Stability Evaluation on Measuring Water-soluble Chloride Anions from Iron Artifacts (철제유물의 수용성 염소이온 측정방법에 대한 안정성 평가)

  • The most ideal method to measure the water-soluble $Cl^-$ ion eluted from iron artifacts is conducting the analysis on desalting solution by Ion Chromatography. But most institutes related to cultural heritages use Cl meter by reason of lack of budget and experts. This study evaluated reliability and stability between Cl meter and Ion Chromatography by doing cross-validation with results from two methods to detect $Cl^-$ ion of desalting solution. From D.I water, extremely small quantities of $Cl^-$ ion was detected by the influence of remaining water-soluble $Cl^-$ ion at the electrode of Cl meter and water-soluble $Cl^-$ which remains in Sodium sesquicarbonate, components of reagent was detected as well. The first desalting solution had the most $Cl^-$ ions, $Cl^-$ ion slightly decreased from the second to the fourth desalting solution and tend to decrease again at the stage of dealkalified in D.I water. Each Cl meter has the standard deviation according to the measured numbers and the higher concentration of $Cl^-$ ion the desalting solution has, the wider the deviation is. But when the concentration of $Cl^-$ ion is low, it was stable to use Cl meter to detect the concentration of $Cl^-$ ion from iron artifacts because there is the small deviation, It is thought that conductivity meter method is not suitable for measuring $Cl^-$ ion, because the electrical conductivity of alkaline solution is too high to measure $Cl^-$ ion.

Antimicrobial & Physiological Characteristics of Ethanol Extract from Pinus rigida Miller Leaves (리기다소나무 잎 에탄올 추출물의 항균 및 생리특성)

  • Pinus rigida Miller leaf extract (PRLE) showed antimicrobial activity remarkably against food pathogenic and spoilage bacteria at concentrations of $100{\sim}250{\mu}g/mL$. Alcohol-soluble PRLE had higher antimicrobial activity against Staphylococcus aureus and E-coli than any other-soluble PRLE such as butanol, ethyl acetate, ether and water. As PRLE concentration increased alcohol-soluble PRLE increased the remarkable inhibitory zone of microbial growth on the microbial media. PRLE showed good stability against temperature and pH in the range of $40{\sim}150^{\circ}C$ and $4{\sim}11$, respectively. This may indicate that PRLE can be a potential anti-microbial agent for industrial application. In addition, SEM of Listeria monocytogenes suggested that it antimicrobial component would perturb the functions of microbial cell membranes synergistically. In the feeding experiment the formaldehyde content in the serum of formalin-fed and PRLE-treated me decreased remarkably due to the lysis of formaldehyde and the rate of hemoglobin biosynthesis was recovered to the orignal state within a short breeding time.

  • Aromatic pelybenzoxazoles(PBOs) display excellent thermal stability plus good solvent and chemical resistance. Wholly aromatic PBOs, in fact, are soluble only in strong acids(e.g., sulfuric, rmethanesulfonic, triflic, and polyphosphoric acids). However, fully heterocyclized polymers have shown some drawbacks in solubility and processing. This problem of processing is currently being exploited to obtain unusual combinations of physical properties in fibers and films. (omitted)

Synthesis of Poly(enaryloxynitriles) Containing Schiff Bases and Their Thermal Properties

  • Poly(enaryloxynitriles) containing Schiff bases were prepared from p-bis(1-chloro-2,2-dicyanovinyl)benzene (2) and various aromatic diols having Schiff base moiety by interfacial polymerization. The chemical structure of the polymers was confirmed through synthesis of their corresponding model compounds. All the polymers were soluble in polar aprotic solvents and their brittle films were cast from DMF solution. They showed a large exotherm around 340 ℃ attributable to the chemical change of dicyanovinyl group. Especially, curing of azomethine group was observed to occur at 390 ℃ by differential scanning calorimetry. According to the thermogravimetric analyses, they exhibited excellent thermal stability with 60-90% residual weight at 500 ℃ in nitrogen.

