A bacterial strain NS-83 isolated from soil was able to produce an extracellular thermostable protease. The strain was identified as Pseudomonas aeruginosa based on its morphological and physiological characteristics. A thermostable protease from this strain has been purified to homogeneity as judged by SDS-PAGE and isoelectric focusing. The purification procedures included hydrophobic interaction, ion exchange, and gel filtration chromatography. The $M_r$ and the pl of the enzyme were 32,000 and 5.9, respectively. The optimal pH at 55$^{\circ}C$ and the optimal temperature at pH 7.0 were 8.0 and 60$^{\circ}C$, respectively. The D-values of the enzyme at 60, 65, and 70$^{\circ}C$ were 22, 2.1, and 0.75 hrs, respectively. The enzyme activity was significantly inhibited in the presence of 1 mM o-phenanthroline or EDTA, suggesting that the enzyme is metalloprotease. The $K_m$, and $V_{max}$ for Hammarsten casein were found to be 3.2 mg/ml and 0.918 unit/ml, respectively. These enzymatic properties were similar to those of elastase produced from P. aeruginosa IFO 3455, but the enzyme was clearly different from the reported elastase, in respect to $Ca^{++}$ effects on enzyme-thermostability. This property, together with amino acid composition analysis, confirmed that the enzyme differs from the known P. aeruginosa elastase.
In general, herbicides have been classified according to selectivity, mobility. time of application, methods of application, mode of action and chemical property and structure. However, there was no generally accepted classification system for practical use in the field. The primary processes affected by the majority of herbicides are the growth process through cell elongation and/or cell division, the photosynthetic process specifically the light reaction, the oxidative phosphorylation and the integrity of the membrane systems. The usual approach in the study of the mechanism by which herbicides kill or inhibit the growth of plants is to initially determine the morphological phototoxicity systems, The mechanism by which a herbicide kills a plant or suppresses its development is actually the resultant effect of primary and secondary(or side) effects. In most instances, the death of the plant is due to the secondary effects. To induce the desired response, a herbicide must be able to gain entry into the plants and once inside, to be transported within the plant to its site(s) of activity in concentrations great enough. Obstacles to the entry and movement of herbicides in plants are generally classified by leaf and soil obstacles, translocation obstacles and biochemical obstacles, and these obstacles are also strongly influenced by plant species and by environmental factors such as light, temperature, rainfall and relative humidity. And hence, in most instances, results obtained from laboratory or greenhous vary from those of field experiment. Author attempted to classify herbicides from the field experiment using the two-dimensional ordination analysis to obtain practical information for selecting effective herbicides or to choose effective herbicide combinations for increasing herbicidal efficacy or reducing the chemical cost. Based on this two-dimensional diagram, desired herbicides or combinations were selected and further investigated for the interaction effects whether these combinations are synergistic, additive or antagonistic. From the results, it was concluded that these new approach could possibly be give more comprehensive informations about effective use of herbicide than any other systems.

New technologies are providing unprecedented knowledge into microbial community structure and functions. Even though nucleic acid based approaches provide a lot of information, metaproteomics could provide a high-resolution representation of genotypic and phenotypic traits of distinct microbial communities. Analyzing the metagenome from different microbial ecosystems, metaproteomics has been applied to seawater, human guts, activated sludge, acid mine drainage biofilm, and soil. Although these studies employed different approaches, they elucidated that metaproteomics could provide a link among microbial community structure, function, physiology, interaction, ecology, and evolution. These approaches are reviewed here to help gain insights into the function of microbial community in ecosystems.

