Effects of Isoflavone Supplementation on Serum Lipids in Hyperlipidemic Postmenopausal Women (일부 고지혈증 폐경 여성의 Isoflavone 공급에 따른 혈중지질 변화에 관한 연구)

  • Intake of soy protein the fisk factors associated with cardiovascular disease in postmenopausal women. This study was designed to effects of isoflavone supplementation on serum lipids in 16 hyperlipidemic postmenopausal women . For this purpose, an intervention study was conducted for 12 weeks. Subjects were healthy, free-living women consuming habitual diets with 0.3g/d of isoflavone. Food and nutrient intake was obtained by 24-hr recall method and anthropometric measurement were made. Systolic and diastolic blood pressure, total serum cholesterol. HDL-cholesterol and LDL-cholesterol were determined before and after the isoflavone supplementation. The results were summarized as follows. The average age, hight, weight and BMI of the subject were 65.3 years, 151.4 cm, 62.2 kg and 27.1, respectively. The systolic blood pressure and diastolic blood pressure were not reduced significantly with isoflavone supplementation. Total cholesterol (p<0.001), HDL-C(p<0.05), and LDL-C(p<0.01) were significantly increased after isoflavone concentration. In conclusion, isoflavone supplementation was not effective to modify risk factors for cardiovascular disease.

  • Few studies have conducted the bone health benefits of usual dietary isoflavone intake in Korean college-student women. To elucidate this benefitial effects and correlations between dietary isoflavone and nutrients intake and bone formation marker (ALPase, osteocalcin), questionnaires, anthropometric measurements, serum mineral (Ca, P, Mg) concentrations were analyzed. Fifty three subjects were used in this study. The average age, height, body weight, BMI, body fat content and triceps skinfold thickness were 21.43 year, 161.07 cm, 52.81 kg, 20.48 kg/$m^2$, 20.72% and 17.59 mm respectively. Soy food intake frequencies were soybean > soybean curd > soypaste stew > soybean milk. The average calorie, protein and Ca intake were 1766.21 $\pm62.54 kcal\;66.45\pm2.00 g\;and\; 549.62 \pm 27.55 mg$ respectively. Serum ALPase activity and osteocalcin concentration were 115.74$\pm3.6u/L$ and 7.15 ng/$m\\ell$ respectively. Usual dietary isoflavone intake was positively related to calory, protein, Ca, P intake and serum Ca, Mg concentration. Serum osteocalcin concentration was negatively correlated with isoflavone intake ($r^2$=0.28, p < 0.05). In these results, usual dietary isoflavone can support an additive effect to bone health and Ca nutrition.

Genetic Diversity of Soybean Landraces in Korea

  • To evaluate the genetic diversity and structure of the South Korean soybean population, 233 landraces collected in various regions of the country were surveyed for 15 allozyme loci and one protein locus. The South Korean population was fixed or nearly fixed at seven of the 16 loci tested. The number of alleles per locus was 2.06 and Nei’s gene diversity was 0.194. These values were lower than the values for the same 16 loci previously reported for the Japanese and Chinese populations. The differences among eight regional groups were not so marked, with only 7.2% of the total variation arising from regional differentiation. Three southern regional groups (Chollabuk-do, Chollanam-do and Kyong-sangnam-do) exhibited a relatively high variability because of frequent occurrence of alleles characteristic of the Japanese population. A marked difference was found in allelic frequencies at the Dial locus between large-seeded landraces and small-seeded ones, suggesting that the latter, which are used mainly for bean sprouts, had been established independently of the former, which are used mostly for soy sauce and cooking with rice. Not only the region but also the usage as food materials should therefore be taken into consideration in designing an efficient collection and preservation method for the Korean soybean landraces.

Properties of Bac W42, a Bacteriocin Produced by Bacillus subtilis W42 Isolated from Cheonggukjang

  • Ten Bacillus strains with antimicrobial activities were isolated from Cheonggukjang produced at different parts in Korea. They all inhibited Listeria monocytogenes ATCC 19111 and nine inhibited Bacillus cereus ATCC 14579. Four isolates (W42, H27, SKE 12, and K21) showing strong inhibiting activities were identified as B. subtilis. B. subtilis W42 was the most inhibiting strain. The antimicrobial activity of culture supernatant from B. subtilis W42 was destroyed completely by proteinase K treatment, indicating that a bacteriocin was the responsible agent. The bacteriocin, Bac W42, was most stable at pH 7 and stable between pH 3-6 and 8-9. Bac W42 was stable up to $80^{\circ}C$. BHI (brain heart infusion) and TSB (tryptic soy broth) were the best media for the activity (320 AU/ml) followed by LB (160 AU/ml). Bac W42 was partially purified by column chromatographies. The specific activity was increased from 1,151.2 AU/ml to 9,043.5 AU/ml and the final yield was 26.3%. Bac W42 was 5.4 kDa in size as determined by SDS-PAGE. Bac W42 showed bactericidal activity against L. monocytogenes ATCC 19111.

