Use of FT-IR to Identify Enhanced Biomass Production and Biochemical Pool Shifts in the Marine Microalgae, Chlorella ovalis, Cultured in Media Composed of Different Ratios of Deep Seawater and Fermented Animal Wastewater

  • Growth rates, photosystem II photosynthesis, and the levels of chlorophyll $\alpha$ and secondary metabolites of Chlorella ovalis were estimated to determine if they were enhanced by the addition of swine urine (BM) or cow compost water (EP) that had been fermented by soil bacteria to deep seawater (DSW) in an attempt to develop media that enabled batch mass culture at lower costs. Growth of C. ovalis in f/2, f/2-EDTA+BM60%, DSW+BM30%, and DSW+EP60% was enhanced and maintained in the log phase of growth for 16 days. The cell densities of C. ovalis in DSW+EP60% ($4.1{\times}10^6$ Cells/ml) were higher than those of f/2 ($2.9{\times}10^6$ Cells/ml), f/2-E+BM60% ($3.7{\times}10^6$ Cells/ml), and DSW+BM30% ($2.7{\times}10^6$ Cells/ml). The growth rate was also more favorable for C. ovalis cultured in DSW+EP60% ($0.15\;day^{-1}$) than that of C. ovalis cultured in the control medium (f/2) ($0.12\;day^{-1}$). Furthermore, the chlorophyll a concentration of C. ovalis cultured in DSW+EP60% (4.56 mg/l) was more than 2-fold greater than that of C. ovalis cultured in f/2 (2.35 mg/l). Moreover, the maximal quantum yields of photo system II at 470 nm (Fv/Fm) were significantly higher in organisms cultured at f/2-E+BM60% (0.53) and DSW+EP60% (0.52) than in the other treatment groups. Finally, Fourier transformation infrared (FT-IR) spectroscopy revealed that C. ovalis grown in DSW+EP60% had more typical peaks and various biochemical pool shifts than those grown in other types of media. Taken together, the results of this study indicate that the use of DSW+EP60% to culture C. ovalis can reduce maintenance expenses and promote higher yields.

Biological Potential of Bioorganic Fertilizer Fortified with Bacterial Antagonist for the Control of Tomato Bacterial Wilt and the Promotion of Crop Yields

  • The application of Bacillus sp. in the biological control of plant soilborne diseases has been shown to be an environmentally friendly alternative to the use of chemical fungicides. In this study, the effects of bioorganic fertilizer (BOF) fortified with Bacillus amyloliquefaciens SQY 162 on the suppression of tomato bacterial wilt were investigated in pot experiments. The disease incidence of tomato wilt after the application of BOF was 65.18% and 41.62% lower at 10 and 20 days after transplantation, respectively, than in the control condition. BOF also promoted the plant growth. The SQY 162 populations efficiently colonized the tomato rhizosphere, which directly suppressed the number of Ralstonia solanacearum in the tomato rhizosphere soil. In the presence of BOF, the activities of defense-related enzymes in tomato were lower than in the presence of the control treatment, but the expression levels of the defense-related genes of the plants in the salicylic acid and jasmonic acid pathways were enhanced. It was also found that strain SQY 162 could secrete antibiotic surfactin, but not volatile organic compounds, to suppress Ralstonia. The strain could also produce plant growth promotion compounds such as siderophores and indole-3-acetic acid. Thus, owing to its innate multiple-functional traits and its broad biocontrol activities, we found that this antagonistic strain isolated from the tobacco rhizosphere could establish itself successfully in the tomato rhizosphere to control soilborne diseases.

A Study on Treatment Conditions of Oil Contaminated Soil by Low Temperature Thermal Desorption (저온 열 탈착에 의한 유류 오염토의 처리 조건의 연구)

  • The objective of this study is to remove BTEX(Benzene, Toluene, Ethylene, Xylene) and TPH(Total Petroleum Hydrocarbon) effectively by using method low thermal desorption. The thermal desorption is frequently selected because it can treat various contaminants effectively. The temperature and heating time are determined by TGA(Thermogravimetric analysis) curve. The experiment result from this research, removal rate of BTEX was up to 100% within 5 minutes and removal rates of TPH were more than 65% at $300^{\circ}C$ and 70% at $500^{\circ}C$ respectively. It was observed that there was a little change of removal rates of TPH.