Nowadays, Korean consumers prefer drum-type washing machines to pulsator-type washers. Washing is a complex process involving the interaction of numerous physical and chemical influences. The main factors in the washing operations are the washing chemistry of the detergent along with the mechanical input, the wash temperature, and the time provided by the washing machine. Heavy-duty detergents that are used in drum-type washing machines contain different components from those used in vertical-axis washing machines. The bath ratio and the mechanical actions to which laundry is subjected are different between the drum-type and the vertical-axis washing machines. In this study we examined the effects of wash temperature, wash time, detergent concentration, and revolution speed on the removal of soils from artificially soiled cloths in a drum-type washing machine with heavy-duty commercial detergent. We used multiple regression analyses to find the relative importance of the factors and the optimum washing conditions. The results of these experiments showed that the washing temperature was the most important factor in the effective removal of most soils. This was followed by the washing time, the detergent concentration, and finally the revolution speed. In this study it was found that superfluous amounts of detergent did not sufficiently increase the soil removal rate. Koreans who are used to washing with cold water should increase the wash time to launder more efficiently.

Bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae (Xoo) is a very serious disease in rice-growing regions of the world. In spite of their economic importance, there are no effective ways of protecting rice plants from this disease. Bacteriophages infecting Xoo affect the population dynamics of the pathogen and consequently the occurrence of the disease. In this study, we investigated the diversity, host range, and infectivity of Xoo phages, and their use as a bicontrol agent on BLB was tested. Among the 34 phages that were isolated from floodwater in paddy fields, 29 belonged to the Myoviridae family, which suggests that the dominant phage in the ecosystem was Myoviridae. The isolated phages were classified into two groups based on plaque size produced on the lawn of Xoo. In general, there was a negative relationship between plaque size and host range, and interestingly the phages having a narrow host range had low efficiency of infectivity. The deduced protein sequence analysis of htf genes indicated that the gene was not a determinant of host specificity. Although the difference in host range and infectivity depending on morphotype needs to be addressed, the results revealed deeper understanding of the interaction between the phages and Xoo strains in floodwater and damp soil environments. The phage mixtures reduced the occurrence of BLB when they were treated with skim milk. The results indicate that the Xoo phages could be used as an alternative control method to increase the control efficacy and reduce the use of agrochemicals.

Subway tunnel structure has been rapidly developed in many cities for its strong transport capacity. The model-based damage detection of subway tunnel structure is usually difficult due to the complex modeling of soil-structure interaction, the indetermination of boundary and so on. This paper proposes a new data-based method for the damage detection of subway tunnel structure. The root mean square acceleration and cross correlation function are used to derive a statistical pattern recognition algorithm for damage detection. A damage sensitive feature is proposed based on the root mean square deviations of the cross correlation functions. X-bar control charts are utilized to monitor the variation of the damage sensitive features before and after damage. The proposed algorithm is validated by the experiment of a full-scale two-rings subway tunnel lining, and damages are simulated by loosening the connection bolts of the rings. The results verify that root mean square deviation is sensitive to bolt loosening in the tunnel lining and X-bar control charts are feasible to be used in damage detection. The proposed data-based damage detection method is applicable to the online structural health monitoring system of subway tunnel lining.

