토양과 작물근계의 유용미생물을 이용하여 작물생산성을 증대하고 병충해의 생물학적 방제를 위해서는 토양-근계의 미생물군집을 분석하고 그 기능을 밝히는 것이 전제가 되어야한다. 그러나 희석평판법으로는 극히 일부분만이 배양된다는 점을 고려할 때, 생존하지만 배양 불가능한 미생물의 군집 분석도 반드시 병행할 필요가 있다. 따라서 본 연구에서는, 고추재배지의 토양, 근권토양, 근면의 세균군집 구조해석을 위해서, 배양을 거치지 않고 각 시료로부터 직접 DNA를 추출하여 PCR증폭, 16S rDNA cloning, sequencing, 계통 해석을 행했다. 그 결과, 토양중에는 근권세균보다 미지의 동정이 되지 않는 세균이 우점하고 있었다. 27 clones 중에서 16 clones이 그램음성세균의 대표격인 Proteobacteria였으며, 방선균 등이 속해있는 고(高) G+C 그램양성세균군은 1 clone이 검출되었다. 그 외는 CFB 군이 2 clones, Verrucomicrobia가 1 clone이었고, Nitrospira가 1 clone이었으며 4 clones은 어느 군에도 속하지 않았다.
Many methods have been developed for the remediation of contaminated soil and groundwater. Among those technologies, in-situ bioremediation is most likely to be cost-effective method for petroleum hydrocarbon contamination. But the in-situ bioremediation can require more time to remediate hydrocarbon-contaminated soil and groundwater than other methods. Therefore we intended to save time of in-situ bioremediation using a biological additive to activate indigenous microbes in soil. The additive, 'Inipol EAP 22' stimulates the growth of specific flora, significantly accelerating the speed at which hydrocarbons are biodegraded. And it hans been tested in accordance with protocol approved by the USEPA and is registered on the National Contingency Plan Product Schedule List. In the experiment, three soil samples contaminated with fuel oil were prepared in the same concentration. Inipol EAP 22 was not added to one sample and was added to the other two samples with 5% and 10% of hydrocarbon by weight respectively. And $CO_2$gas derived from bacterial respiration was analyzed in each samples for 15 days. As a result, 145% and 153% of $CO_2$ evolution (microbial respiration) against the sample without 'Inipol EAP 22' occurred in samples with 'Inipol EAP 22' addition of 5% and 10%, respectively
한국의 토양에서 항생물질을 생성하는 균주를 분리하기 위하여, 토양으로 부터 분리한 균주의 항균력에 대한 Screening을 시행하였다. 그중 충청북도에서 채취한 토양에서 분리한 균주가 그람양성균에 매우 강한 항균력을 나타내어 이 균주에 대한 연구를 진행하였다. International Streptomyces Projeet methods 에 따라 생태학적 및 생리학적 성질을 관찰하여 Streptomyces에 속하는 종임을 확인하였다. 이 균주를 액내배양하여 얻은 항균성물질은 XAD-2와 CM-Sephades를 사용하여 정제하여 quinone류에 속하는 항생물질이라는 것을 밝혔다.
폐광지역 비소오염 토양에 이용될 식물상복원공법으로서 논에서 쉽게 발견되는 피의 적용가능성을 알아보고자 하였다. 식물을 이용한 토양 정화 공법이 가지는 장기간의 처리시간이 소요된다는 단점을 해결하고자 PSM(phosphate-solubilizing microbes)과 EDTA(ethylenediaminetetraacetic acid)를 처리하여 그 상승효과를 관찰하였다. PSM을 처리한 시험구에서는 식물의 바이오매스가 증가하여 비교 시험구에 비하여 비소재거 효율이 약 16% 정도 증가하였다. EDTA 처리구에서는 토양 내 중금속의 이동성야 증가하여 BF(bioaccumulation factor) 는 증가하였으나, EDTA가 식풀의 성장을 저해해 오히려 제거 효율이 20% 정도 감소하는 결과를 가져왔다. 따라서 식물성복원공법에 PSM 및 EDTA의 처리는 식물의 비소 흡수를 어느 정도 증대시킬 수는 있으나, 특히 EDTA를 처리할 경우에는 처리 시기 및 처리량의 선택에 있어서 식물의 성장을 저해하지 않도록 해야 하며, 지속적인 모니터링이 요구된다는 결과를 얻었다.
고온호기성발효장치(TAO system)로 발효 처리한 양돈분뇨고형물을 톱밥과 왕겨로 혼합한 뒤 성형건조장치를 이용하여 펠렛 비료를 제조하였다. 제조된 펠렛 비료(TAO-PF)의 비료이용성을 검토하기 위하여 배추를 이용하여 포장시험을 실시하였으며, 수확 후 생육량 및 토양의 생물, 이화학적 변화를 분석하였다. 생육실험은 펠렛 비료(TAO-PF)를 포함하여 시판 상토(Bed soil), 발효고형물(TAO-S), 화학비료(NPK), 무비료(대조구)로 나누어 실험하였다. 제조된 발효펠렛(TAO-PF)의 상온저장성은 미생물의 증식경향으로 보아 수분 함량이 $18\%,\;25\%$ 처리구보다는 $12\%$ 처리구가 적절한 것으로 판단되었다. 배추를 이용한 각 처리구에서의 생육실험결과, 토양세균의 변화는 TAO-PF 처리구가 NPK, 무비료구 보다 처리기간중 증식하는 경향을 보였으며, 특히 방선균의 증가와 사상균의 감소현상은 NPK, 무비료 처리 에 비해 TAO-S, TAO-PF 처리구에서 현저하여 발효고형물의 토양시비가 토양생물상 변화에 매우 효과적인 것으로 나타났다. 본 생육실험을 통해 배추의 구고, 구폭, 엽수 등으로 보아 TAO-S, TAO-PF 처리구는 각각 화학비료, 시판상토 처리구를 대체 할 수 있을 것으로 판단된다.