A Study on the solid waste of Buk Han Mt National Park (북한산 국립공원의 고형 폐기물에 관한 연구)

  • The number of visiters to Bukhan Mt. national park, generation quantity of solid waste and collection system were researched to consider a counterplan for the pollution control of the national park and study for developing the effective treatment of solid waste was tried through the proximate analysis of each component containing. Results obtained in this study were summerized as follows; The great part of visitors go on an excursion to the Bukhan Mt. national park during July and August and also, the solid waste was generated nearly a half of the total amount at the same period. The major collection facilities in the national park were waste basket and incineration box. But the incineration box was too large in volume and very far in distance, and its collection period was irregular, so it was cause to the congestion of solid waste and bad smell and dirty. Therefore, to complete collection of solid waste, we must set up the waste basket which able to find within 40~50m from the origination place of solid waste and induce the visitors to throw the solid waste. It was obtained as moisture content: 48.5 wt%, volatile solid: 28.4wt%, fixed solid: 23.1 wt%, lower heating value: 1,320kca1/kg from experimental analysis of solid waste. According to this analysis, the incineration operation is possible, but the generation quantity of solid waste was too small to construct incineration plant for heat recovery. It was found that it is suitable for the aerobic composting by mixing with the night soil which generate in the national park after the recovery of resources such as metals, glasses and plastics.

Hairy Vetch Growth-Promoting Rhizobium sp. RH84 and Application to Reclaimed Land (Rhizobium sp. RH84에 의한 간척지 환경에서의 헤어리베치 생육촉진)

  • This study was carried out to investigate the plant growth promoting activity of hairy vetch (Vicia villosa Roth) on reclaimed land. At the previous research, Rhizobium sp. RH84 was isolated and selected for further study from hairy vetch. For the investigation of plant growth promoting effects by the Rhizobium sp. RH84, production of indole acetic acid (IAA), siderophore, phosphate solubilization and nitrogen fixation were tested and other characters were examined. As results, RH84 produced $9.03{\mu}g$ IAA per mL and showed nitrogen fixation activity. With the treatment of Rhizobium sp. RH84 to hairy vetch showed good growth at 0.3% salty reclaimed soil, and the production yield was increased up to 56% at field test. From these results, it was confirmed that the Rhizobium sp. RH84 would be used as a green manure for hairy vetch under the salty condition of reclaimed land.

Nature of a Root-Associated Paenibacillus polymyxa from Field-Grown Winter Barley in Korea

  • Soil or seed applications of plant growth-promoting rhizobacteria (PGPR) have been used to enhance growth of several crops as well as to suppress the growth of plant pathogens. In this study, we selected a PGPR strain, Paenibacillus polymyxa strain E681, out of 3,197 heat-stable bacterial isolates from winter wheat and barley roots. Strain E681 inhibited growth of a broad spectrum plant pathogenic fungi in vitro, and treatment of cucumber seed with E681 reduced incidence of damping-off disease caused by Pythium ultimum, Rhizoctonia solani, or Fusarium oxysporum. When inoculated onto seeds as vegetative cells or as endospores, E681 colonized whole cucumber root systems and root tips. Different temperatures such as $20^{\circ}C\;and\;30^{\circ}C$ did not affect root colonization by strain E681. This colonization was associated with a consistent increase in foliar growth of cucumber in the greenhouse. These results indicate that strain E681 is a promising PGPR strain for application to agricultural systems, particularly during the winter season.

Isolation and Biological Properties of Novel Cell Cycle Inhibitor, HY558, Isolated from Penicillium minioluteum F558

  • In the course of screening for a novel cell cycle inhibitor, a potent Cdk 1 inhibitor, HY558, was found from the culture broth of Penicillium minioluteum F558 isolated from a soil sample. The molecular ion of HY558 was identified at m/z 329 (MH+) with a molecular formula of $C_20H_44ON_2$. HY558 exhibited selective antiproliferative effects on various human cancer cell lines. Its $IC_50$ values were estimated to be 0.29 mM on HepG2, 0.30 mM on HeLa, 0.30 mM on HL6O, 0.33 mM on HT-29, and 0.25 mM on AGS cells. Interestingly, Hy558 demonstrated no antiproliferative effect with normal lymphocytes used as the control, and a low level of inhibition on the proliferation of A549 cancer cells. A flow cytometric analysis of HepG2 cells revealed an appreciable arrest of cells at the G1 and G2/M phases of the cell cycle following treatment with Hy558. furthermore, DNA fragmentation due to apoptosis was observed in HeLa cells treated with 0.46 mM of HY558.

