Conditions for high frequency plant regeneration from cryopreserved embryogenic cell suspension cultures derived from hypocotyl explants of cucumber (Cucumis sativus L.) are described. Cells cryoprotected with a mixture of 2 M DMSO and 0.4 M sucrose exhibited a regeneration frequency of 85%. However, cells cryoprotected with different concentrations of glycerol showed no regeneration after cryopreservation. Pretreatment of cells in a high osmotic medium was not necessary to the process. Upon transfer to MS medium supplemented with 1 mg/L 2,4-dichlorophenoxyacetic acid, regenerated calli gave rise to numerous somatic embryos, then underwent development into plantlets.
This study was conducted to investigate the possibility of obtaining platelets via somatic embryogenesis as a means of in vivo mass propagation in Paeonia lactiflora Pall.. When cultured on MS medium with 0.5 mg/L 2, 4-D, filament explants formed calli. UPon transfer to basal medium the calli gave rise to somatic embryos at a frequency of 38%. Additional of 3g/L activated charcoal enhanced the frequency to 60%. Mature embryos preheated at 5$^{\circ}C$ for 2 weeks had 37% germination on medium with 0.3 mg/L GA$_3$. The germinated embryos developed to complete plantlet when cultured on medium with 2mg/L BA.
This study was carried out to establish a mass production of normal somatic embryos of Oenanthe iavanica (BL.) DC. including examination of nitrogen and sucrose sources, and ABA concentration. Embryogenic cell clumps and embryos were formed on the MS medium devoid of growth regulators. Proliferation of embryogenic cells and clumps was enhanced by 2, 4-D. Meanwhile embryo growth and development occurred on the media containing NAA and IBA. Growth of embryos was generally good in the media containing both 20 mM $KNO_3$ and 20 mM $NH_4NO_3$. The rate of shoot forming embryos was higher on the media containing on1y 20mM $NH_4NO_3$ than on the former. Addition of sucrose at 3-6% enhanced the embryo development, and normal embryos with short hypocotyl was observed on the medium containing $10\mu\textrm{M}$ ABA. Embryogenic cell clumps or globular embryos, when transferred to MS solid media devoid of growth regulators, developed into mature embryos and then into plantlets which had entire primary leaves like zygotic seedlings.
The somatic embryos of Daucus carota L. cv. Hongshim developed in MS basal medium far 4 weeks had varied number of cotyledons. Palisade and spongy parenchyma of cotyledon were not clearly discriminated in all seedlings developed from the embryos. No independent existence of collateral vascular bundle was observed in all seedlings with various types of cotyledon ; instead, vascular bundles were either interconnected or partially connected with one another. Most of the cotyledonary bases on hypocotyl showed short cylinder structure which encircle plumule. The vascular tissues of cotyledonary bases and nodes of seedlings with jar-shaped or 1 cotyledon were connected in ring forms, showing the pattern of ectophloic shiphonostele, and similar ring form structure was also found in the vascular arrangement of 5 cotyledon seedlings. The vascular bundles of seedlings with 2, 3 and 4 cotyledons in many cases had independently arranged within the cotyledonary bases and nodes, showing the pattern of eustele. In hypocotyl, tetrarch or hexarch xylems prevailed in seedlings with jar-shaped cotyledon or 1 and 5 cotyledon; tetrarch xylems prevailed in 2 cotyledon seedlings; and triarch xylems prevailed in 3 cotyledon seedlings. In most of seedlings, cortex vascular bundles were dispensed in the region from cotyledonary node to hypocotyl, but double vascular bundles were also observed occasionally. In roots, diarch xylems were observed in most of seedlings with 2 cotyledons, triarch xylems in half of seedlings with 3 cotyledons, and diarch xylems in most of the remaining seedlings with the occasional occurrences of tetrarch xylems.

