Inhibitory action of adenosine on sinus rate in isolated rabbit SA node (토끼 동방결절 박동수에 대한 아데노신의 작용)

  • The inhibition/influences of adenine compounds on the heart have been described repeatedly by many investigators, since the first report by Druny and Szent-Gyorgyi [1929]. These studies have shown that adenosine and adenine nucleotides have an over-all effect similar to that of acetylcholine [ACh] by slowing and weakening the heartbeat. The basic cellular and membrane events underlying the inhibitory action of adenosine on sinus rate, however, are not well understood. Furthermore, the physiological role of adenosine in regulation of the heartbeat remains still to be elucidated. Therefore, this study was undertaken in order to examine the response of rabbit SA node to adenosine and to compare the response to that of ACh. Isolated SA node preparation, whole atrial pair, or left atrlal strip was used in each experiment. Action potentials of SA node were recorded through the intracellular glass microelectrodes, which were filled with 3M KCI and had resistance of 30-50 M. All experiments were performed in a bicarbonate-buffered Tyrode solution which was aerated with 3% $CO_2-97%$ $O_2$ gas mixture and kept at $35^{\circ}C$. Spontaneous firing rate of SA node at 35C [Mean + SEM, n=16] was 154 + 3.3 beats/min. The parameters of action potentials were: maximum astolic potential [MDP], -731.7mV: overshoot [OS], 9 + 1.4mV; slope of pacemaker potential [SPP], 94 3.0mV/sec.Adenosine suppressed the firing rate of SA node in a dose dependent manner. This inhibitory effect appeared at the concentration of $10^{-6}M$ and was potentiated in parallel with the increase in adenosine concentration. Changes in action potential by adenosine were dose-dependent increase of MDP and decrease of SPP until $10^{-4}$. Above this concentration, however, the amplitude of action potential decreased markedly due to the simultaneous decrease of both MDP and OS. All these effects of adenosine were not affected by pretreatment of atropine [2mg/l] and propranolol [$5{\times}10^{-6}M$]. ACh [$10^{-6}M$] responses on action potential were similar to those of adenosine by increasing MDP and decreasing SPP. These effects of ACh disappeared by pretreatment of atropine [2mg/1]. Inhibition/effects of adenosine and ACh on sinus rate were enhanced synergistically with the simultaneous administration of adenosine and ACh. Marked decrease of overshoot potential was the most prominent feature on action potential. Dipyridamole [DPM], which is known to block the adenosine transport across cell membrane, definitely potentiated the action of adenosine . Adenosine suppressed the sinus rate and atrial contractility in the same dosage range, even in the reserpinized preparation. Above` results suggest that adenosine suppresses pacemaker activity, like ACh, by acting directly on the membrane of SA node, increasing MDP and decreasing SPP.

Regional Ecological Network Design for Wild Animals' Movement Using Landscape Permeability and Least-cost Path Methods in the Metropolitan Area of Korea (경관투과성 및 최소비용경로 분석을 통한 수도권 지역의 광역생태축 구축 연구)

  • As populations inhabiting in natural ecosystem are fragmented by artificial barriers and habitats are destructed by development, extinction possibility of species is getting higher. It is necessary to design and to manage conservation areas and corridors considering animals' movement and migration for sustainable species diversity in present circumstances. 'Least-cost modeling' is one commonly employed approach in which dispersal costs are assigned to distinct habitat types and the last-costly dispersal paths among habitat patches are calculated using a geographical information system (GIS). This study aims to design ecological corridor using least-cost path method and to apply it to a regional ecological network considering movability of medium-large size mammals. This study was carried out over the metropolitan area, which has been deforested by rapid urbanization. Nevertheless there is connected with Gangwon province, Baekdudaegan mountain range and DMZ, considered where many forest species can migrate to this region. This study employs such an approach to develop least-cost path models for medium-large size mammals, have inhabited for this entire region. Considering those species, two forest areas as a source of species supply and forest areas more than 1,000ha are selected as focal forest areas. Movement and migration paths from species supply sources to focal forest areas are calculated by applying landscape permeability theory using land cover map, road density map and land slope map. Results showed least-cost paths from species supply sources to focal forest areas on two species. Wildcat and roe deer are different in some least-cost paths caused by their landscape permeability but paths show generally same specifics. The result of considering regional distribution of expected movement and migration paths to regional ecological network, low altitude mountains of western metropolitan area are evaluated important area for species connectivity. In national or regional levels ecological connectivity is essential to promote species diversity and to preserve integrated ecosystem. This study concludes that developing least-cost models from similar empirical data could significantly improve the utility of these tools.

