질의 부재에 대한 유전적 고정이 확인되었다. 온실에서 6월 14일 파종 시 선발 개체의 성숙일은 10월 3일, 경장은 약 68 cm였고 백립중은 26.5 g 정도였다. 선발 개체는 항영양성분이 부재하면서 녹색자엽을 가진 유색콩 품종 육성을 위한 모본으로 이용될 수 있을 것으로 보인다.

Optimization of the Spreadable Modified Butter Manufacturing by Response Surface Methodology

  • The aim of this study was to optimize the manufacturing condition of spreadable modified butter by RSM. Based on the central composite design, the degree of optimization was expressed as a SFC as a dependent variable (Y, %) determined by NMR with 23 experimental groups. Three independent variables were the contents of butter ($X_1$, 35-75%), the contents of grape seed oil ($X_2$, 15-35%), and the contents of hydrogenated soybean oil ($X_3$, 0-4%). As the result, SFC at $10^{\circ}C$ was ranged from 32.37 to 42.76%. In addition, the regression coefficients were calculated for SFC at $10^{\circ}C$ by RSREG. The regression model equation for the SFC was $Y=39.18-0.04X_1X_3$. Consequently, the optimal contents for manufacturing spreadable modified butter were determined as 55.18% for butter, 40.78% for grape seed oil, and 4.08% for hydrogenated soybean oil, respectively. The predicted response value for SFC at $10^{\circ}C$ was 30.20%, comparable to the actual experimental SFC value as 29.85%. Finally hardness and spreadability in reference butter and spreadable modified butter produced under the optimal conditions was measured. The hardness in spreadable modified butter was 31.80 N as compared to 69.92 N in reference butter. The spreadability in spreadable modified butter was 5.6 point as compared to reference butter. This difference may be due to the contents of solid fat by butter and hydrogenated soybean oil. This study showed that the SFC value at $10^{\circ}C$ could be a suitable indicator for the manufacturing spreadable modified butter to predict important attributes such as mouth feel, hardness and spreadability.

Changes of Germination Rate of Pulses Seed Germplasm after Long-term Conservation

  • The seeds of soybean (Glycine max), adzuki bean (Vigna angularis), mung bean (Vigna radiata), and kidney bean (Phaseolus vulgaris L.) were examined the germination rate after 10 years of long-term storage ($-18^{\circ}C$) conservation. For soybean seeds, 2,313 accessions were examined and germination rate of 1,082 accessions was decreased with below 15% of initial germination rate. For 227 accessions of soybean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 589 accessions was increased and showed no change for 415 accessions after 10 years of long-term storage. For adzuki bean seeds, 2,058 accessions were examined and germination rate of 739 accessions was decreased with below 15% of initial germination rate. For 63 accessions of adzuki bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 535 accessions was increased and showed no change for 721 accessions after 10 years of long-term storage. For mung bean seeds, 438 accessions were examined and germination rate of 139 accessions was decreased with below 15% of initial germination rate. For 5 accessions of mung bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 155 accessions was increased and showed no change for 139 accessions after 10 years of long-term storage. For kdney bean seeds, 366 accessions were examined and germination rate of 7 accessions was decreased with below 15% of initial germination rate. For 65 accessions of kidney bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 201 accessions was increased and showed no change for 93 accessions after 10 years of long-term storage.

대두 녹병에 대한 내병성 및 이병성 품종의 생리적 반응 (Studies on Physiological Reactions of Soybean Cultivars Tolerant and Susceptible to Rust (Phskopsora pachyrhizi Syd.))

  • 열대 및 아열대지역의 대두재배에서 수량을 격감시키고 있는 콩녹병에 대한 내병성품종과 이병성품종을 공시하여, 본 병해의 진전과정과 생리적 및 수량관련형질에 미치는 영향을 검토한바 그 결과를 요약하면 마음과 같다. 1. 이병성품종은 내병성품종에 비하여 병의 조기감염 및 진전이 빨랐으며, 녹병에 의한 낙엽은 콩 꼬투리가 반정도 등숙된 후부터 급격히 증가하였다. 2. LAI의 감소는 이병성품종이 내병성품종에 비하여 빨랐으며, 건전개체와 녹병 이병개체간의 엽녹소함양의 차리는 내병성품종에서 2.04%, 이병성품종에서 16.43%의 차이가 있었다. 3. 지상부 건물중은 녹병무발병구(살균제 처리구)에서는 계속 증가하였으나, 녹병발병구에서는 R$_{6}$ 생육단계 이후부터 감소하였고, 감소정도는 이병성품종이 내병성품종에 비하여 더 컸다. 4. 협과 종실의 건물중은 이병성품종의 녹병무발병구에서는 R$_{6}$생육단계 이후에 급격히 증가하였으나, 녹병발병구에서는 아주 완만하게 증가하였고, 내병성품종에서는 녹병의 영향을 적게 받아 녹병발병구에서 계속 큰 폭으로 증가하였다. 5. 녹병은 불완전립과 빈 꼬투리 수를 증가시키고, 종실의 Oil 함양 및 길이, 폭, 두께를 감소시키며, 협수, 입수, 백립중을 감소시켜 수량을 격감시키지만 단백질 함양에는 영향을 미치지 않았다. 6.수량에 대한 녹병의 영향은 내병성품종에서 22.3%, 이병성품종에서 68.7%의 수양감소가 있었다.

콩의 발아 전 침종처리가 발아에 미치는 영향 (Effect of Pre-Germination by Treatment of Soaking on Germination of Soybean)

  • 우리나라에서 육성된 콩 품종 90개에 대하여 종자의 발아 전 침종처리에 따른 발아능과 발아율을 조사한 결과는 다음과 같다. 침수량과 침종일수에 따른 콩 종자의 발아력은 침수량이 증가하고, 침종일수가 증가할수록 낮아졌는데, 침수량 90ml와 3일간 침종에서 품종간의 차이가 크게 나타났다. 종자의 수분흡수는 침종 후 6시간까지 급격하게 증가한 후 완만하게 증가하다가 24시간 이후부터는 줄어들었는데, 나물콩의 수분 흡수가 가장 낮았고, 밥밑콩의 수분함량이 가장 낮았다. 용존산소량은 침종 후 3시간까지는 급격히 감소하다가 그 이후에는 완만하게 감소하였는데, 풋콩 및 올콩은 침종 3시간 후부터 용존산소량이 가장 낮았으나 24시간이후에는 다른 용도의 콩에 비해 높아졌다. 전기전도율과 TDS는 침종 후 시간이 경과함에 따라 증가하였는데 풋콩 및 올콩이 가장 높았고, 다음으로 장류콩, 밥밑콩이 높았으며 종자의 크기가 작은 나물콩이 가장 낮았다. 침종 후 발아율이 높은 품종은 장류콩에서는 장수콩, 나물콩에서는 소백나물콩, 밥밑콩에서는 선흑콩, 그리고 풋콩 및 올콩에서는 선록콩이었으며, 발아율이 낮은 품종은 장류콩에서는 익산과 송학콩, 나물콩에서는 방사콩, 밥밑콩에서는 진율콩, 풋콩 및 올콩에서는 신록콩이었다. 전기전도율과 TDS가 낮은 품종들은 발아율이 높았고, 높은 품종은 발아율이 낮은 경향이었다. 종자의 수분흡수가 낮은 품종들은 발아율이 높아 수분흡수와 발아율은 부의 상관이 있었다. 종자의 무게가 높은 품종은 전기전도율과 TDS가 높았으며, 종자의 무게가 낮은 품종은 전기전도율과 TDS도 낮아 정의 상관이 있었다.
