Tests of concrete CLWL-DCB specimens had been conducted with displacement-controlled dynamic loading. The crack velocities for 381mm crack extension were 0.80 mm/sec ~ 215m/sec. The external work and the kinetic and strain energies were derived from the measured external load and load-point displacement. The fracture resistance of a running crack was calculated from the fitted curves of the fracture energy required for the tests. The standard error of the fracture energy was less than 3.2%. The increasing rate of the fracture resistance for 28 mm initial crack extension or micro-cracking was relatively small, and then the slope of the fracture resistance increased to the maximum value at 90∼145 mm crack extension depending on crack velocity. The maximum fracture resistance remained for 185 mm crack extension, and then the faster crack velocity showed the faster decreasing rate of the maximum fracture resistance. The maximum fracture resistance increased proportionally to the logarithm of the crack velocity from 142 N/m to 217 N/m when the crack velocity was faster than 0.273 m/sec. The maximum fracture resistance of the fastest tests was similar to the average fracture energy density of 215 N/m. To measure the fracture resistance of concrete, the stable crack extension should be larger than 90∼145 mm depending on crack velocity.
In order to study the morphologic changes of the unfixed odontoblasts suspended in phenol solution of several different concentrations, the author carried out the extraction of lower incisor of S-D strain rats to collect the odontoblasts, and the cells obtained were suspended immediately in saline solution. After observing the odontoblasts in fresh state, the saline solution was substituted with 0.125%, 0.25% 0.5%, 1% and 2% diluted phenol solutions. The morphologic changes were examined with phase contrast microscope at intervals of 10, 30, and 60 minutes. The results were as follows: 1. In saline solution the odontoblast showed cytoplasmic swelling, slender cytoplasmic process, thick rim nuclear membrane with increased dark contrast, and prominent nucleoli and chromatin granules with lapse of time intervals. In accordance with time intervals, blisters appeared in the supranuclear zone and increased its size and moved outward of the cytoplasmic membrane resulting detachment from the cell membrane. The phase dark cytoplasmic granules were increased in its dark contrast and in its size. 2. In 0.125% and 0.25% phenol solution, the odontoblasts and its nucleus shrunk immeidately and its contrast of cellular components was increased. With the lapse of time, the phase-dark granules in cytoplasm were aggregated, and several blisters were formed in and out of the cells. The outline of cytoplasmic membrane was also obscured. 3. In 0.5% phenol solution, the necleus shrunk at once, but soon after it revealed karyolysis accompanying dark contrast of neclear components such as nuclear membrane, nucleoli, and chromatin granules. On the contrary, the cytoplasmic granules showed aggregation and increased dark contrast, small and large blisters were formed in and out of the odontblasts and the outline of cytoplasmic membrane became obscured. 4. In 1% phenol solution, it showed shrinkage of odontblasts and its nuclei with thick rim nuclear membrane, aggregation of chromatin granules and occasional karyorrhexis. The dark contrast of cytoplasmic granules was increased and aggregated each other. But the blister formation could not be found. 5. In 2% phenol solution, it showed the shrinkage of odontoblasts and pyknotic nuclei with increased dark contrast of nucleoli and chromatin granules. The number of cytoplasmic granules was decreased by aggregation. But the blister formation could not be found as in 1% phenol solution.
In order to measure the inter- and intraspecific genetic divergences within the genus Alexandrium, the variations within the internal transcribed spacer (ITS1 and ITS2) regions and 5.85 ribosomal RNA gene of eight Alexandrium species were examined for 33 strains from diverse geographical locations by direct sequencing. Five isolates of A. tamarense (AT-2, AT-6, AT-10, AT-A and AT-B) from Jinhae Bay, Korea were found to be completely identical to a Japanese strain OFX151-A. The length of the amplified ITSI-5.85-ITS2 region varied from 481 nucleotides (in A. margalefi) to 528 nucleotides (in A. affine CU1-1). ITS1 and ITS2 nucleotide lengths were negatively correlated, whereas a positive correlation was found between their G+C content. The degree of sequence divergence ranged from 0.3% (1 bp) to a maximum of 53% (305 Up). Pairwise sequence comparisons revealed a small degree of divergence between A. tamarense and A. Pundyense isolates (1.2 - 2.3% = 6-12 bp), but a high degree of divergence between A. tamarense and A. catenella (19.8% = 102 bp), and between A. catenella and A. Pundyense (19.7%). Although most nodes were weakly supported by bootstrap values, some types tend to form independent molecular groups. A. catenella isolates also formed an independent molecular sub-group, with relaticula strong bootstrap values (94% or 85% and 79% or 98%, respectively in PAUP and NJ trees). Interestingly, A. cohorticula and A. frateculus always clustered within the same sub-group, this result being supported by strong bootstrap values. Our results indicate that the ITS regions provide useful informations on hierarchical population genetic structure and a high phylogenetic resolution in intraspecific and interspecific Alexandrium population.
