Isolation and Evaluation of Protective Effect against Fusarium Wilt of Sesame Plants of Antibiotic Substance from Bacillus polymyxa KB-8

  • An antibiotic compound was isolated from the culture of an antagonist against Fusarium oxysporum f. sp. sesami, Bacillus polymyxa strain KB-8, and tested for the control of Fusarium wilt of sesame in greenhouse conditions. Optimum conditions for culturing the antagonist to obtain the maximum antibiotic activity were determined using different culture media, initial medium acidity, and incubation periods for which yeast -malt extract agar with the initial acidity of pH 5 and over 13 days culture were best. Antibiotic substances extracted by methanol had 2 main fractions, KB-8A and KB-8B, in thin layer chromatography (OLC) with Rf values of 0.35 and 0.67 in a solvent system of chloroform : methanol = 7 : 3. The fraction KB-8A wa purified further by XAD-2, silica gel and Sephadex LH-20 column chromatography, and crystalization. Its minimum inhibitory concentrations (MICs) were $12.8\mu\textrm{g}$/ml for F. oxysporum and Alternaria mali, $6.4\mu\textrm{g}$/ml for Colletotrichum gloeosporioides and Rhizoctonia solani, and $3.2\mu\textrm{g}$/ml for Phytophthora capsici. Soil drenching of antibiotic KB-8A in the concentrations of $13.0\mu\textrm{g}$/ml and $26.0\mu\textrm{g}$/ml effectively inhibited the Fusarium wilt of sesame in a greenhouse test, which appeared to be comparable to the fungicide benlate of $6.5\mu\textrm{g}$ a. i./ml.

  • In the past it has been often observed that the shear stresses at failure are much smaller than the shear strength obtained from traditional laboratory tests and conventional analysis technique is inadequate in stiff soil, such as cemented sand. Many researchers have brought attention to the fact that the presence of flaws i.e. fissures, cracks, joints have a great effect on the strength and overall stress-strain behavior of such materials. They have thought that fracture mechanics may appropriately be adopted as a good tool for analysis of these materials. However, the use of fracture mechanics concept especially for cemented sands is faced with difficulties in obtaining relevant parameters, because fracture parameters and predictions are highly dependent on the material constituents and the size of specimens as well as the size of particles. This paper addresses the effects of sizes which include specimen and aggregate on fracture properties of cemented sand. The results of laboratory tests show that the sizes of specimens and particle have a great effect on the fracture properties such as nominal strength of cemented sand.

Effects of Geosynthetic Reinforcement on Compaction of High Water Content Clay (토목섬유 보강이 고함수비 점성토의 다짐에 미치는 영향)

  • This research was conducted to evaluate the effectiveness of reinforcement for nearly saturated soft clay compaction. The effectiveness was investigated by roller compaction test using nearly saturated clay specimens. The nearly saturated condition was obtained by submerging clay in the water for 12 hours. High water content specimens were compacted in plane strain condition by a steel roller. A specimen was compacted by four 5 cm horizontal layers. Specimens were prepared fur both reinforced and unreinforced cases to evaluate the effectiveness of reinforcement. Used reinforcement is a composite consisted of both woven and non-woven geotextile. The composite usually provides drainage and tensile reinforcement to hi인 water-contented clay so that it increases bearing capacity. Therefore, large compaction load can be applied to reinforced clay and it achieves higher density effectively. The reinforcement also increases compaction efficiency because it reduces the ratio between shear and vertical forces during compaction process. The maximum vertical stress on the base of specimen usually decreased with higher compaction thickness. The reinforcement increases soil stiffness under the compaction roller and it initiates stress concentration. As a result, it maintains higher vertical stress level on the base of specimen that provides better compaction characteristics. Based on test results, it can be concluded that the reinforcement is essential to achieve effective compaction on soft clay.

Characteristics and prediction methods for tunnel deformations induced by excavations

  • The unloading effect from excavations can cause the deformation of adjacent tunnels, which may seriously influence the operation and safety of those tunnels. However, systematic studies of the deformation characteristics of tunnels located along side excavations are limited, and simplified methods to predict the influence of excavations on tunnels are also rare. In this study, the simulation capability of a finite element method (FEM) considering the small-strain characteristics of soil was verified using a case study. Then, a large number of FEM simulations examining the influence of excavations on adjacent tunnels were conducted. Based on the simulation results, the deformation characteristics of tunnels at different positions and under four deformation modes of the retaining structure were analyzed. The results indicate that the deformation mode of the retaining structure has a significant influence on the deformation of certain tunnels. When the deformation magnitudes of the retaining structures are the same, the influence degree of the excavation on the tunnel increased in this order: from cantilever type to convex type to composite type to kick-in type. In practical projects, the deformation mode of the retaining structure should be optimized according to the tunnel position, and kick-in deformation should be avoided. Furthermore, two methods to predict the influence of excavations on adjacent tunnels are proposed. Design charts, in terms of normalized tunnel deformation contours, can be used to quantitatively estimate the tunnel deformation. The design table of the excavation influence zones can be applied to determine which influence zone the tunnel is located in.

3-Dimentional numerical study on dynamic behavior of connection between vertical shaft and tunnel under earthquake loading (3차원 수치해석을 이용한 지진 시 수직구-터널 접속부 동적 거동 분석)

  • 3D time history analysis was performed on vertical shaft-tunnel connection to provide insight into the dynamic stress-strain behavior of the connection considering the effects of soil layers, periodic characteristics and wave direction of earthquakes. MIDAS GTS NX based on FEM (Finite Element Method) was used for this study. From this study, it is revealed that the maximum displacement occurred at the upper part of the connection when the long period seismic waves propagate through the tunnel direction in soft ground. Also, stress concentration occurs due to different behaviors of vertical shaft and tunnel, and the stress concentration could be influence for safety on the connection. The results of this study could be useful for the seismic performance design of vertical shaft-tunnel connection.

