Isolation and Identification of Streptomyces californicus KS-89 Produced Bluish Purple Pigment (청자색 색소를 분비하는 Streptomyces californicus KS-89의 분리 및 동정)

  • The objective intended for this study is that of providing a fairly practical guide to the use of natural pigment in the food industry. Streptomyces isolated from soil were carried out test for the excretion of their bluish purple pigment. One strain of Streptomyces, strain KS-89 showed a high production of bluish purple pigment on the glycerol starch-glutamate medium. The morphological and physiological characteristics of the strain KS-89 were studied according to the methods of Bergey's manual, Nonomura's classification, and Ridham and Lyons classification. Based on the results obtained in these experiments, strain KS-89 was identified as Streptomyces californicus.

$\alpha$-1,3-glucanase를 생산하는 미생물의 분리, 동정 및 배양조건

  • A strain producing $\alpha$-1, 3-glucanase was isolated from soil samples. The strain has a grey aerial mycelium and showed brown color from the other side. The temperature and pH range for growth were from 10$\circ$C to 42$\circ$C and from 4 to 10, respectively. Arabinose, dextrin and mannose were utilized for growth. Analysis of cell wall components revealed that the strain was classified as type I. From the results, the strain was identified as Streptomyces sp. By using this strain, the maximum production of 0.65 units/ml for $\alpha$-1, 3-glucanase was achieved at 37$\circ$C for 48 hrs.

  • This study performed to establish tile characteristics of constant rate of strain consolidation(CRSC) tests. The values obtained by the CRCS tests were compared with oedometer tests. From the comparision of the results of oedometer and CRSC tests for soft soil from Haenam, Korea, it was concluded that. 1. When rate of strain is 0.9mm/hr, compression curve of CRSC test was not coincided with those of tile oedometer test, then preconsolidation stress was larger than those of other rates of strain. 2. Permeabilities from CRSC tests and direct measurements were about same each other, but permeabilities from Oedometer tests were large than those from others.

  • A bacterial strain KN, which highly produced a protease, was isolated from several soil samples and identified to to belong to the genus Bacillus. We selected mutant strain Bacillus sp. KN103N, which was hyperproducer of protease and was resistant to D-cyclowerine, from the strain KN by several steps of mutagenesis. Neutral protease productivity of mutant strain KN103N was about 55 times as much as that of the original strain KN. The optimum pH and temperature for the enzyme activity were 7.0 and 50$^{\circ}C$, respectively and the enzyme was relatively stable at pH6.0~8.0 and below 40$^{\circ}C$. The enzyme was inactivated by EDTA, but not by DFP. These results indicate that the enzyme from Bacillus sp. KN103N was a neutral (metallo-) protease.

  • A soil bacterium was isolated from maize roots cultivated in Korea (KNUC153). The isolate was partially classified on basis of l6S rDNA sequence analysis as Stenotrophomonas maltophilia. By the acetylene reduction assay (ARA), the strain KNUC153 contained nitrogen-fixing abilities. The amount of auxin produced by the strain KNUC153 was $77.6\;{\mu}g/ml$. The strain KNUC153 produced 4 times higher amount of l-amino-cyclopropane-l-carboxy­lic acid deaminase than that of the other known strain Azospirillum sp. KNUC82. Inoculation treatment with the strain KNUC153 for maize seeds showed positive effect on early growth of the plants.

  • A bacterial strain No.71, which produced alkaline protease, was isolated from soil and identified to the genus Bacillus. With the successive mutation, a mutant strain No. M-71, having high alkaline protease productivity, was obtanined from the parental strain No 71. Alkaline protease productivity of mutant strain No. M-71 was about 50 times as much as that of the parental strain No.71. The enzyme preparations showed strong activities toward casein, the optimum pH being 11.0 and the optimum temperature about $55^{\circ}C$.

  • Twenty microbial strains producing the acylase were isolated from soil by using Micrococcus luteus ATCC 9341 as an indicator strain, using either D-($\alpha $)-phenylglycine methylester and 7-aminocephalosporanic acid (7-ACA) or glutaric acid dimethylester and 7-ACA as substrates. Among the isolates, only one strain was turned out to be the 7-ACA producer from either cephalosporin C or glutaryl 7-ACA as the substrates by using the overlay of 7-ACA sensitive strain (SS5). 7-ACA produced from cephalosporin C by an isolate (APS20) was detected by high performance liquid chromatography. The isolated strain (APS20) was identified to Bacillus macerans on the basis of cellular fatty acid profile by gas chromatography. Bacillus macerans APS20 had no $\beta $-lacta-mase activity on cephalosporin C, and that is very important for the enzymatic production process of 7-ACA. However, this strain was resistant up to 100 $\mu $g/ml of cephalosporin C.

  • In this study, obtained large block sample and piston sample of marine clay in korea were performed unconfined compression and consolidation test. Soil properties of two type samples such as failure strain, between two parameter's ratio($E_{50}$ and $q_u$), and volumetric strain were used to evaluate sample disturbance. The result, large block samples show a low disturbance than piston samples. Therefore, we suggest new sample disturbance evaluated method through the relation of OCR and volumaric strain at shallow of marine clay in Korea and suggest new sample disturbance classified method by subdivided grade for failure strain of unconfined compression test.

  • A strain of actinomycetes producing extracellular adenosine deaminase inhibitor, strain V-8, was isolated from soil. Strain V-8 was gam positive and its cell wall chemotype was decided as cell wall chemotype I from analysis of diaminopimelic acid isomers and sugar pattern. This strain had a wide range of sugar utilization as carbon sources. The optimal pH and temperature for growth were $6.8{\sim}7.0$ and $28{\sim}30^{\circ}C$, respectively. From the morphological, chemotaxonomical characteristics and analysis of various physiological characteristics, the strain V-8 was identified Streptomyces sp.

  • From 120 soil and activated sludge, the strains able to grow on benzoate and m-Toluate have been isolated after selective enrichment which were later identified as Psudomonas sp. according to its morphological and biochemical characteristics. Ben-2 strain contained two plasmid DNA having about 120 Kb and below 2.0 Kb by agarose gel electrophoresis. Form the comparative investigation of catechol 1,2-oxygenase and catechol 2,3-oxygenase activities in Ben-2 strain and its cured strain, Ben-2 strain has both of these two enzymes while cured strain has catechol 1,2-oxygenase only.
