Biofilters for the removal of methane using tobermolite, perlite and polyurethane as packing materials have been undergoing recent development. The effects of these packing materials on methane oxidation activity were evaluated in this study. Mixed methanotrophs (consortia A, B, C and D) from wetland and landfill soils were used as the inoculum sources. The influences of packing materials, consisting of tobermolite, perlite, and polyurethane, on the methane oxidation rate and methanotrophic bio-mass, were estimated. When perlite was added into the methanotrophic cultures, the methane oxidation rate was more than twice that of the control (without packing materials), and the methanotrophic biomass increased more than 10 fold. The ratio of methanotrophic bacteria to total bacteria under with tobermolite packing material was higher than the control and the other packing materials, indicating that tobermolite can serve as a specific packing material where dominance of methanotrophs is desired. Therefore, perlite and tobermolite provide habitats which increase the activity of methanotrophic bacteria, and these packing materials are promising for use in methane oxidation processes.
In Western countries, kimchi, the Korean traditional fermented cabbage, is considered to be a healthy. However, it is one of the main sources of the high sodium content of the Korean diet. In order to decrease the sodium content, we manufactured a low-sodium kimchi (LK, salinity 1.0%) and 4 additional low-sodium kimchi starters in which each of 4 lactic acid bacteria (Lb. sakei 1, Lb. sakei 2, Lb. palntarum and W. koreensis) were added. The LKL1 to LKL4 samples were prepared by adding 4 single LAB starters, each with an inoculum size of $10^6CFU/g$, when the cabbage was mixed with kimchi sauce. The kimchi starters were fermented at $10^{\circ}C$ until reaching 0.5% acidity, and then stored at $-1.5^{\circ}C$ until reaching 0.75% acidity. The pH and acidity of the starter kimchi changed more rapidly in the early phase of fermentation (up to 0.75% acidity) than control low-sodium kimchi. After the acidity of the kimchi starters reached 0.75% it remained constant. As the fermentation progressed, the total aerobic and lactic acid bacteria concentrations in the kimchi starter with added Lb. sakei 1 were the same as in the control low-sodium kimchi. The low-sodium kimchi fermentation of the kimchi starter with added Lb. palntarum progressed differently due to a difference in acid resistance. The kimchi starter with added Lb. sakei 2 had an overall liking score that was slightly higher than that of the control low-sodium kimchi due to a lower off-flavor.
Three levels of inoculum concentration from 10 to 30 percent, three kinds of adhesive materials, gum arabic, methyl cellulose and carboxy methyl cellulose, and five different pelleting materials including 4 different sources of lime and calcium carbonate were compared to investigate an optimum condition for seed inoculation by counting the number of viable rhizobium cells. For a peat-cultured Rhizobium inoculant, 10 per cent was found to be an optimum by showing 3.5 $\times$ 10$^9$ viable cells per seed. The highest number of viable cells were observed from gum arabic at 40 per cent, methyl cellulose at 5 per cent and carboxy methyl cellulose at 4 per cent. Among pelleting materials, a dental lime for investment originated from Ransom & Randolph Co. Ohio, U.S.A. resulted best as pelleting material.
Alternative hosts increase the difficulty of disease management in crops because these alternate hosts provide additional sources of primary inoculum or refuges for diversity in the pathogen gene pool. Agropyron cristatum (crested wheatgrass), Bromus inermis (smooth bromegrass), Pascopyrum smithii (western wheatgrass), Stipa viridula (green needlegrass), and Thinopyrum intermedium (intermediate wheatgrass), commonly identified in range, prairie, verge, and soil reclamation habitats, serve as additional hosts for Pyrenophora tritici-repentis, the cause of tan spot in wheat (Triticum aestivum L.). A. cristatum (five lines), B. inermis (seven lines), P. smithii (four lines), S. viridula (two lines), and T. intermedium (six lines) were tested for their reactions to 30 representative P. tritici-repentis isolates from races 1-5. Plants were grown until the two-three-leaf stage in a greenhouse, inoculated individually with the 30 isolates, held at high humidity for 24 h, and rated after 7 days. All lines developed lesion types 1-2 (resistant) based on a 1-5 rating scale. Also, leaves from an additional plant set were infiltrated with two host selective toxins, Ptr ToxA as a pure preparation and Ptr ToxB as a dilute crude culture filtrate. All lines were insensitive to the toxins. Results indicate that these grass hosts have a limited or nonsignificant role in tan spot epidemiology on wheat in the northern Great Plains. Additionally, the resistant reactions demonstrated by the grass species in this research indicate the presence of resistance genes that can be valuable to wheat breeding programs for improving wheat resistance to P. tritici-repentis.
