$Tomato$$spotted$$wilt$$virus$ (TSWV)-infected $Capsicum$$annuum$ plants were collected from open fields during June to July in 2010. The TSWV isolates were designated as Gneung, Ghang-Kjj, Gchang-Njc, Ghae, and Pap. The nucleotide sequence of the nucleocapsid protein (N) and movement protein (NSm) of the five isolates was determined. The pathogenicity of the five isolates was determined on 14 $C.$$annuum$ cultivars two times by using mechanical inoculation. The five isolates induced different response: Both Gneung and Gchang-Kjj did not infect any of the cultivars in the 2nd trial, while Gchang-Njc, Ghae and Pap infected 11, 6 and 13 of 14 cultivars, respectively. The five isolates also were tested on $Solanum$$lycopersicum$ breeding line TGC09-71 and three $Nicotiana$ species. $S.$$lycopersicum$ showed a similar response to the five isolates as did $C.$$annuum$. Both Gchang-Njc and Ghae infected systemically all three $Nicotiana$ species tested. While both Pap and Gneung did not infect any of the $Nicotiana$ species tested. In conclusion, five TSWV isolates induced different infection spectra in $C.$$annuum$ cultivars, $Nicotiana$ species and an $S.$$lycopersicum$ breeding line. Amino acid sequence analysis of the NSm gene could not support or explain the different infection spectra of the five isolates. This study indicated that various isolates must be used as virus inocula for evaluation of $C.$$annuum$ and $S.$$lycopersicum$ cultivars in breeding programs for TSWV resistance.
토마토에서 과형은 과실의 여러 가지 형질 중에서 눈에 가장 잘 띄는 형질이며, 소비자의 토마토를 구매를 결정하는데 많은 영향을 미치는 중요한 형질이다. 토마토의 과형을 결정하는 여러 가지 유전자 중에 OVATE는 둥근 토마토 과일을 서양 배 모양(pear shape)의 과일로 전환하는데 결정적인 역할을 하는 유전자이다. OVATE 유전자에 의해서 과일의 모양이 변하는 것은 조기종결 코돈을 초래하는 열성 돌연변이에 의해서 유도되며, 단백질의 C-말단 영역이 제거됨에 따라 그 기능을 상실하여 나타나는 현상이다. OVATE 유전자는 주로 식물의 생식기관에서 발현되며, 꽃에서는 개악하기 10일전부터부터 전사체가 만들어지고 발달중인 과실에서는 개약 후 8일까지 전사체를 확인할 수 있다. 토마토 분자육종 과정에서 과형 판별을 위해서 OVATE 유전자 연관 분자표지는 보고된 바 있으나 OVATE 유전자 유래 분자표지는 보고된바가 없다. 본 연구에서 국내에서 육성된 육종 라인들의 resequencing을 통해 OVATE 유전자 염기서열간의 SNP를 발견하고 이들을 dCAPS 마커로 전환하여 분자표지를 개발했다. 이러한 분자표지는 둥근 토마토(round)와 서양 배모양(pear shape)토마토 육종 프로그램의 효율성과 정확성을 향상시키는데 활용할 수 있을 것으로 기대한다.
본 연구는 토마토 'Mascara' 품종을 코코피트, 피트모스, 황토와 피트모스 혼합배지에서 양액재배시 생육과 품질에 미치는 영향을 알아보고자 실시하였다. 토마토 묘를 4월 29일에 정식하였고, Yamazaki 토마토 전용 양액을 EC $2.0dS{\cdot}m^{-1}$, pH 6.5로 조정하여 공급하였다. 토마토 과실은 6월 13일부터 8월 24일까지 수확하여 생육과 품질을 조사하였다. 토마토 양액재배에 있어서 배액량은 일사량이 많고 온도가 높았을때 배액량이 감소하였으나, 코코피트, 피트모스, 황토 + 피트모스 등 배지별 배액량은 차이가 없었다. 피트모스 단용 배지에서 재배기간동안 다른 배지에 비해 배액의 EC가 낮게 나타났으며, 황토와 피트모스 혼합배지에서 높게 나타났다. 황토와 피트모스 혼합배지는 다른 배지보다 고상이 많았고, 유효수분함량이 가장 높았다. 토마토의 당도는 황토와 피트모스 혼합배지에서 $5.5^{\circ}Brix$로 가장 높았고, 피트모스 배지에서 $4.7^{\circ}Brix$로 가장 낮게 나타났다. 산도는 황토와 피트모스 혼합배지에서 0.66%로 가장 높았다. 수확량은 황토와 피트모스 혼합배지에서 8,428kg/10a로 가장 많았고, 상품성이 있는 과실수량의 비율도 다른 배지보다 더 높았다.
