Structural Characterization of Non-reducing Oligosaccharide Produced by Arthrobacter crystallopoietes N-08

  • A bacterial strain (Strain N-08) capable of extracellularly producing high level of non-reducing oligosaccharide (NR-OS) isolated from soil. The strain was identified phylogenetically by 16S rDNA sequence analysis and found to be very close to Arthrobacter crystallopoietes. The high production of NR-OS was observed in the basal culture medium containing maltose as a sole carbon source. The NR-OS in culture supernatant was purified by glucoamylase treatment and Dowex-1 (OH.) ion exchange chromatography and its structure was characterized. This oligosaccharide consisted of only glucose. Methylation analysis indicated that this fraction was composed mainly of non-reducing terminal glucopyranoside. Matrixassisted laser-induced/ionization time-of-flight (MALDI-TOF) and electrospray ionization-mass spectrometry (ESI-MS)/MS analyses suggested that this oligosaccharide comprised non-reducing disaccharide unit with 1,1-glucosidic linkage. When this disaccharide was analyzed by $^1H$-NMR and $^{13}C$-NMR, it gave the same signals with $\alpha$-D-glucopyranosyl-(1,1)-$\alpha$-Dglucopyranoside. These results indicated that the NR-OS produced by A. crystallopoietes N-08 was ${\alpha}1$,${\alpha}1$-trehalose. This is the first report of the trehalose which can be produced directly from maltose by A. crystallopoietes N-08.

Optimization of the Sulfur-oxidzing Bacteria, Thiobacillus novellus SRM (황 산화 세균인 Thiobacillus novellus SRM 성장 최적화)

  • The microorganism was isolated from the night soil treatment plant for the removal of sulfur compounds. The growth conditions of the sulfur-oxidizing bacteria were investigated and the isolate characterized as Thiobacillus noveilus SRM. The optimal pH of Thiobacillus novellus SRM on cell growth was pH 7.0 and the optimal temperature was 30$^{\circ}C$ and the optimal air flow rate was 1 vvm, respectively. As a results of cell growth from the Monod plot, the specific growth rate was 0.032 hr$\^$-l/, $V_{max}$ was 1.43 hr$\^$-l/ and $K_{m}$ was 0.32, respectively. The thiosulfate oxidation by Thiobacillus novellus SRM was made of sulfate ion. The sulfate ion reduced pH and decreased cell growth.

Fast Consolidation of Soft Clay due to Ultrasonic Energy (압밀촉진을 위한 초음파의 활용)

  • Various researches have been undertaken to develope a method of enhancing consolidation. This study investigated the effect of ultrasonic energy on consolidation through a series of laboratory experiments. The tests were conducted using a specially designed and fabricated equipment which can apply ultrasonic energy on a soil sample directly during a consolidometer test. Clay specimens were prepared from slurry using a centrifuge facility, and test conditions were varied depending on ultrasonic power and treatment time. The results of the study show that the effect of ultrasonic energy on consolidation is significant. The degree of significance varies with the test conditions. It could be concluded that the study showed potential application of ultrasound to reduce consolidation time.

  • Coleus forskohlii Briq. of the family Lamiaceae yields a valuable secondary metabolite known as forskolin which is a labdane diterpenoid. Coleus forskohlii is the only known source of this compound. Forskolin is used in medicine for the treatment of glaucoma, congestive cardiomyopathy and asthma. Morphogenic callus was induced from young leaves on MS medium augmented with NAA and BA. These calli, when subcultured on MS with KN alone gave rise to shoots. The regenerated shoot developed good root system on MS medium fortified with NAA. The fully grown plantlets were transferred to soil for acclimatization. Coleus plant is mainly infected by a fungi Lasiodiplodia theobromae which causes root of disease. The fungal culture filterate (FCF) of Lasiodiplodia theobromae, has been used in regeneration media to find the MIC and further to select resistant plants to the pathogen. In the present study 40% FCF in the medium showed maximum inhibition and is there fore considered as the MIC level of Coleus forskohlii. This data could prove to be useful for the future for selecting a resistant C,forskohlii plant against the root disease caused by L.theobromae.

Complex odor removal in pilot-scale biofilter with microorganisms immobilized on polymer gel media (미생물 포괄고정화 담체를 적용한 파일럿 스케일 바이오필터에서의 복합악취 제거)

  • A pilot-scale biofilter was constructed to discover degradation characteristics of the complex odor discharged from Ansan wastewater treatment plant. Candida tropicalis for volatile organic compounds, sulfur oxidizing bacteria(SOB) for hydrogen sulfide, and bacteria extracted from feces soil were immobilized on a polymer gel media. According to this study, the EBCT was varied from 36 sec to 18 sec. Toluene was removed as 80% along the variations, but it was recovered as 100% within 1 week. All benzene and xylene were removed during the operation while the efficiency of hydrogen sulfur was temporary decreased at 18 sec of EBCT, thereafter it was recovered to 100% within a week. The maximum elimination capacities of the benzene, toluene, xylene, and hydrogen sulfur were 6.6 g/$m^{3}$/hr, 31.7 g/$m^{3}$/hr, 7.8 g/$m^{3}$/hr, and 133.6 g/$m^{3}$/hr, respectively. There were merits on removal both organic and inorganic complex odor using the pilot-scale biofilter embedded with microorganisms immobilized on polymer gel media.

