These experiments were conducted to evaluate the effects of seed coating with ten environmentally acceptable polymers, on germination percentage, water uptake, respiration, emergence and seedling growth characteristics. The water absorption of polymer-coated seeds in saturation condition was highest in klucel and lowest in polyvinyl pyrrolidone(PVP), and that in water was highest in klucel and lowest in maltrin. Respiration rates of polymer-coated seeds in Hwayoungbyeo and Ilpumbyeo were lower than those of none-coated seeds, and those in Daecheongbyeo and Jinmibyeo were higher than that of none-coated seeds, and those of sepiret coated seeds were higher than PVP coated seeds. The germination polimer-coated seed was reduced by one percentage by coating with seed coating machine. Germination percentage was not affected by any of polymer coating in high quality seed, but there were significant effects in low quality seed coating with waterlock, surelease 46 and sepiret significantly reduced germination some cultivars. Germination percentage after accelated ageing treatment were slightly higher most of polymer-coated seeds than in none-coated seeds, but those of sepiret-and klucel-coated seeds were lower significantly. Germination percentage of seeds coated with daran 8600, rnaltrin, sacrust and opadry were enhanced slightly under cold test other polimers reduced germination. The seedling height of polymer-coated seeds were longer than those of none-coated seeds, but those of waterlock, PVP and maltrin coating seeds were shorter, seedling hight was shortened by polimer coating under cold test. Polymer-coated seeds showed higher emergence percentage, shorter emergence time in field condition. The highest emergence percentage and the shortest emergence time was shown at 90% soil moisture content.
In order to find optimum conditions for somatic embryogenesis from different individual (2-year-old) in Aralia elata were cultured on MS medium supplemented with 1.0 mg/L 2,4-D, 3% sucrose, and 0.3% gelrite. We also investigated the effect of MS medium salt concentration, BA and ABA on the embryo germination and plant regeneration. While noticeable difference was observed on somatic embryo induction among different individual tree, no apparent difference was seen in both germination and regeneration frequencies. Compared with nonembryogenic calli, embryogenic calli tended to look yellow and/or pale brown in color, slowly growing and soft in their texture. Regardless of BA or ABA treatment, half-strength MS salt medium proved to be better than full strength MS medium in both embryo germination and plant regeneration. Both hypocotyl and cotyledon developments were slightly promoted by adding 0.1 mg/L BA. However, its effect on germination and regeneration seemed inferior to control. ABA treatment on somatic embryos at their torpedo and early cotyledonary stages resulted in poor response in germination and regeneration. Although most regenerated plantlets varied greatly in cotyledon number and shape, they could be developed into normal plants after 4 weeks in culture. More than 95% plantlets were acclimatized in an artificial soil mixture, successfully transplanted to nursery bed and grew normally without any phenotypic abnormalty.
In this study, a propagation characteristics of green kyllinga(Kyllinga brevifolia var. leiolepsis H.) was investigated. Two to three rhizomes(1st rhizome) arised from the basal part of main shoot at 4th leaf stage and were grown to extend on the soil surface. Then new shoots(lst tillers) emerged from every node of the 1st rhizome. Second rhizomes also arised from the basal part of lst tiller at the time of 4th leaf openning. With such a regularity, 5th rhizomes and about 688 shoots were produced from one plant during one year-cultivation without competition under the natural condition. The degree of vegetative growth and seed formation was 3 times and 2.5 times higher in rhizomeoriginated plant than in that from seed, respectively. The amount of seed formation and the 1000 seeds weight was highest in one planted on June 1 and Aug. 1, respectively. Short-day treatment of less than 14hr appeared to be necessary for the induction of flowering and it was effective as treated not during germination but since at least 2 leaf stage of green kyllinga. Each shoot individually responded to short-day. When plants were exposed to short-day (9hr, day / 15hr, night) treatment at the stage of 2-3 leaves, more than 7 cycles were required for flowering induction. Bolting in main shoot occurred after emergence of 4 leaves under the short-day condition. Exogenous $GA_3$ slightly accelerated the velocity of bolting only in short-day condition.
