Anti-Inflammatory Effect of Biji (Soybean curd residue) on LPS-Stimulated RAW264.7 Cells (마우스 RAW264.7 세포에 대한 비지 추출물의 항염증 활성)

  • In this study, we evaluated anti-inflammatory effect of biji in LPS-stimulated RAW264.7 cells. Biji inhibited the generation of NO and $PGE_2$ through the suppression of iNOS and COX-2 expression. In addition, biji attenuated the expression of TNF-${\alpha}$ and IL-$1{\beta}$ induced by LPS. Biji blocked LPS-mediated $I{\kappa}B-{\alpha}$ degradation and subsequently inhibited p65 nucleus accumulation in RAW264.7 cells, which indicates that biji inhibits NF-${\kappa}B$ signaling. In addition, biji suppressed p38 phosphorylation induced by LPS. Our results suggests that biji may exert anti-inflammatory activity through blocking the generation of the inflammatory mediators such as NO, $PGE_2$, iNOS, COX-2, TNF-${\alpha}$ and IL-$1{\beta}$ via the inhibiting the activation of NF-${\kappa}B$ and p38. From these findings, biji has potential to be a candidate for the development of chemoprevention or therapeutic agents for inflammatory diseases.

  • This experiment was carried out to evaluate the effect on different levels of crude protein intake according to sex and body weight on the nitrogen retention, and the protein requirement for feeding standard of the Korean Native Goat. The results obtained from the study were as follows. 1. The nitrogen retention rate when the female goat at 6 months age (13kg) had taken native grass hay 40%(126g) and flaking barley 60%(186g) was 36 percent, and when the intake matter was native grass hay 55%(226g) and flaking barley 45%(186g) was 25.6 percent. 2. The nitrogen retention rate when the meal goat at 5 months age (11kg) had taken native grass hay 47%(245g), flaking barley 29%(153g) and soybean meal 24%(126g) was 52.4 percent and when the intake was native grass hay 42%(206g) and flaking barley 58%(279g) was 40.7 percent. But if the in take matter was only (328g) native grass hay (crude protein 8%) it was -23.9 percent. 3. Maintenance nitrogen requirement of Korean native goat were estimated about 5g per day to 11 or 13kg live weight.

The Effects of Dietary Protein Level on Laying Performance and Egg Quality in Japanese Quail (메추리에 있어서 산란 사료 내 단백질 수준이 산란 성적과 난질에 미치는 영향)

  • This experiment was conducted to investigate the effects of various levels of dietary protein on laying performance and egg quality in laying quail. A total of six hundred forty 7-week-old laying type Japanese quail were divided into four groups with eight replicates per group (20 birds per replicate) and fed four diets differing in dietary protein levels (18, 20, 22 and 24%) with isocaloric corn-soybean meal-based diets for 8 weeks. The results showed no significant differences in feed intake and egg weight among the groups. With an increase in dietary protein levels, the egg production and daily egg mass were linearly increased (P<0.05). However, there were no significant differences in eggshell thickness and Haugh unit. The dietary protein did not affect the blood levels of GOT, GPT, BUN, albumin, and creatinine. The ammonia concentration of intestinal digesta was not affected by the dietary treatments. The contents of hepatic triacylglycerol in the groups fed diets containing 20% protein or more were significantly reduced as compared with that of 18% CP group (P<0.05). Overall, this study showed that relatively high levels of dietary CP are needed to sustain the higher egg production and daily egg mass and to avoid the hepatic triacylglycerol accumulation in Japanese quail.

Supplementation of Indigenous Green Microalga (Parachlorella sp.) to Pre-starter Diet for Broiler Chickens (초기 육계 사료내 토착미세조류(Parachlorella sp.) 첨가에 따른 성장 및 면역반응 변화)

  • The present study determined the effect of dietary cultivated microalgae (Parachlorella sp.) on the growth and immune responses of pre-starter broilers. A total of 320 one-day-old birds (Ross 308) were allocated to 4 treatments with 8 blocks in a randomized complete block design. The four experimental diets consisted of a corn-soybean meal-based control diet, and three diets contained 0.5%, 1.0%, and 1.5% microalgae powder at the expense of cornstarch in the control diet. After feeding the experimental diets for 7 days, the body weight and feed intake of all birds were measured, and 8 birds were randomly selected from each treatment. Peripheral blood mononuclear cells (PBMCs) and serum were harvested for immune profile assessment, including cytokines and cell migration receptors. No differences in growth performance were observed among the treatments. The birds that were fed diets containing graded levels of microalga showed a linear increase in the mRNA expression of cytokine genes in PBMCs, including that of IL2, IL1β, and IL18 (P<0.05). With respect to the chemokine receptor genes in PBMCs, mRNA expression of CCR2, CCR9, and ITGA4 changed quadratically (P<0.05), but that of CCR7 increased linearly (P<0.01). Cytokine protein secretion in blood, including that of IL-1β and IL-6, increased linearly (P<0.01) with an increase in the microalgal content. Overall, the present results show that the indigenous microalgae powder used in this study could stimulate immunity with no detrimental effects on the growth performance of pre-starter broiler chickens.

