Influencing of drying-wetting cycles on mechanical behaviors of silty clay with different initial moisture content

  • To get a better understanding of the effect of drying-wetting cycles (DWC) on the mechanical behaviors of silty clay hiving different initial moisture content (IMC), the direct shear tests were performed on sliding band soil taken from a reservoirinduced landslide at the Three Gorges Reservoir area. The results indicated that, as the increasing number of DWC, the shear stress-displacement curves type changed from strain-hardening to strain-softening, and both the soil peak strengths and strength parameters reduced first and then nearly remain unchanged after a certain number of DWC. The effects of DWC on the cohesion were predominated that on the internal friction angle. The IMC of 17% is regarding as the critical moisture content, and the evolution laws of both peak shear strength and strength parameters presented a reversed 'U' type with the rising of the IMC. Based on it, a strength deterioration evolution model incorporating the influence of IMC and DWC was developed to describe the total degradation degree and degradation rate of strength parameters, and the degradation of strength parameters caused by DWC could be counterbalanced to some extent as the soil IMC close to critical moisture content. The microscopic mechanism for the soil strength caused by the IMC and DWC were discussed separately. The research results are of great significance for further understanding the water-weakening mechanicals of the silty clay subjected to the water absorption/desorption.

Identification of Potential Target Genes Involved in Doxorubicin Overproduction Using Streptomyces DNA Microarray Systems

  • Doxorubicin is a highly-valuable anthracycline-family polyketide drug with a very potent anticancer activity, typically produced by a Gram-positive soil bacterium called Streptomyces peucetius. Thanks to the recent development of Streptomyces genomics-based technologies, the random mutagenesis approach for Streptomyces strain improvement has been switched toward the genomics-based technologies including the application of DNA microarray systems. In order to identify and characterize the genomics-driven potential target genes critical for doxorubincin overproduction, three different types of doxorubicin overproducing strains, a dnrI(doxorubicin-specific positive regulatory gene)-overexpressor, a doxA (gene involved in the conversion from daunorubicin to doxorubicin)-overexpressor, and a recursively-mutated industrial strain, were generated and examined their genomic transcription profiles using Streptomyces DNA microarray systems. The DNA microarray results revealed several potential target genes in S. peucetius genome, whose expressions were significantly either up- or down-regulated comparing with the wild-type strain. A systematic understanding of doxorubicin overproduction at the genomic level presented in this research should lead us a rational design of molecular genetic strain improvement strategy.

  • This paper describes research on the prediction of the vertical displacement of surface, horizontal displacements and bending moments in two anchored retaining wall for an excavation by a finite element program. It is very important to consider the appropriate constitutive model for the numerical analysis in excavation behavior. It is shown in this paper that the analyses of excavation considering small strain stiffness gives the more reasonable prediction of the vertical displacement of surface. and the parametric study on the small strain stiffness parameters for excavation analysis has been analysed.

Characterization of Biological Treatment by an Isolated Phenol-Degrading Bacterium (페놀분해세균의 분리 및 생물학적 처리 특성)

  • 20 bacterial strains capable of growing on phenol minimal medium were isolated from soil and wastewater by the enrichment culture technique, and among them, one isolate which was the best in the cell growth was selected and identified as Bacillus sp. SH3 by its characteristics. Strain SH3 could grow with phenol as the sole carbon source up to 15 mM, but did not grow in minimal medium containing above 20 mM of phenol. The optimal conditions of temperature and initial pH for growth and phenol degradation were 30$^{\circ}$C and 7.5, respectively. This strain could grow on various aromatic compounds such as catechol, protocatechuic acid, gentisic acid, o-, m-, p-cresol, benzoic acid, p-hydroxybenzoic acid, anthranilic acid, phenyl acetate and pentachlorophenol, and the growth-limiting log P value of strain SH3 on organic solvents was 3.1. In batch culture, strain SH3 degraded 97% of 10 mM phenol in 48 hours. In continuous culture under the conditions of 20 mM of influent phenol concentration and 0.050 hr$^{-1}$ of dilution rate, the treatment rate of phenol was 94%.

