Urokinase, a plasminogen activator, was conjugated with dextran by the cyanogen bromide activation-coupling method. The resulting water-soluble conjugate was purified by gel permeation chromatography on Sephadex G-200. The maximal activity was obtained when the ratio of urokinase/dextran was 1/20 for the coupling. The final preparation showed 5 CTA units/mg conjugate, 300 CTA units/mg protein, 8.4 % activity retention, and 47 % protein retention. The urokinase-dextran conjugate had good thermal, pH and storage stabilities. In addition, it showed greater resistance to the inhibitory effect of human plasma than native urokinase. Also in vitro biological half-life of urokinase increased 40 times by this conjugation. In view of activity, excellent stability and increased half-life, the conjugate can be a potential fibrinolytic agent in an injectable form.
Various enaminonitriles-terminated reactive polymer precursors containing rigid aromatic and flexible alkyl units were prepared from the corresponding diamines and 1-chloro-1-phenyl-2,2-dicyanoethene (1). All the enaminonitriles-terminated precursors were characterized by spectroscopies and elemental analysis. They were highly soluble in DMF and NMP, and partially soluble in common organic solvents such as THF and acetone. They showed a large exotherm around 350 ℃ attributable to the thermal polymerization by crosslinking of the dicyanovinyl group. Upon heating the precursors, heat-resistant and insoluble network polymers were obtained. Thermogravimetric analyses of the precursors containing rigid aromatic moiety exhibited thermal stability with a 10% weight loss around 420-480 ℃ and 75-88% residual weight at 500 ℃ under nitrogen.

2,3-Di-(2'-hydroxyethoxy)-4'-nitrostilbene (3) was prepared and condensed with terephthaloyl chloride, adipoyl chloride, and sebacoyl chloride to yield novel Y-type polyesters (4-6) containing the NLO-chromophores 2,3-dioxynitrostilbenyl groups, which constituted parts of the polymer backbones. Polymers 4-6 were soluble in common organic solvents such as acetone and N,N-dimethylformamide. Polymers 4-5 showed thermal stability up to $300^{\circ}C$ in thermogravimetric analysis with glass transition temperatures $(T_g)$, obtained from differential scanning calorimetry, in the range $81-95^{\circ}C$. The second harmonic generation (SHG) coefficients $(d_{33})$ of the poled polymer films at the 1064 nm fundamental wavelength were around $3.68{\times}10^{-9}$ esu. The dipole alignment exhibited high thermal stability up to $T_g$, and there was no SHG decay below $T_g$ due to the partial main-chain character of the polymer structure.

New Y-type polyester (3) containing nitrophenylazoresorcinoxy groups as NLO chromophores, which are components of the polymer backbone, was prepared and characterized. Polyester 3 is soluble in common organic solvents such as N,N-dimethylformamide and acetone. It shows a thermal stability up to $240^{\circ}C$ in thermogravimetric analysis with glass-transition temperature ($T_g$) obtained from differential scanning calorimetry near $116^{\circ}C$. The second harmonic generation (SHG) coefficient ($d_{33}$) of poled polymer film at the 1064 nm fundamental wavelength is around $4.63{\times}10^{-9}$ esu. The dipole alignment exhibits a thermal stability even at $4^{\circ}C$ higher than $T_g$, and there is no SHG decay below $120^{\circ}C$ due to the partial main-chain character of polymer structure, which is acceptable for NLO device applications.
2,5-Di-(2'-hydroxyethoxy)benzylidenemalononitrile (3) was prepared and polymerized with 2,4-toluenediisocyanate and 3,3'-dimethoxy-4,4'-biphenylenediisocyanate to yield novel T-type polyurethanes 4 and 5 containing 2,5-dioxybenzylidenemalononitrile, nonlinear optical (NLO)-chromophores as part of the polymer backbones. Polyurethanes 4 and 5 were soluble in common organic solvents such as acetone and N,Ndimethylformamide and showed a thermal stability up to $250{^{\circ}C}$ with glass-transition temperatures ($T_g$) in the range $119-146{^{\circ}C}$. The second harmonic generation (SHG) coefficients ($d_{33}$) of poled polymer films at 1064 nm fundamental wavelength were around $5.82{\times}10^{-9}$ esu. The dipole alignment exhibited high thermal stability up to $T_g$, and there was no SHG decay below $140{^{\circ}C}$ due to the partial main-chain character of the polymer structure, which was acceptable for nonlinear optical device applications.
Repairing damaged DNA and removing all physical connections between sister chromosomes is important to ensure proper chromosomal segregation by contributing to chromosomal stability. Here, we show that the depletion of non-SMC condensin I complex subunit H (NCAPH) exacerbates chromosome segregation errors and cytokinesis failure owing to sister-chromatid intertwinement, which is distinct from the ultra-fine DNA bridges induced by DNA inter-strand crosslinks (DNA-ICLs). Importantly, we identified an interaction between NCAPH and GEN1 in the chromatin involving binding at the N-terminus of NCAPH. DNA-ICL activation, using ICL-inducing agents, increased the expression and interaction between NCAPH and GEN1 in the soluble nuclear and chromatin, indicating that the NCAPH-GEN1 interaction participates in repairing DNA damage. Moreover, NCAPH stabilizes GEN1 within chromatin at the G2/M-phase and is associated with DNA-ICL-induced damage repair. Therefore, NCAPH resolves DNA-ICL-induced ultra-fine DNA bridges by stabilizing GEN1 and ensures proper chromosome separation and chromosome structural stability.
