Plant regeneration was accomplished from protoplast culture of rice (Oryza sativa L. cv. Taebaeg). Embryogenic callus was induced from mature seed on MS medium containing 5 mM proline, 2.5 mg/L 2,4-D, 30 g/L sucrose in the dark at 28$^{\circ}C$ and used to establish embryogenic cell suspension culture. Suspension cells were subcultured every one week in N6 medium supplemented with 5 mM proline, 200 mg/L casein hydrolysate, 2.5 mg/L 2,4-D and amino acids of AA medium. Suspension cultures were composed of cells that were densely cytoplasmic, potentially embryogenic and were at least maintained for more than 6 months in liquid medium. Protoplasts were isolated from fast-growing suspension culture cells and cultured in a slightly modified KpR medium by mixed nurse culture. Isolated protoplasts began to divide within 5~7 days and thereafter, protoplast-derived calli were sequentially transferred to callus proliferating medium that soft agar MS medium contained 2 mg/L 2,4-D and produced distinct embryogenic cells. Microcolonies were then transferred to solid medium which consisted of MS medium containing 5 mg/L kinetin, 1 mg/L NAA, 1 mg/L ABA, 30 g/L sucrose and 10 g/L sorbitol under fluorescent light. Mulitple shoots of 4~5 per callus emerged and were transferred to hormone-free MS medium for root initiation. Thereafter, The plantlets were transferred to pots of soil to mature in the culture room.
This experiment was carried out to analyze the effect of indole on the synthesis of indirubin in suspension culture of Polygonum tinctorium. Adding indole and L-tryptophan into culture media was revealed that indirubin was synthesized in callus grown on solid medium containing indole and proper concentration of indole for indirubin production was decided as 200mg/1. Indirubin content in suspension culture was higher than in solid medium with considerable amount of indirubin secresed into media in suspension culture and highest quantity of indirubin was obtained when indole was added into medium after 20 days suspension culture.
Orostachys japonicus A. Berger is a Perennial herbaceous plant which has been traditionally used as an anti-inflammatory agent to treat hepatitis and as an anticancer agent. The objective of this study was 1) to establish and proliferate in vitro plant of O. japonicus 2) to induce indirect somatic embryogenesis from O. japonicus. General calli and embryogenic calli in all ranges of 2,4-D and BA combination, were induced and were best at 22% (embryogenic cell) in 5.0 mg/L 2,4-D and 0.5 mg/L BA combination. Embryogenic cell line was maintained by subculture at 2 week intervals and transferred to solid and liquid medium for embryo formation. In solid medium culture, globular and heart shaped embryos were observed in MS medium containing 5.0 mg/L 2,4-D and 0.5 mg/L BA combination. The number of embryos was 6.5 per 0.5 g cell, and then the immature embryos transferred to MS basal medium for embryo development. In a suspension culture of embryogenic cells, globular and heart shaped embryos were emerged in MS medium supplemented with 3.0 mg/L 2,4-D and 0.3 mg/L BA combination after 10 days of incubation. The embryo formation rate was about 33% by suspension culture. The ratio of embryo germination was 60.9%, on the other side, the root formation rate was 74.3% in 1/2 MS continuously.

Cultivation conditions for overproduction of lovastatins were investigated from the lovastatin producing strain N-03 which was obtained with NTG (N-methyl-N'-nitro-nitrosoguanidine) treatment from Aspergiliu ferrous ATCC 20542. Produced lactone and acid form of lovastatin were detected, and analyzed by HPLC method. In liquid culture, medium No. 2 containing soy protein produced higher amounts of the lovastatins than medium No. 1 (contained rapeseed oil). In solid culture, maximum production was obtained at 28$^{\circ}C$ for 15 days cultivation using cooked wheat bran. For the overproduction of lovastatin from this strain, solid culture method using plastic bag is more superior than liquid culture.
A study was conducted to investigate in vitro micro propagation of four potato cultivars of Daese, Jasim, Chubaek and Haryeng in MS medium and PM medium (a medium containing half concentration of $NH_4NO_3$ and $KNO_3$, two fold concentration of $CaCl_2$ as compared to MS medium). During $1{\sim}2$ weeks of culture, Daese and Jasim showed better shoot elongation on the solid MS medium than Chubaek and Haryeng whereas Chubaek and Haryeng did better shoot elongation on the solid PM medium. But no difference was observed after 4 weeks of culture. As compared to shoot elongation on the solid medium, it was delayed at early stage of culture in the liquid medium without shaking. Shoot formation ratio of potato (above 4 cm of shoot length) began to increase significantly after 1 week of culture and kept on increasing until 4 weeks. The four cultivars showed the different patterns of shoot growth in bioreactor. The PM medium with a quarter salt strength was effective for the regeneration of axillary buds as well as shoot elongation of Jasim and Chubaek. Daese showed vigorous regeneration of axillary buds in the PM medium with a half salt strength. On the other hand, Haryeng showed slower growth than the other three cultivars.
