It is well known now that near infrared spectroscopy (NIRS) is a fast, no destructive, and inexpensive analytical technique that could be used to classify, identify, and authenticate a wide range of foods and food items. Therefore, the main aims of this study were to provide a new insight into the authentication of five strawberry (Fragaria x ananassa) varieties and to correlate them with geographical zones and the propagating methods used. Three weeks plants of five different strawberry varieties (F. x ananassa Duch. cv Camarosa, Seascape, Chandler, F. Chiloensis, and F. Virginiana) were cultivated in vitro first and then transferred to pots with special soil, and grown in a greenhouse at CINVESTAV, all varieties were acquired from California (USA). After 18 months, ten leaves from each variety were collected. Transmission spectra from each leave were recorded over a range of 10, 000-4, 000 cm$-^{1}$, 32 scans of each strawberry leave were collected using a resolution of 4 cm$-^{1}$ with a Paragon IdentiCheck FT-NIR System Spectrometer. Triplicates of each strawberry leave were used. All spectra were analyzed using principal component analysis (PCA) and soft independent modeling class analogy (SIMCA). The optimum number of components to be used in the regression was automatically determined by the software. Camarosa was the only variety grown from the same shoot but propagated by a different method (direct or in vitro). Five different classes (varieties) or clusters were observed among samples, however, larger inter class distances were presented by the two wildtype samples (F. Chiloensis and F. Virginiana). Camarosa direct and Camarosa in vitro displayed a small overlapping region between them. On the other hand, Seascape variety presented the smallest rejection percentage among all varieties (more similarities with the rest of the samples). Therefore, it can be concluded that the application of NIRS technique allowed the authentication of all strawberry varieties and geographical origin as well. It was also possible to form subclasses of the same materials. The results presented here demonstrate that NIRS is a very powerful and promising analytical tool since all materials were authenticated and classified based on their variety, origin, and treatment. This is of a tremendous relevance since the variety and origin of a plant material can be established even before it gives its typical fruit or flower.

This study was carried out to develop a controlled-release insecticide formulation for the control of rose aphid (Macrosiphum ibarae) in rose and cotton aphid (Myzus persicae) in chrysanthemum (Dendranthema grandiflorum var. Meibung) in greenhouse. Imidacloprid[1-(6-chloro-3-pyridylmethyl)-N-nitroimidazolidin-2-ylideneamine] was chosen as a toxicant. Two synthetic polymers, low density polyethylene(LDPE) and ethylene vinyl acetate(EVA), were used as polymer matrices. The tested plastic sheet formulations were prepared by heat-aided extrusion procedures after mixing imidacloprid technical and the polymers of three different combinations, and physicochemical properties as well as efficacy of the formulations were investigated. The amounts of imidacloprid recovered and incorporated in the formulations were recorded over 90% and 80%, respectively. Release of the active ingredient from the formulations was remarkably affected by mixing rates of polymers. The active ingredient in the formulations was chemically unstable with over 10% degradation rates after 90 day storage at $50{\pm}2^{\circ}C$. The residual amounts of imidacloprid in the soil treated with the formulations were paralleled with the release pattern of the formulations. Efficacy of the formulations on rose and cotton aphid was maintained over 90% even 120days after treatment under greenhouse.
This study was conducted to investigate the effect of fine bubble water treatment on the growth of ginseng seedlings. The growth of ginseng seedlings which were treated with general water and fine bubble water was investigated. The above ground part, the growth of leaf was increased by about 10% and the growth of the stem was about 6%, as the ginseng seedlings which were treated with fine bubble water comparing to the ginseng seedlings treated with general water. Root length was increased about 5%, root width was increased about 8%, roots weight was increased about 9%, and dry weight was increased about 7%. This is not because the whole root growth was increased, but the main root growth was increased about 7%. These results suggested that the physical properties of the fine bubble water stimulated the growth of ginseng seedlings. These results are initial study in the case of ginseng seedlings. Therefore, it can be applicable to the 3-5 years old ginseng plants. Further research will be needed to find out the optimal cultivation condition by controlling the dissolved oxygen amount according to the soil condition and the research applied to the ginseng seedlings.