Partial Safety Factors for Geotechnical Bearing Capacity of Port Structures (항만구조물 지반지지력 산정을 위한 부분안전계수 결정)

  • When eccentric or inclined load acts on foundation of the port & harbor structures, partial safety factors of bearing capacity limit state were estimated using reliability analysis. Current Korean technical standards of port and harbor structures recommend to estimate the geotechnical bearing capacity using the simplified Bishop method. In practice, however, simple method of comparing ground reaction resistance with allowable bearing capacity has been mostly used by design engineers. While the simple method gives just one number fixed but somewhat convenient, it could not consider the uncertainty of soil properties depending on site by site. Thus, in this paper, partial safety factors for each design variable were determined so that designers do perform reliability-based level 1 design for bearing capacity limit state. For these, reliability index and their sensitivities were gained throughout the first order reliability method(FORM), and the variability of the random variables was also considered. In order to verify partial safety factors determined here, a comparison with foreign design codes was carried out and were found to be reasonable in practical design.

Performance and Charging-Discharging Behavior of AGM Lead Acid Battery according to the Improvement of Bonding between Active Material/Substrate using Sand-Blasting Method (Sand-Blasting법을 이용한 활물질/기판간 결합력 향상에 따른 AGM 연축전지의 성능 및 충방전 거동)

  • To cope with automobile exhaust gas regulations, ISG (Idling Stop & Go) and charging control systems are applied to HEVs (Hybrid Electric Vehicle) for the purpose of improving fuel economy. These systems require quick charge/discharge performance at high current. To satisfy this characteristic, improvement of the positive electrode plate is studied to improve the charge/discharge process and performance of AGM(Absorbent Glass Mat) lead-acid batteries applied to ISG automotive systems. The bonding between grid and A.M (Active Material) can be improved by applying the Sand-Blasting method to provide roughness to the surface of the positive grid. When the Sand-Blasting method is applied with conditions of ball speed 1,000 rpm and conveyor speed 5 M/min, ideal bonding is achieved between grid and A.M. The positive plate of each condition is applied to the AGM LAB (Absorbent Glass Mat Lead Acid Battery); then, the performance and ISG life characteristics are tested by the vehicle battery test method. In CCA, which evaluates the starting performance at -18 ℃ and 30 ℃ with high current, the advanced AGM LAB improves about 25 %. At 0 ℃ CA (Charge Acceptance), the initial charging current of the advanced AGM LAB increases about 25 %. Improving the bonding between the grid and A.M. by roughening the grid surface improves the flow of current and lowers the resistance, which is considered to have a significant effect on the high current charging/discharging area. In a Standard of Battery Association of Japan (SBA) S0101 test, after 300 A discharge, the voltage of the advanced AGM LAB with the Sand-Blasting method grid was 0.059 V higher than that of untreated grid. As the cycle progresses, the gap widens to 0.13 V at the point of 10,800 cycles. As the bonding between grid and A.M. increases through the Sand Blasting method, the slope of the discharge voltage declines gradually as the cycle progresses, showing excellent battery life characteristics. It is believed that system will exhibit excellent characteristics in the vehicle environment of the ISG system, in which charge/discharge occurs over a short time.

Application of 3D point cloud modeling for performance analysis of reinforced levee with biopolymer (3차원 포인트 클라우드 모델링 기법을 활용한 바이오폴리머 기반 제방 보강공법의 성능 평가)

  • In this study, a large-scale levee breach experiment from lateral overflow was conducted to verify the effect of the new reinforcement method applied to the levee's surface. The new method could prevent levee failure and minimize damage caused by overflow in rivers. The levee was designed at the height of 2.5 m, a length of 12 m, and a slope of 1:2. A new material mixed with biopolymer powder, water, weathered granite, and loess in an appropriate ratio was sprayed on the levee body's surface at a thickness of about 5 cm, and vegetation recruitment was also monitored. At the Andong River Experiment Center, a flow (4 ㎥/s) was introduced from the upstream of the A3 channel to induce the lateral overflow. The change of lateral overflow was measured using an acoustic doppler current profiler in the upstream and downstream. Additionally, cameras and drones were used to analyze the process of the levee breach. Also, a new method using 3D point cloud for calculating the surface loss rate of the levee over time was suggested to evaluate the performance of the levee reinforcement method. It was compared to existing method based on image analysis and the result was reasonable. The proposed 3D point cloud methodology could be a solution for evaluating the performance of levee reinforcement methods.

