A Correlation Between Fibrinolytic Activity and Microflora in Korean Fermented Soybean Products (된장의 혈전용해효소 활성과 미생물 분포와의 상관관계)

  • A correlation between fibrinolytic activity and microflora in Korean traditional soybean fermented food was investigated. The fibrinolytic activities of traditional soybean pastes and commercially processed samples were 2.42$\pm$1.01 unit/g and 1.58$\pm$0.98 unit/g, respectively. The cell density of Bacillus in traditional soybean pastes was about 10$^{7}$ CFU/g and its commercially processed one was 10$^{6}$ CFU/g. Acid producing bacteria, fungi and yeast group were higher in commercially processed one. The correlations of fibrinolytic activity and microflora in traditional and commercial Doenjang were positively correlated in Bacillus ($R^2$≒ 0.69), negatively correlated in fungal group ($R^2$≒0.40), and there were no significant correlations in acid forming bacteria and yeast group ($R^2$<0.16). Fibrinolytic activities in Meju and Koji were 6.54$\pm$1.97 unit/g and 1.46$\pm$0.43 unit/g respectively, and were positively correlated with Bacillus. Yeast and acid forming bacteria were grown by 5∼6 decimal induction during fermentation period of Doenjang, but Bacillus, fungal cells and fibrinolytic activity were nearly stable. Results indicate that fibrinolytic activity of Doenjang depends on enzyme induction in Meju or Koji processing by Bacillus, Doenjang fermentation process.

Changes of Chemical Composition during Lactic Acid Fermentation of Soy Milk (대두(大豆)요구르트 제조과정중(製造過程中)의 성분변화(成分變化))

  • This experiment was carried out to obtain the fundamental data for development of digestibility and quality enhanced product of soy yogurt. Soy yogurt was processed from raw materials of soybean, defatted soybean and sprouted soybean which inoculated with Lactobacillus acidophilus and Bifidobacterium bifidum as a starter. Changes of chemical compositions, viable cell count and saccharides during processing were investigated including acceptibility of manufactured products. The results were summarized as follows; 1. Defatted soy milk ferment ed with Lactobacillus acidophilus was showed the greatest initial acid productivity and sprouted soy milk was showed the greatest growth of Loctobacillus acidophilus. 2. Acid production was accelerated when 2% glucose was used in soy milk. 3. Addition of reconstituted skim milk in soy milk and defatted roy milk increased acid production but was not showed the effect in the sprouted soy milk. 4. Sprouting soybean, the contents of raffinose and stachyose were decreased but those of glucose was increased. 5. When soy milk was fermented with Lactobacillus acidophilus. the contents of raffinose and stachyose were decreased. 6. As a result of panel test, sprouted soy yogurt which was produced by addition of reconstituted skim milk of 10% showed the greatest flavor and tastes.

Isoflavone Content in Soybean and its Processed Products (콩 및 콩 가공식품의 이소플라본 함량)

  • Daidzein and genistein contents were determined for 46 varieties of soybean and several types of soyfoods using high pressure liquid chromatography. Average content of isoflavones in the soybeans was $809\;{\mu}g/g$ dry matter, ranging from 309 to $1610\;{\mu}g/g$, with Sinpaldal #2 and Dawon showing the highest and lowest amounts, respectively. Isoflavone contents of chonkukjang, soybean paste, and miso were 1045, 781, and $1067\;{\mu}g/g$, respectively. Isoflavone amounts in hard, semi-soft, soft, and fried tofus were 650, 895, 685, and $616\;{\mu}g/g$, respectively.

  • Total aflatoxin ($B_1+B_2+G_1+G_2$) maximum levels of 15 ${\mu}g/kg$ ($B_1=10\;{\mu}g/kg$) were set for grain, beans, peanut, nuts & their processed food (grinding, cutting etc.), processed cereal product & processed bean product, confectionaries (peanut or nut-containing food), soybean paste, red pepper paste, dried red pepper, processed com products for popcorn and steamed rice. The maximum levels for aflatoxin $M_1$ are 0.5 ${\mu}g/kg$ for raw milk and milks before manufacturing processing. The patulin maximum level is 50 ${\mu}g/kg$ in apple juice and apple juice concentrate (including concentrate to use as raw material and converted by concentration multiple). The ochratoxin A is managed at the maximum levels of 5 ${\mu}g/kg$ in wheat, barley, rye, coffee beans and roasted coffee, 10 ${\mu}g/kg$ in instant coffee and raisin, 2 ${\mu}g/kg$ in Grape juice, concentrated grape juice as reconstituted and wine. The fumonisins ($B_1+B_2$) maximum levels are 4000 ${\mu}g/kg$ in com, 2000 ${\mu}g/kg$ in com processed food (grinding, cutting etc.) and com powder, 1000 ${\mu}g/kg$ in processed com products. Standards for mycotoxins in food have been established and the mycotoxin risk in food is managed reasonably and scientifically, based on risk assessment and exposure analysis.

