Monascus purpureus CBS 281.34가 생성하는 홍국 색소의 안정성 (Stability of the Pigments from Monascus purpuresu CBS 281.34)

  • 홍국균이 생산하는 색소를 상업적으로 이용하기 위한 기초연구로서 Monascus purpureus CBS 281.34 80% ethanol 가용성 색소의 열 pH, 유기산 존재 시의 안정성을 알아보았다. 또한 ethanol 가용성 색소 추출물에 증류수를 가해 수분 함량이 0∼80%가 되도록 하고 수용성 색소와 함께 저장하면서 이들의 안정성도 검토하였다. 80% ethanol 가용성 홍국색소는 1$0^{\circ}C$의 저온에서 4주간 저장 시 9.74%의 분해율을 보였으나, 60∼8$0^{\circ}C$의 고온에서는 24시간과 12시간 저장 시 각각 23.06%와 30.36%나 분해되어 색소의 안정성은 단기간 내 크게 저하하였다. pH 2와 pH 10에서의 80% ethanol 가용성 색소 분해율은 pH 4∼8보다 높았고, 0.6% citric acid, 6% acetic acid 및 1.5% lactic acid를 가해 pH 4로 조정하여 4주간 저장 시에는 대조구와 대조구와의 차이가 가장 큰 acetic acid 하에서의 분해율과의 차이도 6.08%로 거의 나타나지 않아 80% ethanol 가용성 홍국 색소는 유기산하에서 비교적 안정한 것으로 사료되었다. 또한 ethanol 가용성 색소에 증류수를 가해 저장하면 색소 안정성은 저하하였는데, 색소 분해율은 수분함량 20%에서 가장 높았고 80%에서 가장 낮았다. 한편 수중에서의 ethanol 가용성 색소는 2$0^{\circ}C$보다 1$0^{\circ}C$에서 현저하게 안정하였다.

Bacillus sp. PY123 균주가 생성하는 수용성 황색수소의 부분 정제 및 그 안정성 (Partial Purification and Stability of a Water-soluble Yellow Pigment from Bacillus sp. PY123.)

  • For application of a yellow pigment as food additives, stability of a water-soluble yellow pigment from Bacillus sp. PY123 was investigated. The yellow pigment from Bacillus sp. PY123 was purified with pH treatment, activated carbon and silica gel column chromatography. The partial purified yellow pigment appeared only one spot on silica gel TLC after 12 evaporation and under irradiation of UV 253nm at dark room. Rf value of the pigment was measured at 0.04 and 0.12 with development of a solvent mixture (Butanol : Acetic acid : water = 4 :1:5) and a solvent mixture (Isopropanol : Ammonia : Water = 9 :1: 2), respectively, The partial purified pigment appeared a white fluorescence under UV365nm irradiation. The partial purified yellow pigment had a main peak and a minor peak on HPLC using 20mM phosphate buffer(pH 7.0) at 1ml/min flow rate. The partial purified pigment was stable at heat treatment, acidic pH, oxide-reductants and surfactants.

Effect of dietary pigment source on shell color of abalone, Haliotis discus hannai

  • This study was conducted to investigate effects of dietary pigment sources on shell color of juvenile abalone, Haliotis discus hannai. Three replicate groups of the abalone, average weight 173 mg, were fed the diets containing various pigment sources such as Porphyra powder, Spirulina, yeast astaxanthin and paprika extract for 16 weeks. Survival and weight gain were not affected by dietary pigment sources (P>0.05). Shell color of abalone fed diets containing Porphyra powder and Spirulina was approached to yellow-red and orange which are similar to shell color of wild abalone. However, shell color of abalone fed the diets containing yeast astaxanthin and paprika extract were similar to that of control diet showing bright green. Porphyra powder and Spirulina contain not only fat-soluble pigment such as chlorophyll and carotenoids but also water-soluble pigment such as phycoerythrin and phycocyanin. These results would be useful information to change shell color of abalone in aquaculture.