본 연구는 환경 친화적 방법인 브러싱을 이용한 기계적 자극의 영향을 받는 오이와 토마토 플러그 묘의 생육 억제 효과를 구명하기 위해 수행되었다. 오이(Cucumis sativus L. 'Joeunbaekdadagi')와 토마토(Solanum lycopersicum L. 'Mini Chal')를 2017년 10월 9일 상업용 혼합 상토가 충진된 40구 플러그 트레이($54{\times}27.5{\times}5cm$)에 파종하였다. 벤로형 유리온실의 재배환경은 $15-25^{\circ}C$의 재배 온도 범위와 $50{\pm}10%$의 상대습도를 유지하였다. 파종 15일후에, 오이와 토마토 묘에 무처리(대조구), $7.5mg{\cdot}L^{-1}$의 diniconazole을 처리하였다. 또한, 오이와 토마토의 brushing 처리는 2, 4, 또는 6시간 간격으로 각각 15일과 20일간 적용되었다. 1회씩 brushing 처리를 하였다. 오이와 토마토의 초장, 하배축, 절간장은 대조구에 비해 diniconazole 처리에서 억제되었다. 잎의 크기는 오이와 토마토 모두 감소하였지만, 반면에 엽록소 값은 diniconazole 처리에서 증가하였다. 그러나 오이의 경경은 2시간 brushing 간격 처리에서 가장 두꺼웠다. 지상부와 지하부의 생체중은 diniconazole 처리에서 유의적으로 낮았다. Brushing의 적용은 토마토 묘의 지상부와 지하부의 건물중, 충실도를 촉진시킴으로써 묘소질을 향상 시켰다. 토마토 묘의 엽록소 형광은 2시간 처리에서 급격히 감소하였으며, 이는 brushing 처리에 의한 기계적 스트레스를 나타낸다. 토마토 묘의 상대 생장률은 diniconazole 처리에서 유의적으로 낮았지만, 오이 묘는 모든 처리에서 유의적인 차이가 없었다. 결과적으로, 오이와 토마토 묘의 생육 억제는 생장조절제의 화학 물질에 의한 diniconazole 처리에서 가장 효과적이었다. 그러나 환경 친화적인 관점에서, 2시간의 brushing 간격 처리는 오이와 토마토 묘의 생장에서 화학적 방법을 대체할 수 있는 응용 가능성을 가지고 있다고 판단된다.

토마토풋마름병에 저항성인 $Solanum$$lycopersicum$ H7996와 극도감수성인 $S.$$pimpinellifolium$ WVa700 간의 교배를 통해 획득한 재조합순계계통 $F_9$ 세대의 188개체를 이용하여 유전자연관지도를 작성하였다. 유전자지도는 DarT 260종, AFLP 74종, RFLP 4종, SNP 1종 및 SSR 22종 등 총 361종의 마커로 구성되었다. 작성된 유전자지도는 총 13개의 연관군(LG)에 2042.7cM을 포함하였으며 마커간의 평균지도거리는 5.7cM이고 이중 DArT마커는 평균 7.9cM당 1개가 분포하였다. SSR 마커의 분포를 기초로 작성된 11개 연관군들은 토마토 염색체의5번과 12번을 제외한 10개 염색체에 해당하였다. DArT 마커는 다른 마커들처럼 토마토 유전체 상에 고르게 분포하였으며, 인접 마커와의 상호분석(${\leq}$ 0.5cM) 결과 클러스터링 빈도가 13.5%인 AFLP 마커보다 3배 정도 높은 38.8%의 빈도로 최고치를 나타내었다. 본 연구를 통해 토마토에서 최초로 DarT 마커를 이용한 유전자연관지도를 작성하였다.
