Design and Optimization of Solid Dispersed Osmotic Pump Tablets of Aceclofenac, A Better Approach to Treat Arthritis

  • The aim of this work was to prepare porous osmotic pump tablets for controlled delivery of Aceclofenac. Aceclofenac solid dispersion was prepared to improve the solubility by using the drug - carrier (Mannitol) ratio of 1:1. The osmotic pump tablets were prepared using the solid dispersed product of Aceclofenac. The formulation contains potassium chloride as osmotic agent, cellulose acetate as semipermeable membrane, poly ethylene glycol (PEG 4000) as pore former and sodium lauryl sulphate (SLS) as solubility enhancer. The formulations were designed by the general factors such as osmotic agent and pore former. All formulations were evaluated for various physical parameters and, the in vitro release studies were conducted as per USP. The drug release kinetic studies such as zero order, first order, and Higuchi and Korsmeyer peppas were determined and compared. All the formulations gave more controlled release compared to the marketed tablet studied. Numerical optimization techniques were applied to found out the best formulation by considering the parameter of in vitro drug release kinetics and dissolution profile standards. It was concluded that the porous osmotic pump tablets (F7) composed of Aceclofenac solid dispersion/Potassium chloride/Lactose/Sodium lauryl sulphate/Magnesium Stearate (400/40/95/10/5, mg/tab) and coating composition with Cellulose acetate/ PEG 4000 (60/40 %w/w) is the most satisfactory formulation. The porous osmotic pump tablets provide prolonged, controlled, and gastrointestinal environment-independent drug release.

  • 세라마이드, 콜레스테롤, 자유지방산은 각질세포간 주요 지질로써 피부장벽의 생성과 유지에 있어 중요한 역할을 담당한다. 그러나, 개에서 각질세포간 지질에 대한 역할은 거의 알려져 있지 않다. 본 연구는 개에서 1.25% sodium lauryl sulphate (SLS)에 의한 피부 장벽손상 유발후 2% 세라마이드, 2%콜레스테롤, 2% 리놀레익산과 이들 세가지 지질의 혼합제 (혼합제)의 장벽손상 복구를 평가하고자 실시하였다. 피부장벽기능은 표피경유수분소실(TEWL), 피부 수화도, 피부 산도, 피부 두께 측정을 통하여 평가하였고 최종적으로 조직학적 분석과 투과 전자 현미경 (TEM)을 통하여 피부 장벽구조를 평가하였다. SLS는 개의 피부에 효과적으로 피부장벽 손상을 유발하였다. 표피경유수분소실은 세라마이드와 혼합제 적용부위에서 계면활성제 및 대조군과 비교하여 유의적인 감소를 나타내었다 (p<0.05, p<0.01). 표피경유수분소실은 12일째에 모든 지질 적용군에서 유의적으로 감소하였으나 특히 세라마이드와 혼합제에서 콜레스테롤과 리놀레익산보다 유의적으로 감소하였다 (p<0.01). 실험 기간 동안, 피부 수화도는 세라마이드와 혼합제에서 유의적 증가를 보였다 (p<0.01). 조직학적 분석에서도 각각의 지질들은 피부손상을 회복시켰다. 특히 세라마이드와 혼합제의 효과가 탁월하였고 각질층의 두께도 대조군에 비해 유의적으로 증가되었다. 피부 산도는 세라마이드, 리놀레익산, 혼합제에서 유의적으로 감소하였다 (p<0.05). 따라서 세라마이드와 혼합제의 국소적용은 계면활성제에 의한 피부 장벽 손상에 효과가 탁월함을 확인하였으며 아토피와 같은 염증성 피부염과 외부 자극에 의한 피부 장벽 손상시에도 세라마이드 또는 혼합제의 병용이 효과적일 것으로 사료된다.

Carrageenan as a Rheology Agent for Mild Cleansing Applications.