The potential of juglone a plant naphthoquinone as a natural herbicide on new target, 7-keto-8-amino pelargonic acid synthetase (KAPAS) in the early step of biotin biosynthesis pathway, was performed in vitro and in vivo. Juglone effectively inhibited KAPAS activities in vitro and the $IC_{50}$ was $9.5{\mu}M$. Foliar application of juglone showed very good herbicidal activity to the eight-tested weed species. Among them, Solanum nigrum was completely controlled at a concentration of $250{\mu}g\;mL^{-1}$ with main symptoms of desiccation or burndown. Digitaria sanguinalis and Aeschynomene indica were also sensitive to juglone treatment. All eight weed species were controlled by 90~100% at a concentration of $500{\mu}g\;mL^{-1}$. However, soil application of juglone to Digitaria sanguinalis did not show any herbicidal symptoms. Cellular leakage from cucumber leaf squares treated with juglone increased depending on the concentrations increased from 6.25 to $100{\mu}M$ after 24 hours incubation with or without light. However, chlorophyll loss in cucumber leaf squares was negligible. Biotin supplements significantly rescued the inhibition of germination rate of Arabidopsis thaliana seeds previously inhibited by the juglone. Our results suggest that the juglone is a possible environmental friendly herbicide candidate with a new target KAPAS inhibiting activity.
The main objective of this research was to improve rooting of M.9 apple rootstock through layering time, shading, layering material and water content of the layering part. Among the treatments, rooting was improved by mother plant shading or an earlier covering (layering in shoot length 2 cm) of shoots. Rooting and root growth of sawdust, chaff or vermiculite treatments were significantly higher than those of the control (soil). In the study of layering material water content, -40 kPa treatment increased rooting ratio, number of roots and root growth compared to -60 kPa treatment and control. These results suggest that early-covering, layering with sawdust, chaff, or vermiculite, and keeping the water content in the layering part at -40 kPa could improve rooting and root growth in M.9 rootstock propagation.
Low seedling growth rates of white clover (Trifolium repens L.) have been limited its good establishment to pastures. The experiment was done to determine the effect of removal of cotyledon and unifoliolate on the growth and morphological characters of contrasting white clover cultivars for 8 weeks after the treatment. Individual plants of cv. Regal (large leaf), Louisiana S-l (medium-large leaf), Grasslands Huia (medium-small leaf) and Aberystwyth S184 (small leaf) were grown in 10cm plastic pot containing a 2:1:1 soil:sand:peat moss mixture until the cotyledon or unifoliolate stage and then removed one (C1) or two cotyledons (C2) at cotyledon stage, and unifoliolate only (U), unifoliolate and one cotyledon (DC1) or unifoliolate and two cotyledons (DC2) at the unifoliolate stage. To measure the removal effect on biomass and morphological characters (leaf area, petiole and stolon lengths, growing tips and leaves), plants were sampled 4, 6 and 8 weeks after the treatment. Intact plants had greater biomass and morphological characters than removal-treated ones, Removal treatments at cotyledon stage, C1 and C2, were decreased more biomass and morphological characters than removal ones at unifoliolate stage while the severer cotyledon removal, the more reduction. Stolon length per plant and petiole length markedly inclined 6 weeks after the treatments although biomass and the other characters continuously did. Relatively large-leaved cultivar had more biomass, leaf area per plant and longer petiole than the other(s) but the reverse results were true in stolon length, growing tips and no. of leaves per plant. Biomass was linearly increased with increased leaf area but the earlier and severer removal, the less slope. This study was carried out to investigate the difference of the content of soil chemical components and growth characteristics in six years old ginseng affected by application of decomposing plant residues in paddy-converted field. The results show that aerial parts of ginseng are no difference between press cake (PC) 200 kg/10a and control but subterranean parts of ginseng PC 200 kg/10a, especially quantity related root fresh weight and tap root diameter, are statically about 1.6 times heavier and about 1.2 times thicker than the ginseng control. Furthermore, the survival rate of PC 200 kg/10a is 67.1% rise significantly compare with the control 50.7%. But compared with the PC 200 kg/10a and the PC 400 kg/10a, ginseng root growth and survival rate of PC 400 kg/10a get worse and that increase physiological disorder occurrence rate than PC 200 kg/10a. Even though there are no significant differences between the ginseng of decomposing plant residues except press cake treatment and the ginseng of control in growth characteristics, it does tend to increase the survival rate and decrease the physiological disorder occurrence rate in most fertilizer treatment except for RSC 2 ton/10a, RSC 4 ton/10a and RH 4 kL/10a. Noted that EC is highly increased and exceeded 1.7 ds/m in RSC 2 ton/10a, RSC 4 ton/10a and RH 4kL/10a. It would be caused physiological disorder in many ways and affected ginseng growth characteristics, survival rate.
