Separation of Antioxidants and Glucose from Grape Skin Extract Using Polyethylene Glycol and Sodium Citrate (폴리에틸렌글리콜과 구연산 나트륨을 이용하여 포도껍질 추출물에서 항산화물질과 포도당 분리)

  • The purpose of this study is to develop a method for separating antioxidants and sugars from grape skin extract. The extract was first mixed with a variety of organic solvents to investigate whether the separation was feasible. When employing acetone, ethanol, dimethylsulfoxide, or dimethylformamide, the organic solvent-extract combination formed a single phase. However, when benzene, ethyl acetate, or n-hexane was added to the extract, the mixture separated into an organic and an aqueous phase and the pigments remained in the aqueous phase. On the other hand, when polyethylene glycol-2,000 (PEG-2000) and sodium citrate were added to the extract, the mixture was separated into three layers, with the majority of the flavonoids migrating to the top layer and 53% of the extract's glucose migrating to the bottom layer. The top layer had significant antioxidant activity, whereas the bottom layer showed no antioxidant activity. The glucose recovery in the bottom layer increased as the molecular weight of PEG increased and the highest recovery (67%) was observed when PEG-8,000 was added. The highest flavonoid separation was observed with PEG-2,000, followed by PEG-8,000 and PEG-400. The flavonoid separation when PEG-2,000 was added resulted in a flavonoid recovery of 48% and 0.2% from the top and bottom layers, respectively. Examining the effect of the separated solution using the agar disc diffusion method on yeast cell growth confirmed that the addition of the extract, the top, and the bottom layer did not inhibit cell growth.

Photocatalytic effect for the carbon-coated TiO2 prepared from different heat treatment temperature (열처리 온도에 따라 제조된 탄소 코팅된 TiO2에 대한 광촉매 효과)

  • Carbon-coated $TiO_2$ was prepared by $CCl_4$ solvent mixing method with different heat treatment temperature (HTT). Since the carbon layers derived from pitch on the $TiO_2$ particles were porous, the carbon-coated $TiO_2$ sample series showed a good adsorptivity and photo decomposition activity. The BET surface area was decreased by the increasing of the heat treatment temperature. The SEM results present to the characterization of surface texture on the carbon-coated $TiO_2$ sample and carbon distributions on the surfaces for all the materials used. The main elements of C, O and Ti were shown from EDX spectra. And the quantity of these elements is very rich in the samples. From XRD data, the pristine anatase peaks were observed in the X-ray diffraction patterns for the carbon-coated $TiO_2$ at the different HTTs. However, the rutile peaks were observed for the carbon-coated $TiO_2$ at HTT of 1073 K and 1123 K. Finally, the excellent photocatalytic activity of carbon-coated $TiO_2$ with UV-vis spectra between absorbance and time could be attributed to the homogeneous coated carbon on the external surface and structural phase transform, and the photocatalytic activity was decreased by the increasing of the HTT.

A study on the Antioxidative and Antimicrobial Activities of the Citrus Unshju peel Extracts (감귤과피 추출물의 항산화 및 항균 효과에 관한 연구)

  • Antioxidative and antimicrobial activities were carried out on the Citrus Unshju peel solvent extracts in order to discover new functional activities. The amounts of polyphenol in 70% metanol extract (MtEx) was measured as 836.8 mg% in Citrus Unshju peel. The EDA (electron donating ability) of 0.01, 0.02 and 0.1% MtEx in Citrus Unshju peel were measured as levels of 81.3, 86.0 and 89.6%. The nitrite scavenging effects of Citrus peel were also determined as the levels of 34.4% (pH 1.2) and 19.5% (pH 7.0). The pH of react solution was more acidic, the nitrite scavenging effect was more increased. The order of antioxidatives was shown as TBHQ > BHT > TOC > ChEx > EaEx > EtEx > WaEx > Control in corn germ oil and TBHQ > ChEx > EaEx > BHT > EtEx > WaEx > TOC > Control in canola oil. A number of the extracts were certified to have antimicrobial activities for a small number of micro-organisms, similar gram negative and positive micro-organisms. According to the results above, it was summerized that Citrus Unshju peel had the higher total polyphenol, EDA, nitrite scavenging effects and antimicrobial activities. Also isolated extract from ChEx and EaEx had high antioxidative, these effects were very similar to that of ${alpha}$-tocopherol and BHT. It would be proposed that Citrus peel can become a new natural source for antioxidative agents in future food industry.