We conducted both the small subunit ribosomal DNA (SSU rDNA) polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and mitochondrial (mt) DNA RFLP analyses for a genetic characterization of Acanthamoeba isolates from contact lens storage cases of students in Seoul, Korea. Twenty-three strains of Acanthamoeba from the American Type Culture Collection and twelve clinical isolates from Korean patients were used as reference strains. Thirty-nine isolates from contact lens storage cases were classified into seven types (KA/LS1 , KA/LS2, KA/LS4, KA/LS5, KA/LS7 KA/LS18, KA/LS31). Four types (KA/LS1 , KA/LS2, KA/LS5, KA/LS18) including 33 isolates were regarded as A. castellanii complex by riboprints. KA/LS1 type was the most predominant (51.3%) in the present survey area, followed by KA/LS2 (20.9%), and KA/LSS (7.7%) types. Amoebae of KA/LS1 type had the same mtDNA RFLP and riboprint patterns as KA/E2 and KA/E12 strains, clinical isolates from Korean keratitis patients. Amoebae of KA/LS2 type had the identical mtDNA RFLP patterns with A. castellanii Ma strain, a corneal isolate from an American patient as amoebae of KA/LS5 type, with KA/E3 and KA/E8 strains from other Korean keratitis patients. Amoebae of KA/LS 18 type had identical patterns with JAC/E1, an ocular isolate from a Japanese patient. Three types , which remain unidentified at species level, were not corresponded with any clinical isolate in their mtDNA RFLP and riboprint patterns. Out of 39 isolates analyzed in this study, mtDNA RFLP and riboprint patterns of 33 isolates (84.6%) were identical to already known clinical isolates, and therefore, they may be regarded as potentially keratopathogenic. These results suggest that contact lens wearers in Seoul should pay more attention to hygienic maintenance of contact lens storage cases for the prevention of Acanthamoeba keratitis.

The structural provisions of rectangular CFT (concrete-filled tubular) columns in the 2005/2010 AISC Specification, ACI 318-08, and EC4 were comparatively analyzed as a preliminary study for establishing the unified standards for composite structures. The provisions analyzed included those related to the nominal strength, the effect of confinement, plate slenderness, effective flexural stiffness, and the material strength limitations. Small or large difference can be found among the provisions of AISC, ACI, and EC4. Generally, the 2010 AISC Specification provides the revised provisions which reflect up-to-date test results and tries to minimize the conflict with the ACI provisions. For example, the 2010 AISC Specification introduced a more finely divided plate slenderness limits for CFT columns. In seismic applications, the plate slenderness limits required for highly and moderately ductile CFT columns were separately defined. However, the upper cap limitations on material strengths in both the AISC and EC4 provisions are too restrictive and need to be relaxed considering the high-strength material test database currently available. This study found that no provisions reviewed in this paper provide a generally satisfactory method for predicting the P-M interaction strength of CFT columns under various material combinations. It is also emphasized that a practical constitutive model, which can reasonably reflect the stress-strain characteristics of confined concrete of rectangular CFT columns, is urgently needed for a reliable prediction of the P-M interaction strength.

The characteristics of shear zone in granular soils largely affect the stability of geo-structures. The goal of this study is to evaluate shear zone in a direct shear test using shear wave, electrical resistivity, and cone tip resistance. Bender elements and electrical resistivity probe are embedded into the wall of a direct shear box made of transparent acrylic material to estimate the shear wave velocities and the electrical resistivity at shear and non-shear zones. At the point of peak and residual strength, micro cone penetration test which can be available to measure tip resistance has been performed. Experimental results show that the shear wave velocities at upper shear zone increase during shearing while the values remain constant at bottom and lower shear zone. Also, resistivities at lower shear zone depend on relative density while resistivities at bottom remain constant. The results of cone penetration test demonstrate the correlation of the cone tip resistance and small strain shear modulus at shear zone. This study suggests that the application of the modified direct shear box including shear wave, electrical resistivity and the micro cone tip resistance may become effective tools for analyzing the characteristics of a shear zone.
