Characteristics of the antibacterial substances produced by Lactobacillus casei subsp. and Streptococcus faecium (Lactobacillus casei subsp. 및 Streptococcus faecium이 생산한 항균성물질의 성상)

  • Antibacterial substances produced by Lactobacillus casei subsp. and Streptococcus faecium were examined for its antibacterial effects against some pathogenic bacteria. They were partially purified with ammonium sulfate precipitation, methanol-acetone extraction, G-50 gel filtration and examined its characteristics. When L casei subsp. and Str faecium were cultivated in MRS broth, stationary phase of L casei is until 24 hours and Str faecium is 20 hours. pH change of the cultured medium was both decreased after 12 hours and then constant at pH 4.5~4.6 after 28 hours. MRS broth culture fluids of L casei subsp. and Str faecium appeared the antibacterial effects by the spot-on-the-lawn method against ETEC, Sal pullorum and Sta aureus. Culture filtrates of L casei subsp. and Str faecium also appeared the antibacterial effects by the disc diffusion method. Culture filtrates of L casei sub. rhamnosus 7469 produced 0.032M of lactic acid and 0.01M of acetic acid. Str faecium 27273 also produced 0.027M of lactic acid and 0.01M of acetic acid. Protein concentrations of culture filtrates produced by L casei sub rhamnosus 7469 and Str faecium 27273 was $495{\mu}g/m{\ell}$ and $594{\mu}g/m{\ell}$, respectively. Antibacterial substances which are partially purified by ammonum sulfate precipitation, methanol-acetone extraction and G-50 gel filtration inhibit the growth of ETEC, Sal pullorum and Sta aureus. Characteristics of purified antibacterial substances was examined. Its molecular weight was about 31Kd, stabilized at $100^{\circ}C/20min.$ and some of proteolytic enzyme treatment.

Correlation between Serum Leptin Levels and BMI in Adults Residing in Pohang, Korea

  • Leptin is a small polypeptide hormone secreted primarily by adipocytes. Leptin regulates energy balance by decreasing food intake and increasing energy expenditure. This study investigated the relationships between serum leptin levels and BMI (body mass index) in 49 adults in Pohang, Korea. The subjects were 25 males and 24 females, aged 21 to 64 years attending an outpatient clinic at Handong University Sunlin Presbyterian Hospital. Values are given +/- the standard error of the mean. Our study shows that the serum leptin levels in these subjects were positively correlated with BMI. The leptin levels were higher in females (2.39+/-1.82 ng/mL) than in males (0.43+/-0.455ng/mL), although lower than previously reported. We therefore compared the serum leptin levels from the male Korean subjects (BMI 24.3+/-0.74㎏/㎡) with serum from six British males with a similar BMI (23.4+/-1.48㎏/㎡). The serum leptin concentrations (1.76+/-0.76 ng/mL) were lower than that of plasma (4.28+/-1.66 ng/mL) in the British subjects. The serum leptin in the British subjects (1.76+/-0.76ng/mL) was higher than that in the Koreans. There was no correlation between leptin levels and BMI in either male (slope 0.018 ± 0.036, p=0.624) or female (slope 0.382±0.433, p=0.417) type 2 diabetic patients in Pohang, Korea. Taken together, our study shows that the serum leptin level in Koreans varies with the BMI, but is lower than that of BMI-matched British subjects.

Expression of eotaxin in 3T3-L1 adipocytes and the effects of weight loss in high-fat diet induced obese mice

  • Eotaxin is an important inflammatory chemokine in eosinophil chemotaxis and activation and, thus, is implicated in asthma. Recently, obesity was associated with an increased prevalence of asthma, but the relationship between obesity and eotaxin expression has only been partially understood in obese mice and human studies. Therefore, we studied the expression patterns of eotaxin in 3T3-L1 preadipocytes/adipocytes to determine whether eotaxin levels are influenced by body weight gain and/or reduction in diet-induced obese mice. First, we investigated eotaxin expression during differentiation in 3T3-L1 adipocytes. Then, we treated 3T3-L1 preadipoeytes/adipoeytes with tumor necrosis factor-alpha (TNF-${\alpha}$), eotaxin, interleukin (IL)-4, IL-5, or leptin. To examine the effects of weight loss in high-fat diet induced obese mice, we fed C57BL/6 mice a high-fat diet or a normal diet for 26 weeks. Then, half of the high-fat diet group were fed a normal diet until 30 weeks to reduce weight. Epididymal adipose tissue, visceral adipose tissue, serum, and bronchoalveolar fluid of mice were examined for eotaxin expression. The results showed that eotaxin expression levels increased with adipocyte differentiation and that more eotaxin was expressed when the cells were stimulated with TNF-${\alpha}$, eotaxin, IL-4, IL-5, or leptin. An in vivo study showed that eotaxin levels were reduced in visceral adipose tissues when high-fat diet fed mice underwent weight loss. Taken together, these results indicate a close relationship between eotaxin expression and obesity as well as weight loss, thus, they indirectly show a relation to asthma.