This experiment was carried out to investigate the effects of different covering materials and methods on heat insulation of a plastic greenhouse, growth and yield of tomato. Night air and soil temperatures in a double-layer greenhouse with external multifold thermal cover (MTC; eight-ounce cassimere+four-fold polyform+double-fold non-woven fabric+single-fold polypropylene covering were about $1^{\circ}C$ lower than in that with internal MTC covering, but about $3^{\circ}C$ higher than in that with an EVA film screen. Tomato yield in the external MTC covering increased by $2\%\;and\;19\%$ as compared to that in the internal MTC covering and the non-covering of MTC, respectively, due to its high light transmission and insulation effect. Night air temperatures in a double-layer greenhouse with external MTC covering and with thermal screen (polyester plus aluminium) were $2.2^{\circ}C\;and\;4.5^{\circ}C$ higher than those in a double-layer greenhouse with an external MTC covering and in a double-layer greenhouse equipped an EVA film screen, respectively. Tomato yield in the treatment with external MTC covering and a thermal screen was $18\%\;and\;37\%$ greater than that in the external MTC covering and in an EVA film screen, respectively. Results indicate that tomato could be grown without heating or with minimal heating in a double-layer greenhouse covered with MTC and a thermal screen during the winter season in sourthern regions of Korea.

The effect of biological control of the root-knot nematode, Meloidogyne incognita, on cucumber and lettuce was evaluated with green manure crop species in greenhouse. Nematicidal effect of sudan grass cultivation in cucumber greenhouse was comparable to that of chemical treatment with fosthiazate GR, showing the high activity of 88.6%. Sudan grass cultivation in lettuce greenhouse significantly reduced the number of M. incognita in soil, showing 93.5% of nematiidal activity. In addition, since growth of sudan grass was superior to other green manure crop species, it is considered that cultivation of sudan grass is proper to control M. incognita in greenhouse.
Paenibacillus polymyxa is a plant growth-promoting rhizobacterium (PGPR) which can be used for biological control of plant diseases. Several bacterial strains were isolated from rotten roots of Korean ginseng (Panax ginseng C. A. Meyer) that were in storage. These strains were identified as P. polymyxa, based on a RAPD analysis using a P. polymyxa-specific primer, cultural and physiological characteristics, an analysis utilizing the Biolog system, gas chromatography of fatty acid methyl esters (GC-FAME), and the 16S rDNA sequence analysis. These strains were found to cause the rot in stored ginseng roots. Twenty-six P. polymyxa strains, including twenty GBR strains, were phylogenetically classified into two groups according to the ERIC and BOX-PCR analyses and 16S rDNA sequencing, and the resulting groupings systematized to the degrees of virulence of each strain in causing root rot. In particular, highly virulent GBR strains clustered together, and this group may be considered as subspecies or biovar. The virulence of the strains seemed to be related to their starch hydrolysis enzyme activity, but not their cellulase or hemicellulase activity, since strains with reduced or no starch-hydrolytic activity showed little or no virulence. Artificial inoculation of the highly virulent strain GBR-1 onto the root surfaces of Korean ginseng resulted in small brown lesions which were sunken and confined to the outer portion of the root. Ginseng root discs inoculated in vitro or two-year-old roots grown in soil drenched with the inoculum developed significant rot only when the inoculum density was $10^{6}-10^{7}$ or more colony-forming units (CFU) per ml. These results suggest that P. polymyxa might induce ginseng root rot if their population levels are high. Based on these results, it is recommended that the concentration of P. polymyxa should be monitored, when it is used as a biocontrol agent of ginseng, especially in the treatment of stored roots.

This study was conducted to evaluate the removal efficiencies of organic matter and pharmaceuticals and to identify the removal mechanism of pharmaceuticals using sand obtained from Hwangryong River in Jangsung. Batch and column studies were used to simulate managed aquifer recharge (MAR) systems. All experiments were performed using field effluent containing pharmaceuticals from Damyang Wastewater Treatment Plant as an influent. Based on the removal results of organic matter and pharmaceuticals from the batch and column experiments, soil organic matter (SOM) and microbial activity were found to effectively remove target contaminants. The removal of organic matter was found to increase under biotic conditions. Neutral and cation pharmaceuticals (iopromide, estrone, and trimethoprim) exhibited removal efficiencies higher than 70% from natural sand and baked sand media in batch and column studies. Carbamazepine persisted in the sand batch and column studies. Anion pharmaceuticals (ketoprofen, ibuprofen, and diclofenac) can be removed under conditions featuring high SOM and adenosine triphosphate (ATP) concentrations in the sand surface. Based on the experimental Batch and column results, biodegradation and sorption were found to be important mechanisms for the removal of pharmaceuticals within the simulated MAR systems.