Microencapsulation of Aster scaber and Aster glehni by Spyay Drying (분무건조법을 이용한 참취 및 섬쑥부쟁이 추출물의 미세캡슐화)

  • Spray drying was used a means for microencapsulation of Aster scaber and Aster glehni. The optimum conditions of spray drying were found to be 210$^{\circ}C$ for the inlet temperature and 5 $m\ell$/min for the feeding rate. The color difference decreased by 12∼25 % far the herb extracts depending on storage temperatures. With an increase in storage temperature, color of the control changed considerably compared to microencapsulated powders. This is due to the fact that wall materials minimized effect of oxygen on the herb extracts. Microencapsulated powders in the amounts of 50 ppm and 250 ppm added to hydrogenated soybean oils led to lower acid values and conjugated diene values during 6 day storage. Compared to the control, the herb extracts displayed 16∼48% increase in antioxidation effect, while the microencapsulated powders exhibited 46∼88% increase. Extracts also showed lower conjugated dienoic acid contents than those of the control. Although antioxidation effect of the microencapsulated powders did not prevail on the significant level in the early stage of storage of oils tested, it became predominant after one week of storage as wall materials were gradually degraded under the high temperature thereby releasing core material from the microcapsule.

Effects of Cheonggukjang Added Phellinus linteus myceria on Lipid Metalbolism in Adult Female Rats (상황버섯균사체청국장이 고지혈증을 유도한 암쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of Cheonggukjang (traditional soybean food, CK) and Cheonggukjang added with Phellinus linteus myceria (CKP) on the lipid metabolism, growth, food intake, and food efficiency ratio(FER) in adult female rats. Sprague-Dawley rats were fed a high fat diet (control diet: 0.1% cholesterol, 10% Lard) for 4 weeks. The rats were randomly assigned to each treatment group: control, and two kinds of CK and CKP (powders of CK and CKP as dietary protein sources). After 4 weeks of experimental diet consumption, the body weights and the uterine fat pad weights of the CK and CKP diet groups were more significantly decreased than those of the control diet group. The hepatic cholesterol, triglyceride and total lipid levels were significantly lower in both the CK and CKP diet fed groups than those in the control group. The concentrations in serum triglyceride and LDL-cholesterol were significantly decreased in the CK and CKP diet groups compared to those in the control group. Fecal total lipid, moisture and wet weight excretion in the CK and CKP diet groups were more increased than those in the control group. These results showed that feeding of cheonggukjang and cheonggukjang added with Phellinus linteus myceria both the triglyceride and LDL-cholesterol in serum, as well as the triglyceride and cholesterol in the livers of the rats.

The Effect of Potassium Phosphate as a pH Stabilizer on the Production of Gellan by Sphingomonas paucibilis NK-2000 (Sphingomonas paucibilis NK-2000에 의한 젤란의 생산에 미치는 pH 안정제로서의 인산칼슐의 영향)

  • Maximal productions of gellan by Sphingomnas paucibilis NK2000 from 20 g/l glucose and 10 g/l soybean pomace were 7.46 g/l in a flask and 7.35 g/l in a 7 l bioreactor, when the initial pH of media was 6.8. Maximal production of gellan in a 7 l bioreactor under pH control by sodium hydroxide was 8.42 g/l, whereas that under control by potassium phosphate was 8.50 g/l. The optimal concentration of potassium phosphate in a medium for production of gellan by S. paucibilis NK2000 was found to be 5.0 g/l. Maximal production of gellan in a medium containing 5.0 g/l potassium phosphate without pH control was 8.93 g/l in a 7 l bioreactor. In this study, a simple process without pH control was developed to enhance the production of gellan, with optimized concentration of potassium phosphate in the medium.