Identification of Lactobacillus ruminus SPM0211 Isolated from Healthy Koreans and Its Antimicrobial Activity against Some Pathogens

  • The intestinal microbiota are important to the host with regard to resistance they impart against bacterial infections and their involvement in mediating metabolic functions. Lactic acid producing bacteria such as Lactobacillus play an important physiological role in these matters. The aim of the present study was to isolate Lactobacillus sp. that inhibits enteric pathogens. Initially, 17 isolates from healthy Koreans were collected on Lactobacillus selective medium. Resistance of the isolates to antibiotics including rifampicin, streptomycin, clindamycin and vancomycin was measured. One of the isolate was identified as Lactobacillus ruminus on the basis of bacterial cell morphology, cultural characteristic and biochemical characteristics, 16S rRNA sequence analysis and PCR-RAPD. Antimicrobial activity of the bacterium against Vancomycin Intermediate Resistant Staphylococcus aureus (VISA) and Vancomycin-Resistant Enterococci (VRE) was measured. About $10^4$ cells of VISA or VRE were mixed with 1, 5, and 9 mL of L. ruminus SPM 0211 and the final volume was adjusted to 10 mL with brain heart infusion (BHI) broth. The cell suspension was incubated for 3, 6, 9, and 24 h, serially diluted and then plated on BHI agar plates. As numbers of L. ruminus SPM 0211 were increased, viable cell count of VISA and VRE decreased. The strongest antimicrobial activity of SPM 0211 was observed after 9 h incubation in any mixture, almost completely inhibiting the growth of these two bacteria. The results suggest that the freshly isolated L. ruminus SPM 0211 may be used as a pro-biotic microbe that prevents the colonization of enteric pathogens and can thereby promote good gastrointestinal health.

Immunomodulatory Activity of Betulinic Acid by Producing Pro-Inflammatory Cytokines and Activation of Macrophages

  • Betulinic acid (BA), a pentacyclic triterpene isolated from Lycopus lucidus, has been reported to be a selective inducer of apoptosis in various human cancer and shown anti-inflammatory and immunomodulatory properties. We postulated that BA modulates the immunomodulatory properties at least two groups of protein mediators of inflammation, interlukin-1$\beta$ (IL-1$\beta$) and the tumor necrosis factor- $\alpha$ (TNF-$\alpha$) on the basis of the critical role of the monocytes and tissue macrophages in inflammatory and immune responses. TNF-$\alpha$ and IL-1$\beta$ were produced by BA in a dose dependent manner at concentration of 0.625 and 10 $\mu$g/mL. The production of NO associated with iNOS was inhibited when treated with LPS at the concentration of 2.5 to 20 $\mu$g/mL of BA whereas COX-2 expression was decreased at 2.5 to 20 $\mu$g/mL. These modulations of inflammatory mediators were examined in LPS-stimulated RAW 264.7 cells and peritoneal macrophages. The morphology of macrophage was also examined and enhanced surface CD 40 molecule was expressed when treated BA at 0.625∼5 $\mu$g/mL with or without LPS. Furthermore, BA (20 $\mu$g/mL) enhanced apoptosis by producing DNA ladder in the RAW 264.7 cells. Our results indicated that BA induced activation of macrophage and pro-inflammatory cytokines. This may provide a molecular basis for the ability of BA to mediate macrophage, suppress inflammation, and modulate the immune response.