Aerobic Liquid Fermentation of Residual Food Waste by Thermophilic Bacteria (고온세균을 이용한 남은 음식물의 호기적 액상발효)

  • For the probiotic feed production from residual food waste, aerobic liquid fermentation was conducted by thermophilic bacteria. 11 Strains of bacteria were isolated from several soil sources and residual food waste. Screening was carried by shaking incubator for the separation of thermophilic strain at $55^{\circ}C$. The isolated strains were tested for enzyme activities such as ${\alpha}$-amylase and protease. 6 Bacterial strains were chosen and were adapted by repeated fermentation processes in food waste substrate. The viable cell count of them at final fermentation stages were shown as $3-7{\times}10^9/ml$ in 2L-jar fermenter. Among them B3, B6 showed higher enzyme activity. By the mixed fermentation of B3, B6 and Bacillus stearothermophilus, the highest viable cell count reached to $1.4{\times}10^{10}/ml$ in 8 hours.

  • For the production of poly(3-hydroxybutyrate-co-3-hydroxyvalerate)(P(3HB-co-3HV)) from butyric acid and valeric acid, 10 strains of bacteria capable of producing P(3HB-co-3HV) were isolated from soil. Among them, the strain HJ-067 showed the best ability of producing P(3HB-co-3HV), and was indentified as a Azotobacter sp. For the production of P(3HB-co-3HV), the optimum concentrations of butyric and valeric acid were 3.0g/l, respectively. The most effective nitrogen source was $(NH_4)_{2}SO_4$ at an optimum concentration of 0.75g/l, which was equivalent to 21.36 in C/N ratio. Deficiency of the cationic metal ions ($Zn^{2+},\;Co^{2+},\;Mn^{2+}$) in the proguction medium had stimulating effect on P(3HB-co-3HV) accumulation, especially in the manganese. deficient medium. The optimum temperature for P(3HB-co-3HV) production was 27$^{\circ}C$ and the optimum initial pH was 7.0. Under the optimum conditions, 1.82g/l of P(3HB-co-3HV) and 3.00g/l of dry biomass were produced after 36 hour cultivation, and the P(3HB-co-3HV) yield and HV% were 60.60% (w/w), 15.92%, respectively.

Purification of Cellulase Produced from Cellulomonas sp. YE-5 (Cellulomonas sp. YE-5가 생산하는 Cellulase의 정제)

  • An extracellular cellulase producing bacterium YE-5 was isolated from soil, and identified as a Cellulomonas sp. by its taxonomical characteristics. The maximal activities of avicelase (0.35 units/ml), CMCase (3.18 units/ml), FPase (0.315 units/ml) and $\beta$-glucosidase (0.882 units/ml) were obtained when this strain was cultured for 48 hrs at $30^{\circ}C$ in a medium containing 0.8% (w/v) Solka floc, 0.06010 (wlv) urea, 0.1% (w/v) $K_2HP0_4$, 0.1% (w/v) $MgS0_4.7H2_0$, 0.2% (w/v) bacto peptone, 0.2% (w/v) yeast extract and pH 6.5. The cellulase was purified by ammonium sulfate fractionation, DEAE-Sepharose column chromatography and Sephadex 6-100 column chromatography from culture filtrate of Cellulomonus sp. YE-5. The molecular weights of purified avieelase, CMCase I, and CMCase II were estimated to be about 95,000 ~ 105,000, 46,000 ~ 47,000 and 120,000 ~ 125,000, respectively.

Antifungal Activity of Pseudomonas sp. DGUM 5051 Against Apple Bitter-rot Causing Fungus, Glomerella cingulata (사과탄저병균(Glomerella cingulata)에 대한 Pseudomonas sp. DGUM 5051의 항진균 활성)

  • The strain DGUM 5051, an antagonistic bacterium against apple-bitter rot causing Glomerella cingulata, was isolated from soil in Kyongju. Based on the morphological and physiological characteristics, the bacterium was identified as Pseudomonas sp. and named as Pseudomonas sp. DGUM 5051. The optimal pH and temperature for cell growth were pH 6.0 and $30^{\circ}C$, whereas those for antifungal activity were pH 7.0 and $24^{\circ}C$, respectively. Among the complex media tested, brucella medium, brain heart infusion medium and Luria-Bertani medium were good for both cell growth and antifungal activity. The high antifungal activity was found in the mineral salts medium, in which sucrose, $KNO_3$ and $K_2HPO_4$ were used as sources of carbon, nitrogen and phosphorus, respectively.

A report of 42 unrecorded actinobacterial species in Korea

  • During a study to discover indigenous prokaryotic species in Korea in 2016, a total of 42 actinobacterial isolates were recovered from various environmental samples collected from natural cave, squid, sewage, sea water, trees, droppings of birds, freshwater, eelgrass, mud flat, sediment and soil. On the basis of a tight phylogenetic clade with the closest species and high level of 16S rRNA gene sequence similarity, it was shown that each isolate was assigned to independent and previously described bacterial species which were assigned to the phylum Actinobacteria. The following 42 species have not been reported in Korea: eight species in two genera n the order Corynebacteriales, 26 species of 16 genera in the Micrococcales, one species of one genus in the Micromonosporales, one species of one genus in the Propionibacteriales, four species of two genera in the Streptomycetales and two species of two genera in the Streptosporangiale. Cell morphology, Gram staining reaction, colony colors and features, the media and conditions of incubation, physiological and biochemical characteristics, origins of isolation and strain IDs of 42 unrecorded actinobacterial species are presented in the species description.
