Binding of Cytokinin to Proteins of Soybean (Glycine max) Leaves (Cytokinin과 대두(Glycine max) 잎단백질의 결합에 대하여)

  • A polyacrylamide gel electrophoresis technique was applied to cytokinin-protein binding assay. Binding of soybean leaf proteins to cytokinin and relative affinities of protein fractions to cytokinin were studied. The electrophoresis technique appeared to be very useful for determination of cytokinin-protein binding, for identification of protein species binding to cytokinin and for comparison of relative affinities of the proteins to cytokinin. The presence of cytokinin-binding proteins in soybean leaves was confirmed from assays with ammonium sulfate precipitation, Sephadex G-25 chromatography, paper chromatography, and electrophoresis. Three groups of cytokinin-binding proteins were identified in the soybean leaf protein extract and two of the three showed low affinity to cytokinin, however, the third one with mobility between $0.0{\sim}0.2$, probably high molecular weight protein (s), showed high affinity in the electrophoretic analysis.

Effect of Ethrel and TIBA on Yield and Yield Components of Soybean (에스렐과 TIBA 처리가 검정콩의 수량 및 수량구성요소에 미치는 영향)

  • This study was conducted to examined the effects of TIBA and ethrel on podding rate, percentage of seed numbers per pod, and seed yield of black soybean. The black soybean variety “Geomjeongkong 1” was used. TIBA of 50, 100, and 150 ppm and ethrel of 500, 1,000, and 1,500 ppm were applied at V6 (fifth leaf stage), V8 (seventh leaf stage), and R1 (flowering stage). The podding rate was increased by the treatment of TIBA at R1 stage while it was decreased by ethrel at all growth stages. Compared with control, TIBA decreased 100 seed weight a little bit but ethrel increased it slightly, on the contrary. Both TIBA of 50 ppm and ethrel of 500 ppm increased the sued yield by 8 to 28% compared with control regardless of growth stages of application; the highest seed yield was obtained at V6 stage of application of 50 ppm in TIBA and 500 ppm in ethrel, respectively.

Effect of Phosphorus Stress on Photosynthesis and Nitrogen Fixation of Soybean Plant under $CO_2$ Enrichment (대기 $CO_2$ 상승시 인산공급이 식물체의 광합성 및 질소고정에 미치는 영향)

  • The objective of this study was to examine the effect of phosphorus deficiency on nitrogen fixation and photosynthesis of nitrogen fixing soybean plant under $CO_2$ enrichment condition. The soybean plants(Glycine max [L.] Merr.) inoculated with Bradyrhizobium japonicum MN 110 were grown with P-stressed(0.05 mM-P) and control(1 mM-P) treatment under control$(400\;{\mu}l/L\;CO_2)$ and enrichment$(800\;{\mu}l/L\;CO_2)$ enviromental condition in the phytotron equipped with high density lamp$(1000\;{\mu}Em^{-2}S^{-1})$ and $28/22^{\circ}C$ temperature cycle for 35 days after transplanting(DAT). At 35 DAT, phosphorus deficiency decreased total dry mass by 64% in $CO_2$ enrichment condition, and 51% in control $CO_2$ condition. Total leaf area was reduced significantly by phosphorus deficiency in control and enriched $CO_2$ condition but specific leaf weight was increased by P deficiency. Phosphorus deficiency significantly reduced photosynthetic rate(carbon exchange rate) and internal $CO_2$ concentration in leaf in both $CO_2$ treatments, but the degree of stress was more severe under $CO_2$ enrichment condition than under control $CO_2$ environmental condition. In phosphorus sufficient plants, $CO_2$ enrichment increased nodule fresh weight and total nitrogenase activity(acetylene reduction) of nodule by 30% and 41% respectively, but specific nitrogenase activity of nodule and nodule fresh weight was not affected by $CO_2$ enrichment in phosphorus deficient plant at 35 DAT. Total nitrogen concentrations in stem, root and nodule tissue were significantly higher in phosphorus sufficient plant grown under $CO_2$ enrichment, but nitrogen concentration in leaf was reduced by 30% under $CO_2$ enrichment. These results indicate that increasing $CO_2$ concentration does not affect plant growth under phosphorus deficient condition and phosphorus stress might inhibit carbohydrate utilization in whole plant and that $CO_2$ enrichment could not increase nodule formation and functioning under phosphorus deficient conditions and phosphorus has more important roles in nodule growth and functioning under $CO_2$ enrichment environments than under ambient condition.