Quality Control(QC) Analysis of Ambient VOCs in Canisters Using GC/MSD Method (Canister와 GC/MSD를 이용한 대기 중 VOCs 분석시스템의 정도관리(QC))

  • In this study, the performance of sampling and of analytical methodology was examined against a number of selected volatile organic compounds (VOCs) in the ambient air. The canister-based sampling method for VOCs has been a viable and widely used approach that is based on research and evaluation performed over the past several years. VOCs were determined using canisters by GCMSD analysis. A total of target analyses in this study included approximately 30 VOCs designated in TO-l4A categories including BTEX. The methodology for QC(quality control) was intensively investigated with a wide range of performance criteria such as; (1) cleanliness of canisters, (2) reproducibility of diluter, (3) instrumental detection limits of preconcentrator and GCMSD, and (4) precision and accuracy of GCMSD. For most of the compounds tested, precision was less than $\pm$5%, IDL was 0.2 ppbv, accuracy was $\pm$5%, respectively. This study demonstrated that the combination of canister sampling and GC/MSD analysis could be reliably applied to the measurements of BTEX in ppbv levels that are typically observed in/outdoor air environment.

Three Freshwater Lembadion Ciliates (Peniculida, Lembadionidae) Newly Reported from Korea

  • Three freshwater ciliates, Lembadion lucens (Maskell, 1887) Kahl, 1931, L. bullinum ($M{\ddot{u}}ller$, 1786) Perty, 1849, and L. magnum (Stokes, 1887) Kahl, 1931, belonging to the genus Lembadion Perty, 1849 are newly reported from Korea with detailed descriptions and illustrations based on live observation and protargol impregnation. Lembadion lucens is characterized by following characteristics: $52-91{\times}33-64{\mu}m$ body size in vivo; ovoid and slightly asymmetric body shape; 27-49 somatic kineties; and 55-80% of whole pellicle covered with meridional lines with rectangular meshes in posterior portion. Lembadion bullinum is distinguishable from its congeners by its body size of $110-130{\times}60-73{\mu}m$ in vivo; ovoid body shape; oral aperture occupying about four-fifths of ventral side; 48-55 somatic ciliary rows; meridional lines with rectangular meshes except anterior portion (about 16% of body) on pellicle. Lembadion magnum is also characterized by its body size of $70-90{\times}37-50{\mu}m$ in vivo; obliquely asymmetric anterior end of body; oral aperture occupying about more than four-fifths of ventral surface, and opened longitudinally entire ventral surface; 45-61 somatic ciliary rows; meridional lines without rectangular meshes on whole pellicle of body. The present study is the first description of the genus Lembadion from Korea.

Current Status and Prospect of Antiobesity Functional Agents

  • The obese population has been increasing over the world wide and obesity became a socioeconomic problems. It is become more serious by the accumulation of the knowledge that the obesity is related directly or indirectly with several diseases like, diabetes, hypertension, etc. With these reasons, many functional food or agents for the purpose of weight loss have been developed. However, most of these remedies are unproven and some have produced even dangerous side effects due to the ephedrine alkaloids contained in Ma-Hang. Because of these reasons, they banned using of these agents in US and regards the antiobesity functional agents as drugs in Europe. Several functional agents are known for weight loss activities like, HCA, L-canitine, CLA, chitosan, calcium supplements and capsaicin containing red pepper, kimchi and kochujang. We describe here about the function, efficacy and mechanism of these antiobesity functional agents. Furthermore, the trial of the mixture of weight loss related herbal ingredients for safe multifunctional antiobesity functional agents are discussed here, as well.

FTIR study of E7 liquid crystals confined to cylindrical cavities of Anodisc membranes

  • Orientation of E7 liquid crystals (LCs) confined to 200 nm-diameter cylindrical cavities of Anodisc membranes are investigated by FTIR dichroism techniques. The cavity walls of the confining pores are chemically modified with different aliphatic acids ($C_nH_{2n+1}COOH$, n=5, 6, 7, 9). From the FTIR spectra of aliphatic acid treated alumina Anodsic membranes, we found the salt formation between -COOH group of aliphatic acid and Anodisc membrane. From the FTIR spectra of LC filled Anodisc membranes, we found abrupt alignment direction change of LC molecules between n=6 and 7 for 2% aliphatic acid treated Anodisc membranes, from parallel to perpendicular direction to the cavity walls. But 4% aliphatic acid treated Anodisc membranes, alignment direction of LC molecules changed between n=5 and n=6, from parallel to perpendicular direction. The same trend was observed for $^2H-NMR$ measurements.

  • The plant of Pedicularis resupinata L. var. oppositifolia (Scrophulariaceae) appeared to be used for the treatment of rheumatoid arthritis, malignant abscess (tumor), urolith, and diuretics in oriental medicinal literatures. Three different compounds were isolated from the aerial part of the plant and characterized by the spectroscopic (UV, IR, NMR, MS) analysis. These compounds were acteoside (compound I), suavissmoside R1 (compound II), and ${_D}-mannitol$ (compound III).