진핵생물의 유전자 발현 과정에서 생성된 단백질은 전사후 변형 과정을 통해 소포체와 골지체에서 당질화가 일어난다. 당질화된 당단백질은 접힘의 오류가 있는 경우를 비롯하여 식물의 분화 조절 등의 경우 당단백질이 분해되며, 이 때 PNGase에 의해 N-당사슬이 단백질의 아스파라긴산 잔기로부터 절단된다. 그러나 식물의 발달과 분화 과정에서 PNGase의 발현 조절에 대해서는 거의 알려진 바가 없다. 기존에 보고된 유전적 정보를 활용하여 토마토의 잎에서 제조된 cDNA library에서 nested RT-PCR을 통하여 PNGase T의 유전자(GenBank Accession number KM401550)를 분리하였는데 이의 ORF는 1,767 bp, 588개의 이미노산으로 이루어졌으며, 분자량은 65.8 KDa이었다. PNGase T의 유전자는 토마토 과육에서 높은 수준으로 항시적으로 발현되었으며, 특히 녹색과보다는 오렌지색으로 숙성되는 과정에서 PNGase T의 전사체량이 크게 증가하였다. 이러한 발현 패턴은 토마토 과육에서 세포죽음의 과정에서 증가하는 단백질 가수분해 효소인 metacaspase의 전사체 증가 페턴과 유사하였으며, 이 시기에는 에틸렌의 생합성 효소 중 노화관련 ACC synthase의 유전자 members (LeACS2, LeACS4, LeACS6)의 발현 패턴과도 유사하였다. 따라서 토마토 과육에서 PNGase T의 유전자 발현은 거대분자가 분해되는 시기에서 과육의 숙성과 노화 과정에서 특이적인 생리적 기능을 나타내는 것으로 판단된다. 향 후 고가의 의약용 재조합단백질의 면역부작용을 완화하기 위하여 식물체 유래의 당단백질의 탈당질화과정에서 PNGase T를 활용함으로써 식물생명공학 분야에서 활용가치가 높을 것으로 사료된다.

The inhabitation and parasitism of root-knot nematodes (RKNs) can be difficult to control, as its symptoms can be easily confused with other plant diseases; hence, identifying and controlling the occurrence of RKNs in plants remains an ongoing challenge. Moreover, there are only a few biological agents for controlling these harmful nematodes. In this study, Xenorhabdus sp. SCG isolated from entomopathogenic nematodes of genus Steinernema was evaluated for nematicidal effects under in vitro and greenhouse conditions. The cell-free filtrates of strain SCG showed nematicidal activity against Meloidogyne species J2s, with mortalities of > 88% at a final concentration of 10%, as well as significant nematicidal activity against the three other genera of plant-parasitic nematodes in a dose-dependent manner. Thymine was isolated as active compounds by assay-guided fractionation and showed high nematicidal activity against M. incognita. Greenhouse experiments suggested that cell-free filtrates of strain SCG efficiently controlled the nematode population in M. incognita-infested tomatoes (Solanum lycopersicum L., cv. Rutgers). In addition, a significant increase in host plant growth was observed after 45 days of treatment. To our knowledge, this is the first to demonstrate the nematicidal activity spectrum of isolated Xenorhabdus species and their application to S. lycopersicum L., cv. Rutgers under greenhouse conditions. Xenorhabdus sp. SCG could be a promising biological nematicidal agent with plant growth-enhancing properties.

