A Study on Microbial Community and Microbial Degradation of Diesel (디젤의 미생물 분해와 군집에 관한 연구)

  • This study investigates characteristics of diesel degradation and variations of microbial community with the soil enrichment cultures. The cultures has yellow(YE-5) and transparent color's(WH-5) colony on solid plate medium. The bacillus type of YE-5 and WH-5 cultures showed diesel degradation at the rate of 99.07mg-Diesel/$L{\cdot}day$ and 57.82mg-Diesel/$L{\cdot}day$ in the presence of 1%(v/v) initial diesel concentration. Diesel degradation was 1.7 times faster than WH-5 culture. YE-5 or WH-5 culture could degrade a wide range of diesel compounds from $C_8$ to $C_24$. Microbial community analysis by PCR-DGGE technique shows that Psedomonas, Klebsiella, Escherichia and Stenotrophomonas as proteobacteria take role on the diesel degradation. uncultured Senotrophomonas sp. was only detected with YE-5 culture. It is concluded that proper combination of the microorganism should be present to stimulate the degradation of diesel and further studies are recommended for the effect of uncultured Senotrophomonas sp. or Escherichia hermannii on diesel degradation.

Development of LC-MS/MS Quantitation Method for Ethoxyquin in Fishery Products (수산물 중 에톡시퀸의 LC-MS/MS 정량분석법 개발)

  • Ethoxyquin (EQ, 1,2-dihydro-6-ethoxy-2,2,4-trimethyl-quinoline) is quinoline-based antioxidant used in the animal feed and food industry to protect the raw materials and final products against oxidation. In recent years the use of synthetic antioxidants in fishmeal ingredients carry-over to farmed fish fillets has received increasing attention in food safety. This study was conducted to develop an analytical method to determine EQ in aquatic products. The analytes were confirmed and quantified via liquid chromatography-tandem mass spectrometry (LC-MS/MS) in the positive ion mode using multiple reaction monitoring (MRM). The sample was extracted with 1 N HCl (in case of flatfish extracted with 1 N HCl containing 10% acetonitrile). Then, solid phase extraction (SPE) was used for the cleanup. Standard calibration curves presented linearity with the correlation coefficient ($r^2$) > 0.99, analyzed at 0.005-0.2 mg/kg concentration. The developed method was validated according to the Codex Alimentarius Commission (CAC) guideline. The limits of quantitation for EQ were 0.01 mg/kg. Average recoveries ranged from 81.3% to 107%. The repeatability of measurements, expressed as the coefficient of variation (CV, %), was below 10%. The analytical method was characterized with high accuracy and acceptable sensitivity to meet CODEX guideline requirements and would be applicable to analyze the EQ residue in aquatic products.

Manufacturing of Wine with Watermelon (수박을 이용한 발효주의 제조)

  • Selection of yeast strains, optimum conditions for alcohol fermentation, sterilization methods, and additives for improving wine quality were investigated to manufacture watermelon wine. Eight yeast strains exhibited significant alcohol fermentation, among which KWS 06 was selected for watermelon wine fermentation, because watermelon wine made by this strain showed the best overall acceptability in sensory evaluation. Sucrose was determined as the best saccharide for alcohol fermentation among sucrose, corn syrup, glucose, fructose, and lactose. Optimum concentration of soluble solid and $(NH_{4})_{2}HPO_{4}$ of nitrogen source were $24^{\circ}Brix and 0.2%, respectively. Addition of raspberries and omija increased wine flavor and alcohol production, respectively, with optimum alcohol production, taste, and color achieved with addition of 20 g/L raspberries and 10 g/L omija. Best sensory quality was obtained by addition of 0.04 % watermelon flavorant to the juice.

