Isolation and Characteristic of the Phosphate Solubilizing Bacteria Klebsiella sp. DA 71-1 (Hydroxyapatite 인산염 가용화 균 Klebsiella sp. DA 71-1의 분리와 가용화특성)

  • To develop high effiency biofertilizer, a bacterium having ability to solubilize inorganic phosphate was isolated from cultivated soils, using a sucrose minimal agar-hydroxyapatite medium. The strain was identified as Klebsiella sp. DA7l-1, based on the physiological and biochemical properties. The activity of solubilizing inorganic phosphate of Klebsiella sp. DA7l-1 against three types of insoluble phosphate such as tri-calcium phosphate, aluminium phosphate, hydroxyapatite were quantitatively determined. The results indicated that the strain solubilized hydroxyapatite. The MPS (mineral phosphate solubilizing) conditions of Klebsiella sp. DA7l-1, were measured to determine the optimal conditions. The optimal temperature and initial pH to solubilize insoluble phosphate in sucrose minimal medium were $30^{\circ}C$ and pH 6.0, respectively.

  • In order to minimize the mycelial pellet size of a high phosphate-solubilizing fungus, Aspergillus sp. PS-104 in liquid media, one of the critical obstacles during the submerged culture of filamentous fungi, an investigation was focused on the culture conditions (media and inoculum size) and additives (different soils, surfactants and polyethylene glycol 200). When the fungus was cultured in PDB, SDB and YPD media. their pellet sizes decreased in the order of SDB=YPD>PDB. At the higher concentrations of initial inoculum ranging from $1{\times}10^3$ to $1{\times}10^7$ conidia/ml, the smaller size of pellet was formed in the PDB medium. In addition, the pellet size was effectively reduced by 1/6${\sim}$1/4 by the addition of 0.1% soil containing zeolite, diatomite, loess, kaoline and talc, excluding bentonite. The addition of 0.1% Tween 80, Triton X-100 and PEG 200 also decreased the pellet size, but SDS completely inhibited the fungal growth.

Induced systemic resistance and plant growth promotion of a phosphate-solubilizing bacterium, Enterobactor intermedium 60-2G (인산가용미생물, Enterobacterium intermedium 60-2G의 식물 생장 촉진 및 전신저항성 유도)

  • A phosphate-solubilizing bacterium, Enterobacter intermedium 60-2G, was examined for plant growth-promotion and induction of systemic resistance using a model system of cucumber and scab disease, caused by Cladosporium cucumerinum. Compared with a nonbacterized control, treatment of cucumber with E. intermedium significantly reduced the severity of scab disease after challenge-inoculation with C. cucumerinum. Treatment of cucumber with E. intermedium also enhanced cucumber growth. The 60-2G strain showed a strong antimicrobial activity against several plant pathogenic fungi including Fusarium soysporum and Magnaporthe grisea. These results suggest the E. intermedium 60-2G is a promising candidate as a biological control agent displaying multiple beneficial properties to promote plant health.

The Effect of Phosphate Solubilizing Microorganism on Cucumber Productivity (미생물비료 인산가용화균이 오이의 생산량에 미치는 영향)

  • The effect of the Penicillium sp. PS-113 with high phosphate solubilizing activity and Lactobacillus sp. was studied on cucumber growing test in green house. Inoculation of Pemicillium sp. Ps113 $5.0{\times}10^6$cfu/ml and Lactobacillus sp. $6{\times}10^6$cfu/ml were longer than control about 3cm length of cucumber. The average weight was also increased 89.2g more than control. Total product of cucumber was 10,681.7g and it was increased 4517.3g as compared to control. Thus, Inoculations of high phosphate solubilizing microorganism increased in productivity on cucumber.

  • Several phosphate-solubilizing microorganisms were isolated in order to enhance the availability of insoluble phosphates accumulated in soils. Among the microorganisms, Aspergillus niger was selected and identified for this study. The phosphate-solubilizing activity. the phosphorus uptake by plants and the changes in soil microbial biomass by inoculation of Aspergillus niger were investigated. The uptake amounts of phosphorus by lettuce and pimiento were increased by inoculation of Aspergillus niger in all experimental treatments. There was negative correlation between the soil microbial biomass P and the soil phosphorus content. However the soil available phosphorus ($Y=-0.0007X^2+0.7126X^2-29.46$, $R=0.8283^{**}$) and the phosphorus absorption of plants ($Y=0.0049X^2-2.2352X+326.34$, $R=0.6350^*$) were significantly correlated to soil microbial biomass C on the positive section of quadric curve.

  • This study was to investigate beneficial effects of microbial mixture on red pepper which was capable of promoting plant growth by solubilizing insoluble phosphate as well as protecting plants from pathogenic attack. Saccharomyces sp. L13 was isolated for phosphate solubilizing activity on aluminium phosphate, tricalcium phosphate, calcium hydrophosphate, and magnesium hydrophosphate. On the other hand, Bacillus sp. L32 was isolated for antagonistic activity against Phytophthora capsisi and Colletotrichum gloeosporioides, causing Phytophthora blight and Anthracnose disease in pepper, respectively. The strain L32 exhibited antagonistic activities both under dual culture assays and detached leaves assays. The each strain under the condition of mixed cultivation exhibited the same growth rates as one under pure cultivation. In greenhouse study, the mixed culture showed the both effect of plant growth promotion and reduction of disease symptom development against P. capsisi and C. gloeosporioides providing a potential as effective microbial agent for plant husbandry.

Characteristics of Insoluble Phosphates Solubilizing by Klebsiella sp. DA71-1/pLYJ (Klebsiella sp. DA71-1/pLYJ의 난용성 인산염 가용화 특성)

  • To develop high efficiency biofertilizer solubilizing insoluble phosphates, lactate dehydrogenase (ldh) gene was isolated from Staphylococcus sp. LJ2. Genetic constructions were carried out using the pGEM-T-easy vector and pHSG398. Recombinant DNA plasmids containing the ldh gene were transferred to Klebsiella sp. DA71-1 by electroporation. The selected transformant was named as a DA71-1/pLYJ. The insoluble phosphates solubilization activity of DA71-1/pLYJ was higher than that of DA71-1 at various culture conditions. Glucose was the best carbon source for insoluble phosphates solubilization among the used carbon sources. Maximal insoluble phosphates solubilizing was found in sucrose minimal (SM) medium containing 3% glucose. The solubilizing activity of DA71-1/pLYJ against three types of insoluble phosphates, such as tri-calcium phosphate, hydroxyapatite, aluminium phosphate, were quantitatively determined. The optimal temperature and initial pH to solubilize insoluble phosphates in the SM medium was $37^{\circ}C$ and pH 5.0, respectively.

  • Studies were carried out on the practical use of Naringinase and some chracteristics of Naringin solublizing enzme which might hydrolyae naringin to purunin. Obtained results were as follows. 1. Selected strain for Naringinase producing was identified to be Aspergillus niger S-1 and its naringinase was applied to chinese citron processing to remove the bitter taste. 2. Of the naringinase, naringin solubilizing enzyme was purified on a DEAE-Sephadex A-50 column and crystalized from acetone and ammonium sulfate. 3. Hydrolized naringin which has higher solubility rather than naringin or naringenin were identified by thin layer chromatography. 4. Hydrolyzed naringin and naringin were separatly determinated by ethylacetate extraction and this result was compared with sensory test.