A virus causing mottle and stunt symptom on Zea mays was observed around Ulleng-do, Korea and identified as Cucumber mosaic virus (CMV-ZM) based upon biological, serological, and molecular characteristics. In host range studies, the CMV-ZM isolate produced local lesions on Datura stramonium, Vigna unguiculata, Cucurbita moschata, Chenopodium amaranticolor, Ch. quinoa, whereas this isolate produced systemic mosaic on Nicotiana tabacum cv. 'Xanthi-nc', Capsicum annuum, Solanum lycopersicum, Solanum melongena, Cucurbita pepo, and Z. mays. In addition, chlorotic local rings on inoculated leaves along with severe mosaic, malformation, and fern leaf symptoms on upper systemic leaves were shown in N. glutinosa plants. Complete nucleotide sequences of each genomic RNA segment was determined and compared to those of the other CMV strains. Comparison of the nucleotide sequence of 1a open reading frame (ORF) revealed approximately 89.2-92.4% sequence identity with each CMV subgroup IA and IB strain, while showing only 78% sequence identity with CMV subgroup II. Nucleotide sequence analysis of RNA2 ORFs revealed 85.3-97.6% sequence identity with subgroup I. In ORFs of RNA3, levels of nucleotide sequence identities were higher than 92-99.2% with CMV subgroup I and lower than 82% with CMV isolates of subgroup II. These results suggest that CMV-ZM isolate is more closely related to subgroup I than subgroup II and therefore, CMV-ZM isolate might be classified into as CMV subgroup I based on biological and molecular analysis.

Physicochemical characteristics and heavy metal content of soils located along the drainage way of an abandoned mine at Busan, Korea ($35^{\circ}31'N$, $129^{\circ}22'E$) (contaminated soil; CS) and uncontaminated soils (50-70 m apart from the drainage way) (NS) were examined. Growth of tomato plants (Solanum lycopersicom cv. Rutgers) in CS and NS, development of the root-knot nematode (Meloidogyne incognita) as root-knot gall formation on tomato plants, and non-parasitic nematode populations in soil were also examined. Growth of tomato plants, root-knot gall formation, and non-parasitic nematode populations were significantly reduced in CS with higher As content, lower pH, higher electrical conductivity (EC), and lower available phosphate (av. $P_2O_5$) than in NS. None of the other physicochemical characters examined differed significantly between CS and NS (low and no significance) and were above or below the critical levels detrimental to plant growth and nematode development, suggesting that As may be the primary hazardous heavy metal in CS. The toxicity of As might be enhanced at low pH in CS because exchangeable forms of some heavy metals increase with the decrease of soil pH. The heavy metals, especially As, may have contributed to increasing EC and decreasing av. $P_2O_5$. Therefore, the effects of mine drainage contamination from the abandoned mine were derived primarily from contamination by heavy metals such as As. These may have been enhanced in toxicity (solubility) by the lowered pH, increased soil salinity (EC) and decreased av. $P_2O_5$. Our results suggest synergistic adverse effects on the plant and the nematode by decreasing osmotic potential and nutrient availability.
The efficacy of plant breeding has been enhanced by application of molecular markers in population screening and selection. Pepper (Capsicum annuum L.) is a major staple crop that is economically important with worldwide distribution. It is valued for its spicy taste and medicinal effect. The aim of this study was to discover single nucleotide polymorphisms (SNPs), microsatellite markers information, and percentage sharing through orthologous analysis of pepper-specific pungency-related genes. Here, we report the results of transcriptome analysis and microsatellite markers for four pepper varieties that possess a pungency-related gene. Orthologous analyses was performed to identify species-specific pungency-related genes in pepper, Arabidopsis thaliana L., potato (Solanum tuberosum L.), and tomato (Solanum lycopersicum L.). Advancements in next-generation sequencing technologies enabled us to quickly and cost-effectively assemble and characterize genes to select molecular markers in various organisms, including pepper. We identified a total of 9762, 7302, 8596, and 6886 SNPs for the four pepper cultivars Blackcluster, Mandarine, Saengryeg 211, and Saengryeg 213, respectively. We used 454 GS-FLX pyrosequencing to identify microsatellite markers and tri-nucleotide repeats (54.4%), the most common repeats, followed by di-, hexa-, tetra-, and penta-nucleotide repeats. A total of 5156 (15.9%) pepper-specific pungency-related genes were discovered as a result of orthologous analysis.

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
