도시쓰레기의 혐기성 소화는 에너지원 및 온난화 가스 저감 문제 등에 의해 최근에 사회적 관심사가 되고 있다. 도시쓰레기는 고형분 함량이 높고 질소성분이 낮으며 셀루로스와 헤미셀루로스가 주성분으로 되어 있다. 도시쓰레기의 메탄 전화율은 대개 50%이며 $0.2m^2/kg$ VS에 해당한다. 고형물 농도가 높을수록 긴 수리학적 체류시간이 필요하며 주입물에 접종슬러지를 혼합하여야 한다. 도시쓰레기의 혐기성 소화 시 C/N비는 25가 상한이고 NH3-N의 적정농도는 700mg/L로 알려져 있다. pH조절을 위하여 흔히 석회와 탄산나트륨이 사용되고있는데 탄산나트륨을 3,500mg/L이상 첨가하면 나트륨 독성이 나타난다. 고온성 혐기 소화조는 운용과 관리가 어려우나 병원성 미생물 억제에 효과적인 것으로 알려져 있다. 도시쓰레기의 혐기성 소화공정의 최적화를 이룩하려면 소화공정에 관여하는 미생물의 작동기전에 대한 이해가 필요하다.
재조합 대장균에서 내포체 형태로 발현시킨 LK68을 생물학적 활성을 가진 native한 단백질로 재생시키기 위해서 PBA 크로마토그래피와 EBA 크로마토그래피를 이용한 고체상 재접힘을 수행하였다. 내포체와 세포파쇄액을 시작물질로 하여 재접힘 공정을 수행하였으며 총 단백질 회수율과 재접힘 수율을 비교한 결과, EBA 공정이 기존의 액상 재접힘이나 PBA를 이용한 재접힘 공정에 비하여 우수함을 확인하였다. 또한 Iysine binding, RP-HPLC, SEC-HPLC, Ellman method 등을 사용하여 분석한 결과 재접힘된 LK68이 native LK68와 동등함을 확인하였다. 본 연구를 통해 EBA 크로마토그래피를 이용한 재접힘 방법은 재접힘 단계의 수율을 향상시킬 뿐 아니라 공정 단계, 시간 등을 감소시켜 공정 성능을 전체적으로 향상시킬 수 있음을 제시하였다.
포말 분리법에 의한 양어장 순환수 중의 고형물 제거 특성을 조사하였다. 장치의 효율에 영향을 미치는 주요인자들은 공탑 공기유속, 체류시간, 그리고 포말층의 높이였다. 측정된 양어장수 시료 내에는 총 단백질량의 약 $50\%$정도가 계면 활성 성분이었다. 공탑 공기유속, 체류시간이 증가함에 따라 단백질, T-N. TA 및 고형물 성분들의 제거율은 증가하였으며, TVS의 제저율은 공탑 공기유속 및 체류시간 (HRT)이 증가함에 따라 증가하였으나 포말층 높이가 높아짐에 따라서는 감소하였다. 포말분리장치는 양어장 순환수 중에서 발생되는 어류 성장에 유해한 성분, 특히 TA 및 TVS의 제거에 우수한 성능을 보임을 알 수 있었다.
The purpose of this study was investigation of quantitative analysis of marker substances in Paeonia lactiflora extracts by solid fermentation. High performance liquid chromatography (HPLC) for the determination of albiflorin and paeoniflorin in P. lactiflora extracts by solid fermentation, the separation method was performed on C18 column ($250\;mm\;{\times}\;4.6\;mm$, $5\;{\mu}m$, RS tech) using gradient solvent mixtures of water-acetonitrile with photodiode array detector (230nm). The flow rate was 1.0 ml/min. Retention time of albiflorin and paeoniflorin was about 28.88, 31.92 min and linearity of calibration was showed good result(r2 = 0.9998, 0.9996), respectively. Content of albiflorin was $0.090\;{\pm}\;0.03%$ in P. lactiflora extract(control), $0.102\;{\pm}\;0.00%$ in P. lactiflora extract fermented with Paecilomyces japonica, $0.056\;{\pm}\;0.01%$ in P. lactiflora extract fermented with Ganoderma lucidum, $0.093\;{\pm}\;0.00%$ in P. lactiflora extract fermented with honey and $0.046\;{\pm}\;0.00%$ in P. lactiflora extract fermented with Nuruk. Content of paeoniflorin was $4.506\;{\pm}\;0.13%$ in control, $2.599\;{\pm}\;0.04%$ in P. lactiflora extract fermented with Paecilomyces japonica, $1.222\;{\pm}\;0.03%$ in P. lactiflora extract fermented with Ganoderma lucidum, $2.750\;{\pm}\;0.05%$ in P. lactiflora extract fermented with honey and $0.847\;{\pm}\;0.00%$ in P. lactiflora extract fermented with Nuruk, respectively. Content of the marker substances did not increase in all fermentation experiment group.