  • For using additives of foods, or cosmetics, a strain A80 producing blue pigment was isolated from soil. The strain A80 was identified as a strain of Azotobacter vinelandii based on morphological and physiological characteristics. The strain A80 was extracellulaly secreted the blue pigment on PYG agar plate, but not secreted it into PYG broth. And then, the strain A80 was extracellulaly secreted the blue pigment in PYG broth containing 2.0% chitin, while the strain A80 was not secreted the blue pigment in PYG broth containing 2.0% chitin and 1% NaCl simultaniously.

  • A strain US50-3 producing water-soluble red pigment was isolated from the pond separating oil from water near the oil storage tanks. The strain US50-3 was identified as a strain of Serratia marcescens considering its morphological and physiological characteristics, and DNA G+C contents. It showed a little difference comparing to the Type strain and was considered to be another biotype strain.

  • 자색고구마 색소 농축액의 유동특성을 원뿔형 회전점도계를 사용하여 가용성 고형분의 농도 25-65% 및 온도 20-6$0^{\circ}C$의 범위에서 조사하였다. 자색고구마 색소 농축액은 뉴톤성유체의 특성을 나타냈다. 자색고구마 색소 농축액의 점도의 온도에 대한 영향은 Arrhenius 식을 따랐으며, 활성화에너지는 14.23-43.00 kJ/mol로서 농도가 증가함에 따라 직선적으로 증가하였다. 자색고구마 색소 농축액의 점도와 온도 및 농도에 대한 관계를 조사한 바, 일정한 온도에서 용액의 점도는 농도가 증가함에 따라 지수함수적으로 증가하였으며, 그 증가 정도는 온도가 낮을수록 증가하였다.

  • To develop a yellow pigment for a food-additive, a strain producing a water-soluble yellow pigment was isolated from phylloplane of tree leaf. The strain PY123 was identified a Bacillus sp. based on morphological and biochemical characterization such as a bacillus form, mortility, spore formation, Gram positive, and catalase production. The pigment production of the strain PY123 was increased about 3 times in patato broth containing 1% sucrose and 0.1 mM CoCl$_{2}$ than in only potato broth after incubation at 30$circ$C, for 2 days. When 0.1 mM CoCl$_{2}$, was added in potato broth containing 1% sucrose at late log phase during incubation of the strain PY123, cell growth was not inhibited, and the period at maximal pigment production of the strain PY123 was about 12hrs faster than in potato broth containing 1% sucrose and 0.1 mM CoCl$_{2}$, simultaniously.

  • The optimal cultural conditions for production of the bluish purple prgment by the cultivation of Streptomyces californicus KS-89 were determined with various substrates. The carbon and nitrogen sources on the production of pigment indicated that soluble strarch and glycerol as carbon sources and sodium glutamate sodium nitrate as nitrogen sources given a mzimum yield of the pigment at 3$0^{\circ}C$ for 7 days. The addition of ferrous sulfate was essential. The highest production of pigment was observed with cultivation in a medium containing 2.0% soluble starch 1% glycerol 0.5% sodium glutamate 0.05% sodim nitrate 0.001% L-proline 0.025% K2HPO4 0.005% MgSO4 .7H2O, 0.04% FeSO4.7H2O, 0.001% thiamine.HCl and pH7.0.

  • Water-soluble browning pigments were artily purified from Korean red ginseng through the several procedures such as fractionation by n-butanol, precipitation by ethanol, dialysis and gel filtration. At least four kinds of water-soluble browning pigment were separated from each other, two kinds of low-molecular-weight and two kinds of high-molecular-weight pigments.

  • 치자로부터 청색 및 황색색소, 고량으로부터 암갈색 색소를 주정을 이용하여 추출ㆍ농축하여 60$^{\circ}$Brix 농도의 추출물을 회수하였는데, 각각의 수율은 0.68, 1.97, 0.63% (w/w)였다. 이들을 대두유: 물: 유화제: 치자청: 치자황: 고량색소=8: 22: 42: 10: 15: 13 (w/w)의 비율로 혼합하고 균질화하여 유용성 천연 검정색소를 얻었다. 이 색소를 대두유에 용해시킨 결과 대두유의 품온이 30∼4$0^{\circ}C$ 범위에서 가장 높은 용해도를 나타내었다. 대두유에 한 번 용해된 색소물질은 혼합 대두유의 품온을 2$0^{\circ}C$ 이하로 유지하여도 재분리현상이 발생하지 않는 것으로 확인되었다