We have established adventitious root culture systems of Codonopsis lanceolata, Codonopsis pilosula and Codonopsis ussuriensis. Root segments of C. lanceolata were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with $0.5mg/{\ell}$ NAA and produced $18.8{\pm}1.9$ roots per explant. Root segments of C. pilosula were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with $1.0mg/{\ell}$ NAA and produced $8.5{\pm}1.8$ roots per explant. Leaf segments of C. ussuriensis were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with $0.5mg/{\ell}$ NAA and produced $7.8{\pm}0.4$ roots per explant. In liquid culture, the best production of adventitious root (fresh weight) was obtained in 1/2 MS medium with $1.0mg/{\ell}$ NAA. This study demonstrated for the first time to produce adventitious roots in C. pilosula and C. ussuriensis.
The most effective conditions of in vitro culture were studied for mass propagation of Pteris cretica 'Wilsonii'. Spores of the species germinated within 7 weeks. The greatest proliferation was obtained with Knop and Hyponex media, but growth was more effective in Hyponex medium. MS medium induced necrosis of prothalli in all strength of nitrogen and sucrose except in case of 0% sucrose. Hyponex medium supplemented with 1% sucrose and 0.6% agar promoted propagation and growth of prothalli. In Hyponex medium, optimal inoculation method was homogenization, but in MS medium dividing colonies of prothalli was more effective. Culturing on solid medium was more effective than liquid culture method. Liquid culture induced necrosis of prothalli. Shaking cultured prothalli showed good growth, but propagation was inhibited compared to those cultured on solid medium.
A fungus, Penicillium sp. PS-113, isolated from soil showed the high phosphate-solubilizing activity in patato dextrose broth-rock phosphate to produce free phosphates to the culture broth with the concentrations of 585 ppm against rock phosphate. In this medium, the optimum temperature and initial pH to solubilize rock phosphate were 30$^{\circ}C$ and pH 7.5, respectively. In order to make the mass production of the conidia from this fungus, we cultured in on various solid-based media like barley, corn, wheat, rice, rice bran, and compost. As a result, the fungus highly produced conidia ranging from 2.1 to $5.1{\times}10_9$ conidia/g${\cdot}$media on these solid media except compost-based medium, which was 0 times less than others. Effects of inoculation of the phosphate solubilizing fungus as a biofertilizer were studied in perlite-based pot cropped with Zea mays Suwon 19. Inoculation of Penicillium sp. PS-113 increased in plant height (1.4 times), plant weight (5.2~8.1 times) and root length (1.1~1.2 times) at 60-day cultivation, compared to Hogland solution either without $NH_4H_2PO_4$ or displace $NH_4H_2PO_4$ to powdered rock phosphate, a phosphorus source for plant growth.
True collagenolytic enzymes in animal tissues were first demonstrated by Gross and Lapiere (1962), who showed the ability of such an enzyme in the culture medium of living explants of tadpole tissue to degrade a specific substrate of undenatured collagen under physiological conditions. Recently, tumor-associated collagenolytic activity has been demonstrated in human neoplasm and in ascites V Carcinoma. This investigation have been peforme to determine whether or not a collagen lytic enzyme could e found in isolated solid Tawa sarcoma of Donryu female rat obtained the culture medium. The results were as follows.
1. 11.5mg% of hydroxyproline contained in Donryu rat skin collagen, which was extracted by 0.5M acetic acid.
2. Cultivation of solid Tawa sarcoma tissues on reconstituted rat skin collagen gels showed lysis of adjacent gel after 18 hours, and much more extensive lysis after 5 days.
3. Collagen substrate was not attacked by the common proteolytic enzymes, trypsin, pepsin, and pronase.
In vitro culture of shoot tip of garlic (Allium sativium L. cv. Seosan) was carried out to find medium condition of the induction of multiple shoots and bulbing for muliproduction of virus-free seed bulbs. For this work, tank culture was introduced. In shoot tip culture on MS solid medium the induction of multiple shoots and bulbing were better by adding 3% sucrose than 8%. Supplementation with 2mg/L 2ip and 0.2 mg/L IAA in this medium was effective. Three point three shoots including 2.7 bulbs were formed from a shoot tip after cultivation for 30 days on this medium. Bulbing of garlic in liquid culture with plastic water tank of 20L supplied air at the side of the lower part was better by adding 3% sucrose than 8% by subculture for 45 days with shoots obtained from shoot tip culture for 30 days on soid MS medium. Shoot growth was vigorous at 3% sucrose however bulb growth was more effective on the medium of 8% sucrose. Because of the effectiveness on solid medium added 3% sucrose, 2 mg/L 2ip and 0.2 mg/L IAA for initial production of multi-shoot in stem tip culture and the effectiveness in liquid culture with water tank for growth of bulbs, the method of two-step culture could be introduced for the multiple production of seed bulb of high quality. It was more desirable by supply of 0.2 mg/L BA and 0.02 mg/L NAA at tank culture time. But growth of the bulbs became poor by increasing concentration of NAA of the medium.