Preparation and Evaluation of Mutivitamin Emulsion (복합비타민 유제의 제조와 평가)

  • Water-lipid soluble multivitamin formulations were widely used to reduce the disease and stress of animals as husbandry has made a remarkable progress in recent. But the efficiency of these formulations is far from satisfactory. So, this study was attempted to develop the physically and chemically stable and useful multivitamin o/w emulsion. Multivitamin o/w emulsion composed of water, soybean oil (10%, v/v), vitamin A, D, E, K, $B_2,\;B_6,\;B_{12}$ and panthenol. To make a stable o/w emulsion, the egg lecithin (2%, w/v) and glycerin (2.5%, w/v) were used for emulsifier and thickening agent, respectively. The oil in water emulsion system was manufactured by microfluidizer and the physicochemical stability of this emulsion was evaluated. The average particle size and interfacial tension were measured. From the result of interfacial tension tested, critical micelle concentration of the egg lecithin was 0.5% (w/v) and optimal concentration for the preparation of emulsion was 2% (w/v). The mean particle size was about $0.6\;{\mu}m$ which was suitable for injections. Short-term accelerated stability as physical stability study was tested by centrifuging and freeze-thawing the emulsion samples. The additions of vitamins resulted in the increment of particle size and reduction of physical stability of emulsion. But it is not an enormous problem for the stability of emulsion. Also, we have performed the long-period preservation stability test for the vitamins. All vitamins were analysed by HPLC. The result of storage under $4^{\circ}C$ and dark conditions demonstrated that all vitamins were maintained stable at least 16 weeks, except for vitamin $B_{12}$.

Charge Transport Properties of Polyaniline-gold/graphite Oxide Composite Films

  • A polyaniline-gold composite was prepared via the polymerization of aniline hydrochloride with or without water-soluble graphite oxide using auric acid as an oxidant. The reaction products were characterized using Xray photoelectron spectroscopy. The thermal stability and embedded crystallinity of the composites were also investigated using thermogravimetric and X-ray diffraction analyses. The electrical properties of the composites were examined using cyclic voltammetric measurements at room temperature and temperature-dependent DC conductivity within 300-500 K. Compared to pure graphene oxide and polyaniline-gold composite, the polyaniline-gold-graphene composite exhibited higher crystallinity and thermal stability, and higher current density response under equivalent conditions.

Stabilization of Aspergillus sp. $\alpha$-Amylase by Modification with $IO_4$-oxidized Starch ($IO_4$-산화전분 변형에 의한 효소의 안정성 증가)

  • The stabilization of Aspergillus sp. $\alpha$-amylase was attained by modification with periodate-oxidized sol-uble starch. The pH stability of modified enzyme was increased at pH 3~4 and 9~11 in the presence of $\alpha$-cyclodextrin($\alpha$-CD) compared with that of native enzyme. Thermal stability of the modified enzyme was increased. After treatment at 6$0^{\circ}C$ for 30min the activity remained 20% for the enzyme modified at pH 9.7 in the presence of $\alpha$-CD and tested in the presence of $\alpha$-CD 10% for the enzyme modified at pH 9.7 in the presence of $\alpha$-CD 0% for the native enzyme. The native enzyme and modified enzyme showed one peak in HPLC. The substrate specificity of the modified enzyme was not changed in HPLC analysis of reaction product.

Biochemical Properties of Recombinant Cyclodextrin Glucanotransferase Expressed in Saccharomyces cerevisiae (Saccharomyces cerevisiae에 발현된 재조합 cyclodextrin glucanotransferase의 생화학적 특성)

  • The cyclodextrin glucanotransferase(CGTase) gene of Bacillus macerans was expressed in Saccharomyces cerevisiae and the recombinant CGTase was partially purified from the yeast culture supernatant. The optimal pH and temperature of the CGTase were found to be 6.0 and 5$0^{\circ}C$, respectively. The pH and temperature stabilities of the recombinant enzyme were significantly enhanced and the half life at 55$^{\circ}C$ was about 60 hr. When the recombinant CGTase was reacted with 5% soluble starch, the conversion yield of total cyclodextrin (CD) from starch was estimated to be 41% at 48 hr, whereas the wild type enzyme showed the yield of 12%. This improvement of conversion yield and thermal stability of CGTase may be useful for the development of low-cost CD production process.

  • Since the advent of recombinant DNA technology coupled with other biotechnology a variety of therapeutically effective proteins and peptides have been extensively invesitigated and many of them are now on clinical trial. They, however, suffer from some problems such as immunogenicity, antigenicity, instability and short half-life in circulation due to their proteinous natures. These drawbacks can be overcome successfully by conjugating proteins and peptides with hydrophilic polymers such as polyethylene glycol (PEG), albumin or dextran. The resulting soluble conjugates showed reduced antigenicity and immunogenicity, increased circulatory half-life, enhanced stability against proteolytic degradation. Comparing with the unmodified proteins and peptides, the therapeutic potential of conjugates is greatly enhanced. Clinical applications of these conjugates have shown promising results for the future use.