Inhibitory Effect of Green-Yellow Vegetables on the Mutagenicity in Salmonella Assay System and on the Growth of AZ-521 Human Gastric Cancer Cells (녹황색 채소류의 돌연변이유발 억제 및 AZ-521 위암세포의 성장 저해효과)

  • The antimutagenic effect of green-ye1low vegetables on the mutagenicities induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and N-nitrosodimethylamine (NDMA) in Salmonella assay system and also their inhibitory effect on AZ-521 human gastric cancer cells were studied. Twenty-four items from twenty six kinds of vegetables(92%) revealed antimutagenic activity toward MNNG (p< 0.0l, 0.05). Perilla leaf, Korean cabbage, cauliflower, lettuce, mustard leaf, water dropwort, small water dropwort, carrot and burdock inhibited the mutagenicity more than 80%. The methanol extracts of the vegetables also showed the antimutagenic activity toward NDMA (p< 0.01, 0.05). Especially, perilla leaf, kale, soybean sprout and onion inhibited more than 80% of the NDMA induced mutagenicity in S. typhimurium TA100. Small water dropwort and perilla leaf exhibited the strong inhibitory effect (97~100%) on the growth of the AZ-521 human gastric cancer cells. Soybean sprout, water dropwort, broccoli, crown daisy, green red pepper, red pepper leaves, spinach, cabbage and sweet potato also inhibited growth of the cancer cells (p < 0.001~0.05).

Functional Properties and Antioxidant Effects of Solanum nigrum-Ethanol Extract (까마중-에탄올 추출물의 기능적 특성과 항산화 효과)

  • Functional properties of Solanium nigrum-ethanol extract were investigated, nitrite scavenging ability(NSA) and antioxidant activities based on thiobarbituric acid(TBA) value were measured. The contents of potassium in fruit and root, calcium in leaf were the highest mineral components in each parts of Solanium nigrum. Total aromatics contents(TAC) was order of fruit>root>leaf, total flavonoid contents(TFC) in leaf was 6 and 10 times, total phenolic compounds contents(TPC) in leaf was 15 and 30 times of those in fruit and root, respectively, and TFC was high with TPC increase. NSA of leaf-ethanol extract was 61.9% on pH 1.2, 53.5% on pH 3.0, 54.8% on pH 4.0 and 14.3% on pH 6.0, where as NSA of root and fruit-ethanol were considerably low to 3.5~7.9% over pH 1.2~6.0. TBA value of soybean oil with the dosage of leaf-extract showed few differences from control within 3 days, but it fairly decreased with considerable antioxidant effect to 47.6% of the control in 8 days.

Analysis of Spectral Reflectance Characteristics Using Hyperspectral Sensor at Diverse Phenological Stages of Soybeans

  • South Korea is pushing for the advancement of crop production technology to achieve food self-sufficiency and meet the demand for safe food. A medium-sized satellite for agriculture is being launched in 2023 with the aim of collecting and providing information on agriculture, not only in Korea but also in neighboring countries. The satellite is to be equipped with various sensors, though reference data for ground information are lacking. Hyperspectral remote sensing combined with 1st derivative is an efficient tool for the identification of agricultural crops. In our study, we develop a system for hyperspectral analysis of the ground-based reflectance spectrum, which is monitored seven times during the cultivation period of three soybean crops using a PSR-2500 hyperspectral sensor. In the reflection spectrum of soybean canopy, wavelength variations correspond with stages of soybean growths. The spectral reflection characteristics of soybeans can be divided according to growth into the vegetative (V)stage and the reproductive (R)stage. As a result of the first derivative analysis of the spectral reflection characteristics, it is possible to identify the characteristics of each wavelength band. Using our developed monitoring system, we observed that the near-infrared (NIR) variation was largest during the vegetative (V1-V3) stage, followed by a similar variation pattern in the order of red-edge and visible. In the reproductive stage (R1-R8), the effect of the shape and color of the soybean leaf was reflected, and the pattern is different from that in the vegetative (V) stage. At the R1 to R6 stages, the variation in NIR was the largest, and red-edge and green showed similar variation patterns, but red showed little change. In particular, the reflectance characteristics of the R1 stage provides information that could help us distinguish between the three varieties of soybean that were studied. In the R7-R8 stage, close to the harvest period, the red-edge and NIR variation patterns and the visible variation patterns changed. These results are interpreted as a result of the large effects of pigments such as chlorophyll for each of the three soybean varieties, as well as from the formation and color of the leaf and stem. The results obtained in this study provide useful information that helps us to determine the wavelength width and range of the optimal band for monitoring and acquiring vegetation information on crops using satellites and unmanned aerial vehicles (UAVs)