Somatic chromosome numbers for 10 taxa and karyotypes analysis for 6 taxa of Korean Vicia were investigated. Somatic chromosome numbers of treated taxa were 2n = 12, 14 or 24 and therefore they proved to be diploid or tetraploid with basic chromosome numbers of x = 6 or 7. The chromosome number of V. hirticalycina (2n = 2x = 12) was reported for the first time in this study. The chromosome numbers of nine taxa were the same as in previous studies; V. angustifolia (2n = 2x = 12), V. cracca (2n = 4x = 24), V. hirsuta (2n = 2x = 14), V. tetrasperma (2n = 2x = 14 + 2B), V. amurensis (2n = 2x = 12), V. chosenensis (2n = 2x = 12, 12 + 2B), V. unijuga (2n = 4x = 24), V. unijuga f. minor (2n = 4x = 24), V. venosa var. cuspidata (2n = 4x = 24). The karyotypes of V. cracca, V. amurensis, V. hirticalycina, V. unijuga, V. unijuga f. minor, V. venosa var. cuspidata were observed as 2 m + 8 sm + 2 st, 2 m + 2 sm + 2 st, 3 m + 1 sm + 2 st, 4 m + 6 sm + 2 st, 4 m + 6 sm + 2 st, 4 m + 8 sm, respectively.
This study was conducted to establish the optimal suspension culture system for both the propagation of embryogenic cells (ECs) and the induction of somatic embryos (SEs) of Kalopanax septemlobus. The proliferation rate of ECs was reduced as the inoculum density was increased; the highest rate was obtained when 0.1 g/100 ml of cells was initially inoculated. According to the analysis of cell growth pattern and cell growth cycle (G1, Sand G2/M), the cell growth started in 5 days culture initiation, grew rapidly until 15 days and then decreased gradually. Distinctive changes of the cell growth cycle by the culture periods was also observed; the growth cycle was doubled from initial 5.6% to 11.7% of S stage in 5 days culture and then reached in stable stages again. Therefore, the results indicated that a 15-day-cycle was the optimal culture period for the propagation of the ECs through the suspension culture. Furthermore, the cell inoculum density was also important for the induction of SE; more than 65% of SEs at the torpedo stage was induced by using the low level of cell inoculum (0.5 g/L), while the higher inoculum densities were rapidly reduced the proportion of SEs at that stage. Although the higher inoculum density delayed the development of SE, it did not affect the proportion of SEs at the globular and heart stage. In conclusion, this study showed that the suspension culture of the Kalopanax septemlobus ECs through the control of inoculum density was an efficient way for both the propagation of ECs and the induction of SEs, suggesting that the development of this system might help to reduce the culture period for the somatic embryo production.

This study was conducted to evaluate effects of various kinds or concentrations in abscisic acid (ABA), reduced nitrogen sources (casein hydrolysate, casamino acid and L-glutamine) and osmoticum for production of somatic embryos (SEs) from pro-embryogenic mass (PEM) in yellow poplar (Liriodendron tulipifera). In comparison of various concentrations of ABA, the highest number (640/10 mg PEM) of SEs was marked in the treatment of 0.5 mg/L. With higher concentration than 0.5 mg/L ABA, number of induced SEs were decreased. And the lowest number of SEs were obtained from the treatment of 20 mg/L ABA. Differences of 8 treatments of the nitrogen sources in the medium were also compared. In the experiment of 8 treatments for SEs production, the highest result showed in the treatment of 500 mg/L casamino acid (223/5 mg PEM). In comparison of different kinds/concentrations of osmotica for SEs induction, the best response was obtained from the treatment of 4% sucrose (317/5 mg PEM). In contrast, no SEs were found from the treatments supplemented with any concentrations of maltose.

High salt concentrations in the ginseng nursery soil environment of Korea is one of important reducing factors for the stable production of quality ginseng. These studies were accomplished for check the response on germination of ginseng seed, somatic embryogenesis of zygotic embryo, and biosynthesis of ginsenoside from ginseng hairy root against NaCl. Ratio of germination was at the $3\%\;and\;84.5\%$ on the basic media with 0.1M and free of NaCl repectedly, but $0\%$ at the upper of 0.2M NaCl. Somatic embryogenesis from zygotic embryo were the highest when immatured embryo was cultured on free of NaCl concentration, and which was intend to decrease at treatment of NaCl. However, in case of using the matured embryo, treatment of 0.05M NaCl resulted in better embryogenesis than NaCl free media. Red pigment was synthesized from ginseng hairy root cultured on the medium with various NaCl concentration(from 0.04 to 0.08M) and its pigment was analyzed as spectrum of anthocyane by spectrophoto- meter scanning. This cell line biosynthesized lots of crude saponin and total ginsenoside than other cell lines, also had 2 times of panaxadiol than panaxatriol.