  • In order to screen dextranase with high dextranolytic activity from microbial origins, dextranase producing fungal isolates were isolated from soil of the Taeion area. 197 strains with dextranolytic activities were isolated, out of which 3 strains with high dextranolytic activities were selected in the first screening. A strain (GR-98) with a best dextranolytic activity was selected in the second screening. The strain was identified to be similiar Aspergillus ustus by the morphological and cultural characteristics. The optimum culture temperature and initial pH for the dextranase production of the strain was 30$\circ$C and 7.0, respectively. The optimum culture medium was composed of 2% dextran, 0.3% KNO$_{3}$, 0.05% K$_{2}$HPO$_{4}$, 0.02% MgSO$_{4}$-7H$_{2}$O, 0.05% KC1, and 2.5 $\mu$g/ml pyridoxamine, and the enzyme production was maximum when the strain was subcultured at 30$\circ$C for 7 days.

  • Twenty-five fungal strains producing an extracellular soybean milk clotting enzyme were isolated from 146 soil samples, and identified as 11 species belonging to six genera of Aspergillus oryzae (5 strains), Aspergillus flavus (2 strains), Aspergillus parasiticus (1 strain), Aspergillus tamarii (2 strains), Aspergillus niger (4 strains), Aspergillus fumigatus (2 strains), Mucor hiemalis (2 strains), Wallemia sebi (4 strains), Scopulariopsis condida (1 strain), Fusarium redolens(1 strain) and Verticillum lecanii (1 strain). Among them, Aspergillus oryzae 020 and Aspergillus tamarii 287 showed relatively high soybean milk clotting activity. The coagulabilities of the enzyme from representative strains of those species decreased as the pH of soybean milk increased from 6.0 to 7.0 The optimum temperature for soybean milk clotting enzymes of those strains were 65$^{\circ}C$.

  • A microorganism, YS-40 strain which powerfully produced penicillinase was isolated from soil. The morphological and cultural characteristics of the strain ; spore chain, spore surface, aerial mass color, melanoid pigment, reverse side pigment, other soluble pigment, and physiological properties were investigated. As the results of various examinations, the strain YS-40 was identified to be similar to Streptemyces bobili except the spore chain and the utilization of L-rhamnose among the various carbon sources.

  • Identification of the Actinomycetes isolate strain No. 497 producing an actinomycin antibiotic MT-497 was performed by ISP and chemotaxonomic methods. The strain Nu. 497 formed various shapes of sclerotia and smooth surface spore. Menaquinone MK-9 ($H_6, H_8$) and iso-, anteiso-branched $C_{15}C_{17}$ fatty acids were detected from whole cell extract. The wall chemotype of stram No. 497 was decided as wall chemotype I from the analysis of DAP isomer, peptidoglycan type and sugar pattern. From these morphological, chemotaxonomic characteristics and analysis of various physiological characteristics. the strain No. 497 was identified as Streptomyces nigrifaciens.

  • Nonlinear cross-anisotropic properties of soil is critical for exact numerical simulation. Theoretically, initial cross-anisotropic properties can be evaluated from triaxial tests with bender element tests, and nonlinear cross-anisotropic properties over initial strain level cannot be evaluated from triaxial tests. In this study, a supposed condition among nonlinear cross-anisotropic properties is suggested to calculate nonlinear cross-anisotropic properties from triaxial tests. Maximum strain and incremental strain energy are applied to combine triaxial test results and theoretical normalized shear modulus curve, respectively Based on combined results, nonlinear cross-anisotropic properties are calculated. Numerical simulation for triaxial tests Is carried out to verify the applicability of the supposed condition with calculated cross-anisotropic properties and simplified nonlinear cross-anisotropic model.

  • Thousand actinomycetes and 50 soil samples were used for the isolation of microorganisms producing yeast cell wall lytic enzymes. Among 493 strains producing large clear zones on autolysed washed yeast (AWY), 117 strains were selected on living yeast cell agar plates. With the method of lytic activity, one strain (St-1702) was selected, which was temporarily identified as Streptomyces eurythermus. The optimal condition for enzyme production of this strain was partially determined as follows: incubation of the strain for 3 days at 30$\circ$C in the medium containing 2% freeze dried yeast cell, 1% glucose, 1% K$_{2}$HPO$_{4}$, 0.01% MgSO$_{4}$'7H$_{2}$O, 0.5% peptone, and 0.2% (NH$_{4}$)$_{2}$CO$_{3}$ with pH 7.0. The protoplast formation of yeast by using the enzyme produced by this strain was compared with commercial enzymes.