  • Viscarin is a tradename given to viscosifying carrageenans manufactured by FMC BioPolymer. The suitability of Vis car ins as rheology agents in mild cleansing applications has been investigated. Rheological properties, foam volume and clarity were measured to determine the impact of including 1 % Viscarin on 10% solutions of the following surfactants: acylglutamate, cocoamidopropyl betaine, PEG-80 laurate, sodium lauryl sulphate and sodium lauryol sarcosinate. Viscosity, pseudoplasticity and thixotropy of Viscarin/surfactant solutions varied with surfactant type. In all cases, the addition of Viscarin substantially increased viscosity. For example, at a shear rate of 1 sol, all surfactant solutions had viscosities <0.1 Pa s while viscosities of Viscarin/surfactant solutions ranged from 10 to 60 Pa s. By comparison, a solution of 1 % Viscarin had a viscosity of 0.3 Pa s. Clarity of surfactant solutions decreased in all cases on the addition of Viscarin. However, it was found that by including a mild solubilizing surfactant, such as PEG 40 hydrogenated castor oil, crystal clarity could be maintained in Viscarin/surfactant solutions. Viscarin increased the foam volume of sodium lauryolsarcosinate solutions from 10 ml to 220 ml and had no impact on the foam volume of the other surfactants tested. These results were used to formulate a clear, ultra-mild foaming cleansing gel based on sodium lauryol sarcosinate and Viscarin without the need for a secondary, foam-boasting surfactant. A mild shampoo was also formulated. Both products have excellent skin-feel and are capable of suspending bubbles and solid inclusions.

Inhibitory Effect of {Surfactant- MnO4-} Aggregation in KMnO4 Oxidation of Proline and Methionine: A Kinetic Study

  • Anionic (sodium lauryl sulphate, NaLS) cationic (cetyl ammonium bromide, CTAB) and non-ionic (Tween-80) surfactants have been found to inhibit the rate of oxiadation L-proline and L-methionine by alkaline $KMnO_4$. A first order dependence of rate of oxidation was observed with respect to $MnO_4{^-}$. The order of reaction in substrate and alkali was found to be fractional nearby 0.65 and 0.55 in Aminoacid and $OH^-$, respectively. An aggregation/association between $MnO_4{^-}$ and surfactant has been confirmed spectrophotometrically. A mechanism, involving kinetically inactive [$MnO_4{^-}$ surfactant] aggregate and consistent with kinetic data, has been proposed. The effect of surfactants has been discussed in terms of hydrophobic and electrostatic interactions.

조제 첨가 음이온 발포제의 거품 효율 (Foaming Efficiency of Anion Foaming Agent Solution to Add Dyeing Assistants)

  • To optimize the foam dyeing procedures for polyester and polyester/nylon blended non-woven fabrics, the effects of organic solvents and surfactants were investigated by measuring the foam heights and surface tensions of the foam dyeing solution. The results of the experiments can be summarized as follows: 1) Organic solvents and surfactants (sodium lauryl sulphate: SLS) solution lower the surface tension (ST) of the water, but ST lowering rate of SLS solution is greater than that of solvents. For a 0.25% SLS solution, the minimum surface tension was 30.3 dyne/cm, which is nearly the same value for organic solvents. 2) For 0.25% SLS solutions, additional adding of a 4.0% organic solvent makes the foam height (FH) be its maximum. 3) At 0.6 g/ι dye concentration, incorporation of 0.4% SLS makes the surface tension of the foam solution be its minimum. The foam height did not show any trend due to the dye type. 4) The effect of foam stabilizers (sodium alginate (Alg-Na) and hydroxy ethyl cellulose (HEC)) were also investigated. The foam height of the foaming solution with HEC was greater than that with Alg-Na. The foam stability of the foaming solution with Alg-Na was better than that with HEC.

미립잠열슬러리의 유체역학적 특성연구 (Fluid dynamical characteristics of microencapsulated phase change material slurries)

  • An experimental study was peformed to measure the viscosity of microencapsulated PCM slurries as the functions of its concentration and temperature, and also influence to its fluid dynamics. For the viscosity measurement, a rotary type viscometer, which was equipped with temperature control system, was adopted. The slurry was mixed with water and Sodium Lauryl Sulphate as a surfactant by which its suspended particles were dispersed well without the segregation of particles during the experiment. The viscosity was increased as the concentration of MicroPCM particle added. The surfactant increased 5% of the viscosity over the working fluid without particles. Experiments were proceeded by changing parameters such as PCM particles'concentration as well as the temperature of working fluid. As a result, a model to the functions of temperature for the working fluid and its particle concentration is proposed. The proposed model, for which its standard deviation shows 0.8068, is agreed well with the reference's data. The pressure drop was measured by U-tube manometer, and then the friction factor was obtained. It was noted that the pressure drop was not influenced by the state of PCM phase, that is solid or liquid in its core materials at their same concentration. On the other hand, it was described that the pressure drop of the slurry was much increased over the working fluid without particles. A friction factor was placed on a straight line in all working fluids of the laminar flow regardless of existing particles as we expected.

