Effect of Phosphorus Removal by Oyster Shell on Longevity of Constructed Wetlands (굴패각에 의한 인 처리가 인공습지의 수명에 미치는 영향)

  • BACKGROUND: Constructed wetlands are low-cost alternatives for treating domestics sewage. However, previous study has reported that the removal of phosphorus in constructed wetlands was limited. Therefore, a new alternative was needed to extend the life of the constructed wetlands. The purpose of this study was to evaluate the effect of total phosphorus removal by oyster shell on longevity of constructed wetlands for treating domestic sewage. METHODS AND RESULTS: The changes of total phosphorus concentration and treatment efficiency in two constructed wetlands (CWs) classified as system A (coarse sand 100%) and system B (coarse sand 90%+oyster shell 10%) were investigated for 6 years. The actual saturation time of total phosphorus in the systems A and B was estimated to be longer than that of theoretical saturation by adsorption isotherm experiment. In particular, the saturation pattern of phosphorus in system A was maintained at a certain concentration level in the initial stage of operation, and finally saturation was reached as the saturation gradually progressed from the breaking point. In system B, the saturation period of phosphorus was prolonged as compared with system A due to the addition of oyster shells. CONCLUSION: Our results suggest that the longevity of the constructed wetlands can be extended due to the phosphorus saturation by adding the oyster shells to the coarse sands in constructed wetlands.

Hydrogen Peroxide- and Nitric Oxide-mediated Disease Control of Bacterial Wilt in Tomato Plants

  • Reactive oxygen species (ROS) generation in tomato plants by Ralstonia solanacearum infection and the role of hydrogen peroxide ($H_2O_2$) and nitric oxide in tomato bacterial wilt control were demonstrated. During disease development of tomato bacterial wilt, accumulation of superoxide anion ($O_2{^-}$) and $H_2O_2$ was observed and lipid peroxidation also occurred in the tomato leaf tissues. High doses of $H_2O_2$ and sodium nitroprusside (SNP) nitric oxide donor showed phytotoxicity to detached tomato leaves 1 day after petiole feeding showing reduced fresh weight. Both $H_2O_2$ and SNP have in vitro antibacterial activities against R. solanacearum in a dose-dependent manner, as well as plant protection in detached tomato leaves against bacterial wilt by $10^6$ and $10^7$ cfu/ml of R. solanacearum. $H_2O_2$- and SNP-mediated protection was also evaluated in pots using soil-drench treatment with the bacterial inoculation, and relative 'area under the disease progressive curve (AUDPC)' was calculated to compare disease protection by $H_2O_2$ and/or SNP with untreated control. Neither $H_2O_2$ nor SNP protect the tomato seedlings from the bacterial wilt, but $H_2O_2$ + SNP mixture significantly decreased disease severity with reduced relative AUDPC. These results suggest that $H_2O_2$ and SNP could be used together to control bacterial wilt in tomato plants as bactericidal agents.

Effects of Environmental Infrastructure Regeneration in Urban Region - A Case Study of M Apartment Complex in Daejeon (도시 단지내 환경 인프라 재생 효과 고찰 - 대전시 M 아파트 단지 사례연구)

  • This study introduces a resource recycling system in urban apartment complex using four different technologies. The four technologies, called 4G, include a production technology for reclaimed water (Green water), a biogas production technology from organic waste (Green biogas), a reuse technology of rainwater (Green rainwater), and urban agropark (Green pyramid). Green water is the technology for producing the reclaimed water from wastewater, rainwater and underground water, and the average concentrations of BOD, SS, T-N and coliform of reclaimed water were 7.8mg/L, ND (not detected), 4.9mg/L and ND, respectively. Green biogas is the technology for producing biogas and effluent after treating organic wastes (e.g. food waste and night soil) discharged from households, and the average production rates of hydrogen and methane were $0.33m^3/m^3/d$ and $0.24m^3/m^3/d$, respectively. Green pyramid, agricultural farm operated by biogas and reclaimed water, provides a healthy and recreational space for residents, and plant growth rates using treated water and reclaimed water showed height of 1.32cm and weight of 112.8g. Therefore, 4G technologies can improve the recycling rate and treatment efficiencies of waste and wastewater in an apartment complex.