Antioxidant, Anti-inflammation, and Anti-aging Effects of Ethyl Acetate Fraction from Cynanchum paniculatum Extracts (산해박 분획물의 항산화, 항염증 및 항노화 활성 효과 연구)

  • In this study, we prepared fractions from Cynanchum paniculatum (C. paniculatum) with ethyl acetate and identified the structure and content of paeonol, an indicator substance. Fractions were obtained by sequential solvent fractions using n-hexane, methylene chloride, ethyl acetate, n-butanol, and distilled water, and showed the highest paeonol content in ethyl acetate (EtOAc). The structure of the ethyl acetate fraction (EA) was confirmed by nuclear magnetic resonance (NMR) and the content was determined by high performance liquid chromatography (HPLC) analysis. The antioxidant activity of EA was tested through its ability to scavenge 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and its ability to inhibit nitric oxide (NO) production in RAW 264.7 cells induced by lipopolysaccharide (LPS) was tested for its ability to inhibit NO production in a concentration-dependent manner. Furthermore, MMP-1 production induced by ultraviolet B (UVB) was decreased in a concentration-dependent manner by the EA, and the ability to produce procollagen type-I was increased in a concentration-dependent manner. In addition, as a result of conducting clinical trials using cosmetic formulations containing EA, the effect of improving wrinkles around the eyes was confirmed. Thus, it is expected that Cynanchum paniculatum can be used as a natural functional cosmetic material with antioxidant and anti-aging effects.

Effect of zinc oxide nanoparticle types on the structural, mechanical and antibacterial properties of carrageenan-based composite films (산화아연 나노입자 유형이 카라기난 기반 복합 필름의 구조, 기계적 및 항균 특성에 미치는 영향)

  • In this study, zinc oxide nanoparticles (ZnONPs) were synthesized using three distinct zinc salts: zinc acetate, zinc chloride, and zinc nitrate. These ZnONPs were subsequently utilized in the fabrication of carrageenan-ZnONPs (Car-ZnONPs) composite films. The study assessed influence of the various ZnONPs on the morphological, water vapor barrier, color, optical, and antimicrobial properties of the Car-ZnONPs composite films. The surface morphology and UV-blocking attributes of the composite films were affected by the type of ZnONPs used, but their surface color, transparency, and chemical structure remained unaltered. The composite film's thickness and elongation at break (EB) significantly increased, while the tensile strength significantly decreased. In contrast, film's elastic modulus (EM) and water vapor permeability coefficient (WVP) showed no significant difference. All the composite films with added ZnONPs demonstrated potent antibacterial activity against Escherichia coli O157:H7 and Listeria monocytogenes . Among the carrageenan-based composite films, Car-ZnONPsZC showed the highest antibacterial and UV-blocking properties, and its elongation at break was significantly higher than that of the pure carrageenan films. This suggests that ZnONPs composite films have the potential to be used as an active packaging film, preserve the safety of the packaged food and extend shelf life.