Host rock or the standing sculptured Buddha in the Yongamsa temple is macular porphyritic biotite granite, which has gone through mechanical and chemical weathering. The rock around the Buddha statue is busily scattered with steep inclinations that are almost vertically discontinuous planes with the strikes of $N8^{\circ}E$. Especially the development of the joints that cross the major joints causes the structural instability of the rock. The rock of the Buddha statue is separated into several rock blocks because of many different discontinuity. Thus it is estimated that the bed rock has not only plane and toppling failure but also wedge failure in all the sides. Since the differential pressure is imposed on the body of the Buddha in the host rock, it is urgent to give a reinforce treatment of geotechnical engineering for the safe of its structural stability. Very contact area of joints have turned into soil, which promotes the growth of weeds and plant roots, then aggravates the mechanical weathering of the rock. Thus conservational treatments should also be considered to get rid of secondary contaminants and vegetation along the discontinuities and to prevent further damages.
An actinomycetes F-97 producing tyrosinase inhibitor was isolated from soil samples. Isolation and purification of tyrosinase inhibitor produced by F-97 was performed as follows: IRC-120 ($NH_4^+$ type) column chromatography, silica gel column chromatography, $C_{18}$ column chromatography and Sephadex LH-20 column chromatography were used successively after the centrifuged supernatant was adjusted to pH 4.0. To identify the purity of the inhibitor, octadecylsilyl(ODS) HPLC was carried out with 5% methanol as a mobile phase. Finally, the purification yield of a tyrosinase inhibitor was 5.24%. The inhibitor was very soluble in water, methanol and ethanol but insoluble in acetone, butanol, ethylacetate and chloroform. The ${\lambda}_{max}$ value of this inhibitor in water was 194nm under UV light. The biochemical test of the inhibitor was positive in Molish, Benedict, cone. $H_2SO_4$, and $KMnO_4$ tests but negative in iodine, ninhydrin, Million, Sakaguchi, xanthoproteic and Emerson tests. The tyrosinase inhibitor was stable against heat treatment of $100^{\circ}C$ for 50 minutes and pH $4{\sim}9$. The $IC_{50}$ value of this inhibitor was $19.2{\mu}g/ml$ for mushroom tyrosinase. In $1,000{\mu}g/ml$ inhibitor concentration, inhibition zone was 27 mm for Streptomyces bikiniensis NRRL B-1049. The inhibition of F-97 against mushroom tyrosinase was competitive with tyrosine.
Paenibacillus sp. JB-13 producing the cyclodextrin glucan-otransferase(CGTase) [EC 2.4.1.19] that glucosylated ascorbic acid(AA) at the C-2 position was isolated form soil and the optimal conditions for the production of 2-O-$\alpha$-D- Glucopyranosl L-Ascorbic acid(AA-2G) with CGTase were investigated. CGTase produced AA-2G efficiently using dextrin as a substrate and AA as an aceptor. Several AA-2-oilgosaccharides(AA-2Gs) were also produced in this reaction mixture, and these were efficiently hydro-lyzed to AA-2G and glucose by the treatment with glucoamylase. The optimal temperature for AA-2G production was $37^{\circ}C$ and the optimal pH was around 6.5. CGTase also utilized $\alpha$-,$\beta$-,${\gamma}$-CDs, soluble starch, com statch, dia-static solution from rice and diastatic solution from malt as substrate, but not glucose. The reaction mixture for the maximal production of AA-2G was following; 15% total substrate concentration, 2,500 units/ml of CGTase and a mixing ration of 3:2(g of AA: g of dextrin). Under this condition, 56 mM of AA-2G ,which corresponded to 12.4% yield based on AA. was produced after incubation for 44 hrs at $37^{\circ}C$ and pH 6.5.