In the year of 2002 annual nationwide survey of virus diseases occurring in the pepper fields and greenhouses in Korea, the distribution and the incidence of viral diseases was investigated. The pepper samples from both greenhouses (155 samples) and open fields (227 samples) were collected and further analyzed to detect eleven different viruses by RT-PCR. The results indicate that no sample collected from both greenhouse and open field seems to be infected by TMV, RMV, PVY, AMV, and TSWV. On the other hand, CMV, BBWV2, PepMoV, PMMoV, TMGMV and ToMV are readily identified from greenhouse and open field samples by RT-PCR. The infection rates of the collected samples between greenhouse and open field are largely different. Comparing with 10% of virus-infected pepper samples grown in greenhouse, approximately one third of pepper samples collected from open field are infected. The mixed-infection rates in the virus-infected greenhouse and open field samples are 16% and 61%, respectively. The dominant virus occuring in greenhouse is PMMoV, indicating that virus-infected seed stocks and infected plant debris in the growing area may be important sources of inocula. On the other hand, both CMV and BBWV2 are dominant viruses in open field. This may indicate that the migration of viruliferous insect vectors into pepper fields may be the most important source of inoculum. Also, the survey shows that BBWV2 is newly immerging virus to be controlled in Korea. The discrepancies on the distribution and the occurrence of viral diseases between field and greenhouse may provide a fact that the accumulation and distribution of inoculum by successive cultivation and the migration of viruliferous vectors into growing areas are likely to be important factors to determine the incidence of viral diseases. Therefore, the further studies on epidemiology and the consideration of new breeding program of pepper are essential to minimize virus diseases.
The present study was carried out to investigate morphological characteristics of pseudosclerotia of Grifola umbellata formed by artificial cultures. Isolate G. umbellata DUM GUS-01 was obtained from sclerotium cultivated in field. The fungal isolate was cultured on PDYM broth, PDYMA(potato dextrose yeast malt agar) and oak sawdust media at $20^{\circ}C$ under the dark condition. G. umbellata DUM GUS-01 showed a volumetric increment of fungal lumps rather than mycelial growth. Particularly, G. umbellata DUM GUS-01 produced a large amount of melanin pigments in all culture treatments. The color of the fungal mass has been changed into grey gradually, and then formed melanized rind-like structure on its superficial part. The fungal structures which were covered with melanized rind-like layer were named as pseudosclerotia of G. umbellata. The pseudosclerotia of G. umbellata DUM GUS-01 formed a new white mycelial mass, which was swollen out of the melanized rind structure for its volumetric increment. When the pseudosclerotia were sectioned, their structure was discriminated from two structures such as a melanized rind-like structure layer formed by aggregation of aged mycelia and a white mycelial mass with high density. As results of scanning electron microscopic examination, the pseudosclerotia of G. umbellata DUM GUS-01 which were formed in in vitro conditions were similar to the sclerotia of G. umbellata cultivated in natural conditions except for the crystals formed in medula layer of natural sclerotia. Although size, solidity of rind structure and mycelial compactness of pseudosclerotia were more poor than those of natural sclerotia, the morphological structure and growth pattern of pseudosclerotia were very similar to those of natural sclerotia. Therefore, it is probable to induce pseudosclerotia to sclerotia of G. umbellata in in vitro conditions. Consequently, it seems that the induced pseudosclerotia can be used as inoculum sources to substitute natural sclerotia in field cultivation.
Present studies were conducted to evaluate the effects of medium strength(MS and Hyponex), carbon sources and their concentrations, agar concentrations, and inoculation amounts on prothallus propagation of Pterdium aquilinum var. latiusculum(Desv.) Underw. ex Hell in vitro. The optimum MS medium strength for prothallus propagation was 2MS concentration. Phosphate source was most effective for prothallus growth of P. aquilinum var. latisculum. The addition of 1% sucrose or glucose to MS medium promoted prothallus multiplication. Growth of prothallus was not affected by agar concentration. Propagation of homogenized prothallus was vigorous even in liquid medium. Chopped gametophytes(100 and 200 mg) were inoculated on 250 ml ${\Delta}$flask with 100 mL of 2MS concentration medium and suspension culture was done at 100 rpm for 22 days. After 20 days, prothallus multiplication slowed down, so 100 mg of chopped prothalli is recommended for initial inoculation, since initial amount of inoculum did not affect subsequent prothallus multiplication. Consequently after 20 days of suspension culture, prothallus should be subcultured or transplanted outside of growing vessels.