Effects of extraction conditions on color quality and antioxidant properties of persimmon (Diospyros kaki Thunb.) leaf tea (침출 조건이 감잎차의 색 및 항산화 특성에 미치는 영향)

  • This study was carried out to investigate the effect of the extraction conditions for persimmon (Diospyros kaki Thunb.) leaf tea (PLT) on its color quality and antioxidant properties. The amount of persimmon leaf (PL) powder and pH influenced the PLT's color and antioxidant capacity. As the amount of PL powder in tea increased, lightness decreased while yellowness increased. The PLT with the highest amount of PL (10 mg/mL) exhibited the highest 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity and ferric reducing antioxidant power (FRAP). In addition, the PLT with the highest PL showed the highest total polyphenol and flavonoid contents. Subsequently, PLT was prepared using 10 mg/mL PL powder under varying pH conditions. As pH increased from 4 to 7, lightness decreased while redness and yellowness increased. Antioxidant capacity was determined using 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity and FRAP assay showed that lower pH exhibited higher antioxidant capacity. The PLT extracted under the lowest pH of 4 showed higher polyphenol and flavonoid contents than that extracted under higher pH conditions. Overall, PLT extraction using a solvent with pH 4.0 showed better antioxidant activities and higher amounts of polyphenolic compounds. Simultaneously, lesser lightness, redness, and yellowness were detected in PLT extracted under pH 4 conditions. In conclusion, to acquire a better functional health benefit in terms of antioxidant capacity, preparing PLT under pH 4 conditions is suggested.

Flavonol glycosides from the flowers of Carthamus tinctorius and their anti-diabetic activity (잇꽃(Carthamus tinctorius)으로부터 Flavonol glycoside 화합물들의 분리 및 항당뇨 효과)

  • The flowers of Carthamus tinctorius (Safflower) were extracted with 80% aqueous methanol (CTex) and the concentrates were partitioned into EtOAc (CTE), n-BuOH (CTB), and H2O (CTW) fractions. Repeated silica gel (SiO2) and octadecyl silica gel column chromatographies for the EtOAc and n-BuOH fractions led to isolation of four flavonol glycosides. Nuclear magnetic resornance, infrarad spectroscopy, and mass spectroscopy revealed the chemical structure of the isolated compounds, astragalin (1), isoquercetin (2), nicotiflorin (3), and rutin (4). Quantitative analysis of four isolated compounds in CTex was performed by HPLC. CTex was found to contain 1 at 0.107, 2 at 0.367, 3 at 6.752, and 4 at 0.991 mg/g, respectively. Through this study, an experiment was conducted to evaluate the protective effect on pancreatic islets of the extract, solvent fractions, and all isolated compounds using a zebrafish larvae damaged by alloxan. Pancreatic islet size treated with EtOAc (CTE), n-BuOH (CTB), and H2O (CTW) fractions and compounds 1-4 significantly increased compared to the alloxan-induced group. These results indicate that C. tinctorius flowers and its isolated compounds are used as potential anti-diabetic agents.

Effect of Ethanol Fractionation of Lignin on the Physicochemical Properties of Lignin-Based Polyurethane Film

  • Lignin, a prominent constituent of woody biomass, is abundant in nature, cost-effective, and contains various functional groups, including hydroxyl groups. Owing to these characteristics, they have the potential to replace petroleum-based polyols in the polyurethane industry, offering a solution to environmental problems linked to resource depletion and CO2 emissions. However, the structural complexity and low reactivity of lignin present challenges for its direct application in polyurethane materials. In this study, Kraft lignin (KL), a representative technical lignin, was fractionated with ethanol, an eco-friendly solvent, and mixed with conventional polyols in varying proportions to produce polyurethane films. The results of ethanol fractionation showed that the polydispersity of ethanol-soluble lignin (ESL) decreased from 3.71 to 2.72 and the hydroxyl content of ESL increased from 4.20 mmol/g to 5.49 mmol/g. Consequently, the polyurethane prepared by adding ESL was superior to the KL-based film, exhibiting improved miscibility with petrochemical-based polyols and reactivity with isocyanate groups. Consequently, the films using ESL as the polyol exhibited reduced shrinkage and a more uniform structure. Optical microscope and scanning electron microscope observations confirmed that lignin aggregation was lower in polyurethane with ESL than in that with KL. When the hydrophobicity of the samples was measured using the water contact angle, the addition of ESL resulted in higher hydrophobicity. In addition, as the amount of ESL added increased, an increase of 7.4% in the residual char was observed, and a 4.04% increase in Tmax the thermal stability of the produced polyurethane was effectively improved.