Physicochemical changes and plant growth effect on composting of spent mushroom substrates (버섯수확후배지의 퇴비화에 따른 물리 화학적 변화 및 식물 생육 효과)

  • This study aimed to assess the feasibility of composting spent mushroom substrate (SMS) materials of Lentinula edodes (Le), Hericium erinaceus (He), and Pleurotus ostreatus (Po). The different SMSs were composted for 7 to 10 days at high temperatures over 50℃; the composting procedure was completed in 30 days. A maturity test was conducted using the radish seed germination index and CoMMe-100. The composted SMS (CSMS) from Le and He showed gemination indices of 130% and 81%, respectively, that satisfied the criteria of maturity standard (gemination index over 70%) and the CoMMe-100 analysis. The physicochemical changes of CSMSs included an increase in the pH range from 4-5 to 6-7, slight reduction in the EC to 1-1.4 dS/m, and an organic content of 36.9% in LeCSMS. In LeCSMS, the contents of N, P, and K were 1.2%, 2.3%, and 0.77%, respectively, and heavy metals were detected below the standard value in all CSMSs; the Ca and Mg contents in the CSMSs were increased from 30% to 60% when compared to those in the SMSs. The C/N ratio (from 26-33) in LeSMS and HeSMS decreased to 15.3-15.9 in CSMSs. The growth effect of LeCSMS treatment on pepper seedlings was 60% higher than that in the control groups, one of which was treated with commercial organic compost; the former showed a superior growth effect on the leaf width, leaf length, and leaf number compared to other control groups. In conclusion, LeCSMS and HeCSMS could be utilized as compost resources capable of efficient soil amendment and plant growth promotion.

  • From the tuberous root of Aconitum carmichaeli Debx.(Ranunculaceae), the main root is called as common monkhood mother root and the later root is called as the prepared aconite root. From the prepared aconite root. Looking at the processing method of the prepared aconite root, it is divided into Yeombuja (prepared aconite root processed in salt) and heuksoonpyeon (baekbupyeon) following the processing method after removing the soil and this is a way of processing the prepared aconite root without damage it. The recently produced raw prepared aconite root is easily damaged, thus it shall be preserved in salt to have the crystal shape on the surface of the prepared aconite root and store and transport in firmly solidified yeombuja condition. Therefore, yeombuja shall remove the salt before use and requires processing for use but heuksoonpyeon or baekbupyeon may use immediately. For the succession of the unique processing techniques of our ancestors, there has to be studies on the techniques. Prepared aconite root is generally used as holy medicines to cure the yang depletion syndrome, kidney-yang deficiency syndrome, and obstruction of qi in the chest syndrome. However, they are the substances with toxicity. It is contemplated that the contents of processing are broadly understood through the document on the processing method, and based on such foundation, the systematic set and proof on the documents are made along with the addition of the contemporary scientific theory and technology to develop the traditional processing technology to maximize the treatment effect and safety of prepared aconite root. In this study, the historic data and records on the processing method of latteral root of aconitum carmichaeli Debx will be rearranged to contribute to the standardization of medicinal herbs, maximization of efficacy and minimization of the side effects.

Study on the Application Method of Fertilizers and Leaves and Stems of Irish Potato on Rice Crop Cultivated After Potato (감자후작 벼에 대한 감자경엽 및 비료 사용 방법 개선에 관한 연구)

  • The study was carried out to find out the reasonable fertilization method for rice in the area of Irish potato cultivated paddy where has various nutritional disorders and pest problems. The treatment consisted of three different application methods of potato sterns and leaves with 4 levels of nitrogen application in each method. The application of potato sterns and leaves caused more release of $NH_4$ -N in the soils but the growth of rice were slightly hindered by the strongly reduced soil conditions when the nitrogen had been applied more than 10kg/10a. The application of half-dried or semi-decomposed stems and leaves of potatoes were caused to increase ripening ratio and 1,000 grain weight of rice with a relatively high $SiO_2$/N ratio in the rice plant, so the yield of rice increased 3-5% than the plot without potato sterns and leaves. The potash and silicate fertilizer would seem to be effective in the area of Irish potato cultivating paddy field since the contents of extractable potassium and soluble silicate in the soils is lacked. The suitable amount of nitrogen on rice cropping seems to be about 7kg/10a.