Isoflavone Contents in Some Varieties of Soybean and on Processing Conditions (콩의 종류와 가공 조건에 따른 isoflavone의 함량 변화)

  • enistein (G), and daidzein (D), the major isoflavones, were analyzed in 14 varieties of Korean soybean and various processed soybean products by using high performance liquid chromatography. Isoflavone contents (G+D) were greatly variable among varieties ranged from 308.2 $\mu\textrm{g}$/g to 1,134.2 $\mu\textrm{g}$/g and highest in Danyopkong and Jinpumkong. Among hypocotyl, cotyledon and hull of soybean the concentration of the isoflavone (G+D) in the hypocotyl was highest ranged from 2,971.7 $\mu\textrm{g}$/g to 5,704.9 $\mu\textrm{g}$/g. The distributions of genistein and daidzein were also different in hypocotyl, cotyledon and hull. Higher ratio of daidzein to genistein (D/G) was found in the hypocotyl (4-12) compared to cotyledon and hull (0.1-4). Isoflavone (G+D) contents of soymilks (Sinpaldal#2, Eunhakong) prepared at 16 hour hydration were decreased to 1.1-1.2 times compared with that at 8 hour hydration. Commercial soymilks contained much lower isoflavone (G+D) than laboratory soymilks. Soybean curd (Eunhakong) prepared with MgCl$_2$ showed higher isoflavone (G+D) contents than that with CaSO$_4$. But these values of two different soybean curds made at laboratory were similar to those of 3 commercial curds. The concentration of the isoflavones in soybean sprout separated with 3 parts revealed highest in the head and lowest in the stem. Compared with non-fermented soybean foods the fermented soybean produfts, Kochujang and soybean paste, Duen Jang, showed very low contents of isoflavone (G+D),2.8-3.0 $\mu\textrm{g}$/g, 35.9-63.6 $\mu\textrm{g}$/grespectively.

The Measurement of Folacin Content in Korean Foods -Part 3. Folate Distribution in Various Foods- (한국 상용 식품의 엽산 분석에 관한 연구 -제 3 보-)

  • In continuation of the previous $studies^{2{\sim}3)}$, the folate activity levels in 226 Korean food items were determined by a modified microbiological assay with Lactobacillus casei. There was a large variation in folate activity between the different food groups as well as between each individual food. There was also a wide variation in the biologic availability of folate in foods and the different forms of the folate with different foods in varying amounts. Data showed that almost always, foods cooked and/or processed were lower in folate activity than fresh or raw food and the amount of the loss varied greatly in each food. In calculating dietary intake, total rather than free folate activity levels should he used. In addition, loss of folate activity during cooking and processing of foods should be considered as a major concern for appraising diets and food supplies. Among all assayed food items, including Part $I^{2)}}$ and $I^{3)}$, yeast 2800. ug total per 100g the highest folate level. Soybean, spinach, Shepherd's purse and liter of beef and pork had over 100 ug total per 100 g folate activity. Folate ranging over 50 ug total per 100 g was found in all dried legumes, nuts and seeds assayed, Garland Chrysanthemum, leek, mugwort, wafer cress, asparagus, e99 folk and beef kidney. Wheat, sweet Potatoes,dried fungus, green onion, hotrod pepper, lettuce, radish and some fermented soybeen products had considerably higher folate content ranging around 40 ug total per 100 g. Substantial amounts of folate were not found in many food groups, and among specific groups, in part in starch, sweets, fruits, meat, fish, milk, and cooked and processed foods. Soused fish, oils and fats, beverages, liquor and seasonings, other than fermented soybean products, had almost no folate.