$Pyoverdin_{2112}$ of Pseudomonas fluorescens 2112 Inhibits Phytophthora capsici, a Red-Pepper Blight-Causing Fungus

  • A bacterium, Pseudomonas fluorescens 2112, that is antagonistic against a red-pepper blight-causing fungus, Phytophthora capsici, was isolated from the local soil of Gyongju, Korea. This strain formed an orange-colored clear halo zone on chrome azurol S (CAS) blue agar, suggesting the production of a siderophore in addition to an antifungal antibiotic. The optimal culture conditions for siderophore production by P. fluorescens 2112 were 30-h cultivation at $25^{\circ}C$ and pH 6.5 in King's B medium. The presence of $20{\mu}g/ml\;of\;Fe^3+$ ion or EDDHA promoted the production of siderophore in King's B medium. The siderophore was purified from culture broth by CM-Sephadex C-25 and Sephadex G-25 column chromatographies. The UV spectra of the purified siderophore was the same as that of pyoverdins or pseudobactins. The molecular mass was 1,958 Da determined by FAB-rlass spectrometer, and the amino acid composition analysis showed that the purified siderophore consisted of glycine/threonine/serine/glutamic acid/alanine/lysine with the molar ratio of 3:2:1:1:1:1, DL-Threo-${\beta}$-hydroxyaspartic acid and $N^{\delta}$-hydroxyornithine, two of the essential constituents of pyoverdin, were also found. The purified siderophore pyoverdin showed strong in vitro and in vivo antagonistic activities against phytophthora blight-causing P. capsici. Especially in an in vivo pot test, the siderophore protected red-pepper Capsicum annum L. very well from the attack of P. capsici. These results indicated that the purified siderophore of P. fluorescens 2112 played a critical role in the biocontrol of the red-pepper blight disease, equivalent to treatment by P.fluorescens 2112 cells.

Identification of Stenotrophomonas maltophilia LK-24 and its Degradability of Crystal Violet

  • A number of soil and wastewater samples were collected from the vicinity of an effluent treatment plant for the chemical industry. Several microorganisms were screened fur their ability to decolorize the triphenylmethane group of dyes. As a result, a novel crystal violet dye-degrading strain LK-24 was isolated. Taxonomic identification including 16S rDNA sequencing and phylogenetic analysis indicated that the isolate had a $99.5\%$ homology in its 16S rDNA base sequence with Stenotrophomonas maltophilia. The triphenylmethane dye, crystal violet, was degraded extensively by growing cells of Stenotrophomonas maltophilia LK-24 in agitated liquid cultures, although their growth was strongly inhibited in the initial stage of incubation. This group of dyes is toxic, depending on the concentration used. The dye was significantly degraded at a relatively lower concentration, below $100{\mu}g\;ml^-1$, yet the growth of the cells was totally suppressed at a dye concentration of $250{\mu}g\;ml^-1$. The degradation products of crystal violet were identified as 4,4'-bis(dimethylamino)-benzophenone and ${\rho}$-dimethylaminophenol by Gas chromatography-Mass spectrometry. The 4,4'-bis(dimethylamino)-benzophenone was easily obtained in a reasonable yield, as it was not metabolized further by S. maltophilia LK-24; however, the ${\rho}$-dimethylaminophenol was not easily identifiable, as it was further metabolized.

A Proposal of Standard Method for the Analysis of Total Petroleum Hydrocarbons (TPHs) in Marine Sediments and Biota (해양환경시료(해저퇴적물과 해양생물)에 축적된 석유계총탄화수소(TPHs) 분석에 대한 공정시험기준 제안)

  • Regulatory test method for Total Petroleum Hydrocarbons (TPHs) in the marine sediment and biota has not still been established even though TPHs are one of the major pollutants in marine environment. Based on the Korean Soil Standard Method (SSM) for TPHs, we considered a new treatment method for determining TPHs in marine environmental samples by using a Gas chromatography coupled with Mass spectrometric detector. We suggested an improved recovery test for quality control procedures and introduced analytical procedures of removing sulfur, polar organic materials, water and saponification for removing neutral lipids in marine bottom sediments and biota.