들깨와 들깨 증자를 $25{\sim}28^{\circ}C$ 에서 $2{\sim}3$일간 발아시켜 얻은 발아들깨로부터 n-hexane으로 들기름을 추출한 후 이들 2종류의 틀기름에 함유되어 있는 산화방지 성분을분리하고, 발아에 외한 산화방지 성분의 함량 변화가 틀기름의 산화안정성에 미치는 영향을 검토하였다. 들기름을 acetone 및 methanol에 용해시켜 동결처리한 다음 분리된 불용성 획분의 산화안정성을 측정한 결과 들기름에 존재하는 산화방지성 물질은 methanol에 용해성이 강한 성분임을 확인하였다. 들기름에서 분리된 methanol 가용성 획분을 silica gel column chmmato-graphy로 분획하여 산화방지 활성을 검정한 결과 n-hexane : ethyl acetate의 비가 7 : 3인 획분에서 가장 높은 산화방지 활성을 나타내었다. 산화방지 활성이 가장 높았던 획분을 TLC로 분리한 결과 3개의 band가 확인되었다. 이중 $R_f$치 0.71인 band핀가 가장 높은 산화방지 활성을 나타내었으며 이 band에 존재하는 산화방지 성분은 phenol성 화합물로 추청되었다. TLC에 의하여 분리된 산화방저 성분을 다시 HPLC에 의하여 분러한 결과HPLC chromatogram상에서 4가지 활성물질 peak가 분리되었다. 이들 peak중 들깨에서는 P 1, P 2의 함량이 각각 46.5%, 25.6%이었고 P 3의 함량은 22.6%이었으나, 발아들깨에서는 P 1, P 2의 함량이 각각43.8%, 20.6%로 약간 감소한 반면 P 3 함량은 29.8%로 상당량 중가하였다. 이와 같이 HPLC 분석에서 분리된 4가지 peak중 발아에 의해 P 1, P 2의 활성물질 함량은 상대적으로 감소하고 P 3의 활성물질 함량은 증가하는 것으로 보아 이들 물질외 함량 변화에 의해서도 들깨 및 발아들깨로부터 추출한 들기름의 산화안정성이 다르게 나라나는것으로 추청되었다.
Systematic electrophoretic analysis of alcohol-soluble proteins and salt-soluble proteins of 247 Panax ginseng (P.g) and Panax quinquefolium (P.q) germplasms seed was carried out on an improved lactate-polyacrylamide gel electrophoresis, a method with high resolving power, good reproducibility and stability. The electrophoregrams of proteins, according to their migration rate, were classified into four groups such as ${\alpha}$, ${\beta}$, ${\gamma}$ and $\omega$ for the alcohol-soluble proteins and three such as I, II and III for the salt-soluble ones. Panax ginseng or Panax quinquefolium had their own unique band pattern distinguishable from each other, regarding as their specific "fingerprint". In this study, 3 of 168 (1.8%) P.g germplasms and 1 of 79 (1.3%) P.q germplasms had their own unique band pattern, showing that P.g and P.q germplasms have poor genetic diversity in species. The band patterns of dry seed and stratified seed (embryo rate=60%) were basically the same. The band number of the F, hybrid of p.gx p.q was exactly equivalent to the number of the common bands plus the specific bands of the two parents, indicating that the difference of band patterns was a genetic trait con- trolled by the nuclear genes. The electrophoregram of F1 of P.g x P.q could be predicted by that of the two parents and the band pattern of the F1 hybrids could be demnonstrated by that of the mixed seed extract from the two parents.
방향족 액정 폴리에스테르는 열방성 액정 중합체의 대표저인 예로 지금까지 구조-물성 관계가 많이 연구되었다. 본 연구에서는 유기 용매에 녹일 수 있는 가용성 방향족 액정 폴리에스테르를 합성하고, 아세틸렌 말단기를 갖는 열경화성 수지와 복합화하여 제반 물성을 비교하였다. 가용성 방향족 액정 폴리에스테르는 6-hydroxy-2-naphthoic acid, terephthalic acid, isophthalic acid 및 4-aminophenol을 단량체로 하여 에스테르 및 아미드 교환 반응에 기초로 한 축합 중합법을 이용하여 합성하였다. 아세틸렌 말단기를 갖는 열경화성 수지는 4-ethynylaniline과 terephthaloyl dichloride, isophthaloyl clichloride, 4,4'-biphenyldicarbonyl dichloride 등과 각각 반응시켜 합성하였다. 가용성 방향족 액정 폴리에스테르와 열경화성 수지를 용액 블렌딩하고 열처리하여 복합체를 제조하였으며, 복합체의 열적 안정성, 열팽창계수, 유전특성 등을 조사하였다.
항균제 대체제로 응용하기 위하여 다양한 저분자량 수용성 키토산(LMWSC; MW1, MW3, MW5, MW10)을 제조하였으며, 이들의 항균제 대체제 사용 가능여부와 그 작용 기작에 대하여 연구하였다. 먼저, 다양한 분자량 형태의 LMWSC를 이용하여 사람에게 유해한 각종 박테리아를 이용하여 항균효과를 확인하였고, 그 중 MW10의 항균효과가 가장 우수한 것으로 확인되었다. 그 반면 사람의 적혈구를 이용한 용혈활성 실험에서 독성을 나타내지 않았다. MW10의 항균 효과가 세균의 어느 부분에서 일어나는지 확인하기 위해 박테리아의 세포막 조건(PE/PG=7/3, w/w)으로 인공리포좀을 만들었고, 여기에 MW10을 처리한 결과 세균 막에서 항균효과를 나타냄을 추론할 수 있었다.
