Production and Characterization of Lipopeptide Biosurfactant from Bacillus subtilis A8-8

  • A biosurfactant-producing bacterial strain was selected from oil-contaminated soil because of its ability to degrade crude oil and tributyrin $(C_{4:0})$. The strain was identified as Bacillus subtilis A8-8 based on its morphological, biochemical, and physiological characteristics. When B. subtilis A8-8 was grown with crude oil as the sole carbon source, the biosurfactant from the strain emulsified crude oil, vegetable oil, and hydrocarbons. Soybean oil was the optimum substrate for the emulsifying activity and emulsion stability of the biosurfactant, both of which were superior to those of several commercially available surfactants. The biosurfactant was purified by a procedure including HCl precipitation, methanol treatment, and silica-gel chromatography. The partially purified biosurfactant was analyzed by TLC (thin-layer chromatography), SDS-PAGE, and HPLC and it reduced the surface tension of water from 72 mN/m to 26 mN/m at a concentration of 30 mg/l. Therefore, the purified lipopeptide biosurfactant has strong properties as an emulsifying agent and acts as an emulsion-stabilizing agent.

  • A strain of Streptomyces sp. (YS-40) which was able to produce penicillinase, was isolated from soil and the enzymatic characteristics of this enzyme were investigated. The crude enzyme was obtained with the fractionation by 80 % cold acetone. The optimal temperature and pH of this enzyme was 45$^{\circ}C$ and 5.0 respectively. The stable pH range of the enzyme was between pH 5.5 and 8.0 at 37$^{\circ}C$. By heat treatment at 6$0^{\circ}C$ and 8$0^{\circ}C$ for 10 min, the remained relative activities were about 50%, 30% respectively. The activity of the enzyme was inhibited by Cu$^{++}$, $_Mn^{++}$, Zn$^{++}$ but Co$^{++}$, Li$^{++}$, $Ca^{++}$, $Mg^{++}$ $Ba^{++}$ did not affect. Among 11 chemical reagents, ethylenedi aminetetra-acetic acid disodium salt (EDTA-2Na), sodium dodecyl sulfate (SDS) and sodium fluoride inhibited the enzyme activity.

Insight into Norfloxacin Resistance of Acinetobacter oleivorans DR1: Target Gene Mutation, Persister, and RNA-Seq Analyses

  • Antibiotic resistance of soilborne Acinetobacter species has been poorly explored. In this study, norfloxacin resistance of a soil bacterium, Acinetobacter oleivorans DR1, was investigated. The frequencies of mutant appearance of all tested non-pathogenic Acinetobacter strains were lower than those of pathogenic strains under minimum inhibitory concentration (MIC). When the quinolone-resistance-determining region of the gyrA gene was examined, only one mutant (His78Asn) out of 10 resistant variants had a mutation. Whole transcriptome analysis using a RNA-Seq demonstrated that genes involved in SOS response and DNA repair were significantly up-regulated by norfloxacin. Determining the MICs of survival cells after norfloxacin treatment confirmed some of those cells were indeed persister cells. Ten colonies, randomly selected from among those that survived in the presence of norfloxacin, did not exhibit increased MIC. Thus, both the low mutation frequency of the target gene and SOS response under norfloxacin suggested that persister formation might contribute to the resistance of DR1 against norfloxacin. The persister frequency increased without a change in MIC when stationary phase cells, low growth rates conditions, and growth-deficient dnaJ mutant were used. Taken together, our comprehensive approach, which included mutational analysis of the target gene, persister formation assays, and RNA sequencing, indicated that DR1 survival when exposed to norfloxacin is related not only to target gene mutation but also to persister formation, possibly through up-regulation of the SOS response and DNA repair genes.

  • This studies were conducted to investigate the effects of hexaconazole on growth of watermelon(Citrullus lanatus Schrad) and ethylene evolution from gourd(Lagenaria siceraria Standl). While foliar application of hexaconazole exhibited inhibition effect, little or no inhibition was found in the plants treated with soil drench application. The plant height and number of internode in watermelon were significantly inhibited at 7 days after treatment(DAT), but no significant difference was found at 42 DAT. No significant difference was found in site of female flower setting when treated with hexaconazole. The length of branch was significantly inhibited at the 2nd or the 3rd internode. It was suggested that the inhibition of stem elongation was related with application timing of the chemical. Hexaconazole increased ethylene evolution more effectively at earlier stage of gourd, but after 5 DAT ethylene evolution was decreased.

  • Plant growth-promoting rhizobacteria (PGPR) are beneficial native soil bacteria that colonize plant roots and result in increased plant growth. The objective of this study was to determine the plant growth promotion in canola plants by selected PGPR strain 13-1 under low temperature condition. The seed treatment of strain 13-1 was enhanced plant height and root elongation on canola plant at low temperature condition. This result determined that a selected strain of PGPR can enhance plant growth and root propagation under extremely low temperature conditions. Thus, this PGPR strain extends their role on plant growth promotion on canola until low temperature condition for practical applications.