Inhibition of Cyclooxygenase-2 Activity and Prostaglandin E2 Production through Down-regulation of NF-κB Activity by the Extracts of Fermented Beans (발효 콩의 NF-κB 활성 억제를 통한 cyclooxgenase-2 활성과 prostaglandin E2 생성 억제)

  • Cyclooxygenase (COX)-2 is generally known as an inducible enzyme, and it produces arachidonic acid to prostaglandin $E_2$ ($PGE_2$), which has been demonstrated to play a critical role in inflammation. In the present study, we investigated the effects of the extracts of fermented beans including soybean (FS), black agabean (FBA) and yellow agabean (FYA), on the expression of COXs and production of $PGE_2$ in U937 human promonocytic cells. Treatment of phorbol 12-myristate 13-acetate (PMA) significantly induced pro-inflammatory mediators such as COX-2 expression and $PGE_2$ production, whereas the levels of COX-1 remained unchanged. However, pre-treatment with FS, FBA and FYA significantly decreased PMA-induced COX-2 protein as well as mRNA, which is associated with inhibition of $PGE_2$ production. Moreover, FS, FBA and FYA markedly prevented the increase of nuclear translocation of nuclear factor kappa B (NF-${\kappa}B$) p65 by PMA. Our data indicate that the extracts of fermented beans exhibits anti-inflammatory properties by suppressing the transcription of pro-inflammatory cytokine genes through the NF-${\kappa}B$ signaling pathway.

Isolation of Mannanase-producing Bacteria, Bacillus subtilis WL-6 and WL-11, and Cloning and Characterization of Mannanase (Bacillus subtilis 분리균 2주 유래 mannanases의 특성 비교)

  • Two bacterial strains producing extracellular man nanase were isolated from doenjang, a traditionally fermented soybean paste in Korea. The isolates, WL-6 and WL-11, were identified as Bacillus subtiis on the basis of their 16S rRNA gene sequences, morphological, and biochemical properties. Two genes encoding the mannanase of both B. subtilis WL-6 and B. subtilis WL-11 were each cloned into Escherichia coli, and their nucleotide sequences were determined. Both mannanase genes consisted of 1,086 nucleotides, encoding polypeptides of 362 amino acid residues. The deduced amino acid sequences of the two WL-6 and WL-11 mannanases, designated Man6 and Man11, respectively, differed from each other by eight amino acid residues, and they were highly homologous to those of mannanases belonging to the glycosyl hydrolase family 26. The 26 amino acid stretch in the N-terminus of Man6 and Man11 was a predicted signal peptide. Both Man6 and Man11 were localized at the level of 94–95% in an intracellular fraction of recombinant E. coli cells. The enzymes hydrolyzed both locust bean gum and mannooligosaccharides, including mannotriose, mannotetraose, mannopentaose, and mannohexaose, forming mannobiose and mannotriose as predominant products. The optimal reaction conditions were 55°C and pH 6.0 for Man6, and 60°C and pH 5.5 for Man11. Man11 was more stable than Man6 at high temperatures.

Isolation and Characterization of a Novel Calcium/Calmodulin-Dependent Protein Kinase, AtCK, from Arabidopsis

  • Protein phosphorylation is one of the major mechanisms by which eukaryotic cells transduce extracellular signals into intracellular responses. Calcium/calmodulin ($Ca^{2+}/CaM$)-dependent protein phosphorylation has been implicated in various cellular processes, yet little is known about $Ca^{2+}/CaM$-dependent protein kinases (CaMKs) in plants. From an Arabidopsis expression library screen using a horseradish peroxidase-conjugated soybean calmodulin isoform (SCaM-1) as a probe, we isolated a full-length cDNA clone that encodes AtCK (Arabidopsis thaliana calcium/calmodulin-dependent protein kinase). The predicted structure of AtCK contains a serine/threonine protein kinase catalytic domain followed by a putative calmodulin-binding domain and a putative $Ca^{2+}$-binding domain. Recombinant AtCK was expressed in E. coli and bound to calmodulin in a $Ca^{2+}$-dependent manner. The ability of CaM to bind to AtCK was confirmed by gel mobility shift and competition assays. AtCK exhibited its highest levels of autophosphorylation in the presence of 3 mM $Mn^{2+}$. The phosphorylation of myelin basic protein (MBP) by AtCK was enhanced when AtCK was under the control of calcium-bound CaM, as previously observed for other $Ca^{2+}/CaM$-dependent protein kinases. In contrast to maize and tobacco CCaMKs (calcium and $Ca^{2+}/CaM$-dependent protein kinase), increasing the concentration of calmodulin to more than $3{\mu}M$ suppressed the phosphorylation activity of AtCK. Taken together our results indicate that AtCK is a novel Arabidopsis $Ca^{2+}/CaM$-dependent protein kinase which is presumably involved in CaM-mediated signaling.
