Responses of Soybean Cultivars to Excessive Soil Moisture Imposed at Different Growth Stages

  • Soybean [Glycine max (L.) Merrill] crops, grown in a rice soybean rotation, can suffer when grown in soil with excessive moisture. The objective of this work were to determine the reduction in growth and yield, responses of vegetative and reproductive growth of soybean to excessive soil moisture achieved by prolonged irrigation. Responses of different cultivars were determined at growth stages from V6 to R8 to clarify the sensitive growth stages or characteristics to excessive soil moisture. Cultivar differences in response to excessive soil moisture condition were conspicuous in seed dry weight and harvest index (HI) but not in the response of seed number or pod number per plant. The timing of irrigation causing the condition of excessive soil moisture influenced the vegetative or reproductive traits. Soybean plants were more affected by irrigation commencing at the pre-flowering than at the post-flowering stage. Post-flowering irrigation did not reduce growth of vegetative organs significantly; in fact the growth of stems and leaves was facilitated by the prolonged irrigation commencing at flowering. Differences between cultivar response to prolonged irrigation were assumed to relate to the reduced amount of assimilates translocated to the reproductive organ.

  • Lipoxygenase might be associated with seed deterioration by catalyzing the incorporation of molecular oxygen into fatty acids and generating free radicals. This study was performed to determine whether seed lipoxygenase activity would alter soybean seed longevity. In this study, germination percentage of lipoxygenase-lacking cultivar Jinpumkong2 (lx1lx1lx2lx2lx3lx3) was lower than that of Taekwangkong (Lx1Lx1Lx2Lx2Lx3Lx3). Segregation ratio for the three lipoxygenase isozymes of the F2-derived from the cross between Taekwangkong and Jinpumkong2 was fitted to 9 (Lx1Lx2Lx3) : 3 (Lx1Lx2lx3) : 3 (lxllx2Lx3) : 1 (lx1lx2lx3), suggesting the tight linkage between the Lx1 and Lx2 loci. Germination percentages varied widely but not differed among lipoxygenase isozyme types of F$_3$ seeds before and after accelerated aging. Seed coat of Jinpumkong2 was damaged severely following accelerated aging, whereas that of Taekwangkong was not. Thus, seed of lipoxygenase-lacking soybean cultivar, Jinpumkong2 showed greater deterioration compared with that of the normal Taekwangkong. However, the presence or absence of lipoxygenase activity had no effect on soybean germination.

  • Improvement of crop yield can be achieved through understanding genetic variation in reproductive characters and its impact on yield components. The present study was performed to evaluate genetic diversity for reproductive growth characters in exotic germplasm resources and to determine the relationships between developmental and growth periods with yield and yield components in soybean cultivar groups. For phenotypic evaluation such as reproductive and agronomic traits, a total of 80 indigenous and exotic soybean cultivars collected from four different geographical regions (China, Japan, Korea, and Vietnam) were grown from May to November of 2003 at the Seoul National University Farm, Suwon, Korea ($127^{\circ}02'E$ longitude, $37^{\circ}26'N$ latitude). Most of all the characters exhibited wide range of phenotypic variation, of which pod number, seed number, and plant yield showed greater range as compared to other characters. Korean cultivar groups showed greater diversity than the other cultivar groups in seven characters. Correlation analysis showed that days to flowering (DTF) and days to maturity (DTM) had close association with agronomic traits as well as yield and yield components. Both DTF and DTM had positive correlation with the other characters except one hundred seed weight. Stepwise multiple linear regression revealed that seed and pod number were identified as being significant for plant yield. The results in this study indicated wide variation in agronomic traits including DTF and DTM, suggesting the valuable genetic resources in a soybean breeding program.

Quantitative Variation of Total Seed Isoflavone and its Compositions in Korean Soybean Cultivars (Glycine max (L.) Merr.)