Cytokine Release from Organotypic Human Oral Tissue Following Exposure to Oral Care Product Chemicals

  • Purpose: Measuring viability of a three-dimensional in vitro organotypic human oral tissue model has been suggested as an alternative test method to the oral mucosa irritation test of oral care products. The aim of this study was to investigate the production of two different cytokines using organotypic human oral tissue model following exposure to chemicals that are commonly used in oral care products. Materials and Methods: The organotypic human oral tissues were exposed to ethanol, sodium lauryl sulphate or hydrogen peroxide for 90 minutes. Following exposure, interleukin (IL)-1α and IL-8 productions were assessed and correlated with cell viability testing as well as histology of the organotypic human oral tissues. Result: High levels of IL-8 were released from organotypic human oral tissues in all of the test and control groups without any significant differences between them. In contrast, differences were found in IL-1α release between the test and control groups. Additionally, the trend of IL-1α release corresponded to the phenotypes observed in histological analysis while different trend existed between IL-1α release and cell viability. Conclusion: The study concluded the non-specific release of IL-8 for the assessment of oral care product chemicals' toxicity, while potential of measuring IL-1α cytokine level as the possible alternative test method.

  • Bacillus thuringiensis var. alesti (H. Serotype III)로부터 분리된 단백질 독소의 물리화학적 성질 및 누에에 대한 생물학적 검정에 있어서 다음과 같은 결과를 얻었다. 1. Nutrient agar 배지를 사용하여 항온기에서 3$0^{\circ}C$에 세균을 배양했을 때 완전한 Bacterial lysis는 30일 걸렸고 액체 배지를 사용하여 발효기(Fermentation Jar)에 배양했을 때는 그 기간이 반으로 단축되었다. 2. 분리된 결정체들만으로부터 추출된 단백질 독소와 포자-결정체의 혼합들로부터 분리된 독소와는 단백질 독소의 순수도에 있어서 차이가 없었다. 3. 단백질 결정체를 알카리(pH 9.5)에 용해시켜 Sodium Lauryl Sulphate Polyacrylamide gel을 사용한 전기영동 을 행했을 때 분자량이 120,000, 87,000, 74,000의 3개의 단백질의 분리되었다. 4. 분리된 독소의 누에에 대한 생물학적 검정에 있어서 4령기 누에 체중 g당 LD$_{50}$는 0.080$\mu\textrm{g}$이었다.

Studies on Solvent Sublation of Trace Heavy Metals by Continuous Flow System as Ternary Complexes of 1,10-Phenanthroline and Thiocyanate Ion

  • A continuous flow system has been developed to determine trace Cu(II), Mn(II), Ni(II) and Zn(II) in a large volume of water samples by a solvent sublation technique. The mixed solution of 1,10-phenanthroline(phen) and thiocyanate ion was used as ligands for the formation of their ternary complexes. The continuous system was constructed in this laboratory with a peristaltic pump, a mini shaker, three mixing bottles and a flotation cell by connecting each part with a polyethylene tube. The flotation conditions such as the flow rate of sample solution and the injection rates of ligand, buffer and surfactant solutions have been investigated to obtain the best sublation efficiencies. Each solution flowed into the flotation cell through each polyethylene tube by the peristaltic pumps. The ternary complexes were floated and extracted into MIBK in a flotation cell of 2 L by bubbling a nitrogen gas. The absorbances of extracted analytes in MIBK were directly measured by graphite furnace-AAS. The concentrations of 1,10-phenanthroline and thiocyanate ion were $2.6\;{\times}\;10^{-3}$ M and $2.3\;{\times}\;10^{-2}$ M in the mixed solution, respectively. The pH of sample solution was adjusted to 5.0 with a buffer solution and 1%(m/v) sodium lauryl sulphate solution was added as a surfactant to support the effective flotation of the complexes. The $N_2$ gas was bubbled at 30 mL/min for 90 minutes for 20 L of sample. Reproducible results of less than 10% RSD and recoveries of 80-120% could be obtained in real samples.