  • Dredged fills have been widely used to secure a land for the engineering activities. Before the useful application of the area, the soils should be consolidated to acquire the aquate shear strength. Several research projects have attempted to develop a method fur accelerating the consolidation of soft clay. Our study examined the effect of ultrasonic energy on the consolidation of soft clay, Tests were conducted using specially designed and fabricated equipment that was capable of directly applying ultrasonic energy to soil samples during consolidation tests. The specimens were prepared from slurry using a centrifuge facility, and tests were conducted at various levels of ultrasonic power and treatment time. The study showed that ultrasonic energy had a considerable effect on consolidation time, suggesting that ultrasound can be used to reduce the consolidation time of soft clay.

  • The Seokdae Waste Landfill is a middle-sized site used from June, 1987 to May, 1993. Many joints and faults are developed in andesitic rocks and rhyolitic rocks distributed at the landfill. The chemical analyses of leachates, streams and groundwaters sampled in July, 1996 and June, 1997 show that the concentrations of leachates and streams were decreased, and that the groundwater qualities became worse. The groundwater contamination is deeply extended to not only shallow groundwater but also bedrock-groundwater around the Seokdae Waste Landfill Area. The range of groundwater contamination by the leachates is about 500 m to the west and about 1 km to the south from the boundaries of the waste landfill. The development of monitoring wells and pumping wells, the construction of a leachate-treatment facilities, and the adjustment of the existing grout curtains are necessary for the control of water pollution at the Seokdae Waste Landfill Area.

  • The purpose of this study was to investigate the rural housewives' recognition and practice on environmental preservation. The subjects of this study were 300 rural housewives living in Chung-buk province. Questionaires were distributed to them during a week from August $25^{th}$ to August $31^{th}$ in 1995. Major findings were as follows: 1. The saved water quite well in washing their faces and the dishes compared with urban housewives, but they did not save water so well in using the lavatory. They used synthetic detergent, one of water pollutants, more often than natural soaps in shampooing and laundering. In addition, they did not use a measuring cup. It resulted in the waste of detergent and aggravation of water pollution. 2. Food waste was disposed of properly, but used batteries were not collected separately. A disposable vinyl bag was more commonly used than a reusable shopping basket. The reason for not using a shopping basket was bothering or uncustomary. Since most of them did not know refillable products of goods with environmental mark, they did not use those. Therefore, housewives should be educated in order to prevent soil pollution. 3. The use of spry-type pesticide and hair treatment, one of air pollutants, was wide-spread among them. They hardly recognized and practiced saving of electric power when they used household electric appliances. 4. The major source of environmental knowledge was mass media like TV of radio. Many of them did not know the practical ways to solve the environmental education program. In conclusion, low level of recognition and practice of rural housewives on environmental preservation indicated that a systematic environmental education program should be developed and implemented.

Evaluation of the Impacts of Water Quality Management in Kyongan Stream Watershed using SWAT Model (SWAT 모델을 이용한 경안천 유역의 수질관리 영향 평가)

  • SWAT model would be applied to evaluate the pollutant removal capacity with various best management practices (BMPs) in Kyongan stream watershed which plays an important role in water quality conservation and improvement of Paldang reservoir. The methods for the representation of various BMPs scenarios with SWAT is developed and evaluated. Riparian buffer strip, agricultural conservation practices to reduce fertilizer, sediment, and nutrients occurring from farm field (Grassed swale, Contour farming/Parallel terrace, Field border, Farm retention pond, Grade stabilization structure), and washland such as wetland and pond to extend detention and improve water quality are represented in SWAT. And to represent the expansion of existing Waste Water Treatment Plants (WWTPs) in Soil and Water Assessment Tool (SWAT), reduction effect for point source pollutants was simulated. As the result of simulation, the removal rates of SS, TN, TP from scenarios of Kyongan stream watershed are the average annual SS yield by 5.2% to 69.2%, the average annual TN yield by 0.5% to 26.3%, and the average annual TP yield by 1.3% to 32.5%, respectively. This study has demonstrated that the SWAT is a very reliable and useful water quality and quantity assessment tool, and the BMPs representation in SWAT for watershed management is able to effectively simulate in Kyongan Stream watershed.

  • Experiments were conducted to know the ecology of injurious paddy field weeds, Scirpus hotarui and aneilema japonica in 1982-1983. Almost all emergences of the both species were on the soil surface in emergence depth. The vigorous growth stage was before Aug. 18, and the plant height and branch numbers were decreased and the days to flower were diminished by the shortday length treatment. A. japonica was not renewed by once cutting, but cutting S. hotarui on vigourous tillering stage was the most effective renew the growth and all of overwintered plants were renewed in emergence. The more these weeds emergence, the less rice yields.