  • The variation of content of 12 soybean seed isoflavone components was determined in the aglycone, glucoside, malonylglucoside and acetylglucoside groups of 44 Korean soybean cultivars grown in 2016 as well as in 2017. The total isoflavone content of the 44 cultivars averaged at $2935.4{\mu}g/g$ and was in the range of 950.6 to $5226.3{\mu}g/g$ for two years. Malonylglucoside group averaged at $2437.2{\mu}g/g$ with the highest proportion of isoflavone composition (83.0%). Significant differences were observed between cultivars, years and their interactions for both the total isoflavone and each composition group contents (P < 0.0001); however, no year-wise differences were observed for daidzein and genistin. The broad-sense heritability ($h^2$) within the set of 44 Korean soybean cultivars was as high as 0.93 for the total isoflavone content and was in the range of 0.8-0.92 for each composition group of isoflavone except for acetylglucoside. The total isoflavone content in cultivar group for soy-sprout was higher ($3850.4{\mu}g/g$) than that for the other cultivar groups of soy-paste and tofu ($3082.8{\mu}g/g$), black or green soybean cooked with rice ($2345.8{\mu}g/g$), and early maturity group ($1298.6{\mu}g/g$). The total isoflavone content of 'Sowonkong', a soybean cultivar for soy-sprout, was the highest ($5226.3{\mu}g/g$). In the cultivar group for soy-paste and tofu, the average isoflavone contents of 'Daepung', 'Daepung2ho', 'Saegeum', 'Uram', and 'Jinpung' were higher than $4000{\mu}g/g$. With the exception of small seeded cultivars with low isoflavone contents such as 'Sohwang' and 'Socheongja', the seed size and total isoflavone content were significantly negatively correlated in 2016 and 2017, respectively ($r=-0.47^{**}$ and $-0.49^{**}$). The number of days of growth from flowering to maturity did not affect the variations observed in isoflavone content.

  • Soybean rhizobia were isolated from 101 soybean (Glycine max.) cultivar which had been grown for the breeding experiment in Korea. Seven strains of the fast-growing soybean rhizobia and nine strains of the slow-growing soybean rhizobia were selected on the basis of their growth rate in AMA medium and their high ability of nodulation. The slow-growing soybean rhizobia were identified as Bradyrhizobium japonicum in the acetylene-reducing activity, microbial characteristics, and biochemical characteristics whereas the fast-growing soybean rhizobia were very similar to Rhizobium fredii.

Isolation of soybean mutants with high and low inorganic phosphorus

  • In soybean (Glycine max (L.) Merr.) seeds, phosphorus (P) is primarily stored in the form of phytate, which is generally indigestible by monogastric animals such as human, pig, poultry, and fish. Thus, this study was conducted to isolate soybean mutants with high available P. Inorganic P content was assessed in a total of 1,266 ethyl methanesulfonate (EMS) $M_4$ lines from the Pungsannamul cultivar. Among the tested lines, four EMS lines (PE379, PE432, PE2205, and PE2503) showed higher mean inorganic P ($1.21-1.56gkg^{-1}$) than did the Pungsannamul cultivar ($0.90gkg^{-1}$). Additionally, six EMS lines (PE718, PE828, PE1466, PE1552, PE3378, and PE3386) showed lower mean inorganic P ($0.38-0.60gkg^{-1}$). The high inorganic P mutants isolated in this study will be further investigated for phytate and total P levels. Moreover, the high and low inorganic P lines will be utilized in a future study of the biochemical pathway of phytate.

Polyphenol and Phytate Contents and Their Relationship to Antioxidative Activity in Soybeans (콩의 폴리페놀, 피테이트 함량과 항산화능과의 관계)

  • In this study we evaluated polyphenol contents, phytate contents, and antioxidant activities (DPPH radical and hydroxyl radical scavenging activity) in several soybean cultivars. The polyphenol contents of the cultivars ranged from 0.0446 mg/g to 0.0652 mg/g, with the highest level found in the WS82 cultivar, whereas and the lowest in Duyukong. The phytate content of the WS2906 cultivar was the highest at 22.28 mg/g whereas Duyukong showed the lowest level. $IC_50$ values (concentration of soybean extract exerting 50% scavenging of radicals) for DPPH radical and hydroxyl radical scavenging activity showed the highest in WS82. A positive correlation between DPPH radical scavenging activity and polyphenol contents was observed in the soybean cultivars, whereas not significant relationship between antioxidant activity and phytate content was found.