In greenhouse farming, a variety of humic acids have been applied to improve soil conditions and plant growth. However, it is still unclear that how humic acids combined with chemical fertilizers affect growth and quality of fruit vegetable crops. This study was conducted to determine the combination effect of humic acids and chemical fertilizers on the growth and fruit quality of tomato (Lycopersicon esculentum MILL.) grown under greenhouse conditions. Three different formulation types of humic acid were used: liquid type A, liquid type B and solid type C. The tomato plants were grown in three treatment combination plots and in conventional fertilizer (CF) plot with recommended levels of nitrogen, phosphorus and potassium: HA combined with CF (HA+CF), HB combined with CF (HB+CF) and HC combined with CF (HC+CF). For most of growth characteristics (i.e. leaf number, internode length, maximum leaf length, leaf width and chlorophyll contents) determined in this experiment, no significant differences were observed between all combination treatments and CF. However, integrated fruit qualities (i.e. averaged weight, sugar contents and acidity) were slightly improved in the humic acid combined with CF treatments when compared with CF alone treatment. No phytotoxicity was observed with humic acid treatments. However, further studies will probably be needed to use widely and safely these humic acids, in order to ensure a maximizing growth, fruit yield and quality of tomato.
This study was conducted to examine the effect of row covering materials on the growth and yield of broccoli for early production in the open meld. Materials for plastic tunnel were row-covering (ventilating non-woven fabric), perforated film (2.3% pore space) and P.E. film. The types of row covers were tunnel and covering period was one months after transplanting. The row cover and perforated film increased daily air and soil temperature by 3~4$^{\circ}C$ and 2~3$^{\circ}C$, respectively as compared to no covering. Also, relative humidities in those treatments were higher than those in P.E. and no covering. The row covering and perforated film enhanced plant growth. Broccoli grew best and abnormal head appeared the least with row covering treatment. Days taken for head formation, however, did not differ between treatments but the weight of curd was over 400 g in the treatment of row covering and perforated film covering. Marketable yield doubled under row covering as compared to no covering. The first harvest time was earlier by two weeks in row-covering. The results of this experiment indicated that covering row-cover film in spring cultivation was effective fer not only yield increase but also quality evaluation in early production of broccoli.
Bacillus sp. YBL-7 which had been isolated from ginseng root-rot suppressive soil was able to antagonize Fusarium solani causing ginseng root-rot by their antibiotic substance. In order to develop multifunctional antagonist on Bacillus sp. YBL-7 as a biocontrol agent against Fusarium salam', optimal conditions for protoplast transformation system of Bacillus sp.YBL-7 by the vector plasmid pE194 were investigated. The protoplasts of Bacillus sp. YBL-7 were obtained at best efficiency by treatment with 200${\mu}g$/ml of lysozyme in the pH 7.0 of SMM buffer for 90 minutes at $40^{\circ}C$. The cell wall of the protoplast was regenerated on the agar plate containing 1.2% agar and 0.7 M mannitol. Under the best condition for protoplast formation and regeneration, the optimal transformation was achieved with 40% polyethylene glycol (M.W. 4000) treatment for 10minutes. The vector plasmid pE194 showed the best transformation frequency at 5$\mu$g/ml of final concentration. The pE194 was very stable over 80% in the transformants.