Effect of Fines Content Including Clay on Liquefaction of Silt (점토를 포함한 세립분 함유량이 실트의 액상화에 미치는 영향)

  • Liquefaction behaviors of two natural silty samples containing fines including clay of 50% or more (Sample No.1 was silt 44.1% and clay 8.8%, sample No.2 was silt 57.2% and clay 12.4%) were examined by a cyclic triaxial test. According to the results on samples containing 50% or more of fine particles, an increase in the fine content decreases the liquefaction resistance of the sample. In other words, when the fine content increases, the liquefaction state of sample is reached with a small number of cyclic loads. In the relationship between the excess pore water pressure ratio and the number of cycles, the slope of the excess pore water pressure ratio increases more steeply as the fine content increases. As a result of analyzing the liquefaction behavior of the two silts with the content of clay contained in the fines, liquefaction occurred more easily in the No.2 silt with high clay content. This result shows that the clay contained in the fines affects the liquefaction behavior of the silt.

  • Many 1g shaking table tests with an SDOF structure supported by a single pile were performed to evaluate the soil-structure interaction (SSI) effect. Since most structures supported by group piles are MDOF structures with columns, the SSI effect is simulated using a large 1g shaking table test and numerical analysis. According to the results, the movement in the piles tends to increase with input acceleration and when the input frequency is similar to the natural frequency. Furthermore, the slope of the dynamic p-y curve remains constant regardless of the variation of acceleration and input frequency. According to the results of the dynamic p-y backbone curve and the moment of group piles, a center pile with a leading pile has more soil resistance than side piles with a trailing pile, and the effect of group piles is observed above the 7D center to center pile distance.

  • This study describes the results of model experiment to analyze the effect of backfill material types on the behavior of underground facility. In the model experiment, backfill materials around the existing underground facility were applied with soil (Jumunjin standard sand) and CLSM. The displacement of underground facility was analyzed for each excavation stage considering the separation distance between the excavation surface and the backfill area based on the experimental results. When soil was applied as a backfill material, the soil on the back of the excavation surface collapsed by excavation and formed an angle of repose, and the process of slope stability was repeated at each excavation stage. In addition, the displacement of underground facility began to occur in the excavation stage that the failure line of soil passes the installation location of the underground facility. When CLSM was applied as a backfill material, there was almost no horizontal and vertical displacement of the ground regardless of the separation distance from the excavation surface even when excavation proceeded to the backfill depth. Therefore, this result showed that it can have a resistance effect against the lateral earth pressure generated and the collapse of the original ground by adjacent excavation, if a backfill material with high stiffness such as CLSM is applied.

Fabrication and Characterization of NiCo2O4/Ni Foam Electrode for Oxygen Evolution Reaction in Alkaline Water Splitting (알칼라인 수전해 산소 발생 반응을 위한 NiCo2O4/Ni foam 전극 제조 및 특성 평가)

  • Environmental issues such as global warming due to fossil fuel use are now major worldwide concerns, and interest in renewable and clean energy is growing. Of the various types of renewable energy, green hydrogen energy has recently attracted attention because of its eco-friendly and high-energy density. Electrochemical water splitting is considered a pollution-free means of producing clean hydrogen and oxygen and in large quantities. The development of non-noble electrocatalysts with low cost and high performance in water splitting has also attracted considerable attention. In this study, we successfully synthesized a NiCo2O4/NF electrode for an oxygen evolution reaction in alkaline water splitting using a hydrothermal method, which was followed by post-heat treatment. The effects of heat treatment on the electrochemical performance of the electrodes were evaluated under different heat-treatment conditions. The optimized NCO/NF-300 electrode showed an overpotential of 416 mV at a high current density of 50 mA/cm2 and a low Tafel slope (49.06 mV dec-1). It also showed excellent stability (due to the large surface area) and the lowest charge transfer resistance (12.59 Ω). The results suggested that our noble-metal free electrodes have great potential for use in developing alkaline electrolysis systems.

  • In this research, a kinematic wave model was applied for the runoff analysis, Regulation of streamflow was estimated by the calibration of roughness coefficient as a parameter. The data analyzed were obtained from Ananomiya and Shirasaka experimental basins at Tokyo University Forest in Aichi. Estimation methods and characteristics of roughness coefficient as a evaluation method of hydrological function of forest are summarized as follows ; 1. Roughness coefficient($N_s$) indicates the resistance of hillslope to the flowing water of surface runoff. There exists an hypothesis that resistance of hillslope to flowing water increase with the growth forest and development of the $A_o$ layer. 2. Roughness coefficient($N_s$) was estimated by the parameter when the stream direct runoff was calculated by using the kinematic wave. 3. Secular change of '$N_s$' in ananomiya has a curve which has an upper limit and increases exponentially near the limit. The curve quickly increased from 1935 to 1945 when results of afforestation for erosion control were thought to be effective. On the other hand, slight increase of '$N_s$' in Shirasaka indicates that there was not such a big change in the surface of soil layer. 4. The increase of '$N_s$' was related with decrease of direct runoff and increase of base flow. It was recognized that the rate of direct runoff decreased with the improvement of forest physiognomy and the rate of base flow was increased. But absolute value of water runoff per one storm decreased in chronological order.