Content of nodakenin was $0.76\;{\pm}\;0.02%$ in control, $0.31\;{\pm}\;0.00%$ in Angelicae Gigantis Radix extract fermented with Paecilomyces japonica(SDT)(p<0.01), $0.51\;{\pm}\;0.02%$ in Angelicae Gigantis Radix extract fermented with Ganoderma lucidum(SYT)(p<0.01), $0.82\;{\pm}\;0.03%$ in Angelicae Gigantis Radix extract fermented with honey(SST)(p<0.05) and $0.88\;{\pm}\;0.01%$ in Angelicae Gigantis Radix extract fermented with Nuruk(SNT)(p<0.01). Content of decursin was $4.50\;{\pm}\;0.08%$ in control, $2.90\;{\pm}\;0.05%$ in Angelicae Gigantis Radix extract fermented with Paecilomyces japonica(SDT)(p<0.01), $2.65\;{\pm}\;0.08%$ in Angelicae Gigantis Radix extract fermented with Ganoderma lucidum(SYT)(p<0.01), $4.46\;{\pm}\;0.11%$ in Angelicae Gigantis Radix extract fermented with honey(SST) and $4.73\;{\pm}\;0.04%$ in Angelicae Gigantis Radix extract fermented with Nuruk(SNT)(p<0.05), respectively.
$ZnMn_2O_4$ has been prepared by a mechanochemical process using a mixture of $Mn_2O_3$ and ZnO as starting materials, and investigated as a possible anode material for lithium-ion batteries. The phase evolution and morphologies of the ball-milled and annealed powders are characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM) with energy dispersive microanalysis (EDX), respectively. The solid-state reaction for the formation of $ZnMn_2O_4$, under the given experimental conditions, is achieved in a short time (30 min), and the prepared samples exhibit excellent electrochemical performances including an enhanced initial coulombic efficiency, high reversible capacity, and stable capacity retention with cycling.
Submerged Membrane Bio-Reacter(SMBR) process was used to food wastewater treatment. From laboratory pilot-scale experiment data, it was confirmed that this process was very effective process for organics, suspended solid, and N, P treatment. It was found that BOD and COD removal rate were obtained 90% and 92%, respectively, for 150 days operation. Organics loading rate did not affect to the removal efficiency because MLSS concentration in aerobic tank was highly maintained. IN the case of first reactor operated with anoxic and second reactor operated as aerobic, T-N, T-P removal rate were obtained 93% and 95%, respectively. It was shown that removal efficiency could be maintained stable due to the complete removal of SS and sludge production decreased with increasing of sludge retention time.
A bench-scale continuous activated sludge system was operated in order to study the effects of solid waste landfill leachate on municipal wastewater treatment. The leachate sample was collected from Nanjido landfill. During the 7 weeks of operational period, the leachate content in the influent fed to the system was increased stepwise from 3% to 5% and 7%. The solids retention time was the major process control parameter, and it was 10 days. With the municipal wastewater alone, COD removal was 75%. The removal percents, however, decreased to 60% and 45% when the leachate content was 3% and 7%, respectively. For the wastewater spiked with the leachate, the sludge production was higher than for the municipal wastewater alone. Sludge settleability determined by SVI deteriorated with the increase of leachate content. The specific oxygen uptake rate, however, was measured higher as leachate content increased.
This work experimentally determined the effect of microaerobic condition on anaerobic digestion of thickened waste activated sludge in semi-continuous mesophilic digesters at hydraulic retention time of 20 days. The concentration of hydrogen sulfide was $7{\pm}2ppm$ at the microaerobic condition and $14{\pm}2ppm$ at the anaerobic condition. Removal efficiency of volatile solid was not significantly different between microaerobic ($40{\pm}8%$) and anaerobic ($38{\pm}8%$) conditions. There was no important difference between microaerobic ($1,352{\pm}98ml/d$) and anaerobic ($1,362{\pm}104ml/d$) conditions in the biogas production, either. Therefore, it could be concluded that the application of the microaerobic condition was an efficient method of the hydrogen sulfide removal from the biogas.

Improper management and unsanitary approaches are implemented in disposal of leachate, which has resulted in groundwater pollution at village Uruli Devachi, Pune, India. Various physico-chemical treatment methods are commercially available for leachate treatment. However, the application of biological methods viz. phytoremediation to the municipal solid waste landfill leachate has been limited. We report the remediation ability of Typha aungstifolia and Acrorus calamus that is capable of reducing hazardous constituents from the landfill leachate. After 96 h of hydraulic retention time (HRT), it was observed that T. aungstifolia-treated sample showed high reduction potential in reducing biochemical oxygen demand, chemical oxygen demand, hardness, total dissolved solids, Na, Mg, Ca and Ni whereas A. calamus showed greater reduction capacity for alkalinity, Cl, Cu, Zn and Cr. Furthermore, it was also observed that T. aungstifolia withstood longer HRT than A. calamus. In situ application of T. aungstifolia and A. calamus for remediation of landfill leachate carries a tremendous potential that needs to be further explored.