Immobilization and Characterization of Tannase from a Metagenomic Library and Its Use for Removal of Tannins from Green Tea Infusion

  • Tannase (Tan410) from a soil metagenomic library was immobilized on different supports, including mesoporous silica SBA-15, chitosan, calcium alginate, and amberlite IRC 50. Entrapment in calcium alginate beads was comparatively found to be the best method and was further characterized. The optimum pH of the immobilized Tan410 was shifted toward neutrality compared with the free enzyme (from pH 6.4 to pH 7.0). The optimum temperature was determined to be $45^{\circ}C$ for the immobilized enzyme and $30^{\circ}C$ for the free enzyme, respectively. The immobilized enzyme had no loss of activity after 10 cycles, and retained more than 90% of its original activity after storage for 30 days. After immobilization, the enzyme activity was only slightly affected by $Hg^{2+}$, which completely inhibited the activity of the free enzyme. The immobilized tannase was used to remove 80% of tannins from a green tea infusion on the first treatment. The beads were used for six successive runs resulting in overall hydrolysis of 56% of the tannins.

Isolation, Purification and Characterization of Phytase from Asperfillus sp. (Aspergillus속 균주가 생산하는 Phytase의 분리 정제 및 특성)

  • To extract insoluble proteins and to improve funtional properties of abolished proteins, an phytase producing Aspergillus sp. SM-15 was isolated from soil. The enzyme was purified and its enzymological characteristics were investigated. Phytase production reached to maximum when the wheat bran medium containing 1% mannose, 1% yeast extract, 1% (NH4)2HPO4 and 0.2% calcium chloride was cultured for 4 days. Phytase was purified 17.1 fold and specific activity was 244.32unit/mg by a sequencial process of ammonium sulfate fraction, ion exchange chromatography and gel filtrations Pruified enzyme was confirmed as a single band by the polyacrylamide gel electro-phoresis. The molecular weight of phytase was estimated to be 46,000. The optimum pH and temperature for the phytase activity were 5.5 and 5$0^{\circ}C$. The enzyme is stable in pH 4.5~5.5, 6$0^{\circ}C$. The activity of purified enzyme was inhibited by Hg2+ whereas activited by Pb2+ and Fe2+. The activity of phytase was inhibited by the treatment with iodine. The result indicate the possible involvement of histidine at active site. Km and Vmax of the puridied phytase were 37.037mM/L and 159.87umol/min, respectively.

Improved Cleaning Method for Dental Instruments

  • We searched at the "PubMed.gov" and "jendodon.com" sites to conduct a literature review on dental instruments that are reused in clinical settings and on infection control involving pre-disinfection or sterilization cleaning/rinsing. The keyword "dental clean" was used for the Web search. We found the present official definition of instrument cleaning performed prior to disinfection or sterilization rather limiting ("removal of foreign matter (soil, organism, etc.) from the instruments"). Thus, we proposed to expand the definition to include the removal of oils applied to protect the metallic instruments and from corrosion, stains, and rust resulting from the frequent reuse of the instruments. Clinicians are found to clean their dental instruments (a) immediately after treating their patients or (b) following their treatment but not immediately afterward. In the latter case, we recommend presoaking to be added. Ultrasonic sterilization of 5~15 minutes is found to be more effective in terms of eliminating residual matter from the instruments compared to other methods. To check on the cleaning results, we recommend visual inspection, which can be quick and practical in clinical settings. The latest products being developed and marketed on the market address the related problems. Nonetheless, research must be continued on the effects of presoak, cleaning/rinsing, disinfection, and high-temperature or heating-based sterilization on the dental instruments and on dental clinicians' practices in cleaning, disinfection, and sterilization. We advise dental clinicians to select the proper cleaning methods and detergents for their instruments to help eliminate or prevent corrosion, staining, and rusting, to reduce the maintenance costs, and to ensure user-friendly instruments/apparatuses.

Characteristics of Reinforced Drainage Geotextile for Waste Treatment System (폐기물매립지용 보강형배수재의 배수특성에 관한 연구)

  • The settlement occurring during landfill construction often causes a damage of drainage system. Clogging can reduce the hydraulic conductivity of the Leachate Collection and Drainage System, which results in the increase of leachate level within the landfill. Consequently, the insulation ability of leachate will be decreased. The main purpose of this project is to estimate a newly designed reinforced drainage geotextile (RDG) combining non-woven fabrics with geogrid for minimizing the destruction of drainage layer as well as evaluating RDG's application in the leachate collection and drainage system. Thus, the project observed the permittivity changes of RDG, and evaluated the drainage ability using RDG in the leachate collection and drainage system.

  • A method of construction with a basis treatment that uses a latticed bamboo mat puts bamboo's peculiarity and bamboo mat's bending substance to use, and a latticed bamboo mat equally distributes to embankment loading in a basis ground. Therefore it prevents ground destruction by an ill-balanced load and an irregular ground subsidence, and it makes safety system. This investigation make progress a soil box model by a dredged reclamation ground of west and south sea at domestic area, and make a comparative study and analyze the strengthening effect based on a grid size of a bamboo-mat and ground failure that it will use in the field.