Identification of the Volatile Compounds in Polyethylene Terephthalate Bottles and Determination of Their Migration Content into Mineral Water (PET 생수병 내 휘발성 물질의 동정 및 이행량 분석)

  • This study was carried out to identify the volatile organic compounds (VOCs) in polyethylene terephthalate (PET) bottles and to determine the extent to which VOCs migrate into mineral water during the bottling process and storage. A greater amount of nonanal and decanal was generated from the PET bottles than from the PET preforms. Benzene, ethylbenzene, nonanal, and vinyl benzoate were identified from the PET bottles when the incubation temperature of the headspace solid-phase microextraction (HS-SPME) sampler was set to 60, 80, and $100^{\circ}C$. As the incubation temperature increased, the concentrations of nonanal, vinyl benzoate, and decanal increased significantly. When the high-density polyethylene (HDPE) PET bottle caps were extracted with dichloromethane, the level of Irgafos 168 was found to be $206{\pm}20.1\mu}g/g$. The concentration of 2,4-di-tert-butylphenol in water was $4.80{\pm}0.2{\mu}g/L$. Therefore, it is necessary to avoid exposing PET and HDPE resins to high temperatures during the manufacturing process and storage of bottled water.

Cytotoxicity and Antitumor Effects of Insambaekhaptang on C57BL/6 Mice Melanoma-induced Lung Metastasis (인삼백합탕(人蔘百合湯)이 B16세포에 대한 세포독성능 및 C57BL/6계 생쥐의 폐전이암의 억제에 미치는 영향)

  • Oriental medicine as a candidate for effective cancer treatment recently gain positive concerns in fields of therapeutic oncology. that is why some herbal medicines have been empirically safer in toxicity than anticancer drugs used in western medicine, and to show excellent therapeutic efficacy in human trial. Thus, these effects by clinically applied-herbs have not yet fully demonstrated in experimental tumor model. This study was initiated to evaluate the antitumor effect of Insambaekhaptang as candidate of antitumor-herbal agent against B16 melanoma metastasized into C57BL/6 mice lung. In experiment to test whether Insambaekhaptang can directly kill cancer cells in vitro or not, Insambaekhaptang showed direct killing action in concentration or higher against B16 melanoma cells using MTT assay, and showed lower IC50. Another experiment to know whether Insambaekhaptang can inhibit growth and metastasis of cancer cell or not, Insambaekhaptang significantly inhibited Solid tumor by intraperiperal injected-melanoma and lung metastasis induced by intravenous injected-melanoma in inbred C57BL/6 mice. When quantitative survival time increasing, we could obtain results that increased 113% in treated by Insambaekhaptang. These results show that Insambaekhaptang can inhibit growth of B16 melanoma cells through various biological mechanisms.

EPerformance of high-rate anaerobic sequencing batch reactor treating sewage sludge and food waste (연속 회분식 혐기성 공정을 이용한 하수슬러지와 음식물쓰레기의 혼합소화 거동 특성)

  • Temperature-phased anaerobic digestion (TPAD), anaerobic sequencing batch reactor (ASBR), and co-digestion technologies were combined together in order to overcome low efficiencies of conventional anaerobic sewage sludge digestion processes. In the performance, TPAD-ASBR process showed high VS removal efficiency over 60% up to the organic loading rate (OLR) of 2.7 g VS/L/d. The first-stage of TPAD-ASBR and control system played a most significant role in VS destruction and methane production. Methane production rate (0.79 l $CH_4/L/d$) of the system was higher than that (0.59 l $CH_4/L/d$) of the control system. The substrate characteristics of the sewage sludge, such as low VS concentration (1.5%, w/w) and biodegradability, were properly improved by the addition of food waste as a co-substrate, leading to more efficient VS removal and methane production. With several track studies, it was revealed that the independent solid retention time (SRT) of those systems prevented untreated particles from outflowing and also, extended the retention time of the active biomass for further degradation. Consequently, it was confirmed that the sequencing batch operation of the TPAD process using co-substrate was a promising alternative for the recycling of sewage sludge with low VS content.