  • The development of an enrichment method for the rapid and effective identification of Salmonella spp. in sewage or food was studied. As a growth factor for Salmonella, 10 mM cyclic adenosine monophosphate (cAMP) in trypticase soy broth with 0.6% yeast extract (TSBYE) increased cell number five-folds and 0.6% yeast extract in selenite broth increased cell number ten-folds of control. Bile salts in selenite broth was tested for the selection of S. enteritidis in a mixture with Staphylococcus aureus, Pseudomonas aeruginosa, Lactobacillus plantarum and Escherichia coli. The latter four strains were effectively inhibited at 0.1% bile salt. A two-step culture method was used to enrich Salmonella spp.; a primary-enrichment and secondary- enrichment culture. At a primary-enrichment step, selenite broth with 0.6% yeast extract and 10 mM cAMP was used, and at a secondary-enrichment step, 0.1% bile salt was additionally used. Culture times of a primary- enrichment and a secondary-enrichment step were 8 hr and 6 hr, respectively. In this procedure, cell number increased from 10$^{0.3}$ to 10$^{8.5}$ with inhibition of other strains within 14 hr. In the case of an initial cell concentrarion as low as 10$^{-2}$ cfu/ml, a cell number increased to 10$^{7}$ cfu/ml by using a 10 hr primary-enrichment and 6 hr secondary-enrichment procedure.

Recipe Standardization of Native Local Foods in Gijang Region(The First Report) - Myeolchijjigae, Galchijjigae, Maejipjjim - (기장 향토음식의 조리표준화(제1보) - 멸치찌개, 갈치찌개, 매집찜 -)

  • The purpose of this study was to contribute to merchandising of native local foods. This study was designed to standardize the recipes and to analyze the nutrients of native local foods in Gijang region such as 'Myeolchijjigae', 'Galchijjigae' and 'Maejipjjim'. The test recipe for each food was prepared according to the information obtained from the personal interview of Gijang natives and then supplied for the sensory evaluation. After that, CAN Pro 2.0 was used for the nutritional evaluation. The results are as follows : Myeolchijjigae was preferred when radishes and traditional soybean paste were added. Galchijjigae was more liked when young pumpkins, mixed soy sauce and soybean paste were added. Maejipjjim was preferred with Aeji(Codium adhaerens), Warty sea squirt(Styela clava), soybean paste and powdered perilla seed(Perilla frutescens var. japonica) added. The analysis of nutrient composition revealed that in general energy content was low whereas protein content was as high as recommended requirement in Myeolchijjigae and Maejipjjim. Calcium and phosphorus were high in Myeol-chijjigae, vitamin A in Galchijjigae, and vitamin A, B_6$, folic acid, phosphorus and iron in Maejipjjim.

Changes of Soyasaponin Contents in Soybean Sprouts (콩나물 생육기간 중 사포닌 함량의 변화)

  • Soyasaponin I, II, III and V contents were investigated in seed, cotyledons and sprouts of soybean (Glycine max (L.) Merill) subjected to germination over five days. High-performance liquid chromatography coupled with tandem mass spectrometric (HPLC-MS/MS) method was used to evaluate the content of soyasaponins. Soyasaponins contents were different according to the varieties. Germination of soybeans dramatically increased soyasaponin contents in soybean sprouts in a time-dependent manner. Cotyledons had a higher contents of soyasaponins compared to dried seed (p<0.05). After five days of germination, Soyasaponin I and II increased 10 times higher after germination. Soyasaponin I and II are major metabolites in cotyledons and hypocotyls. Soyasaponin III and V were also detected in seed and increased depended on the germination stage. Soyasaponin V was at its highest levels in the hypocotyl, almost 7 times higher than the initial content in soybean seeds. Therefore, the germination of soybean sprouts significantly increased soyasaponin content.

Production and Characterization of a New ${\alpha}$-Glucosidase Inhibitory Peptide from Aspergillus oryzae N159-1

  • An ${\alpha}$-glucosidase inhibitor was developed from Aspergillus oryzae N159-1, which was screened from traditional fermented Korean foods. The intracellular concentration of the inhibitor reached its highest level when the fungus was cultured in tryptic soy broth medium at $27^{\circ}C$ for five days. The inhibitor was purified using a series of purification steps involving ultrafiltration, Sephadex G-25 gel permeation chromatography, strong cation exchange solid phase extraction, reverse-phase high performance liquid chromatography, and size exclusion chromatography. The final yield of the purification was 1.9%. Results of the liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis indicated that the purified ${\alpha}$-glucosidase inhibitor was a tri-peptide, Pro-Phe-Pro, with the molecular weight of 360.1 Da. The IC50 value of the peptide against ${\alpha}$-glucosidase activity was 3.1 mg/mL. Using Lineweaver-Burk plot analysis, the inhibition pattern indicated that the inhibitor acts as a mixed type inhibitor.

Screening and Identification of a Streptomyces platensis YK-2, a New Transglutaminase Producer

  • A bacterial strain, YK-2, was isolated as a producer of trans glutaminase from a forest soil sample of Daegu, Korea. The isolate showed a G+C content of 72.7 mol%, contained meso-$A_2pm$ as the cell-wall amino acid, and possessed menaquinone MK-9 ($H_6$) and menaquinone MK-9 ($H_8$) at a ratio of 6:4. The chemotaxonomic analysis, as well as phylogenetic analysis based on the 16S rDNA sequence, identified the isolate as a member of Streptomyces platensis. For transglutaminase production, the optimum medium composition was determined to be 2% glucose, 1% polypeptone, 1% soy tone, and 0.1% $MnCl_2$. The transglutaminase was stable within the pH range of 5.0-9.0 and $30-45^{\circ}C$, and the optimum pH and temperature were pH 8.0 and $45^{\circ}C$, respectively, without any requirement for $Ca^{2+}$.

  • The purpose of this study is to obtain the basic information for developing new product and marketing strategies of salted wild vegetable. This study analyzes consumer's consumption behavior and preference on salted Allium victorialis using the conjoint analysis. The result shows that 'Container' is the most important factor among various attributes of salted Allium victorialis, followed by 'Origin of Salting Sauce Soy', 'Price', 'Traditional Food Quality Certification', and 'Area of production'.