In our greenhouse experiment, soil heat treatment groups (50, 80, and 121℃) significantly promoted growth and disease suppression of Panax notoginseng in consecutively cultivated soil (CCS) samples (p < 0.01), and 80℃ worked better than 50℃ and 121℃ (p < 0.01). Furthermore, we found that heat treatment at 80℃ changes the microbial diversity in CCS, and the inhibition ratios of culturable microorganisms, such as fungi and actinomycetes, were nearly 100%. However, the heat-tolerant bacterial community was preserved. The 16S rRNA gene and internal transcribed spacer (ITS) sequencing analyses indicated that the soil heat treatment had a greater effect on the Chao1 index and Shannon's diversity index of bacteria than fungi, and the relative abundances of Firmicutes and Proteobacteria were significantly higher than without heating (80 and 121℃, p < 0.05). Soil probiotic bacteria, such as Bacillus (67%), Sporosarcina (9%), Paenibacillus (6%), Paenisporosarcina (6%), and Cohnella (4%), remained in the soil after the 80℃ and 121℃ heat treatments. Although steam increased the relative abundances of most of the heat-tolerant microbes before sowing, richness and diversity gradually recovered to the level of CCS, regardless of fungi or bacteria, after replanting. Thus, we added heat-tolerant microbes (such as Bacillus) after steaming, which reduced the relative abundance of pathogens, recruited antagonistic bacteria, and provided a long-term protective effect compared to the steaming and Bacillus alone (p < 0.05). Taken together, the current study provides novel insight into sustainable agriculture in a consecutively cultivated system.
Forest soils are often nitrogen-limited, and nitrogen input to forest soils can cause substantial changes in the structure and functions of a soil ecosystem. To determine the effects of nitrogen input on methane oxidation and the microbial enzyme activities, manipulation experiments were conducted using nitrogen addition to soil samples from Mt. Jumbong. Our findings suggested that the addition of nitrogen to the soil system of Mt. Jumbong did not affect the microbial enzyme activities. Conversely, the addition of nitrogen affected the rate of methane oxidation. Inorganic nitrogen in soils can inhibit methane oxidation via several mechanisms, such as substrate competition, toxic effects, and competition with other microbes, but the inhibitory effects are not always the same. In this research, seasonal changes were found to produce different inhibitory factors, and these different responses may be caused from differences in the methantrophic bacteria community structure.
After two highly weathered soils were treated with glucose, ammonium nitrate, monobasic potassium phosphate and biocides, and incubated for 4 or 6 weeks, adsorption tests were carried out to determine their effect on P adsorption. Glucose addition generally decreased P adsorption. The addition stimulated microbial activity, which might contribure to the reduced adsorption, probably through chelation and anion competition. Consistent endency was not observed with N treatment. Addition of P initially decreased P adsorption, probably through blockage of adsorption sites. Biocides generally decreased adsorption, probably because the microbes that 몬 been killed. Soil 1 with naturally lower levels of C and higher levels of aluminium adsorbed more P than soil 2. These results suggest that in highly weathered soils, which are low in available P and high in exchangeable Al, cultivation techniques which increase soil organic matter will also result in higher levels of plant-available P.
This study presents a technical perspective to the fate characteristics of phenol and m-cresol, which represent some of the most common organic chemicals found in chemical spill accidents, and likely to persist in soil and groundwater due to their highly stable physicochemical properties. Some cases of domestic and foreign chemical accidents linked to phenol and m-cresol contamination were compiled. Due to their low organic carbon-water partitioning coefficient (Koc), phenol and m-cresol tend to migrate into groundwater and remained as dissolved phase. On the other hands, phenol and m-cresol can be readily decomposed by microbes in soil and groundwater under appropriate conditions. Therefore, the fate characteristics of these chemicals are highly contingent on environmental conditions. Thus, if a great quantity of leakage is occurred by chemical accidents, the up-to-date and correct information about fate characteristics taking into account both the chemical and environmental conditions is greatly needed to minimized the potential hazards from these chemicals.
인광석을 처리한 토양에서 인산 가용화균 Penicillium oxalicum CBPS-3F-Tsa이 식물의 인산 흡수와 생육에 미치는 효과를 온실조건에서 조사하였다. 인광석을 처리한 토양에서 담배 식물체중의 인산 농도는 증가하였으며, 인산 가용화균을 접종한 경우에는 인산 농도가 더욱 증가하였다. 식물체의 총 인산 흡수량도 인산 가용화균을 접종한 경우 증가하였다. 담배 식물의 생육 또한 인광석과 인산 가용화균을 동시에 처리한 경우 인광석만 처리한 경우에 비하여 통계적으로 유의성 있게 증가하였는데, 이러한 인산 가용화균의 생육 촉진 효과는 인산 흡수에 미치는 영향과 밀접한 관계를 보였다. 따라서 인산 가용화균과 인광석 처리에서 나타난 식물의 인산 흡수량 및 생육 증대 효과는 결국 접종한 인산 가용화균에 의한 난용성 인광석의 가용화에 따른 결과로 판단된다.
