  • Geotrichum sp. MF01, isolated from oil-contaminated soil, utilized methyl ethyl ketone(MEK) as the sole source of carbon and energy. The strain MF01 showed a Michaelis-Menten kinetics on MEK, and the kinetic parameters determined for MEK degradation were; specific removal rate, $r_{max}$ = 0.14 $h^{-1}$; half-saturation constant, $K_m$ = 5.88 mM. The adsorption of MEK by heat-killed strain was 0.62 mg at 8.07 mg MEK indicating that the degradation was the primary removal mechanism over adsorption. Biodegradation of MEK was studied in a biofilter using perlite, vermiculite 0:1, v/v) as supporting material. During 57 days of biofilter operation, $^3h^{-1}$.

Enzymatic saccarification of cellulosic wastes by pectinase

  • The study was aimed to saccharify callulosic waste by pectinase produced from strain KL34 isolated from soil. The enzyme activity in the culture using 1%(w/v) fruit waste as carbon source reached to 3.8 U/ml. In the enzymatic hydrolysis of cellulosic waste, we obtained 9.5g/L reducing sugar in the condition of supernatant containing 5 U/ml enzyme and 10%(w/v) apple rind as substrate. Additionally, in enzymatic hydrolysis of food waste using pectinase from KL34, reducing sugar of 12.7g/L was obtained, indicating enhancement of 1.6 fold compared with that of only cellulase employment.

  • A bacterial strain YC4963 with antifungal activity against Colletotrichum orbiculare, a causal organism of cucumber anthracnose was isolated from the rhizosphere soil of Siegesbeckia pubescens (Siegesbeckia pubescens Makino;Family:Compositae) in Korea. Based on physiological and biochemical characteristics and 16S ribosomal DNA sequence analysis, the bacterial strain was identified as Pseudomonu aureofaciens. The bacteria also inhibited mycelial growth of several plant fungal pathogens such as Botrytis cinerea, Fusarium oxysporum and Rhizoctonia solani on PDA and 0.1 TSA media. The antibiotic activity was found from the culture filtrate of TSB(tryptic soy broth) and its active compounds were quantitatively bound to XAD adsorber resin. The antibiotic spectrum was broad and growth of C. orbiculare and F. oxysporum, B. cinerea were inhibited at very low concentration. The chemical data from various chromatographic procedures showed that active fraction consisted of at least two phenazine derivatives. However, the metabolites had no inhibitory effect on Pythium ultimum which was reported to be sensitive to phenazine antibiotics. The compounds responsible for the activity are now under investigation.

  • Settlement with crack on the hardened liners may occur in the weak clay due to waste load since the stiffness of the hardened liner is greater than that of the clay layers. Way of reducing deformation crack in the hardened liner is investigated using two computer programs, CONSOL and FLAC. The computer program CONSOL estimates the magnitude of settlement with time in clay layers and FLAC analyses the stress and deformation relationship between the foundation of landfill and waste load. The results show that a representative block of the analyzed area reaches the consolidation settlement of 1.32m, 8.8 years after the disposal of waste started with the degree of consolidation U=90%. The stress within the hardened liner exceeds the allowable vertical stress of 5kg/$\textrm{cm}^2$ and horizontal stress of 1.67kg/$\textrm{cm}^2$ at the concave part of the liner where the main and branch drainage pipes of leachate are located. It was recognized that the thickness of the interested area should be enlarged or the strength of the same area should be improved to tolerate the planned waste load.

  • Microorganisms, including the strain IJ-3 isolated from soil, were found to secrete an extracellular soybean protein coagulating enzyme and the strain, IJ-3 was identified as Genus Bacillus according to the Bergey's manual . The enzyme coagulated protein in soymilk , thus forming a curd at pHs 5.8-6.4 and at 55-75℃. The optimum temperature for soybean protein coagulating activity was 65-75℃ and the enzyme was stable at temperature below 50℃ and was found to be stable with about 60-100% of the original activity at a with pH ranges(pH6-7). The molecular weight of enzyme was estimated to be 28,000 by SDS-PAGE. The curd formed with the enzyme from Bacilus sp. IJ-3 has a smooth texture, and a mild taste without any bitterness or a beany flavor.