Somatic chromosome counts and karyotype analyses were carried out for eight taxa of Korean Allium sect. Sacculiferum. The basic chromosome number of sect. Sacculiferum was x = 8, and they could be cytologically divided into two groups, that is, a diploid group (2n = 2x = 16) containing A. thunbergii var. thunbergii, A. thunbergii var. deltoides, A. thunbergii var. teretifistulosum, A. deltoidefistulosum, A. longistylum, A. linearifolium and A. taqueti, and a tetraploid group (2n = 4x = 32) with only A. sacculiferum. All observed chromosomes were classified into metacentric, submetacentric and subtelocentric. The metacentric ones appeared in all treated taxa. One or two pairs of submetacentric chromosomes were observed in most taxa except A. sacculiferum, the unique taxon with subtelocentric chromosomes. All taxa had a pair of homologous chromosomes with satellites, and the B-chromosomes found in A. thunbergii var. thunbergii, A. deltoidefistulosum, A. sacculiferum and A. longistylum, were metacentric or telocentric. The karyotypes of A. longistylum and A. linearifolium were firstly investigated in this study. In conclusion, the somatic chromosome numbers and karyotypes for members of the sect. Sacculiferum were valuable characters in identifying taxa, investigating interspecific relationships and delimiting taxa. In addition, A. thunbergii var. teretifolium, an invalid name (homonym), was renamed as A. thunbergii var. teretifistulosum H. J. Choi & B. U. Oh.
Recently successful induction of haploid plant by means of anther culture method has become a big topic among geneticists and plant breeders. The haploid plant can be used as a precious material for such basic researches as mutation or genetics. Once the haploid is obtained, production of homozygous plant is not a difficult problem. The method of producing homozygous plant can, also, be applied to the practical breeding works. When applied to the hybridization of self-fertilizing breeding period would be greatly shortened and in cross-fertilizing vegetables production of uniform hybrid seed would be very easily obtained. Last few years many scientists attempted anther cultures using various plant species, but it was successful only in several species. Unlike the other tissue cultures which use somatic organs or tissues as explants, anther culture seems to be very difficult because the plants or calli have to be induced from the haploid microspores or pollen grains. In the present experiment anther culture of fruit trees and ornamental shrubs of four genera and seven species was attemped. Anthers of Various stages ranging from tetrad and late microspore were cultured on the modified Murashige and Skoog's medium supplemented with various concentrations of auxins and kinetin as growth regulators. Handling of materials, sterilization, and other operations of culture were done by routine methods. The results were summarized as follows: 1. Calli were induced in the anthers of Forsythia Koreana Nak., Rhododendron mucronuratum Turcz., R. yedoense Max. var. Poukhanense Nak., and Prunus armeniaca L. var. ansu Max. No signs of callus were observed in Prunus persica Sieb. et Zucc. var. vurgaris Max., Pyrus ussuriensis var. macrostipes (Nak.), and Prunus salcina Lindley. 2. Calli were easily formed in any of the media with differing concentrations of auxins and kinetin. 3. In F. Koreana calli developed from anther surface and connective. Callus emerging out of anther locule was not observed. 4. Somatic calli arose from filament, connective, and inside of anther wall in R. mucronulatum. Many of the microspores accumulated starch grains. 5. The anther lobes located opposite the filament of R. yedoense turned easily to calli. This phenomenon was not observed in R. mucronulatum. Microspore embedded for a period in the medium became starch pollen. No callus was observed arising from microspore. 6. In P. armeniaca calli were not induced from somatic anther tissues. Instead, callus emerged out of anther locule rupturing the anther slit. Starch was not formed in the microspore. 7. In P. persica, Pyrus ussuriensis, and P. salcina, calli were not observed in the anthers examined more than 60 days after culture. Microspores of these species, however, were free of starch grains even after long period of subculture. 8. It was learned that somatic calli of the species examined arose usually from endothelium of anther wall, septum of two neighboring anther locules, parenchyma tissues of connectives, or anther lobes. 9. In the anther locule of P. armeniaca cultured long in medium, swollen microspores, polynucleate microspores, multicellular pollen grains, or callus mass were frequently observed, this indicating that the callus of this species was microspore-origin. 10. It was clarified that in P. armeniaca production of haploid plant by anther culture might be possible.