  • The objective of this study was to estimate corn and soybean yield in Illinois and Iowa in USA using satellite and meteorological data. MODIS products for NDVI were downloaded from a NASA website. Each layer was processed to convert projection and extract layers for NDVI. Relations of NDVI from 2002 to 2012 with corn and soybean yield were investigated to find informative days for rice yield estimation. Weather data for the county of study state duration from 2002 to 2012 to correlate crop yield. Multiple regression models based on MODIS NDVI and rainfall were made to estimate corn and soybean yields in study site. Corn yields estimated for 2013 were $10.17ton\;ha^{-1}$ in Illinois, $10.21ton\;ha^{-1}$ in Iowa and soybean yields estimated were $3.11ton\;ha^{-1}$ in Illinois, $2.58ton\;ha^{-1}$ in Iowa, respectively. Corn and Soybean yield distributions in 2013 were mapped to show spatial variability of crop yields of the Illinois and Iowa state.

Optimization of Average Food Consumption Data for Koreans in 1990s (한국인을 위한 식품 평균소비량(1990년대) 자료의 최적화)

  • In Korea, food balance sheet and national nutrition survey have been conducted in parallel by Central Government for several decades. However, the gap between the two statistics became larger to reach an intolerable level in recent years, likely due to the limitation of dietary survey method in spite of the drastic change in dietary habits after industrialization and urbanization during last three decades. The currently available data from national nutrition survey was optimized by taking in to account of the food balance sheet. Adjusted consumption data for individual food items during the period of 1991-95 were given in tabular forms for groups of cereals potatoes sugars & sweets, legumes, nuts & seeds, fruits, vegetables, meats, eggs, dairy products oils & fats and fishery foods. Conversion factors of processed foods including wheat products, soybean products, Kimchi and dairy products into their raw materials were tentatively proposed. Great differences between supply data and intake data of food commodities were pointed out to be likely due to the seasonal variation and ignorance of processed foods and dining-out in the national nutrition survey. It is recommended to utilize these optimized food intake data as the nationally-representative one until new data from the national health and nutrition survey undertaken after modified procedures since 1998 are obtained and verified in future.

Screening of Antibrowning Agents for Minimally Processed Vegetables (최소가공채소류에 적합한 갈변방지제의 선발)

  • Peeled garlic, soybean sprouts, cut onion and cut green pepper were treated by dipping in solutions of different antibrowning agents, and then stored at $10^{\circ}C$. Surface color and polyphenol oxidase activity of produce were measured through the storage. Tested antibrowning agents include ascorbic acid, citric acid and allyl isothiocyanate. The commodities showed different responses to the antibrowning agent solutions; 1% citric acid for peeled garlic, 1% ascorbic acid for soybean sprouts, 2% citric acid for cut onion, and 1% ascorbic acid for cut green pepper showed better retardation in browning than the other treatments for respective product. For peeled garlic, surface browning was concomitant with increase in polyphenol oxidase activity during storage. On the other hand, there was no positive correlation between surface browning and polyphenol oxidase activity for the other stored products.

Study on the promotion of inflammation and whitening of natural materials using bioconversion technology

  • Bioconversion, the enzymatic process by microorganism on organic precursor to desired products. The natural extract is converted into a form that can be easily absorbed into the skin, while scaling up of to higher quantity is possible. Selection of naturally processed raw material rather than chemically processed is preferred. Therefore, fermentation was carried out by mixing Rubus coreanus Miquel, soybean, Zanthoxylum schinifolium as bioconverting materials, the possibility of inflammation, whitening material were checked. In this study, useful microorganisms were isolated from various salted fish, and 16S rDNA sequence was analyzed to confirm their genetic characteristics. Combining the three natural materials using bioconversion technology to study their activity before and after fermentation. To evaluate the antioxidant activity and the active ingredient content the DPPH radical scavenging activity and the total polyphenol content were examined. Raw 264.7 cells were used to evaluate MTT assay, NO and $TNF-{\alpha}$ production inhibitory activity. Also, to evaluate the whitening activity, tyrosinase inhibitory activity and melanin formation inhibitory activity were measured using B16F10 cells. In total 34 strains were obtained from various salted fish. The effective strains useful for the bioconversion process, showed that DPPH radical scavenging ability and polyphenol content were increased in the two kinds of microbial treatment groups compared to the untreated group. 16S rDNA sequencing analysis of the strains showed excellent in Pediococcus pentosaceus B1 in comparison. An increase of up-to 156% in anti-oxidative activity and 141% in polyphenol content was observed after bioconversion. In addition, after mixed fermentation the toxidty of Raw 264.7 and B16F10 cells tended to decrease and a significant increase was observed in anti-inflammatory activity as well. Also, tyrosinase activity and melanin significantly. synthesis decreased significantly.