Milk Conjugated Linoleic Acid (CLA) Profile and Metabolic Responses of Dairy Cows Fed with High-temperature-micro-time (HTMT) Treated Diets Containing High Quantity Extruded Soybean (ESB)

  • A feeding trial was conducted to examine the effect of high-temperature-micro-time (HTMT) processing of diets containing extruded soybean (ESB) in high quantity on milk fat production, metabolic responses, and the formation of conjugated linoleic acid (CLA) and trans-vaccenic acid (TVA). Twenty-one multiparous Holstein cows in mid-lactation were blocked according to milk yield in the previous lactation. Cows within each block were randomly assigned to either normal concentrate or HTMT treated diets containing ESB (7.5% HTMT-ESB and 15% HTMT-ESB). It was hypothesized that the HTMT-ESB would affect the undegradable fatty acids in the rumen and, thus, would modify the fatty acid profile of milk fat. Both 7.5% and 15% HTMT-ESB did not affect milk yield, fat, protein, lactose and solid-not-fat (SNF), but the proportion of cis-9, trans-11 CLA in milk fat was significantly increased by these treatments. Content of TVA in milk fat was not affected by HTMT-ESB. The HTMT-ESB influenced the fatty acid profile in milk fat, but there was little difference between 7.5% and 15% of supplementation. HTMT-ESB feeding significantly decreased the concentration of plasma insulin and glucose, while plasma growth hormone (GH), triglyceride (TG), non-esterified fatty acid (NEFA) and HDLcholesterol were increased by 7.5% and 15% ESB-HTMT supplementation in comparison to the control group (p<0.05). However, no significant difference was observed in plasma LDL-cholesterol, insulin like growth factor (IGF)-1, T3, T4, and leptin concentrations among treatments (p>0.05). The present results showed that cis-9, trans-11 CLA production was increased by HTMT treatment of dietary ESB without reduction of milk fat, and the unchanged milk fat and yield was assumed to be associated with the constant level of thyroid hormones, leptin, and IGF-1.

Effects of Feeding Extruded Soybean, Ground Canola Seed and Whole Cottonseed on Ruminal Fermentation, Performance and Milk Fatty Acid Profile in Early Lactation Dairy Cows

  • Four ruminally cannulated Holstein cows averaging 43 days in milk (DIM) were used in a $4{\times}4$ Latin square to determine the effect of feeding extruded soybean, ground canola seed and whole cottonseed on ruminal fermentation and milk fatty acid profile. One hundred and twenty lactating Holstein cows, 58 (${\pm}31$) DIM, were assigned to four treatments in a completely randomized block design to study the effects of the three types of oilseeds on production parameters and milk fatty acid profile. The four diets were a control diet (CON) and three diets in which 10% extruded soybean (ESB), 5% ground canola seed (GCS) and 10% whole cottonseed (WCS) were included, respectively. Diets consisted of concentrate mix, corn silage and Chinese wild rye and were balanced to similar concentrations of CP, NDF and ADF. Ruminal fermentation results showed that ruminal fermentation parameters, dry matter intake and milk yield were not significantly affected by treatments. However, compared with the control, feeding cows with the three oilseed diets reduced C14:0 and C16:0 and elevated C18:0 and C18:1 concentrations in milk, and feeding ESB increased C18:2 and cis9, trans11 conjugated linoleic acid (CLA). Production results showed that feeding ESB tended to increase actual milk yield (30.85 kg/d vs. 29.29 kg/d) and significantly decreased milk fat percentage (3.53% vs. 4.06%) compared with CON. Milk protein (3.41%) and solid non-fat (13.27%) from cows fed WCS were significantly higher than from cows fed CON (3.24% and 12.63%, respectively). Milk urea N concentrations from cows fed the ESB (164.12 mg/L) and GCS (169.91 mg/L) were higher than cows fed CON (132.31 mg/L). However, intake of DM, 4% fat corrected milk, energy corrected milk, milk fat and protein yields, milk lactose percentage and yield, somatic cell count and body condition score were not affected by different treatments. The proportion of medium-chain fatty acid with 14 to 16 C units in milk was greatly decreased in cows fed ESB, GCS and WCS. Feeding ESB increased the concentration in milk of C18:1, C18:2, C18:3 and cis9, trans11-CLA content by 16.67%, 37.36%, 95.24%, 72.22%, respectively, feeding GCS improved C18:0 and C18:1 by 17.41% and 33.28%, respectively, and feeding WCS increased C18:0 by 31.01% compared with feeding CON. Both ruminal fermentation and production trial results indicated that supplementation of extruded soybean, ground canola seed and whole cottonseed could elevate the desirable poly- and monounsaturated fatty acid and decrease the medium chain fatty acid and saturated fatty acid content of milk fat without negative effects on ruminal fermentation and lactation performance.