Nitrogen Uptake and Growth of Soybean Seedlings under Flooding Stress

  • This experiment was carried out on plastic pots ($40cm{\times}25cm{\times}30cm$) filled with sand soil at greenhouse using two soybean cultivars with small seed; one was Pungsannamulkong (PSNK) recognized as a tolerant cultivar against excessive water stress and the other one was Sobaeknamulkong (SBNK) recognized as a susceptible cultivar. Seed was sown with 30 plants of 2 hills, and the amount of applied fertilizer was N; 3.0 g, P; 3.0 g, and K; 3.4 g per $m^2$ with all basal fertilizations. Plants were grown under photoperiod of natural light with day temperature of $31{\pm}5^{\circ}C$ and night temperature of $22{\pm}1^{\circ}C$. The flooding treatment was done for 3, 5, 7 and 10 days by filling pots with tap water up to 1 cm above the level of the soil surface when plants were 2 days after emerging. Nitrogen uptake by leaves of soybeans decreased significantly by the flooding after 6 days. This significant reduction of N uptake by flooding was evidently recognized from the chlorosis of leaves. The dry matter of flooded soybean seedlings significantly decreased compared to non-flooded soybean seedlings at 10 days. The dry matter of roots also showed similar result of the shoot. Shoots had more N reduction than roots under the flooding. This N reduction was more pronounce in SBNK than in PSNK. Chlorophyll content of flooded soybeans showed decreasing or non-increasing tendency, and the reduction of chlorophyll content was more in SBNK than in PSNK from the flooding stress. Nitrate content of soybean seedlings with flooding stress showed decreasing tendency in shoot and root parts. Ammonium content, however, was higher in flooding stress compared to the non-flooding. Flooding caused a remarkable change in the AA (amino acid) composition and TAA (total amino acid) concentration in the leaves of soybean seedlings.

A TMT-based quantitative proteomic analysis provides insights into the protein changes in the seeds of high- and low- protein content soybean cultivars

  • The presence of high amounts of seed storage proteins (SSPs) improves the overall quality of soybean seeds. However, these SSPs pose a major limitation due to their high abundance in soybean seeds. Although various technical advancements including mass-spectrometry and bioinformatics resources were reported, only limited information has been derived to date on soybean seeds at proteome level. Here, we applied a tandem mass tags (TMT)-based quantitative proteomic analysis to identify the significantly modulated proteins in the seeds of two soybean cultivars showing varying protein contents. This approach led to the identification of 5,678 proteins of which 13 and 1,133 proteins showed significant changes in Daewon (low-protein content cultivar) and Saedanbaek (high-protein content cultivar) respectively. Functional annotation revealed that proteins with increased abundance in Saedanbaek were mainly associated with the amino acid and protein metabolism involved in protein synthesis, folding, targeting, and degradation. Taken together, the results presented here provide a pipeline for soybean seed proteome analysis and contribute a better understanding of proteomic changes that may lead to alteration in the protein contents in soybean seeds.

Physicochemical Characteristics of Fermented Soybean Products with Aspergillus Strain (Aspergillus속 균주를 이용한 콩 발효물의 이화학적 특성)

  • In this study, the properties of the substances fermented soybean producted by manufacturing with several Aspergillus strains were investigated. The five soybean cultivar that includes miso, jinpung, pyeongwon, cheonga and chamol were used in this experiment. The pH and total acidity were 6.30~6.66%, and 0.27~0.48%, respectively with differences depending on the samples. The color values for L value, a, and b value were 60.28~69.80, 0.64~3.68, and 11.48~31.21, respectively. The amylase activities, protease activities, and amino-type nitrogen of the jinpung samples were the highest among all cultivars. The mold counts the fermented soybean products by cultivars were 6.18~9.14 log CFU/g, and miso was the highest. Free amino acid and organic acid contents were highest in the jinpung and showed different composition depending on each sample. A total of 18 volatile aroma-compounds, including two acids, four alcohols, four ketones, three phenols, one furan, three pyrazines, and one miscellaneous compounds. In conclusion, it is expected that manufacturing A. oligze inoculation fermented soybean products using jinpung cultivar will improve quality.