  • A bacterium(Y-5) which has potent antibacterial activity against swine atrophic rhinitis bacteria (Bordetella bronchiseptica, Pasteurella multocida) was isolated from soil and identified as a strain of Bacillus sp. Y-5 upon investigation of the morphological and physiological characteristics. The culture broth obtained from incubation of the Y-5 strain at $30^{\circ}C$ for 21 h in tryptose-bouillon agar medium (pH 6.0) showed active antibacterial activity against Bordetella bronchiseptica and the culture broth that of $30^{\circ}C$, pH 6.5, 18 h showed active antibacterial activity against Pasteurella multocida.

Characteristics of Biosurfactants produced by Bacillus sp. LSC11 (Bacillus sp. LSC11가 생산하는 biosurfactant의 특성)

  • Several bacterial strains producing biosurfactants were isolated from polluted marine and soil by oil. One of the strains named LSC11 showed strong production activity of biosurfactants. This strain was identified as a Bacillus sp. LSC11 based on the morphological, biochemical, and physiological characteristics. The biosurfactant, produced by the strain, emulsified crude oil, vegetable oil, and hydrocarbons. The surface tension of the culture broth of Bacillus sp. LSC11 decreased to 32 mN/m. The crude biosurfactant was obtained from the culture broth by acid precipitation, freeze drying, solvent extraction, and evaporation. The emulsifying activity of the biosurfactant showed better than the chemically synthesized surfactant (SDS, Span40, Span 85). The biosurfactants had strong properties as an emulsifying agent and as an emulsion-stabilizing agent.

Isolation of Chitin-utilizing Bacterium and Production of Its Extracellular Chitinase

  • A bacterial strain, designated as WY22, producing extracellular chitinase was isolated from the soil around the Youngduck area, after enrichment culture in a medium containing $1{\%}$ (w/v) wet colloidal chitin as a sole carbon source. The isolate was identified as a strain of Bacillus sp. based on its morphological and physiological characteristics. It was observed that Bacillus sp. WY22 could inhibit the growth of Fusarium oxysporum with hyphal extention-inhibition assay on potato dextrose agar plate supplemented with $1{\%}$ collidal chitin. Optimum culture conditions of Bacillus sp. WY22 were examined for chitinase production in a chitin medium. High level production of chitinase was observed not only in the chitin medium but in a medium supplemented with $1{\%}$ N-glucosamine or lactose instead of chitin. The optimum concentrations of colloidal chitin and yeast extract were 3.0 and $0.5{\%}$, and the optimum culture conditions for initial pH of medium and temperature were 7.0 and $30^{\circ}C$, respectively, for the production of chitinase.

  • Flexible pipes take advantage of their ability to move, or deflect, under loads without structural damage. Common types of flexible pipes are manufactured from polyethylene (PE), polyvinyl chloride (PVC), steel, glass fiber reinforced thermosetting polymer plastic (GFRP), and aluminum. In this paper, we present the result of an investigation pertaining to the short-term behavior of buried polyethylene pipe. The mechanical properties of the polyethylene pipe produced in the domestic manufacturer are determined and the results are reported in this paper. In addition, vertical ring deflection is measured by the laboratory model test and the finite element analysis (FEA) is also conducted to simulate the short-term behavior of polyethylene pipe buried underground. Based on results from soil-pipe interaction finite element analyses of polyethylene pipe is used to predict the vertical ring deflection and maximum bending strain of polyethylene pipe.

  • The Screening of fungi producing naringinase was done. A strain of Aspergillus midulans showed the highest naringinase activity among 447 strains those were isolated from soil, spoiled citrus friuts and stock cultures. The cultural Conditions of Asp. nidulans for production of naringinase were studied. A strain of Asp. nidulans showed higher activity when it was cultivated at 30$^{\circ}C$ for 3 days on wheat bran media supplemented with 2.0% naringin, 0.2% (NH$_4$)$_2$ SO$_4$ and 0.2% CaCO$_3$.