Changes in spectroscopic characteristics and pyrene binding coefficients of terrestrial dissolved organic matters(DOM) were investigated during microbial incubation. The incubation studies were conducted for 21 days using a leaf litter DOM and a soilderived DOM with an inoculum from a river. The dissolved organic carbon(DOC), the specific UV absorbance(SUVA), the synchronous fluorescence spectra, and the pyrene organic carbon-normalized binding coefficient(K$_{oc}$) of the DOM were measured at the incubation days of 0, 3, 7, 14 and 21. After the 21-day incubation, DOC were reduced to 61% and 51% of the original concentrations of the litter DOM and the soil-derived DOM, respectively. Comparison of the spectroscopic characteristics before and after the incubation revealed that the SUVA, the fulvic-like fluorescence(FLF), the humic-like fluorescence(HLF) of the different DOM were enhanced by the incubation whereas the protein-like fluorescence(PLF) was reduced. This indicates that more aromatic and humic-like compounds were enriched during the biodegradation process while biodegradable and weak carbon structures were depleted. Irrespective of the DOM sources, SUVA values showed a positive relationship with pyrene K$_{oc}$ with a correlation coefficient of 0.97. The FLF and HLF also exhibited good correlations with K$_{oc}$ values although different regression equations were obtained from the different DOM. Our results suggest that the selected spectroscopic characteristics could be good estimation indices for the changes of the binding reactivity of DOM for hydrophobic organic contaminants during biodegradation process.

CH4-oxidizing and N2O-reducing bacterial consortia were enriched from the rhizosphere soils of maize (Zea mays) and tall fescue (Festuca arundinacea). Illumina MiSeq sequencing analysis was performed to comparatively analyze the bacterial communities of the consortia with those of the rhizosphere soils. Additionally, the effect of root exudate on CH4 oxidation and N2O reduction activities of the microbes was evaluated. Although the inoculum sources varied, the CH4-oxidizing and N2O-reducing consortia derived from maize and tall fescue were similar. The predominant methanotrophs in the CH4-oxidizing consortia were Methylosarcina, Methylococcus, and Methylocystis. Among the N2O-reducing consortia, the representative N2O-reducing bacteria were Cloacibacterium, Azonexus, and Klebsiella. The N2O reduction rate of the N2O-reducing consortium from maize rhizosphere and tall fescue rhizosphere increased by 1.6 and 2.7 times with the addition of maize and tall fescue root exudates, respectively. The CH4 oxidization activity of the CH4-oxidizing consortia did not increase with the addition of root exudates. The CH4-oxidizing and N2O-reducing consortia can be used as promising bioresources to mitigate non-CO2 greenhouse gas emissions during remediation of oil-contaminated soils.
The optimum culture conditions for tropane alkaloid production in hairy root cultures of Korea native Scopolia paviflora Nak. were investigated. Hairy root was induced from the rhizome of the mother plant on B5 medium containing 1.0 mg/L IBA. Among the culture media examined, 1/2 B5 medium was the best for tropane alkaloid production, whereas the growth of hairy root increased in SH medium. The best result on the growth of hairy root was obtained in 1.0 mg/L NAA, and tropane alkaloid production was obtained in plant growth regulator-free medium. Of the carbone sources tested, 3% sucrose promoted the growth of hairy root, whereas 5% sucrose increased tropane alkaloid production. Optimum inoculum densities for root growth and tropane alkaloid production were 0.5 g and 1 g, respectively. The addition of XAD resins (1 % w/v) to hairy root cultures led to increases in tropans alkaloid production, and the release of alkaloid into the medium and its adsorption by the resin accounted for about 50 to 80% of total production. It is concluded that optimized culture conditions and the addition of XAD resins could be used in the development of a bioprocess for tropane alkaloid production in hairy root cultures of S. paviflora Nak.