Lipid Peroxidation of Ginseng Thylakoid Membrane (인삼 틸라코이드 막의 지질과 산화)

  • In order to elucidate the mechanism of the leaf-burning disease of ginseng (Panax ginseng C.A. Meyer), the relationships between thylakoid membrane peroxidation and chlorophyll bleaching were investigated in comparison with the ones of soybean (Glycine max L). When I measured the rate of lipid peroxidation in the thylakoids of ginseng and soybean by irradiation of light(60 w.m-2), it was identified that, the remarkably lower rate of lipid peroxidation was found in the ginseng thylakoid than the case of soybean. When lipid peroxidation of ginseng thylakoid was induced in the dark, chlorophyll contents of thylakoid was not changed. The results suggest that lipid peroxidation does not affect the chlorophyll bleaching in ginseng thylakoid. Thylakoid membrane peroxidation as well as chlorophyll bleaching was closely related with photosynthetic electron transport. But, according to the quenching experiment active oxygen species induced lipid peroxidation may be different species in the case of chlorophyll bleaching.

  • Photosynthetic characteristics of soybean leaves infected with Pseudomonas syringae pv. glycinea were investigated for 8 days. The difference in photosynthesis rate between healthy and diseased soybean leaves decreased for 2 to 4 days after inoculation and then increased. In respiration rate, healthy and diseased leaves showed the same tendency as photosynthetic rate. The stomatal resistance increased following Pseudomonas syringae pv. glycinea infection. The total chlorophyll content of the infected leaf was less than that of the uninfected. Pseudomonas syringae pv. glycinea infection induced the malformation of stacked grana in chloroplast. Dry matter production declined after infection.

DNA, RNA, Protein and Yield of the Soybean Plant, Glycine max Merr., as Affected by Phosphorus Nutrition (대두의 핵산, 단백질 및 물질생산에 미치는 인산비료의 효과)

  • The effect of phosphorus nutrition on the content of deoxyribonucleic acid(DNA), ribonucleic acid(RNA), crude protein and plant growth of soybean plant(Glycine max, Merr.) was studied. Yields of the above-and under-ground parts of the soybean plant in terms of dry weight, the amounts of crude protein, RNA and DNA continued to increase with increasing phosphorus supply. The amounts of RNA and crude protein were highest in the leaf tissues where most intensive growth was taking place. The relationships among DNA, RNA, crude protein and plant growth appeared to consist of the central dogma which has immortalized, while DNA in plant tissue was subject to charges cuased by external environmental facters such as phosphorus nutrition.

A Study on Making Meju (Molded Soybean) for Traditional Jang (전통장의 메주 제조에 관한 연구)

  • In this study, we analyzed the utensils, covers and mats that were used for making meju, the shape of meju, and the heating method used for making meju from the 225 ways of preparing jang mentioned in the 32 volumes of the ancient cook books from 530 AD to 1950. The heating method of traditional meju bean and starch included 57 kinds of steaming, 59 of boiling, 21 of roasting + boiling, and 2 of cooking. The shape of meju included 41 kinds of egg, 27 of ball, 22 of lump, a kind of doughnut, 8 kinds of hilt, 6 of flat, 4 of chip, and a kind of square. Among the 72 gochoojang meju, the heating method of bean included 9 kinds of boiling, and 6 kinds of steaming; whereas the heating method of starch included 19 kinds of steaming of dough, 11 of rice cooking, and 5 of boiling of dough. The utensils for molding of bean meju were 49 kinds of straw sack, 14 of round straw container, 11 of heating bed, 7 of large straw bowl or Japanese-snailseed, 5 of jar, 4 of ditch, 3 of straw bowls, 2 of pottery steamer of dough, 2 of gourd, and a kind of long round bamboo bowl and sack of straw. The cover and the mat used for molding of meju included 36 kinds of straw, 17 kinds of paper mulberry leaf, 15 of wide straw seat, 14 of mugwort, 11 of pine tree leaf, 10 of soybean leaf, 6 of cocklebur leaf, 6 of sumac leaf, 6 of barley straw, 6 of mulberry leaf, 5 of fallen leaf, 5 of cogon grass, 4 of reed seat, 3 of scrap of cloth, 2 of Indian bean tree leaf, a kind of reed. There were only 5 kinds of hanging.