Aspergillus niger S-1 was proved to be a good hesperidinase producer which have been selected for naringinase utilization. Enzyme of this strain had good characteristics and purified relative high degree with good recovery by ammonium sulfate or aceton treatment. Results obtained were summarized as follows (1) The enzyme was most active at 60$^{\circ}C$, when the reaction was performed in the pH 4.0 for 30min. Optimum pH for enzyme activity was 5.0 and activity was retained 78% at pH value 3.5. (2) Hesperidinase activity retained 95% of its full activity after treatment at 60$^{\circ}C$ for 30min at pH value 4.0., 70% at 70$^{\circ}C$ and 65% at 80$^{\circ}C$. Most stable pH of this enzyme was showed 5.0 after treatment for 24hr at 4$^{\circ}C$ (3) Only Magnesium ion activated enzyme reaction, while other metallic ions, Cu$\^$++/, Mn$\^$++/, Pb$\^$++/, Mo$\^$++/, Ag$\^$++/, Hg$\^$++/ inhibited. (4) Eleven fold purification with 35% recovery was obtained in the case of 60% aceton treatment and 10-fold purification with 5.6% recovery was showed with 40% aceton comparing to the crude extract Enzyme. (5) Crude enzyme precipitated with 0.4-0.6 saturated ammonium sulfate contained 13f6 of the original enzyme activity with 48-fold increase in specific activity and enzyme has been purified 25 fold with a yield 19% by 0.6-5.8 saturation. (6) Hesperidinase formation was noticeably increased by addition of small amount of orange-peel extraction on the wheat bran medium.
Tensile constitutive stress-strain model of steel fiber reinforced concrete (SFRC) in fib MC2010 was investigated. In order to model tensile behavior of SFRC, three point loading flexural tests were conducted on notched small beams according to BE-EN-14651. Design parameters for the constitutive model were determined from the flexural tests. Flexural test and finite element analysis were conducted on large SFRC beam without steel reinforcements and compared with each other. In addition, parametric study on the effect of compressive and tensile model, and characteristic length on flexural behavior of the SFRC beam was conducted also. In results, pre-peak load-displacement curves from the FE analysis was close to experimental curves but significant difference was shown in post-peak behavior. The reason of the difference is originated from the fact that the fiber distribution and orientation were not being properly considered in the MC2010 model. This study shows that modification and detail explanations on the orientation factor K in MC2010 might require to better reproduce the behaviour of large scale SFRC beams.

A fifth equation is required to determine the five independent elastic constants of a transversely isotropic rock from compression test of a single specimen. As an approximation proposed by Saint-Venant has been used for long time, it may cause an erroneous result in some cases, especially for specimen with low angle of anisotropy. Three equations were suggested replacing this traditional equation and proved to be applicable by the model analysis in the previous studies. As Saint-Venant's approximation is turned out the same as the first one of them, it has the characteristics that the apparent Young's modulus is monotonously increasing according to the anisotropic angle. The methodology to analyze the elastic constants from four independent strain measurements by uniaxial compressive test of a single standard specimen is concisely described, and the necessity and compatibility of new suggested equations are discussed. Saint-Venant's approximation can determine the elastic constants close to true values and other equations may be unnecessary in specimens with medium to large angle. Nevertheless, they may become applicable because they can produce the almost same amount. For the specimens of small angle of anisotropy, Saint-Venant's approximation may result in out of general ranges or thermodynamic constraints, but other suggested equations can produce the almost true value. Thus they can be applied before other alternative equation is known. The guide map constructed by model study may decide the most compatible one of the three equations.
Four strains of water mold, ChS-E0511, RaT-E0511, RaT-A0512 and MaS-F0512, were isolated from salmonid fish and/or their eggs taken from culture farms in Yangyang, Milyang and Pyeongchang, Korea in 2005. Descriptions of their morphological aspects, the results of the phylogenetic analysis conducted, and the sequence of the small sub-unit 18S rRNAs of the isolates confirmed that they all belong to the species Saprolegnia parasitica. Only one species, ChS-E0511, which was isolated from fertilized eggs of the chum salmon, was classified as part of the S. parasiticaGroup 1 according to its oogonia and gemmae production. The chemotherapeutic effects of various chemicals on the ChS-E0511 strain were assessed from the inhibitory effects of growth in GY media and the relative ratio of eyed eggs to fertilized eggs of the rainbow trout. Malchite green, a prohibited substance in food animals, was better than others, such as the Opuntia ficus-indicaextract, 2-bronopol, and sodium chloride. These results suggest that the fungi isolated from salmonids and/or their eggs identified as S. parasitica were composed of more than two groups. These isolates will be useful in an intensive evaluation of therapeutic agents.