Geochemistry of metasedimentary groundwaters and spar waters has been studied in comparison with that of granitic groundwaters in Korea. Metasedimentary groundwaters show $Ca^{2+]$-${HCO_3}^-$ type at depth and low sodium concentrations compared with granitic groundwaters, which is due to the lack of plagioclase in their aquifer mineralogy and, therefore, the predominant reaction of calcite dissolution. According to factor analysis, metasedimentary groundwaters at 100~300 m depth are represented by 1) the dissolution of calcite and Mg-carbonates, 2) transformation of kaolinite to illite, and 3) the presence of sodium as not the product of plagioclase dissolution but a artificial pollutant. Discriminant function between the granitic and metasedimentary groundwaters shows a good discriminating ability with 81.8%, and groundwaters of volcanic aquifer, which has abundant plagioclase, are included in the granitic group by this function. Spa water samples show the result of active water-rock interaction due to high temperature.
This paper is results of experimental and nunerical works on the behavior of the cut-and-cover tunnel. Centrifuge model tests were performed to simulate the behavior of the cut-and-cover tunnels having cross sections of national road and subway tunnels. Model experiments were carried out with changing the cut slope and the slope of filling ground surface. Displacements of tunnel lining resulted from artificially accelerated gravitational force up to 40g of covered material used in model tests, were measured during centrifuge model tests. In model tests, Jumunjin Standard Sand with the relative density of 80 % and the zinc plates were used for the covered material and the flexible tunnel lining, respectively. Basic soil property tests were performed to obtain it's the property of Jumumjin Standard Sand. Shear strength parameters of Jumunjin Standard Sand were obtained by performing the triaxial compression tests. Direct shear tests were also carried out to find the mechanical properties of the interface between the lining and the covered material. Numerical analysis with the commercially available program of FLAC were performed to compare with results of centrifuge model experiment In numerical modelling. Mohr-Coulomb elasto-plastic constitutive model was used to simulaye the behavoor of Jumunjin Standard Sand and the interface element between the lining and the covered material was implemented to simulate the interaction between them. Compared results between model tests and numerical estimation with respect to displacement of the lining showed in good agreements.
The hormonal requirement suiting micropropagation of Morus alba during any season throughout the year was studied. Sprouting frequency from axillary buds of M. alba was greatly influenced by the time of explant collection, the highest value was achieved when nodal explants were collected at the end of bud dormancy period (late in March) and cultured on Murashige and Skoog (MS) medium supplemented with low concentration (0.5 mg/L) of BAP, kinetin or IBA (85-68%). In addition, they showed higher axillary bud sprouting on growth-regulators-free medium (49%) than others collected in autumn or winter and cultured on medium supplemented with various growth regulators (47-48%). Regardless of that period, young explants with greenish buds collected in summer exhibiting high sprouting frequency (66%) on MS medium supplemented with 0.5 mg/L kinetin and 0.5 mg/L GA3. Shoot multiplication via adventitious bud formation was achieved when the nodal explants were cultured on MS medium supplemented with 2 mg/L BAP and 0.2 mg/L IBA. Further multiplication via nodal explants of in vitro grown shoots was obtained on MS medium supplemented with 0.5 mglL BAP and 0.5 mg/L GA3. While half strength MS medium supplemented with low concentration (0.5 mg/L) of IBA, IAA or 2,4-D stimulated adventitious root formation, IBA was the best. After transfer the plantlets to the soil, acclimatization for three weeks was essential prerequisite for survival in high frequency (92%). Peroxidase activity is related to break of bud dormancy where maximum enzyme activity was detected when the lateral buds were induced to commence growth under field condition (early in spring) or in vitro.
Investigations related with factors influencing root and crown rot are rare and mainly related to farming practice and soil management. The main objective of this study was to examine broader range of factors influencing stem-base infestation of winter wheat in the field conditions. The effect of spatial distribution of infected plants on disease index (DIs) assessments was also investigated. Analysis of factors influencing DIs of crown rot of wheat demonstrated significant influence of the growing seasons (P < 0.001) and extreme fluctuations in winter temperatures (P < 0.001). In addition to that, localities together with their interaction with the growing season also significantly influenced DIs (P < 0.001). Aggregation of infected plants influenced variability of DI estimations, and it was pointed out that more extensive investigation should be conducted on broad range of DI in order to establish sampling method giving uniform sampling precision. Fusarium graminearum was shown to be predominant Fusarium species in Serbia (72.6%) using sequence-characterized amplified region analysis. Interestingly F. oxysporum was isolated in higher frequencies (27.4%) than it was reported in the literature. Given that there were no reports on the diversity of Fusarium species causing crown rot of wheat in Serbia, this study presents first report on this important subject. It also indicated that more attention should be focused on combined effects of abiotic and biotic factors influencing stem-base infestation of winter wheat. This knowledge will contribute to better understanding of factors influencing root and crown rot of wheat which would ensure sustainable disease management in the future.