Review of the study on the surfactant-induced foliar uptake of pesticide (계면활성제에 의해 유도되는 농약의 엽면 침투성 연구 현황)

  • Research trends in the measurement of foliar uptake of pesticides and the recently proposed action mechanism of the surfactant-induced uptake of pesticides were reviewed with the related reports and studies. Major techniques used in those fields are bioassay, radiotracer techniques with leaves or cuticular membrane. Recently, a new method using Congo Red as a tracer was proposed. The limiting factor in the pesticides uptake into leaves is the waxy layer which consists of the epicuticular and cuticular wax. Physico-chemical parameters such as molar volume, water solubility and partition coefficient of pesticides have limited influences on the pesticide uptake into leaves. Polydisperse ethoxylated fatty alcohol surfactants are well known as the good activator for many pesticides. It is now generally agreed that uptake activation is not related to the intrinsic surface active properties of surfactants such as surface activity, solvent property, humectancy and critical micelle concentration. Recent studies using ESR-spectroscopy revealed that the surfactants have an unspecific plasticising effect on the molecular structure of the wax and cuticular matrix, leading to increased mobilities of pesticides. Penetration of surfactants into waxy layer altered the pesticide mobility in wax and the partition coefficient of pesticide, and then the pesticides penetration into leaves was enhanced temporally. The enhancing effect of surfactant could be significantly different depending on the carbon number of aliphatic moiety and the number of ethoxy group in polyoxyethylene chain of surfactants. It is suggested that the rate of penetration of surfactants should have a significant relationship with the rate of penetration of pesticides.

The Antimelanogenic Effects of Compounds Extracted from Bamboo Inner Film (죽황(竹黃)으로부터 분리한 미백활성 성분의 멜라닌생성 억제효과)

  • In order to develop a new depigmenting agent, extracts were obtained from 60 native plants and their antimelanogenic activities were screened by evaluating the inhibitory effect on tyrosinase which is a major enzyme responsibles for the melanin synthesis. The extracts of Trichosanthes kirilowii fruits, Phyllostachys bambusoides inner films (BIF), Clerodendrum trichotomum leaves, and Acer okamotoanum leaves showed relatively high inhibitory effect on tyrosinase and their $IC_{50}$ values were $50{\sim}100{\mu}g/mL$. The extract of BIF inhibited melanin synthesis of B16F10 melanoma cells by 52%, which was the highest among those of various extracts. Furthermore, the effect of BIF extract is 10% higher than that of arbutin (42%), a popular depigmenting agent in Korea. Ten compounds having antimelanogenic activity were isolated from the BIF extract by solvent extraction and chromatography. These compounds were identified as phenolic derivatives: SM701, SM702, SM703, and BPR211 were hydroquinone derivatives; SM707 a gallic acid derivative; SM704, SM705, SM706, SM708 and SM709 ferulic acid derivatives. The free radical scavenging activities of these compounds were measured and compared to those of hydroquinone and vitamin C. The $SC_{50}$ values scavenging 50% DPPH of SM702 and SM709 were $60{\sim}70{\mu}M$ similar to that of hydroquinone and those of SM701 and SM708 were $30{\sim}40{\mu}M$ slightly lower than that of vitamin C. These results suggest the presence of components having high antioxidant activity in the BIF extract. The SM709, identified as 1,2-O-diferulylglycerol, inhibited the activities of tyrosine hydroxylase and dopa oxidase by 18 and 60%, respectively. The SM709 also inhibited the melanin synthesis of B16F10 melanoma cells by 62% and this was the highest antimelanogenic activity among those obtained from the various purified compounds. Therefore, antimelanogenic activity of the BIF extract was concluded to be due to both inhibition of DOPA oxidase and antioxidant activity.