A Study on the Applicability of Levee Leakage Monitoring System Using Movable TDR Sensor (제방 누수 모니터링을 위한 이동식 TDR 센서의 적용성 평가)

  • Several types of methods such as resistivity survey, ground penetration radar, etc are used for detection of levee leakage and according to the river design guidelines detection of levee leakage is performed by measuring the hydraulic conductivity of levee soil. But, the former can not verify the leakage point and degree of saturation, the latter is an after treatment method. Movable sensor, which is a high-tech TDR system developed since 2000, can obtain directly the dielectric constant profile covering the whole depth of levee. In this study, laboratory and field model experiments were carried out using movable TDR sensor in order to evaluate the applicability as detection system of levee leakage, As the result, movable TDR system has proven to be 3 times more sensitive to water contents than dry unit weight, and the results conclude that the dielectric constant, water contents and density of the ground proved to have a correlation among them, and the dielectric constant is expected to be a basic data on detection of levee leakage.

Enhanced drought and oxidative stress tolerance in transgenic sweetpotato expressing a codA gene (CodA 고발현 형질전환 고구마의 산화 및 건조 스트레스 내성 증가)

  • Glycine betaine (GB) is one of the compatible solutes that accumulate in the chloroplasts of certain halotolerant plants under salt or cold stress. The codA gene for choline oxidase, the enzyme that converts choline into GB, has been cloned from a soil bacterium Arthrobacter globiformis. We generated transgenic sweetpotato plants [Ipomoea batatas (L.) Lam] expressing codA gene in chloroplasts under the control of the SWPA2 promoter (referred to as SC plants) and evaluated SC plants under oxidative and drought stresses. SC plants showed enhanced tolerance to methyl viologen (MV)-mediated oxidative stress and drought stress due to induced expression of codA. At $5{\mu}M$ of MV treatment, all SC plants showed enhanced tolerance to MV-mediated oxidative stress through maintaining low ion leakage and increased GB levels compared to wild type plants. When plants were subjected to drought conditions, SC plants showed enhanced tolerance to drought stress through maintaining high relative water contents and increased codA expression compared to wild type plants. These results suggest that the SC plants generated in this study will be useful for enhanced biomass production on global marginal lands.

Development of Biofungicide Using Bacillus sp. KBC1004 for the Control of Anthracnose of Red Pepper (길항세균 Bacillus sp. KBC1004를 이용한 고추탄저병의 생물학적 방제제 개발)

  • To develop an effective biopesticide to control pepper anthracnose disease, an isolate which showed strong inhibitory effect on the mycelial growth and conidial germination of Colletotrichum acutatum was selected among the antagonistic bacterial isolates collected from pepper grown soil. The bacterial isolate was identified as Bacillus sp. KBC1004 using 16S rRNA sequence analysis. The liquid culture of KBC1004 was freeze-dried and formulated as a wettable powder(WP). The wettable powder form of KBC1004 required at least 24 hours to activate and to inhibit the conidial germination of C. acutatum. In vitro bioassay using the detached green pepper fruits, biocontrol activity of the WP was not recognizable in simultaneous inoculation, but significant disease suppression was observed pre-treatment (24 hr) of the WP before pathogen inoculation. In field experiment, 4 times foliar applications of the 1/500 diluted wettable powder from the end of June showed great control efficacy similar to that of the chemical fungicide application. These results suggest that the formulated WP product could be an alternative mean to control of pepper anthracnose disease in environmentally friendly farming practices.

Groundwater Environment of the Okmyong Waste Landfill in the Pohang City (포항 옥명 폐기물 매립지의 지하수 환경)

  • The Okmyong Waste Landfill which is one of major industrial waste landfills in Korea has been used for 10 years since January, 1988. The groundwater in range of 2∼3 km from the landfill is a little acidic and has high concentrations of EC, NO$_3$, and SO$_4$ because of acidic rain in Pohang. The groundwater pattern in Piper's trilinear diagram belongs to Ca-SO$_4$and Na-SO$_4$types, but Ca-SO$_4$type is more predominant. The groundwater of five monitoring wells at the landfill area is a litle alkaline and has high concentrations of TS, COD, Na, Cl, SO$_4$, Alkalinity and F because of the effect of the leachates. They also have high heavy metals of Mn, Zn, Pb, Cr and Ni but toxic organic compounds are not detected. Their groundwater pattern is Na-SO$_4$type and distinguished from the groundwater pattern Na-Cl types of the Nanjido and the Seokdae Municipal Waste Landfills. The range of groundwater contamination which is validated up to now is about 120m west from the entrance of the Okmyong Waste Landfill. To prevent the dispersion of the leachates to the outside, cutoff walls should be constructed at the boundary of the landfill. Several pumping wells should also be developed at the landfill so that the contaminated groundwater can be pumped and treated at a leachate-treatment plant in the landfill.