Induction of Apoptotic Cell Death in Human Jurkat T Cells by a Chlorophyll Derivative (Cp-D) Isolated from Actinidia arguta Planchon

  • The chloroform and methanol (2;1, v/v) extract from an edible plant, Actinidia arguta Planchon, appeared to possess antitumor activity against human leukemias Jurkat T and U937 cells through inducing apoptosis. The substance in the solvent extract was purified by silica gel column chromatography, preparative TLC, and Sephadex LH-20 column chromatography. Characteristics of the substance analyzed by UV scanning analysis, $^1H$ and $^{13}C$ NMR spectra suggested that the substance belongs to the chlorophyll derivatives-like group. The $IC_{50}$ value of the chlorophyll derivative (Cp-D) determined by MTT assay was $15\mu\textrm{g}/ml$ for Jurkat, $10\mu\textrm{g}/ml$ for U937, and $11.4\mu\textrm{g}/ml$ for HL-60m and was more toxic to these leukemias than to solid tumors or normal fibroblast. In order to elucidate cellular mechanisms underlying the cytotoxicity, the effect of the Cp-D on Jurkat T cells was investigated. When cells were treated with the Cp-D at a concentration of $15\mu\textrm{g}/ml$, [3H]thymidine incorporation declined rapidly and wa undetectable in 1h. However, no significant changes were made in the cell cycle distribution of the cells by 24h. The sub-Gl peak representing apoptotic cells began to be detectable in 36h, at which time apoptotic DNA fragmentation was also detected on agarose gel electrophoresis, demonstrating that the cytotoxic effect of the Cp-D is attributable to the induced apoptosis. Under the same conditions, although the protein level of cyclin-dependent kinases such as cdc4, csk6, cdk2, and cdc2 was not significantly changed until 24h, the kinase activity of all c안 rapidly declined and reached a minimum level within 1-6h and then recovered to the initial level by 12h and sustained until 24h. These results suggest that inactivation of cdks at an inappropriate time during the cell cycle progression in jurkat T cells following a treatment with the Cp-D leads to induction of apoptotic cell death.

  • This study is aimed to develop the adsorbent which can effectively remove toxic hydrophobic organic compounds from the aqueous phase. The emphasis was made to elucidate the adsolubilization behavior of sparingly soluble organic compounds (SSOCs) into the cetyltrimetylammonium bromide(CTAB) layer formed on anthracite by the partition coefficient. The amount of SSOCs removed from aqueous solution was increased with increase of the amount of CTAB coated on the surface and wich increase of SSOCs's hydrophobicity. With the surface-modified solid shown in above. chloroform and benzene at the initial concentration of $6{\times}10^{-4}M$ were removed over 95%. Experimentally determined partition coefficient($K_d$) values between organo-anthracite and organics were 4~25 times higher than theoretical $K_d$ values of same organics Organo-anthracite formed by the addition of the CTAB can effectively immobilize organic contaminants dissolved in landfill leachate and can also be applicable to wastewater treatment containing toxic hydrophobic organic compounds such as chloroform and benzene.
